1The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 4Newly discovered hydrothermal fields along the ultraslow-spreading Southwest Indian Ridge around 63°E显示文摘The ultraslow-spreading Southwest Indian Ridge(SWIR) to the east of the Melville fracture zone is characterized by very low melt supply and intensive tectonic activity. Due to its weak thermal budget and extremely slow spreading rate, the easternmost SWIR was considered to be devoid of hydrothermal activity until the discovery of the inactive Mt. Jourdanne hydrothermal field(27°51′S, 63°56′E) in 1998. During the COMRA DY115-20 cruise in2009, two additional hydrothermal fields(i.e., the Tiancheng(27°51′S, 63°55′E) and Tianzuo(27°57′S, 63°32′E)fields) were discovered. Further detailed investigations of these two hydrothermal sites were conducted by Chinese manned submersible Jiaolong in 2014–2015. The Tiancheng filed can be characterized as a lowtemperature(up to 13.2°C) diffuse flow hydrothermal field, and is hosted by fractured basalts with hydrothermal fauna widespread on the seafloor. The Tianzuo hydrothermal field is an inactive sulfide field, which is hosted by ultramafic rocks and controlled by detachment fault. The discovery of the three hydrothermal fields around Segment #11 which receives more melt than the regional average, provided evidence for local enhanced magmatism providing heat source to drive hydrothermal circulation. 5Classification of four distinct osteoarthritis subtypes with a knee joint tissue transcriptome atlas显示文摘The limited molecular classifications and disease signatures of osteoarthritis(OA)impede the development of prediagnosis and targeted therapeutics for OA patients.To classify and understand the subtypes of OA,we collected three types of tissue including cartilage,subchondral bone,and synovium from multiple clinical centers and constructed an extensive transcriptome atlas of OA patients.By applying unsupervised clustering analysis to the cartilage transcriptome,OA patients were classified into four subtypes with distinct molecular signatures:a glycosaminoglycan metabolic disorder subtype(C1),a collagen metabolic disorder subtype(C2),an activated sensory neuron subtype(C3),and an inflammation subtype(C4).Through ligand-receptor crosstalk analysis of the three knee tissue types,we linked molecular functions with the clinical symptoms of different OA subtypes.For example,the Gene Ontology functional term of vasculature development was enriched in the subchondral bone-cartilage crosstalk of C2 and the cartilage-subchondral bone crosstalk of C4,which might lead to severe osteophytes in C2 patients and apparent joint space narrowing in C4 patients.Based on the marker genes of the four OA subtypes identified in this study,we modeled OA subtypes with two independent published RNA-seq datasets through random forest classification.The findings of this work contradicted traditional OA diagnosis by medical imaging and revealed distinct molecular subtypes in knee OA patients,which may allow for precise diagnosis and treatment of OA.Chunhui Yuan Zongyou Pan Kun Zhao Jun Li Zixuan Sheng Xudong Yao Hua Liu Xiaolei Zhang Yang Yang Dongsheng Yu Yu Zhang Yuzi Xu Zhi-Yong Zhang Tianlong Huang Wanlu Liu Hongwei Ouyang 2020Bone Research2020,8,4:4

10Effect of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV 6) for the treatment of diarrhea-dominant irritable bowel syndrome: a patient-blinded pilot trial with orthogonal design显示文摘OBJECTIVE: To investigate the effects of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV6) in patients with diarrhea-dominant irritable bowel syndrome(D-IBS) by assessing predominant factors, and determining the best factor combinations and their interactions. To identify the optimal quantity of warming moxibustion Tianshu(ST 25, bilateral) and Qihai(CV 6) to achieve optimal efficacy.METHODS: An L9(3~4) orthogonal design was applied to 233 confirmed subjects based on the three most influential factors and the three most frequently used levels. Nine programs were designed.Subjects were assigned to four subgroups according to age and gender; each subject underwent one of the nine programs randomly. We selected Tianshu(ST 25, bilateral) and Qihai(CV 6). The scores of symptoms and the IBS Quality of Life(IBS-QOL) were evaluated after the final treatment.RESULTS: After treatment, clinical symptoms of all patients were significantly improved(P = 0.001).Quality of life also improved significantly(P < 0.05).For all the participants, frequency was the dominating factor regarding symptoms(P = 0.01). Duration was the dominating factor for quality of life(P =0.0001). The best combination for improving symptoms in 30-40-year-old male and female patients was two courses of 10 min each once a day, and for41-50-year-old male and female patients it was three courses of 10 min(males) or 30 min(females)twice a day. The best combination for quality of life was three courses of 20 min once a day. No interaction was found between frequency and course, or frequency and duration.CONCLUSION: Warming moxibustion Tianshu(ST25, bilateral) and Qihai(CV 6) is a promising therapy for D-IBS. 18Advanced duodenal carcinoma:Chemotherapy efficacy and analysis of prognostic factors显示文摘Objective This study aimed to determine the efficacy of chemotherapy and to identify potential chemotherapy agents to treat advanced primary duodenal carcinoma(PDC). Methods Seventy-three patients with advanced PDC were included in the study. Response rate(RR), disease control rate(DCR), progression-free survival(PFS), overall survival(OS) and prognosis were compared among patients using the Cox proportional hazards model. Results The overall RR and DCR of 52 patients were 21.15% and 69.23%, respectively. The median PFS and OS times were 4.51 and 11.47 months, respectively. Palliative chemotherapy improved the OS of patients with advanced PDC compared with patients who did not receive chemotherapy(14.28 months vs. 5.20 months, HR = 0.205, 95% CI: 0.077 to 0.547, P = 0.0016). Multivariate analysis indicated mucinous histology and liver metastasis as factors predictive of poor prognosis in patients with advanced PDC. Conclusion Palliative chemotherapy may improve the OS of patients with advanced PDC.
