1A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. 4The APR triangle:A practical zone in the Glissonean approach to laparoscopic anatomical right hepatectomy显示文摘Background:An understanding of vascular anatomy is crucial for the safe performance of laparoscopic anatomical liver excision.We discovered a triangular zone during the laparoscopic right liver surgery and termed this zone the APR triangle.The purpose of this study was to determine the probability of the existence of the APR triangle and elucidate its various forms.Methods:Analyzed three-dimensional image reconstructions of 66 individuals who underwent liver surgery and calculated the statistics for various types of APR triangles under various grouping settings.Results:The APR triangle was present in the majority of cases,with right hepatic vein trunk type in 68%and right hepatic vein branch type in 21%,respectively.The angle between the right anterior and right posterior hepatic pedicles(AP&PP)was at most between 45 and 90°(74%).There was a 35%chance that at least one of the AP&PP was longer than 2 cm,and a 39%chance that both were.The right posterior pedicle first branch would appear at the bifurcation of AP&PP in 13%only.Conclusions:The APR triangle is objectively present and may represent a practical zone for performing laparoscopic right hepatic anatomical resection more simply and safely.Jie Chen Ziyu Zhang Rui Zhou Anzhi Wang Jun Cao Yajin Chen 2022iLIVER2022,1,3:2
5Differential transcriptomic landscapes of multiple organs from SARS-CoV-2 early infected rhesus macaques显示文摘SARS-CoV-2 infection causes complicated clinical manifestations with variable multi-organ injuries,how-ever,the underlying mechanism,in particular immune responses in different organs,remains elusive.In this study,comprehensive transcriptomic alterations of 14 tissues from rhesus macaque infected with SARS-CoV-2 were analyzed.Compared to normal controls,SARS-CoV-2 infection resulted in dysregulation of genes involving diverse functions in various examined tissues/organs,with drastic transcriptomic changes in cerebral cortex and right ventricle.Intriguingly,cerebral cortex exhibited a hyperinflammatory state evidenced by sig-nificant upregulation of inflammation response-related genes.Meanwhile,expressions of coagulation,angio-genesis and fibrosis factors were also up-regulated in cerebral cortex.Based on our findings,neuropilin 1(NRP1),a receptor of SARS-CoV-2,was significantly elevated in cerebral cortex post infection,accompanied by active immune response releasing inflammatory factors and signal transmission among tissues,which enhanced infection of the central nervous system(CNS)in a positive feedback way,leading to viral encephalitis.Overall,our study depicts a multi-tissue/organ tran-scriptomic landscapes of rhesus macaque with early infection of SARS-CoV-2,and provides important insights into the mechanistic basis for COVID-19-asso-ciated clinical complications.Chun-Chun Gao Man Li Wei Deng Chun-Hui Ma Yu-Sheng Chen Yong-Qiao Sun Tingfu Du Qian-Lan Liu Wen-Jie Li Bing Zhang Lihong Sun Si-Meng Liu Fengli Li Feifei Qi Yajin Qu Xinyang Ge Jiangning Liu Peng Wang Yamei Niu Zhiyong Liang Yong-Liang Zhao Bo Huang Xiao-Zhong Peng Ying Yang Chuan Qin Wei-Min Tong Yun-Gui Yang 2022Protein & Cell2022,13,12:1

7A survey of Intel SGX and its applications显示文摘This paper presents a comprehensive survey on the development of Intel SGX(software guard extensions)processors and its applications.With the advent of SGX in 2013 and its subsequent development,the corresponding research works are also increasing rapidly.In order to get a more comprehensive literature review related to SGX,we have made a systematic analysis of the related papers in this area.We first search through five large-scale paper retrieval libraries by keywords(i.e.,ACM Digital Library,IEEE/IET Electronic Library,SpringerLink,Web of Science,and Elsevier Science Direct).We read and analyze a total of 128 SGX-related papers.The first round of extensive study is conducted to classify them.The second round of intensive study is carried out to complete a comprehensive analysis of the paper from various aspects.We start with the working environment of SGX and make a conclusive summary of trusted execution environment(TEE).We then focus on the applications of SGX.We also review and study multifarious attack methods to SGX framework and some recent security improvements made on SGX.Finally,we summarize the advantages and disadvantages of SGX with some future research opportunities.We hope this review could help the existing and future research works on SGX and its application for both developers and users.Wei ZHENG Ying WU Xiaoxue WU Chen FENG Yulei SUI Xiapu LUO Yajin ZHOU 2021Frontiers of Computer Science2021,15,3:1
8Ultra-broadband spatial light modulation with dual-resonance coupled epsilon-near-zero materials显示文摘It has been found that the dielectric constants of transparent conductive oxides(TCOs)can be adjusted in an extremely large range by tuning the carrier density.Due to the remarkable light confinement property of the epsilon-near-zero(ENZ)effect of TCOs,it has attracted extensive interests of light modulation.However,the operation wavelength bandwidth is usually limited by optical resonance that is applied to enhance the light-TCOs interaction.In this work,a dual-resonance light coupling scheme is proposed to expand the modulation depth-bandwidth product with almost one order-of-magnitude improvement.In a metallic subwavelength grating structure with deep trenches backed by a ground plane,the ENZ mode can be coupled to both magnetic resonance and Fabry-Perot resonance respectively by tuning the bias.Decent light modulation can be obtained in a large operation wavelength band covering two resonances by optimizing the dual-resonance configuration.Such a reconfigurable efficient broadband modulation is important for robust communication link and possesses remarkable capacity for wavelength division multiplexing.Qin Chen Shichao Song Huacun Wang Li Liang Yajin Dong Long Wen 2021Nano Research2021,14,8:0

10Splenic autotransplantation and oesophageal transection anastomosis in patients with portal hypertension (26 years clinical observation)显示文摘The surgical treatment methods for cirrhosis patients complicated with portal hypertension are complicated.In this study,we evaluated the effectiveness of a new treatment strategy:splenic auto-transplantation and oesophageal transection anastomosis on 274 patients from three aspects:clinical observation,splenic immunology and portal dynamics.From 1979 to 2005,274 cirrhosis patients with portal hypertension who underwent the new treatment strategy were followed up to observe different clinical indexes,which were then compared with those of the traditional surgery treatment.From 1999 to 2002,a randomized control trial(RCT)was performed on 40 patients to compare their immune function after operation.From 1994 to 2004,another RCT was carried out on 28 patients to compare the portal dynamics through three-dimensional dynamic contrast enhanced MR angiography(3D DEC MRA)investigation after operation.Among 274 patients(mean age 41.8 years),the emergency operative mortality(4.4%),selective operative mortality(2.2%),complication rate(17.9%),morbidity of hepatic encephalopathy(<1%),bleeding rate of portal hypertension gastritis(PHG)(9.1%),and morbidity of hepatic carcinoma(8%)were similar to those under traditional operation;the spleen immunology function(Tuftsin,IgM)decreased among the groups 2 months after operation.Through 3D DCE MRA,the cross section area,the velocity and volume of blood flow of main portal vein decrease significantly after operation in both groups,the auto transplantation group was significantly lower in velocity and volume of blood flow than in the control group.Splenic auto transplantation and esophageal transection anastomosis are a safe,effective,and reasonable treatment strategy for portal hypertension with varicial bleeding.It can not only correct hypersplenism but also completely stanch blood,and auto transplanted spleen in the retroperitoneal space can preserve immune function and establish abroad collateral circulation.CHEN Jisheng HUO Jinshan ZHANG Hongwei SHANG Changzhen CHEN Rufu ZHANG Jie Obetien Mapudengo CHEN Yajin ZHANG Lei 2007Frontiers of Medicine2007,1,1:0
