1Low-dose decitabine enhances the effect of PD-1 blockade in colorectal cancer with microsatellite stability by re-modulating the tumor microenvironment显示文摘PD-1 blockade has demonstrated impressive clinical outcomes in colorectal cancers that have high microsatellite instability.However,the therapeutic efficacy for patients with tumors with low microsatellite instability or stable microsatellites needs further improvement.Here,we have demonstrated that low-dose decitabine could increase the expression of immune-related genes such as major histocompatibility complex genes and cytokine-related genes as well as the number of lymphocytes at the tumor site in CT26 colorectal cancer-bearing mice.A more significant inhibition of tumor growth and a prolongation of survival were observed in the CT26 mouse model after treatment with a combination of PD-1 blockade and decitabine than in mice treated with decitabine or PD-1 blockade alone.The anti-tumor effect of the PD-1 blockade was enhanced by low-dose decitabine.The results of RNA sequencing and whole-genome bisulfite sequencing of decitabine-treated CT26 cells and tumor samples with microsatellite stability from the patient tumor-derived xenograft model have shown that many immune-related genes,including antigen-processing and antigen-presenting genes,were upregulated,whereas the promoter demethylation was downregulated after decitabine exposure.Therefore,decitabine-based tumor microenvironment re-modulation could improve the effect of the PD-1 blockade.The application of decitabine in PD-1 blockade-based immunotherapy may elicit more potent immune responses,which can provide clinical benefits to the colorectal cancer patients with low microsatellite instability or stable microsatellites.Ganjun Yu Yanfeng Wu Wenying Wang Jia Xu Xiaoping Lv Xuetao Cao Tao Wan 2019Cellular & Molecular Immunology2019,16,4:14
2Heterotic Group Classification of 63 Inbred Lines and Hybrid Purity Identification by Using SSR Markers in Winter Cabbage(Brassica Oleracea L. var. capitata)显示文摘Winter cabbage is an important crop cultivated through winter in the region near the Yangtze River, enabling the supply of fresh cabbage there at that time of year. However, a problem has emerged regarding the newly developed parents of winter cabbage, which is completely different from spring and autumn cabbage, namely, how to combine these parents to breed an elite hybrid. To classify the heterotic groups and improve the efficiency of parent selection in winter cabbage breeding, 20 polymorphic SSR markers were selected to screen 63 winter cabbage inbred lines. Seventeen pairs among the 20 SSR markers amplified polymorphic bands. These primers amplified two to six bands,with an average of 2.8 bands per primer, and a total of 47 polymorphic bands were generated in the 63 inbred lines. These lines included flatheaded morphotype and round-headed morphotype, thus they were separately classified into heterotic groups based on the SSR markers. The flat-headed morphotype contained 21 inbred lines and was classified into three heterotic groups, named Hanchun 4, Jiali, and Dongsheng, in accordance with the representative germplasm contained in each group. The round-headed morphotype contained 42 inbred lines and was classified into five heterotic groups, named Parte, Bejo1039, YK-143, SCA002, and Golden B90. Meanwhile, parent analysis of 20 developed elite combinations showed that their parents were all distributed in different heterotic groups, indicating that the group classification was reasonable,which can provide a basis for further parent selection in winter cabbage breeding. 4Analysis on Dam-Breaking Mode of Tangjiashan Barrier Dam in Beichuan County显示文摘Tangjiashan landslide is a typical high-speed consequent landslide of medium-steep dip angle. This landslide triggered by earthquake took place in about semi-minute. The relative sliding displacement is 900 meters, so average sliding speed is about 30 meters per second. The longitudinal length of barrier dam which is formed by high-speed landslide along river is 803.4 meters; and maximum width crossing river is 611.8 meters. And its volume is estimated about 20.37 million steres. Through detailed geological investigation of the barrier dam, together with early geological information before earthquake, geological structures of the barrier dam and its stability of upstream and downstream slopes are studied when water level reaches different elevations in condition of continual after shocks with seismic intensity of 7 or 8 Richter scale. On this basis, dam-breaking mode of barrier dam is discussed deeply. 8Intramedullary schwannoma of the upper cervical spinal cord:a case study of identification in pathologic autopsy显示文摘Intramedullary schwannoma of the upper cervical spinal cord is rarely reported in forensic medicine.We herein report a case involving a patient who died of compression from an intramedullary schwannoma in the upper cervical spinal cord.A 30-year-old man initially presented with a five-day history of pain in the left chest that progressed to weakening in the left arm.Although the patient was treated with analgesic poultices,he developed inspiratory dyspnoea and died while working the next day without having undergone any medical imaging examination or surgical treatment.Anatomical and histopathological examinations revealed an intramedullary schwannoma in the left cervical spinal cord(C_(3)-C_(5))underneath the spinal nerve root.The cause of death might have been asphyxia secondary to the tumour,which interfered with the nerve function in the respiratory muscles.This finding suggests that an autopsy is essential for pathologists and medicolegists to comprehensively undertake their due obligation to obtain“the first evidence”,especially when there is a lack of directly related evidence.As part of the central nervous system,the spinal cord could be systematically included in a routine pathological autopsy in some cases.Xianxian Li Guangtao Xu Ruibing Su Junyao Lv Xiaoping Lai Xiaojun Yu 2017Forensic Sciences Research2017,2,1:2
9原发性肝癌超声造影特征及其与病理相关性的研究(英文)显示文摘Objective:The aim of this study was to evaluate the value of contrast-enhanced ultrasound(CEUS) for blood perfusion of primary liver cancer(PHC) and investigate the correlation between microvascular architecture of PHC and pathological differentiation.Methods:Two hundred and seventy-eight patients with 329 PHC lesions were examined by CEUS and analysised the contrast enhancement pattern and correlation with pathology.Results:1.CEUS patterns of PHC:71.7%(236/329) showed 'swift enhancement in the arterial phase and swift expurgation in the portal phase',13.4%(44/329) for as 'swift enhancement and slow expurgation',7.3%(24/329) as 'swift enhancement and simultaneity expurgation',4.3%(14/329) for the 'slow enhancement and swift expurgation',2.1%(7/329) as 'slow enhancement and expurgation',1.2%(4/329) as 'not fast forward'.2.90.3%(297/329) of PHC lesions were hypervascular liver cancer and 9.7%(32/329) were hypovascular.Hepatocellular carcinoma(HCC) were hypervascular lesions and intrahepatic cholangiocarcinoma(ICC) were hypovascular lesions.3.PHC size had a significant difference on the contrast media purfusion pattern(P < 0.05),but not on the contrast media expurgation pattern.4.The accuracy of PHC by CEUS were 97.3% and compared to pathology,9 lesions of PHC were misdiagnosed.Conclusion:CEUS can show the different blood perfusion characteristics of PHC with closely related to pathological differentiation,which be valuable to diagnose liver cancer.Qin Si Xiaoli Qian Shengxi Huang Xiaoping Lv Wei Tong Yanli Huang Jian Chen 2011The Chinese-German Journal of Clinical Oncology2011,10,9:1
10N6-methyladenosine-modified DBT alleviates lipid accumulation and inhibits tumor progression in clear cell renal cell carcinoma through the ANXA2/YAP axis-regulated Hippo pathway显示文摘Background:The mechanism of metabolism reprogramming is an unsolved problem in clear cell renal cell carcinoma(ccRCC).Recently,it was discovered that the Hippo pathway altered tumor metabolism and promoted tumor progression.Thus,this study aimed at identifying key regulators of metabolism reprogramming and the Hippo pathway in ccRCC and pinpointing potential therapeutic targets for ccRCC patients.Methods:Hippo-related gene sets and metabolic gene sets were used to screen potential regulators of the Hippo pathway in ccRCC.Public databases and samples from patients were applied to investigate the association of dihydrolipoamide branched chain transacylase E2(DBT)with ccRCC and Hippo signaling.The role of DBT was confirmed by gain or loss of function assays in vitro and in vivo.Mechanistic results were yielded by luciferase reporter assay,immunoprecipitation,mass spectroscopy,and mutational studies.Results:DBT was confirmed as a Hippo-related marker with significant prognostic predictive value,and its downregulationwas caused bymethyltransferaselike-3(METTL3)-mediated N6-methyladenosine(m6A)modification in ccRCC.Functional studies specified DBT as a tumor suppressor for inhibiting tumor progression and correcting the lipid metabolism disorder in ccRCC.Mechanistic findings revealed that annexin A2(ANXA2)interacted with the lipoyl-binding domain of DBT to activate Hippo signaling which led to decreased nuclear localization of yes1-associated transcriptional regulator(YAP)and transcriptional repression of lipogenic genes.Conclusions:This study demonstrated a tumor-suppressive role for the DBT/ANXA2/YAP axis-regulated Hippo signaling and suggested DBT as a potential target for pharmaceutical intervention in ccRCC.Daojia Miao QiWang Jian Shi Qingyang Lv Diaoyi Tan Chuanyi Zhao Zhiyong Xiong Xiaoping Zhang 2023Cancer Communications2023,43,4:1
11A MODEL FOR THE RELEASE BEHAVIOR OF DRUGS FROM HYDROGEL NANOPARTICLE显示文摘A model to correlate and predict the release behavior of drugs from hydrogel nanoparticles is presented in this paper. The nanoparticle is considered as a combination of a shell of an elastic semipermeable membrane and a core of a fluid phase (After swelling equilibrium). The fluid core consists of network building materials and other components that are able to partition in hydrogel nanoparticle phase and surrounding coexisting liquid phase, and is enveloped by the membrane shell. The excess Gibbs energies of the hydrogel nanoparticle phase and the surrounding coexisting fluid phase are expressed e.g. using UNIQUAC equation with “free-volume” contribution for non-ionic solution and VERS-model for ionic one. 14Managed grassland alters soil N dynamics and N2O emissions in temperate steppe显示文摘Reclamation of degraded grasslands as managed grasslands has been increasingly accelerated in recent years in China. Land use change affects soil nitrogen(N) dynamics and nitrous oxide(N_2O) emissions. However, it remains unclear how large-scale grassland reclamation will impact the grassland ecosystem as a whole. Here, we investigated the effects of the conversion from native to managed grasslands on soil N dynamics and N_2O emissions by field experiments in Hulunber in northern China. Soil(0–10 cm), nitrate(NO_3^-),ammonium(NH_4^+), and microbial N were measured in plots in a temperate steppe(Leymus chinensis grassland) and two managed grasslands(Medicago sativa and Bromus inermis grasslands) in 2011 and 2012. The results showed conversion of L. chinensis grassland to M.sativa or B. inermis grasslands decreased concentrations of NO_3~–-N, but did not change NH_4~–N . Soil microbial N was slightly decreased by the conversion of L. chinensis grassland to M.sativa, but increased by the conversion to B. inermis. The conversion of L. chinensis grassland to M. sativa(i.e., a legume grass) increased N_2O emissions by 26.2%, while the conversion to the B. inermis(i.e., a non-legume grass) reduced N_2O emissions by 33.1%. The conversion from native to managed grasslands caused large created variations in soil NO?3^-+–N and NH_4~–N concentrations. Net N mineralization rates did not change significantly in growing season or vegetation type, but to net nitrification rate. 16PREPARATION AND CHARACTERIZATION OF DOXORUBICIN-LOADED MAGNETIC ANTICANCER NANOPARTICLES显示文摘Doxorubicin(ADM)-loaded magnetic anticancer nanoparticles,using Fe3O4 as core, doxorubicin as model drug and polyvinylpyrrolidone (PVP) as matrix, were prepared by inverse emulsion polymerization. The experimental results showed that the average diameter of Fe3O4 particles was 19.8nm. The X-ray diffraction indicated that the prepared Fe3O4 particle was pure cubic Fe3O4. The results obtained by SEM showed the magnetic nanoparticles under optimal operating condition had a smooth spherical surface , LLS showed an average size of 78.7nm. 20Observation of oxygen evolution over a {Ni12}-cluster-based metal-organic framework显示文摘The development of efficient electrocatalysts based on non-noble metals for oxygen evolution reaction(OER) remains an important and challenging task. Multinuclear transition-metal clusters with high structural stability are promising OER catalysts but their catalytic role is poorly understood. Here we report the crystallographic observation of OER activity over robust {Ni12}-clusters immobilised in a porous metal-organic framework, NKU-100, by single-crystal X-ray diffraction as a function of external applied potential. We observed the aggregation of confined oxygen species around the {Ni12}-cluster as a function of applied potential during the electrocatalytic process. The refined occupancy of these oxygen species shows a strong correlation with the variation of current density.
