1Structures of EV71 RNA-dependent RNA polymerase in complex with substrate and analogue provide a drug target against the hand-foot-and-mouth disease pandemic in China显示文摘Enterovirus 71(EV71),one of the major causative agents for hand-foot-and-mouth disease(HFMD),has caused more than 100 deaths among Chinese children since March 2008.The EV71 genome encodes an RNAdependent RNA polymerase(RdRp),denoted 3D^(pol),which is central for viral genome replication and is a key target for the discovery of specific antiviral therapeutics.Here we report the crystal structures of EV71 RdRp(3D^(pol))and in complex with substrate guanosine-5'-triphosphate and analog 5-bromouridine-5'-triphosphate best to 2.4Åresolution.The structure of EV71 RdRp(3D^(pol))has a wider open thumb domain compared with the most closely related crystal structure of poliovirus RdRp.And the EV71 RdRp(3D^(pol))complex with GTP or Br-UTP bounded shows two distinct movements of the polymerase by substrate or analogue binding.The model of the complex with the template:primer derived by superimposition with foot-and-mouth disease virus(FMDV)3D/RNA complex reveals the likely recognition and binding of template:primer RNA by the polymerase.These results together provide a molecular basis for EV71 RNA replication and reveal a potential target for anti-EV71 drug discovery.Yang Wu Zhiyong Lou Yi Miao Yue Yu Hui Dong Wei Peng Mark Bartlam Xuemei Li Zihe Rao 2010Protein & Cell2010,1,5:30

3Postoperative adjuvant antiviral therapy for hepatitis B/C virus-related hepatocellular carcinoma:A meta-analysis显示文摘AIM:To investigate the impact of postoperative antiviral treatment on tumor recurrence and survival of patients with chronic hepatitis B virus(HBV) or hepatitis C virus(HCV) infection-related primary hepatocellular carcinoma(HCC) after curative therapy.METHODS:We performed a meta-analysis of randomized and non-randomized control trials from electronic search and manual search.The fixed effect model of Mantel-Haenszel method and the random effect model of Der Simonian and Laird method were used for homogeneous and heterogeneous studies,respectively.Seven HCV-related studies,three HBV-related studies and three studies on HBV or HCV-related HCC were identified.RESULTS:A total of 1224 patients were included in this analysis.The estimated odds ratios(OR) for the 1-,2-,3-and 5-year recurrence were 0.54 [15.4% vs 24.1%,95% confidence interval(CI):0.32-0.89,P=0.02],0.42(36.9% vs 58.0%,95% CI:0.19-0.90,P=0.03),0.37(47.9% vs 63.8%,95% CI:0.19-0.71,P=0.003),and 0.32(66.7% vs 74.3%,95% CI:0.15-0.66,P=0.002),respectively;and the OR for the 1-,2-,3-,5-and 7-year mortality were 0.23(1.2% vs 9.1%,95% CI:0.07-0.71,P=0.01),0.31(6.4% vs 22.1%,95% CI:0.12-0.79,P=0.01),0.43(12.7% vs 20.8%,95% CI:0.21-0.89,P=0.02),0.42(25.1% vs 42.0%,95% CI:0.27-0.66,P=0.0002) and 0.28(31.9% vs 52.2%,95% CI:0.13-0.59,P=0.0008).CONCLUSION:This meta-analysis indicates the postoperative antiviral therapy,interferon in particular,may serve as a favorable alternative to reduce recurrence and mortality in patients with HBV/HCV related HCCs.Miao, Ruo-Yu Zhao, Hai-Tao Yang, Hua-Yu Mao, Yi-Lei Lu, Xin Zhao, Yi Liu, Chang-Ning Zhong, Shou-Xian Sang, Xin-Ting Huang, Jie-Fu 2010World Journal of Gastroenterology2010,16,23:26
4Nomogram to predict overall survival after gallbladder cancer resection in China显示文摘AIM To integrate clinically significant variables related to prognosis after curative resection for gallbladder carcinoma(GBC) into a predictive nomogram.METHODS One hundred and forty-two GBC patients who underwent curative intent surgical resection at Peking Union Medical College Hospital(PUMCH) were included. This retrospective case study was conducted at PUMCH of the Chinese Academy of Medical Sciences and Peking Union Medical College(CAMS & PUMC) in China from January 1, 2003 to January 1, 2018. The continuous variable carbohydrate antigen 19-9(CA19-9) was converted into a categorical variable(cCA19-9) based on the normal reference range. Stages 0 to IIIA were merged into one category, while the remaining stages were grouped into another category. Pathological grade X(GX) was treated as a missing value. A multivariate Cox proportional hazards model was used to select variables to construct a nomogram. Discrimination and calibration of the nomogram were performed via the concordance index(C-index) and calibration plots. The performance of the nomogram was estimated using the calibration curve. Receiver operating characteristic(ROC) curve analysis and decision curve analysis(DCA) were performed to evaluate the predictive accuracy and net benefit of the nomogram, respectively.RESULTS Of these 142 GBC patients, 55(38.7%) were male, and the median and mean age were 64 and 63.9 years, respectively. Forty-eight(33.8%) patients in this cohort were censored in the survival analysis. The median survival time was 20 months. A series of methods, including the likelihood ratio test and Akaike information criterion(AIC) as well as stepwise, forward, and backward analyses, were used to select the model, and all yielded identical results. Jaundice [hazard ratio(HR) = 2.9; 95% confidence interval(CI): 1.60-5.27], cCA19-9(HR = 3.2; 95%CI: 1.91-5.39), stage(HR = 1.89; 95%CI: 1.16-3.09), and resection(R)(HR = 2.82; 95%CI: 1.54-5.16) were selected as significant predictors and combined into a survival time predictive nomogram(C-index = 0.803; 95%CI: 0.766-0.839). High prediction accuracy(adjusted C-index = 0.797) was further verified via bootstrap validation. The calibration plot demonstrated good performance of the nomogram. ROC curve analysis revealed a high sensitivity and specificity. 8Neuroprotective effects of bloodletting at Jing points combined with mild induced hypothermia in acute severe traumatic brain injury显示文摘Bloodletting at Jing points has been used to treat coma in traditional Chinese medicine. Mild induced hypothermia has also been shown to have neuroprotective effects. However, the therapeutic effects of bloodletting at Jing points and mild induced hypothermia alone are limited. Therefore, we investigated whether combined treatment might have clinical effectiveness for the treatment of acute severe traumatic brain injury. Using a rat model of traumatic brain injury, combined treatment substantially alleviated cerebral edema and bloodbrain barrier dysfunction. Furthermore, neurological function was ameliorated, and cellular necrosis and the inflammatory response were lessened. These findings suggest that the combined effects of bloodletting at Jing points(20 μL, twice a day, for 2 days) and mild induced hypothermia(6 hours) are better than their individual effects alone. 9Effects of acid deposition on terrestrial ecosystems and their rehabilitation strategies in China显示文摘South China has become the third largest region associated with acid deposition following Europe and North America, the area subject to damage by acid deposition increased from 1.75 million km 2 in 1985 to 2.8 million km 2 in 1993. Acid deposition has caused serious damage to ecosystem. Combined pollution of acid rain and SO 2 showed the obvious multiple effects on crops. Vegetable was more sensitive to acid deposition than foodstuff crops. Annual economic loss of crops due to acid deposition damage in eleven provinces of south China was 4.26 billion RMB Yuan. Acid deposition caused serious damage to forest. Annual economic loss of wood volume was about 1.8 billion RMB Yuan and forest ecological benefit loss 16.2 billion in eleven provinces of south China. Acid deposition in south China was typical “sulfuric acid type”. 10Roles of Smad3 and Smad7 in rat pancreatic stellate cells activated by transforming growth factor-beta 1显示文摘BACKGROUND:Pancreatic stellate cells(PSCs) play a major role in promoting pancreatic fibrosis.Transforming growth factor beta 1(TGF-β1) is a critical mediator of this process.This study aimed to determine the expression of the Smad3 and Smad7 genes in the process of PSC activation,and explore the mechanisms of chronic pancreatitis.METHODS:The expressions of Smad3 and Smad7 in PSCs before and after TGF-β1 treatment were detected by reverse transcription-polymerase chain reaction and Western blotting analysis.Smad3 expression was detected in PSCs after treatment with 5 ng/ml of TGF-β1 for 24 hours.RESULTS:Smad7 expression was decreased in TGF-β1-activated PSCs(P<0.05) in a dose-dependent manner.When TGF-β1 concentration reached 10 ng/ml,the expression of p-Smad3 Smad3,and Smad7 was inhibited(P<0.05).CONCLUSIONS:TGF-β1 promotes the expression of Smad3 and inhibits the expression of Smad7 during the activation of PSCs.In contrast,high-dose TGF-β1 downregulates the expression of Smad3 in completely activated PSCs.Qian, Zhu-Yin Peng, Quan Zhang, Zheng-Wei Thou, Long-An Miao, Yi 2010Hepatobiliary & Pancreatic Diseases International2010,9,5:13
11Roux-en-Y gastric bypass for Chinese type 2 diabetes mellitus patients with a BMI,28kg/m^2:a multi-institutional study显示文摘Roux-en-Y gastric bypass surgery(RYGB) has been demonstrated to be successful for treating type-II diabetes2mellitus(T2DM) patients with a body mass index(BMI),30 kg/m,but reports of RYGB for T2 DM patients with22 a BMI,28 kg/mare lacking.T2 DM patients with a BMI,28 kg/mwere prospectively recruited to participate in this study in four hospitals.The endpoint was T2 DM remission(defined by fasting blood glucose(FBG) level,110 mg/d L and hemoglobin(Hb)A1c level,6.0% at 12 months postoperatively).Predictors of remission were investigated by univariate and multivariate analyses.Eighty-six patients were assessed.Eighty-five patients underwent RYGB,with one conversion to open surgery.We compared the values of various variables before and after2 surgery.The mean BMI decreased from 24.68±2.12 to 21.72±2.43 kg/m(P,0.001).Fifty-eight(67.4%) patients were not treated by drugs or insulin after surgery,and 20 patients(23.3%) had complete remission of T2 DM at12 months after surgery with an acceptable number of complications.The mean Hb A1 c level in the remission group was significantly lower than that in the non-remission group.Patients with a higher weight,lower Hb A1 c level,higher C-peptide level,and higher FBG level were more likely to have T2 DM remission in multivariate2 analyses.In conclusion,RYGB was effective and safe for treating T2 DM patients with a BMI,28 kg/m.Complete remission can be predicted by cases having a higher weight,lower Hb A1 c level,higher C-peptide level,and higher FBG level.Hui Liang Wei Guan Yanling Yang Zhongqi Mao Yijun Mei Huan Liu Yi Miao 2015The Journal of Biomedical Research2015,29,2:13

15Essential roles of exosome and circRNA_101093 on ferroptosis desensitization in lung adenocarcinoma显示文摘Background:Resistance to ferroptosis,a regulated cell death caused by irondependent excessive accumulation of lipid peroxides,has recently been linked to lung adenocarcinoma(LUAD).Intracellular antioxidant systems are required for protection against ferroptosis.The purpose of the present studywas to investigate whether and how extracellular system desensitizes LUAD cells to ferroptosis.Methods:Established human lung fibroblasts MRC-5,WI38,and human LUAD H1650,PC9,H1975,H358,A549,and H1299 cell lines,tumor and matched normal adjacent tissues of LUAD,and plasma from healthy individuals and LUAD patients were used in this study.Immunohistochemistry and immunoblotting were used to analyze protein expression,and quantitative reverse transcription-PCR was used to analyze mRNA expression.Cell viability,cell death,and the lipid reactive oxygen species generationwere measured to evaluate the responses to ferroptosis.Exosomes were observed using transmission electron microscope.The localization of arachidonic acid(AA)was detected using click chemistry labeling followed by confocal microscopy.Interactions between RNAs and proteins were detected using RNA pull-down,RNA immunoprecipitation and photoactivatable ribonucleoside-enhanced crosslinking and immunoprecipitation methods.Proteomic analysis was used to investigate RNA-regulated proteins,and metabolomic analysis was performed to analyze metabolites.Cellderived xenograft,patient-derived xenograft,cell-implanted intrapulmonary LUAD mouse models and plasma/tissue specimens from LUAD patients were used to validate the molecular mechanism.Results:Plasma exosome from LUAD patients specifically reduced lipid peroxidation and desensitized LUAD cells to ferroptosis.A potential explanation is that exosomal circRNA_101093(cir93)maintained an elevation in intracellular cir93 in LUAD to modulate AA,a poly-unsaturated fatty acid critical for ferroptosisassociated increased peroxidation in the plasma membrane.Mechanistically,cir93 interacted with and increased fatty acid-binding protein 3(FABP3),which transported AA and facilitated its reaction with taurine.Thus,global AA was reduced,whereas N-arachidonoyl taurine(NAT,the product of AA and taurine)was induced.Notably,the role of NAT in suppressing AA incorporation into the plasma membrane was also revealed.In pre-clinical in vivo models,reducing exosome improved ferroptosis-based treatment.Conclusion:Exosome and cir93 are essential for desensitizing LUAD cells to ferroptosis,and blocking exosome may be helpful for future LUAD treatment.Xiao Zhang Yunhua Xu Lifang Ma Keke Yu Yongjie Niu Xin Xu Yi Shi Susu Guo Xiangfei Xue Yikun Wang Shiyu Qiu Jiangtao Cui Hong Wang Xiaoting Tian Yayou Miao Fanyu Meng Yongxia Qiao Yongchun Yu Jiayi Wang 2022Cancer Communications2022,42,4:10
16Rosmarinic acid elicits neuroprotection in ischemic stroke via Nrf2 and heme oxygenase 1 signaling显示文摘Rosmarinic acid(RA) can elicit a neuroprotective effect against ischemic stroke, but the precise molecular mechanism remains poorly understood. In this study, an experimental ischemic stroke model was established in CD-1 mice(Beijing Vital River Laboratory Animal Technology, Beijing, China) by occluding the right middle cerebral artery for 1 hour and allowing reperfusion for 24 hours. After intraperitoneally injecting model mice with 10, 20, or 40 mg/kg RA, functional neurological deficits were evaluated using modified Longa scores. Subsequently, cerebral infarct volume was measured using TTC staining and ischemic brain tissue was examined for cell apoptosis with TUNEL staining. Superoxide dismutase activity and malondialdehyde levels were measured by spectrophometry. Expression of heme oxygenase-1(HO-1), nuclear factor erythroid 2-related factor 2(Nrf2), Bcl-2, Bax, Akt, and phospho-Ser473 Akt proteins in ischemic brain tissue was detected by western blot, while mRNA levels of Nrf2, HO-1, Bcl-2, and Bax were analyzed using real time quantitative PCR. In addition, HO-1 enzyme activity was measured spectrophotometrically. RA(20 and 40 mg/kg) greatly improved neurological function, reduced infarct volume, decreased cell apoptosis, upregulated Bcl-2 protein and mRNA expression, downregulated Bax protein and mRNA expression, increased HO-1 and Nrf2 protein and mRNA expression, increased superoxide dismutase activity, and decreased malondialdehyde levels in ischemic brain tissue of model mice. However, intraperitoneal injection of a HO-1 inhibitor(10 mg/kg zinc protoporphyrin IX) reversed the neuroprotective effects of RA on HO-1 enzyme activity and Bcl-2 and Bax protein expression. The PI3 K/Akt signaling pathway inhibitor LY294002(10 mM) inhibited Akt phosphorylation, as well as Nrf2 and HO-1 expression. 17Review of the fabrication and application of porous materials from silicon-rich industrial solid waste显示文摘Porous materials have promise as sound insulation, heat barrier, vibration attenuation, and catalysts. Most industrial solid wastes, such as tailings, coal gangue, and fly ash are rich in silicon. Additionally, a high silicon content waste is a potential raw material for the syn- thesis of silicon-based, multi-porous materials such as zeolites, mesoporous silica, glass-ceramics, and geopolymer foams. Representative sil- icon-rich industrial solid wastes (SRISWs) are the focus of this mini review of the processing and application of porous silicon materials with respect to the physical and chemical properties of the SRISW. The transformation methods of preparing porous materials from SRISWs are summarized, and their research status in micro-, meso-, and macro-scale porous materials are described. Possible problems in the application of SRISWs and in the preparation of functional porous materials are analyzed, and their development prospects are discussed. 19Simulated Microgravity Conditions and Carbon Ion Irradiation Induce Spermatogenic Cell Apoptosis and Sperm DNA Damage显示文摘Objective To investigate the effect of simulated microgravity and carbon ion irradiation(CIR) on spermatogenic cell apoptosis and sperm DNA damage to the testis of male Swiss Webster mice,and assess the risk associated with space environment.Methods Sperm DNA damage indicated by DNA fragmentation index(DFI) and high DNA stainability(HDS) was measured by sperm chromatin structure assay(SCSA).Apoptosis of spermatogenic cells was detected by annexin V-propidium iodide assay.Bax(the expression levels of p53) and proliferating cell nuclear antigen(PCNA) were measured by immunoblotting;p53 and PCNA were located by immunohistology.Results HDS,DFI,apoptosis index,and the expression levels of p53 and Bax were detected to be significantly higher in the experimental groups(P<0.05) compared with those in the control group;however,the PCNA expression varied to a certain degree.p53-and PCNA-positive expression were detected in each group,mainly in relation to the spermatogonic cells and spermatocytes.Conclusion The findings of the present study demonstrated that simulated microgravity and CIR can induce spermatogenic cell apoptosis and sperm DNA damage.Sperm DNA damage may be one of the underlying mechanisms behind male fertility decline under space environment.These findings may provide a scientific basis for protecting astronauts and space traveler's health and safety.LI Hong Yan ZHANG Hong MIAO Guo Ying XIE Yi SUN Chao DI Cui Xia LIU Yang LIU Yuan Yuan ZHANG Xin MA Xiao Fei XU Shuai GAN Lu ZHOU Xin 2013Biomedical and Environmental Sciences2013,26,9:10
