| 2 | Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. | 3 | Electrochemical and spectroscopic characteristics of dissolved organic matter in a forest soil profile显示文摘Dissolved organic matter(DOM) represents one of the most mobile and reactive organic compounds in ecosystem and plays an important role in the fate and transport of soil organic pollutants, nutrient cycling and more importantly global climate change.Electrochemical methods were first employed to evaluate DOM redox properties, and spectroscopic approaches were utilized to obtain information concerning its composition and structure. DOM was extracted from a forest soil profile with five horizons. Diferential pulse voltammetry indicated that there were more redox-active moieties in the DOM from upper horizons than in that from lower horizons.Cyclic voltammetry further showed that these moieties were reversible in electron transfer. Chronoamperometry was employed to quantify the electron transfer capacity of DOM, including electron acceptor capacity and electron donor capacity, both of which decreased sharply with increasing depth. FT-IR, UV-Vis and fluorescence spectra results suggested that DOM from the upper horizons was enriched with aromatic and humic structures while that from the lower horizons was rich in aliphatic carbon, which supported the findings obtained by electrochemical approaches. Electrochemical approaches combined with spectroscopic methods were applied to evaluate the characteristics of DOM extracted along a forest soil profile. | 5 | NbMoTiVSi_(x) refractory high entropy alloys strengthened by forming BCC phase and silicide eutectic structure显示文摘In order to improve mechanical properties of refractory high entropy alloys,silicide was introduced and NbMoTiVSi_(x)(x=0,0.1,0.2,0.3,and 0.4,molar ratio) refractory high entropy alloys are prepared by vacuum arc melting.Phase composition,micro structure evolution and mechanical properties were systematically studied.Results show that the silicide phase is formed in the alloys with addition of silicon,and the volume fraction of silicide increases from 0 to 8.3 % with increasing of silicon.Microstructure observation shows that the morphology of dendrite changes from columnar to near equiaxed,eutectic structure is formed at grain boundaries and composed of secondary BCC phase and silicide phase.The average length of the primary and second dendrites decreases with the increasing of silicon.Whereas,the ratio of eutectic structure increases from 0 to 19.8 % with the increment of silicon.The refinement of microstructure is caused by heterogeneous nucleation from the silicide.Compressive tests show that the yield and ultimate strength of the alloys increases from 1141.5 MPa to 2093.1 MPa and from 1700.1 MPa to 2374.7 MPa with increasing silicon content.The fracture strain decreases from 24.7 %-11.0 %.Fracture mechanism is changed from ductile fracture to ductile and brittle mixed fracture.The improvement of the strength is caused by grain bounda ry strengthening,which includes more boundaries around primary BCC phase and eutectic structure in grain boundary,both of them is resulted from the formation of silicide. | Qin Xu Dezhi Chen Chongyang Tan Xiaoqin Bi Qi Wang Hongzhi Cui Shuyan Zhang Ruirun Chen | 2021 | Journal of Materials Science & Technology2021,,1: | 9 |
| 6 | Subtyping Animal Influenza Virus with General Multiplex RT-PCR and Liquichip High Throughput (GMPLex)显示文摘This study developed a multiplex RT-PCR integrated with luminex technology to rapidly subtype simultaneously multiple influenza viruses. Primers and probes were designed to amplify NS and M genes of influenza A viruses HA gene of H1, H3, H5, H7, H9 subtypes, and NA gene of the N1 and N2 subtypes. Universal super primers were introduced to establish a multiplex RT-PCR (GM RT-PCR). It included three stages of RT-PCR amplification, and then the RT-PCR products were further tested by LiquiChip probe, combined to give an influenza virus (IV) rapid high throughput subtyping test, designated as GMPLex. The IV GMPLex rapid high throughput subtyping test presents the following features: high throughput, able to determine the subtypes of 9 target genes in H1, H3, H5, H7, H9, N1, and N2 subtypes of the influenza A virus at one time; rapid, completing the influenza subtyping within 6 hours; high specificity, ensured the specificity of the different subtypes by using two nested degenerate primers and one probe, no cross reaction occurring between the subtypes, no non-specific reactions with other pathogens and high sensitivity. When used separately to detect the product of single GM RT-PCR for single H5 or N1 gene, the GMPLex test showed a sensitivity of 10-5(= 280ELD50) forboth tests and the Luminex qualitative ratio results were 3.08 and 3.12, respectively. When used to detect the product of GM RT-PCR for H5N1 strain at the same time, both showed a sensitivity of 10-4(=2800 ELD50). | 9 | Multivariate predictive model for asymptomatic spontaneous bacterial peritonitis in patients with liver cirrhosis显示文摘BACKGROUNDSpontaneous bacterial peritonitis (SBP) is a detrimental infection of the asciticfluid in liver cirrhosis patients, with high mortality and morbidity. Earlydiagnosis and timely antibiotic administration have successfully decreased themortality rate to 20%-25%. However, many patients cannot be diagnosed in theearly stages due to the absence of classical SBP symptoms. Early diagnosis ofasymptomatic SBP remains a great challenge in the clinic.AIMTo establish a multivariate predictive model for early diagnosis of asymptomaticSBP using positive microbial cultures from liver cirrhosis patients with ascites.METHODSA total of 98 asymptomatic SBP patients and 98 ascites liver cirrhosis patients withnegative microbial cultures were included in the case and control groups,respectively. Multiple linear stepwise regression analysis was performed toidentify potential indicators for asymptomatic SBP diagnosis. The diagnosticperformance of the model was estimated using the receiver operatingcharacteristic curve.RESULTSPatients in the case group were more likely to have advanced disease stages,cirrhosis related-complications, worsened hematology and ascites, and higher mortality. Based on multivariate analysis, the predictive model was as follows: y (P) = 0.018 + 0.312 × MELD (model of end-stage liver disease) + 0.263 × PMN(ascites polymorphonuclear) + 0.184 × N (blood neutrophil percentage) + 0.233 ×HCC (hepatocellular carcinoma) + 0.189 × renal dysfunction. The area under thecurve value of the established model was 0.872, revealing its high diagnosticpotential. The diagnostic sensitivity was 73.5% (72/98), the specificity was 86.7%(85/98), and the diagnostic efficacy was 80.1%.CONCLUSIONOur predictive model is based on the MELD score, polymorphonuclear cells,blood N, hepatocellular carcinoma, and renal dysfunction. | 10 | IL-17A-dependent gut microbiota is essential for regulating diet-induced disorders in mice显示文摘The gut microbiota plays a key role in obesity and related metabolic disorders, and multiple factors including diet, host genotype, and age regulate it. Many studies have examined the contribution of extrinsic factors to the regulation of the gut microbiota, but the importance of the host genetic constitution cannot be ignored. Interleukin 17A(IL-17A), a pro-inflammatory cytokine, is important in the defense against infection and diseases. Here, we investigated the association among IL-17, a high-fat diet(HFD), and the gut microbiota. Mice deficient in IL-17A were resistant to diet-induced obesity and related diseases. Compared with the Ⅱ-17^(-/-)mice, wild-type(WT) mice challenged with HFD showed obvious weight fluctuations, such as those seen in type 2 diabetes, and hematological changes similar to those associated with metabolic syndrome. However, housing WT mice and Ⅱ-17a^(-/-)mice together significantly alleviated these symptoms in the WT mice. A metagenomic analysis of the mouse feces indicated that the microbial community compositions of these two groups differed before HFD feeding. The HFD mediated shifts in the gut microbial compositions, which were associated with the mouse phenotypes.We also identified potentially beneficial and harmful species present during this period, and drew networks of the most abundant species. A functional analysis indicated pathway changes in the WT and Ⅱ-17a^(-/-)mice when fed the HFD. | 12 | Microstructure,tensile properties and thermal stability of AlMgSiScZr alloy printed by laser powder bed fusion显示文摘The new generation of Sc and Zr modified Al alloys has been attracted wide concerns in aerospace industry,owing to the excellent mechanical performances and superior thermal stability than other normal Al alloys.By microalloying with Sc and Zr,the Al3(Sc,Zr)particle forms as the grain refiner during the solidification,which is extremely beneficial for the laser powder bed fusion(LPBF)processed Al alloys.In this study,a new type Al-14.1 Mg-0.47 Si-0.31 Sc-0.17 Zr alloy was additively manufactured by LPBF,and the microstructure,tensile properties and thermal stability were studied in detail.By using a single melt-67°scanning strategy,the LPBF-processed specimen with a relative density of 99.4%and tensile strength of 487.7 MPa was obtained at 160 W-200 mm/s.And this AlMgSiScZr alloy can still exhibit an excellent tensile strength of 393.9 MPa at a moderate temperature of 473 K.After the aging treatment,the tensile properties further increased due to the precipitate hardening of Mg2Si and Al3(Sc,Zr),and the maximum value(580 MPa)was reached at an aging time of 10 h.The average crystal size was almost unchanged after aging treated at 325℃and 24 h,indicating this AlMgSiScZr alloy has an improved thermal stability.The AlMgSiScZr alloy is recommended to substitute some particular titanium alloys in aerospace field afterwards. | Jiang Bi Zhenglong Lei Yanbin Chen Xi Chen Ze Tian Nannan Lu Xikun Qin Jingwei Liang | 2021 | Journal of Materials Science & Technology2021,,10: | 4 |
| 13 | Ultraflat, broadband, and highly coherent supercontinuum generation in all-solid microstructured optical fibers with all-normal dispersion显示文摘High flatness, wide bandwidth, and high-coherence properties of supercontinuum(SC) generation in fibers are crucial in many applications. It is challenging to achieve SC spectra in a combination of the properties, since special dispersion profiles are required, especially when pump pulses with duration over 100 fs are employed. We propose an all-solid microstructured fiber composed only of hexagonal glass elements. The optimized fiber possesses an ultraflat all-normal dispersion profile, covering a wide wavelength interval of approximately 1.55 μm. An SC spectrum spanning from approximately 1030 to 2030 nm(corresponding to nearly one octave) with flatness<3 dB is numerically generated in the fiber with 200 fs pump pulses at 1.55 μm. The results indicate that the broadband ultraflat SC sources can be all-fiber and miniaturized due to commercially achievable 200-fs fiber lasers. Moreover, the SC pulses feature high coherence and a single pulse in the time domain, which can be compressed to 13.9-fs pulses with high quality even for simple linear chirp compensation. | 16 | Detection of Porcine Epidemic Diarrhea Virus in Guangxi Province from 2011 to 2014 and Sequence Analysis of Its M Gene显示文摘Detection of pigs epidemic diarrhea virus( PEDV) was conducted on 331 piglets diarrhea fecal samples collected in Nanning,Yulin and other 12 areas of Guangxi Province from January of 2011 to April of 2014 by the method of reverse transcription-polymerase chain reaction( RT-PCR). The results showed that the positive samples of PEDV were 210 and the positive rate was 63. 44%. The clone and sequencing of M gene was carried out on 25 positive samples. PEDV reference strains were selected from Gene Bank to conduct the sequence homology alignment analysis and the phylogenetic tree of M gene. The M gene homology and amino acid sequence identity between 25 isolated strains and 51 reference strains were 96. 0%- 99. 6% and 94. 3%- 99. 6%,respectively. The genetic variation analysis of M gene showed that the genetic relationship of PEDV prevalent strains in Guangxi Province from 2013 to 2014 was close to that of the prevalent strains in Beijing,Anhui,Wuhan,Hebei and Guangdong from 2010 to 2013,and which were far from that of the Chinese early isolates CH / S( Gen Bank number: JN547228),vaccine strain CV777( Gen Bank number: AF353511) and Attenuated DR13( Gen Bank number: JQ023162). | 17 | Biodistribution and Toxicity Assessment of Superparamagnetic Iron Oxide Nanoparticles In Vitro and In Vivo显示文摘Biodistribution and toxicity assessment are critical for safe clinical use of newly developed medicines.Superparamagnetic iron oxide nanoparticles (SPION)are effective carriers for targeted drug delivery.This study aimed to examine the toxicity and biodistribution of SPION coated with polyethylenimine (PEI)(SPION-PEI)designed for small interfering RNA (siRNA) delivery both in vitro and in vivo.SPION-PEI/siRNA complexes were prepared at different weight ratios.Cytotoxic effects of SPION-PEI/siRNA on HSC-T6 cell viability were determined by using 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT).Rats were divided into three groups:a control group,a normal-saline group and a SPION-PEI/siRNA group.After a single intravenous injection,in vivo nanoparticle biodistribution and accumulation were evaluated by Prussian blue staining in the heart,liver,spleen,lung and kidney 8 h,24 h,and 7 days after the injection.Their distribution was histologically studied at the three time points by measuring ironpositive areas (μm2)in organ sections stained with Prussian blue.The same organs were analyzed by H&E staining for any possible histopathological changes.Furthermore,biochemical indexes such as alanine amino transaminase (ALT),aspartate transaminase (AST),blood urea nitrogen (BUN)and creatinine (CREA)were also assessed at all experimental time points.Electrophoresis exhibited that the SPION-PEI could retard siRNA altogether at weight ratios above 4.MTT assay showed that SPION-PEI loaded with siRNA had low cytotoxicity.In vivo study revealed that the liver and spleen were the major sites of SPION-PEI/siRNA deposition.The iron content was significantly increased in the liver and spleen,peaking 24 h after intravenous injection and then declining gradually.No evidence was found of irreversible histopathological damage to any of the organs tested.These results suggested that most SPION-PEI/siRNA complexes were distributed in the liver and spleen,which might be the target organs of SPION-PEI/siRNA complexes.SPION- PEI/siRNA may serve as in vivo carrier for biomedical medicines. | Qin YU Xiao-qin XIONG Lei ZHAO Ting-ting XU Hao BI Rong FU Qian-hua WANG | 2018 | Current Medical Science2018,38,6: | 3 |
| 18 | Fusobacterium nucleatum enhances the efficacy of PD-L1 blockade in colorectal cancer显示文摘Given that only a subset of patients with colorectal cancer(CRC)benefit from immune checkpoint therapy,efforts are ongoing to identify markers that predict immunotherapeutic response.Increasing evidence suggests that microbes influence the efficacy of cancer therapies.Fusobacterium nucleatum induces different immune responses in CRC with different microsatellite-instability(MSI)statuses.Here,we investigated the effect of F.nucleatum on anti-PD-L1 therapy in CRC.We found that high F.nucleatum levels correlate with improved therapeutic responses to PD-1 blockade in patients with CRC.Additionally,F.nucleatum enhanced the antitumor effects of PD-L1 blockade on CRC in mice and prolonged survival.Combining F.nucleatum supplementation with immunotherapy rescued the therapeutic effects of PD-L1 blockade.Furthermore,F.nucleatum induced PD-L1 expression by activating STING signaling and increased the accumulation of interferon-gamma(IFN-γ)^(+)CD8^(+)tumor-infiltrating lymphocytes(TILs)during treatment with PD-L1 blockade,thereby augmenting tumor sensitivity to PD-L1 blockade.Finally,patient-derived organoid models demonstrated that increased F.nucleatum levels correlated with an improved therapeutic response to PD-L1 blockade.These findings suggest that F.nucleatum may modulate immune checkpoint therapy for CRC. | Yaohui Gao Dexi Bi Ruting Xie Man Li Jing Guo Hu Liu Xianling Guo Juemin Fang Tingting Ding Huiyuan Zhu Yuan Cao Meichun Xing Jiayi Zheng Qing Xu Qian Xu Qing Wei Huanlong Qin | 2021 | Signal Transduction and Targeted Therapy2021,6,12: | 3 |

| 20 | An additively manufactured Al-14.1Mg-0.47Si-0.31Sc-0.17Zr alloy with high specific strength,good thermal stability and excellent corrosion resistance显示文摘A novel Al-14.1 Mg-0.47 Si-0.31 Sc-0.17 Zr alloy was applied in the printing process of selective laser melting(SLM),and the corresponding microstructural feature,phase identification,tensile properties and corrosion behavior of the Al Mg Si Sc Zr alloy were studied in detail.As fabricated at 160 W and 200 mm/s,the Mg content of bulk sample decreased to 11.7 wt%due to the element vaporization at high energy density,and the density of this additively manufactured Al Mg Si Sc Zr alloy was 2.538 g/cm^(3),which is4.2%8.5%lighter than that of other SLM-processed Al alloys.After heat-treated(HT)at 325℃and 6 h,the microstructure was almost unchanged with an alternate distribution of fine equiaxed crystals and coarse columnar crystals.Nano-sized Al3(Sc,Zr)and Mg_(2)Si phases precipitated dispersedly in the Al matrix,and the tensile strength increased from 487.6 MPa to 578.4 MPa for precipitation strengthening and fine grain strengthening.With a fine grain size of 2.53μm,an excellent corrosion resistance was obtained for the as-printed(AP)Al Mg Si Sc Zr alloy.While the corrosion resistance of HT sample decreased slightly for the formation of non-dense oxide layer and pitting corrosion induced by diffuse precipitation distribution.This SLM-printed Al Mg Si Sc Zr alloy with high specific strength,good thermal stability and excellent corrosion resistance has broad prospects for the aerospace and automotive applications. | Jiang Bi Zhenglong Lei Yanbin Chen Xi Chen Nannan Lu Ze Tian Xikun Qin | 2021 | Journal of Materials Science & Technology2021,,8: | 2 |
