1Simulation of shock wave buffet and its suppression on an OAT15A supercritical airfoil by IDDES显示文摘In the present paper,extremely unsteady shock wave buffet induced by strong shock wave/boundary-layer interactions (SWBLI) on the upper surface of an OAT15A supercritical airfoil at Mach number of 0.73 and angle of attack of 3.5 degrees is first numerically simulated by IDDES,one of the most advanced RANS/LES hybrid methods.The results imply that conventional URANS methods are unable to effectively predict the buffet phenomenon on the wing surface;IDDES,which involves more flow physics,predicted buffet phenomenon.Some complex flow phenomena are predicted and demonstrated,such as periodical oscillations of shock wave in the streamwise direction,strong shear layer detached from the shock wave due to SWBLI and plenty of small scale structures broken down by the shear layer instability and in the wake.The root mean square (RMS) of fluctuating pressure coefficients and streamwise range of shock wave oscillation reasonably agree with experimental data.Then,two vortex generators (VG) both with an inclination angle of 30 degrees to the main flow directions are mounted in front of the shock wave region on the upper surface to suppress shock wave buffet.The results show that shock wave buffet can be significantly suppressed by VGs,the RMS level of pressure in the buffet region is effectively reduced,and averaged shock wave position is obviously pushed downstream,resulting in increased total lift.HUANG JingBo XIAO ZhiXiang LIU Jian FU Song 2012Science China(Physics,Mechanics & Astronomy)2012,55,2:24
2Numerical investigation of unsteady vortex breakdown past 80°/65° double-delta wing显示文摘An improved delayed detached eddy simulation(IDDES) method based on the k-x-SST(shear stress transport) turbulence model was applied to predict the unsteady vortex breakdown past an 80 /65 double-delta wing(DDW), where the angles of attack(AOAs) range from 30 to 40. Firstly, the IDDES model and the relative numerical methods were validated by simulating the massively separated flow around an NACA0021 straight wing at the AOA of 60.The fluctuation properties of the lift and pressure coefficients were analyzed and compared with the available measurements. For the DDW case, the computations were compared with such measurements as the mean lift, drag, pitching moment, pressure coefficients and breakdown locations.Furthermore, the unsteady properties were investigated in detail, such as the frequencies of force and moments, pressure fluctuation on the upper surface, typical vortex breakdown patterns at three moments, and the distributions of kinetic turbulence energy at a stream wise section.Two dominated modes are observed, in which their Strouhal numbers are 1.0 at the AOAs of30, 32 and 34 and 0.7 at the AOAs of 36, 38 and 40. The breakdown vortex always moves upstream and downstream and its types change alternatively. 3Numerical investigation of an advanced aircraft model during pitching motion at high incidence显示文摘An unsteady Reynolds averaged Navier–Stokes(URANS) method combined with a rigid dynamic mesh technique was developed to simulate unsteady flows around complex configurations during pitching motion. First, a test case with the NACA0012 airfoil was selected to validate the numerical methods and our in-house codes. Then, we evaluated the unsteady flows around an advanced aircraft model during harmonic pitching motion at high incidence. The effects of pitching motion on the hysteresis of aerodynamic force, the evolution of the leading-edge vortex, and the distribution of pressure on the model's surface were analyzed in detail. The roles of several significant parameters such as the reduced frequency and pitching amplitude were revealed. Several conclusions were found: pitching motion would delay the initiation of the leading-edge vortex, strengthen the vorticity, postpone the occurrence of vortex breakdown, and weaken the massively separated flows, thus causing additional aerodynamic force. Two categories of critical reduced frequency have been found, which divide the influence of reduced frequency on aerodynamic force into three stages, called the linear increasing range, slowly increasing range, and constant range. The first-order phase lag between the aerodynamic force and the incidence is a constant that is independent of the amplitude when the reduced frequency is sufficiently high. 4Numerical studies of reverse flows controlled by undulating leading edge显示文摘The improved delayed detached-eddy simulation(IDDES) method is used to simulate the reverse flows past an NACA0012 airfoil at medium(10°) and large(30°) angles of attack. The numerical results of the baseline configuration are compared with the available measurements. The effects of the undulating leading edge with four different amplitudes are compared and analyzed at angle of attack of 10°. Based on these analyses, the amplitude of A/C=0.04 yields the best performance. Compared with the uncontrolled case, the performances of the undulating leading edge are greatly improved with reducing of the aerodynamic fluctuations. 8DeepNitro: Prediction of Protein Nitration and Nitrosylation Sites by Deep Learning显示文摘Protein nitration and nitrosylation are essential post-translational modifications(PTMs)involved in many fundamental cellular processes. Recent studies have revealed that excessive levels of nitration and nitrosylation in some critical proteins are linked to numerous chronic diseases.Therefore, the identification of substrates that undergo such modifications in a site-specific manner is an important research topic in the community and will provide candidates for targeted therapy. In this study, we aimed to develop a computational tool for predicting nitration and nitrosylation sites in proteins. We first constructed four types of encoding features, including positional amino acid distributions, sequence contextual dependencies, physicochemical properties, and position-specificscoring features, to represent the modified residues. Based on these encoding features, we established a predictor called DeepNitro using deep learning methods for predicting protein nitration and nitrosylation. Using n-fold cross-validation, our evaluation shows great AUC values for DeepNitro, 0.65 for tyrosine nitration, 0.80 for tryptophan nitration, and 0.70 for cysteine nitrosylation, respectively,demonstrating the robustness and reliability of our tool. Also, when tested in the independent dataset, DeepNitro is substantially superior to other similar tools with a 7%à42% improvement in the prediction performance. Taken together, the application of deep learning method and novel encoding schemes, especially the position-specific scoring feature, greatly improves the accuracy of nitration and nitrosylation site prediction and may facilitate the prediction of other PTM sites. 9The recent trend of ethnic intermarriage in China: an analysis based on the census data显示文摘Interethnic marriage is a key field in ethnic relations study.A high level of interethnic marriage only occurs in a multiethnic society in which the political system,economic structure,laws,and cultural environment allow young people with different ethnic backgrounds to mingle with each other.This is why the interethnic marriage rate can be used to measure the degree of interactions among different ethnic groups.It is the most reliable indicator for a trend of long-term ethnic relations.Due to the lack of relevant data in official statistics,there are limited studies focusing on interethnic marriage at the macro level.Based on the 1990,2000,and 2010 census data and the special volume on ethnicity edited by the State Ethnic Affairs Commission,this paper attempts to determine the basic trend of interethnic marriages in China over the past three decades.It discusses some basic characteristics of interethnic marriages among different ethnic groups.How to strengthen national unity,ethnic solidarity,and improve the relationships among different ethnic groups are major challenges faced by China in the twenty-first century.Studying the interethnic marriage from sociological and demographic approaches may help us understand the basic trend of interethnic integration in today’s China.Zhixiang Jian 2017The Journal of Chinese Sociology2017,4,1:0
10In situ performance and stability tests of large-area flexible polymer solar cells in the 35-km stratospheric environment显示文摘Flexible organic solar cells(FOSCs)are one of the most promising power sources for aerospace aircraft due to their atractive advantages with high power-per-weight ratio and excellent mechanical flexibility.Understanding the performance and stability of high-performance FOSCs is essential for the further development of FOSCs for aerospace applications.In this paper,after systematic investigations on the performance of the state-of-the-art high-performance solar cells under thermal cycle and intensive UV irradiation conditions,in situ performance and stability tests of the solar cells in the 35 km stratospheric environment were carried out through a high-altitude balloon uploading.The encapsulated FOSCs with an area of 0.64 cm^(2)gave the highest power density of 15.26 mW/cm^(2)and an efficiency over 11%,corresponding to a power-per-weight ratio of over 3.32 k W/kg.More importantly,the cells showed stable power output during the 3-h continuous flight at 35 km and only 10%performance decay after return to the lab,suggesting promising stability of the FOSCs in the stratospheric environment.Zihan Xu Guoning Xu Qun Luo Yunfei Han Yu Tang Ying Miao Yongxiang Li Jian Qin Jingbo Guo Wusong Zha Chao Gong Kun Lu Jianqi Zhang Zhixiang Wei Rong Cai Yanchu Yang Zhaojie Li Chang-Qi Ma 2023National Science Review2023,10,4:0

12An Open-Label Study of Pegylated Liposomal Doxorubicin,Vincristine, and Reduced-Dose Dexamethasone Combination Therapy in Newly Diagnosed Multiple Myeloma Patients in the Chinese Population显示文摘OBJECTIVE Though doxorubicin is highly activein the treatment of multiple myeloma, its toxicityprofile limits its therapeutic index. We performed thisstudy to evaluate the efficacy and safety of pegylatedliposomal doxorubicin (PLD, Caelyx^(?)), vincristine,and reduced-dose dexamethasone combinationtherapy in newly diagnosed multiple myeloma (MM)patients in a Chinese population.METHODS This was an open-label, single-armstudy in which newly diagnosed patients with MMreceived PLD 40 mg/m^2 intravenously on Day 1,vincristine 1.4 mg/m^2 intravenously (maximum 2 mg)on Day 1, and 40 mg of dexarnethasone (intravenouslyor orally) from Day 1 to Day 4. Treatment wasrepeated every 28 days for at least 4 cycles.RESULTS In the intent-to-treat (ITT) analysis, theoverall response rate was 68.29%, and the completeremission rate was 10.98%. The incidence of alladverse events was 46.34%. 133-Epi-betulinic acid 3-O-β-D-glucopyranoside(eBAG)induces autophagy by activation of AMP-activated protein kinase in hepatocellular carcinoma显示文摘3-Epi-betulinic acid 3-O-β-D-glucopyranoside(eBAG)is a pentacyclic triterpene mainly distributed in food and medicinal plants,which exhibits various pharmacological properties.However,whether these functions are attributed to eBAG or additional components in these plants remain unknown.Herein,we report that eBAG exerted an inhibitory activity against hepatocellular carcinoma and esophageal cancer cells.EBAG induced non-apoptotic cell death in hepatocellular carcinoma cells.The eBAG-induced cell death was inhibited by knock-down of autophagy related gene(ATG)5 and ATG7,by administration of 3-methyladenine,a selective autophagy inhibitor that suppresses phosphoinositide 3-kinase(PI3K),and by chloroquine,a classic autophagy flux inhibitor.We demonstrated that eBAG induced an autophagy-mediated cell death.Application of eBAG mimicked cellular bioenergetics depletion leading to the reduction of intracellular ATP,activation of AMP-activated protein kinase(AMPK),and inhibition of mTOR.Co-treatment with compound C,an AMPK inhibitor,abrogated cell death induced by eBAG.We further validated the anti-tumor effect of eBAG in the murine xenograft model of hepatocellular carcinoma and found that eBAG treatment promoted the induction of autophagy and reduction of tumor growth in mice.As a functional food ingredient,eBAG is a potential therapeutic agent for the treatment of hepatocellular carcinoma and esophageal cancer.Mengjie Liu Xuqiang Liu Kaiyue He Yongping Jian Yujia Li Jinrong Guo Jingyu Yang Zhixiang Xu Wenyi Kang 2024Food Science and Human Wellness2024,13,3:0
14VSOLassoBag:a variable-selection oriented LASSO bagging algorithm for biomarker discovery in omic-based translational research显示文摘Screening biomolecular markers from high-dimensional biological data is one of the long-standing tasks for biomedical translational research.With its advantages in both feature shrinkage and biological interpretability,Least Absolute Shrinkage and Selection Operator(LASSO)algorithm is one of the most popular methods for the scenarios of clinical biomarker development.However,in practice,applying LASSO on omics-based data with high dimensions and low-sample size may usually result in an excess number of predictive variables,leading to the overfitting of the model.Here,we present VSOLassoBag,a wrapped LASSO approach by integrating an ensemble learning strategy to help select efficient and stable variables with high confidence from omics-based data.Using a bagging strategy in combination with a parametric method or inflection point search method,VSOLassoBag can integrate and vote variables generated from multiple LASSO models to determine the optimal candidates.The application of VSOLassoBag on both simulation datasets and real-world datasets shows that the algorithm can effectively identify markers for either case-control binary classification or prognosis prediction.In addition,by comparing with multiple existing algorithms,VSOLassoBag shows a comparable performance under different scenarios while resulting in fewer features than others.In summary,VSOLassoBag,which is available at http://gffzza891166c5c83403bsvwnk9of9c5cb6ovk.ffgz.tsg.suse.edu.cn/VSOLassoBag/under the GPL v3 license,provides an alternative strategy for selecting reliable biomarkers from high-dimensional omics data.For user’s convenience,we implement VSOLassoBag as an R package that provides multithreading computing configurations.Jiaqi Liang Chaoye Wang Di Zhang Yubin Xie Yanru Zeng Tianqin Li Zhixiang Zuo Jian Ren Qi Zhao 2023Journal of Genetics and Genomics2023,50,3:0

16Numerical studies of undulation control on dynamic stall for reverse flows显示文摘The delayed detached-eddy simulation with adaptive coefficient(DDES-AC)method is used to simulate the baseline and leading-edge undulation control of dynamic stall for the reverse flow past a finite-span wing with NACA0012 airfoil.The numerical results of the baseline configuration are compared with available measurements.DDES and DDES-AC perform differently when predicting the primary and secondary dynamic stalls.Overall,DDES-AC performs better owing to the decrease of grey area between the strong shear layer and the fully three-dimensional separated flow.Moreover,the effects of the undulating leading-edge on the forces,lift gradients,and instantaneous flow structures are explored.Compared with the uncontrolled case,the lift gradient in the primary dynamic stall is reduced from 18.4 to 8.5,and the secondary dynamic stall disappears.Therefore,periodic unsteady air-loads are also reduced.Additionally,the control mechanism of the wavy leading edge(WLE)is also investigated by comparison with the straight leading edge(SLE).No sudden breakdown of strong vortices is the main cause for WLE control.Biao Wang Jian Liu Yunjun Yang Zhixiang Xiao 2020Acta Mechanica Sinica2020,36,2:0

18Multiomics analysis reveals metabolic subtypes and identifies diacylglycerol kinase α (DGKA) as a potential therapeutic target for intrahepatic cholangiocarcinoma显示文摘Background:Intrahepatic cholangiocarcinoma(iCCA)is a highly heteroge-neous and lethal hepatobiliary tumor with few therapeutic strategies.The metabolic reprogramming of tumor cells plays an essential role in the develop-ment of tumors,while the metabolic molecular classification of iCCA is largely unknown.Here,we performed an integrated multiomics analysis and metabolic classification to depict differences in metabolic characteristics of iCCA patients,hoping to provide a novel perspective to understand and treat iCCA.Methods:We performed integrated multiomics analysis in 116 iCCA samples,including whole-exome sequencing,bulk RNA-sequencing and proteome anal-ysis.Based on the non-negative matrix factorization method and the protein abundance of metabolic genes in human genome-scale metabolic models,the metabolic subtype of iCCA was determined.Survival and prognostic gene analy-ses were used to compare overall survival(OS)differences between metabolic subtypes.Cell proliferation analysis,5-ethynyl-2’-deoxyuridine(EdU)assay,colony formation assay,RNA-sequencing and Western blotting were performed to investigate the molecular mechanisms of diacylglycerol kinaseα(DGKA)in iCCA cells.Results:Three metabolic subtypes(S1-S3)with subtype-specific biomarkers of iCCA were identified.These metabolic subtypes presented with distinct prog-noses,metabolic features,immune microenvironments,and genetic alterations.The S2 subtype with the worst survival showed the activation of some special metabolic processes,immune-suppressed microenvironment and Kirsten ratsar-coma viral oncogene homolog(KRAS)/AT-rich interactive domain 1A(ARID1A)mutations.Among the S2 subtype-specific upregulated proteins,DGKA was further identified as a potential drug target for iCCA,which promoted cell proliferation by enhancing phosphatidic acid(PA)metabolism and activating mitogen-activated protein kinase(MAPK)signaling.Conclusion:Viamultiomics analyses,we identified three metabolic subtypes of iCCA,revealing that the S2 subtype exhibited the poorest survival outcomes.We further identified DGKA as a potential target for the S2 subtype.Weiren Liu Huqiang Wang Qianfu Zhao Chenyang Tao Weifeng Qu Yushan Hou Run Huang Zimei Sun Guiqi Zhu Xifei Jiang Yuan Fang Jun Gao Xiaoling Wu Zhixiang Yang Rongyu Ping Jiafeng Chen Rui Yang Tianhao Chu Jian Zhou Jia Fan Zheng Tang Dong Yang Yinghong Shi 2024Cancer Communications2024,44,2:0
19TIGER:A Web Portal of Tumor Immunotherapy Gene Expression Resource显示文摘Immunotherapy is a promising cancer treatment method;however,only a few patients benefit from it.The development of new immunotherapy strategies and effective biomarkers of response and resistance is urgently needed.Recently,high-throughput bulk and single-cell gene expression profling technologies have generated valuable resources.However,these resources are not well organized and systematic analysis is difficult.Here,we present TIGER,a tumor immunotherapy gene expression resource,which contains bulk transcriptome data of 1508 tumor samples with clinical immunotherapy outcomes and 11,057 tumor/normal samples without clinical immunotherapy outcomes,as well as single-cell transcriptome data of 2,116,945 immune cells from 655 samples.TIGER provides many useful modules for analyzing collected and user-provided data.Using the resource in TIGER,we identified a tumor-enriched subset of CD4^(+)T cells.Patients with melanoma with a higher signature score of this subset have a significantly better response and survival under immunotherapy.We believe that TIGER will be helpful in understanding anti-tumor immunity mechanisms and discovering effective biomarkers.Zhihang Chen Ziwei Luo Di Zhang Huiqin Li Xuefei Liu Kaiyu Zhu Hongwan Zhang Zongping Wang Penghui Zhou Jian Ren An Zhao Zhixiang Zuo 2023Genomics, Proteomics & Bioinformatics2023,21,2:0
