1Rice Brittleness Mutants: A Way to Open the ‘Black Box’ of Monocot Cell Wall Biosynthesis显示文摘Rice is a model organism for studying the mechanism of cell wall biosynthesis and remolding in Gramineae.Mechanical strength is an important agronomy trait of rice (Oryza sativa L.) plants that affects crop lodging and grain yield.As a prominent physical property of cell walls,mechanical strength reflects upon the structure of different wall polymers and how they interact.Studies on the mechanisms that regulate the mechanical strength therefore consequently results in uncovering the genes functioning in cell wall biosynthesis and remodeling.Our group focuses on the study of isolation of brittle culm (bc) mutants and characterization of their corresponding genes.To date,several bc mutants have been reported.The identified genes have covered several pathways of cell wall biosynthesis,revealing many secrets of monocot cell wall biosynthesis.Here,we review the progress achieved in this research field and also highlight the perspectives in expectancy.All of those lend new insights into mechanisms of cell wall formation and are helpful for harnessing the waste rice straws for biofuel production.Baocai Zhang Yihua Zhou 2011Journal of Integrative Plant Biology2011,53,2:23
2Genetic Contribution of Paleopolyploidy to Adaptive Evolution in Angiosperms显示文摘Ancient whole-genome duplications (WGDs or polyploidy) are prevalent in plants, and some WGDs occurred during the timing of severe global environmental changes. It has been suggested that WGDs may have contributed to plant adaptation. However, this still lacks empirical evidence at the genetic level to support the hypothesis. Here, we investigated the survivors of gene duplicates from multiple ancient WGD events on the major branches of angiosperm phylogeny, and aimed to explore genetic evidence supporting the significance of polyploidy. Duplicated genes co-retained from three waves of independent WGDs (~120 million years ago [Ma], ~66, and <20 Ma) were investigated in 25 selected species. Gene families functioning in low temperature and darkness were commonly retained gene duplicates after the eight independently occurring WGDs in many lineages around the Cretaceous-Paleocene boundary, when the global cooling and darkness were the two main stresses. Moreover, the commonly retained duplicates could be key factors which may have contributed to the robustness of the critical stress-related pathways. In addition, genome-wide transcription factors (TFs) functioning in stresses tend to retain duplicates after waves of WGDs, and the coselected gene duplicates in many lineages may play critical roles during severe environmental stresses. 4The plant cell wall: Biosynthesis, construction, and functions显示文摘The plant cell wall is composed of multiple biopolymers, representing one of the most complex structural networks in nature. Hundreds of genes are involved in building such a natural masterpiece. However, the plant cell wall is the least understood cellular structure in plants. Due to great progress in plant functional genomics,manyachievementshavebeenmadein uncovering cell wall biosynthesis, assembly, and architecture, as well as cell wall regulation and signaling. Such information has significantly advanced our understanding of the roles of the cell wall in many biological and physiological processes and has enhanced our utilization of cell wall materials. The use of cutting-edge technologies such as single-molecule imaging,nuclear magnetic resonance spectroscopy, and atomic force microscopy has provided much insight into the plant cell wall as an intricate nanoscale network, opening up unprecedented possibilities for cell wall research. 5Constructing a high-density linkage map for Gossypium hirsutum × Gossypium barbadense and identifying QTLs for lint percentage显示文摘To introgress the good fiber quality and yield from Gossypium barbadense into a commercial Upland cotton variety, a high-density simple sequence repeat(SSR) genetic linkage map was developed from a BC1F1 population of Gossypium hirsutum×Gossypium barbadense. The map comprised 2,292 loci and covered 5115.16 centi Morgan(c M) of the cotton AD genome, with an average marker interval of 2.23 c M. Of the marker order for 1,577 common loci on this new map, 90.36% agrees well with the marker order on the D genome sequence genetic map. Compared with five published high-density SSR genetic maps, 53.14% of marker loci were newly discovered in this map. Twenty-six quantitative trait loci(QTLs) for lint percentage(LP) were identified on nine chromosomes. Nine stable or common QTLs could be used for marker-assisted selection. Fifty percent of the QTLs were from G. barbadense and increased LP by 1.07%–2.41%. 6A quality evaluation strategy for Rhizoma coptidis from a variety of different sources using chromatographic fingerprinting combined with biological fingerprinting显示文摘Most of herbal medicines of different sources are used for the same treatment goal in clinic despite their tremendous differences of the quality.Because the clinical efficacy is closely related to the quality of medicines,the implementation of an effective quality control(QC)process is vital.Chemical methods based on content determination and chromatographic fingerprints(CFs)have been recommended as powerful tools.In spite of advances in QC,it is still difficult to determine that which of the chemical compositions can best reflect the correlation with the desired biological activity.Herein,we have focused on the biological activities of multi-source herbal medicines to establish an analytical method.By looking for correlations between the biological activities and their chemical compositions,the method can identify reasonable chemical indicators or chemical methods with biological activity information.In this study,high-performance liquid chromatography(HPLC)and microcalorimetry are combined as a useful method for evaluating the quality of Rhizoma coptidis from different sources.The results of biological fingerprints(BFs)showed that the antibacterial activity sequence of R.coptidis of different sources was Coptis teeta Wall.(CT)>Coptis chinensis Franch.(CC)>Coptis deltoidea C.Y.Cheng et Hsiao(CD).The'spectrum-activity'relationships between CFs and BFs were investigated using the canonical correlation analysis(CCA).Compared with the CCA result of CFs and BFs,the canonical correlation coefficient r1and r2were 0.9974 and 0.9752,respectively.It revealed that there are a close correlation between the spectrum-activity relationships and several of the major anti-bacterial components in the CFs,including jatrorrhizine,coptisine,palmatine,and berberine.Of course,R.coptidis was only a representative of multisource medical plants.The result of the current study will provide an effective reference for evaluating the quality of multisource herbal medicines.LI JunXian YAN Dan MA LiNa XIONG Yin YAN ChunXia LI BaoCai PENG Cheng XIAO XiaoHe 2013Chinese Science Bulletin2013,58,33:5

8Stimulation impact of electric currents on heterotrophic denitrifying microbial viability and denitrification performance in high concentration nitrate-contaminated wastewater显示文摘Electric current stimulation has been shown to have a positive influence on heterotrophic denitrifying microbial viability and has the potential to improve wastewater denitrification performance. This study investigated the effects of varying current densities on microbial activity and NO_3^- removal efficiency under heterotrophic conditions.NO_3^-removal rate was highest at an applied current density of 400 mA/m^2. However, the optimum removal efficiency of total inorganic nitrogen(TIN; 99%) was achieved when the current density was fixed at 200 m A/m^2. Accumulation of NH_4^+-N and NO_2^--N byproducts were also minimized at this current density. The activity of heterotrophic denitrifying microorganisms was much higher at both 200 and 400 mA/m^2. Moreover, the average adenosine-5′-triphosphate(ATP)content(an indicator of cell metabolism) at a current density of 1600 mA/m^2 was lower than that under no current, indicating heterotrophic denitrifying microbial activity can be inhibited at high current densities. Hence, direct electrical stimulation on the activity of heterotrophic denitrifying microorganisms in the developed system should be lower than 1600 mA/m^2. 11Phenylpropanoid Derivatives Are Essential Components of Sporopollenin in Vascular Plants显示文摘The outer wall of pollen and spores,namely the exine,is composed of sporopollenin,which is highly resistant to chemical reagents and enzymes.In this study,we demonstrated that phenylpropanoid pathway derivatives are essential components of sporopollenin in seed plants.Spectral analyses showed that the autofluorescence of Lilium and Arabidopsis sporopollenin is similar to that of lignin.Thioacidolysis and NMR analyses of pollen from Lilium and Cryptomeria further revealed that the sporopollenin of seed plants contains phenylpropanoid derivatives,including p-hydroxybenzoate(p-BA),p-coumarate(p-CA),ferulate(FA),and lignin guaiacyl(G)units.The phenylpropanoid pathway is expressed in the tapetum in Arabidopsis,consistent with the fact that the sporopollenin precursor originates from the tapetum.Further germination and comet assays showed that this pathway plays an important role in protection of pollen against UV radiation.In the pteridophyte plant species Ophioglossum vulgatum and Lycopodium clavata,phenylpropanoid derivatives including p-BA and p-CA were also detected,but G units were not.Taken together,our results indicate that phenylpropanoid derivatives are essential for sporopollenin synthesis in vascular plants.In addition,sporopollenin autofluorescence spectra of bryophytes,such as Physcomitrella and Haplocladium,exhibit distinct characteristics compared with those of vascular plants,indicating the diversity of sporopollenin among land plants.Jinp-Shi Xue Baocai Zhang HuaDong Zhan Yong-Lin Lv Xin-Lei Jia TianHua Wang Nai-Ying Yang Yu-Xia Lou Zai-Bao Zhang Wen-Jing Hu Jinshan Gui Jianguo Cao Ping Xu Yihua Zhou Jin-Feng Hu Laigeng Li Zhong-Nan Yang 2020Molecular Plant2020,13,11:2

13Regulating the morphology of fluorinated non-fullerene acceptor and polymer donor via binary solvent mixture for high efficiency polymer solar cells显示文摘Fluorinated non-fullerene acceptors(NFAs) usually have planar backbone and a higher tendency to crystallize compared to their non-fluorinated counterparts, which leads to enhanced charge mobility in organic solar cells(OSCs). However, this selforganization behavior may result in excessive phase separation with electron donors and thereby deteriorate device efficiency.Herein, we demonstrate an effective approach to tune the molecular organization of a fluorinated NFA(INPIC-4 F), and its phase separation with the donor PBDB-T, by varying the casting solvent. A prolonged film drying time encourages the crystallization of INPIC-4 F into spherulites and consequently results in excessive phase separation, leading to a low device power conversion efficiency(PCE) of 8.1%. Contrarily, a drying time leads to fine mixed domains with inefficient charge transport properties,resulting in a moderate device PCE of 11.4%. An intermediate film drying time results in the formation of face-on π-π stacked PBDB-T and INPIC-4 F domains with continuous phase-separated networks, which facilitates light absorption, exciton dissociation as well as balanced charge transport towards the electrode, and achieves a remarkable PCE of 13.1%.
