5A highly sensitive SPE-liquid/liquid extraction-RPLC analytical method for the determination of 6β-hydroxycortisol and cortisol in cancer patients' urine显示文摘Objective: A highly sensitive SPE-liquid/liquid extraction RPLC method has been developed for the analysis of 6β-hydroxycortisol and cortisol in the urine of cancer patients. Methods: After SPE column purification and liquid-liquid extraction, the sample test solutions were analyzed with RPLC using a C18 analytical column. This improved analytical method has been validated for linearity, accuracy (recovery from urine), repeatability (within-day and between-day precision), specificity, sensitivity, and stability. This SPE-liquid/liquid extraction-RPLC is rapid, simple, accurate and reproducible. The technique is particularly useful for monitoring the CYP3A activity of cancer patients in clinical settings. The results are expressed as the ratio of 6β-hydroxycortisol to cortisol. Results: The CYP3A activity from a total of 153 samples was measured using this improved method. Considerable variation in the CYP3A activity of different cancer patients has been documented. Thus, personalized medical treatment based on the individual metabolic enzyme activity level is necessary. 7Development and Characterization of Microsatellite Markers for the Pacific Abalone (Haliotis discus) via EST Database Mining显示文摘The EST database of the Pacific abalone (Haliotis discus) was mined for developing microsatellite markers. A total of 1476 EST sequences were registered in GenBank when data mining was performed. Fifty sequences (approximately 3.4%) were found to contain one or more microsatellites. Based on the length and GC content of the flanking regions, cluster analysis and BLASTN, 13 microsatellite-containing ESTs were selected for PCR primer design. The results showed that 10 out of 13 primer pairs could amplify scorable PCR products and showed polymorphism. The number of alleles ranged from 2 to 13 and the values of Ho and He varied from 0.1222 to 0.8611 and 0.2449 to 0.9311, respectively. No significant linkage disequilibrium (LD) between any pairs of these loci was found, and 6 of 10 loci conformed to the Hardy-Weinberg equilibrium (HWE). 11Evidence of the predominance of passive symplastic phloem loading and sugar transport with leaf ageing in Camellia oleifera显示文摘Phloem loading and transport of sugar from leaves to sink tissues such as fruits are crucial for yield formation.Camellia oleifera is an evergreen horticultural crop with high value;however,its low production limits the development of the C.oleifera industry.In this study,using a combination of ultrastructural observation,fluorescence loss in photobleaching(FLIP)and inhibitor treatment,we revealed that C.oleifera leaves mainly adopt a symplastic loading route from mesophyll cells to the surrounding vascular bundle cells in minor veins.HPLC assays showed that sucrose is the main sugar transported and only a small amount of raffinose or stachyose was detected in petioles,supporting a passive symplastic loading route in C.oleifera leaves.Compared to leaves grown this year(LT),the carbohydrate synthesis capacity in leaves grown last year(LL)was decreased while LL retained more soluble sugar,suggesting a decrease in transport capacity with leaf ageing.TEM and tissue staining showed that a reduction in plasmodesmata density leads to a decline in the degree of cellular coupling and is responsible for the weakening transport capacity in older leaves.RNA-seq revealed several differentially expressed genes(DEGs)including CoPDCB1-1,CoSUT1 and CoSWEET12,which are likely involved in the regulation of phloem loading and sugar transport.An expression correlation network is constructed between PD-callose binding protein genes,sugar transporter genes and senescence-associated genes.Collectively,this study provides the evidence of the passive symplastic phloem loading pathway in C.oleifera leaves and constructs the correlation between sugar transport and leaf ageing.Shiwen Yang Kehao Liang Yongjiang Sun Jinshun Zhang Yibo Cao Jing Zhou Aibin Wang Lingyun Zhang 2023Horticultural Plant Journal2023,9,4:0
12Comparative transcriptome profiling reveals the defense pathways and mechanisms in the leaves and roots of blueberry to drought stress显示文摘Blueberry is an important horticultural plant that is very susceptible to drought.However,the molecular regulation mechanisms of blueberry tolerance to drought remain elusive.In this study,the transcriptome profile of blueberry by RNA-seq under different degrees of drought treatment were conducted and drought-related genes and pathways were screened using weighted gene co-expression network analysis(WGCNA)and Venn analysis.Interestingly,the leaves and roots of blueberry exhibited different expression patterns under drought treatment and differentially expressed genes(DEGs),GO terms and KEGG pathways were more enriched in leaves than in roots.The majority of DEGs were associated with signal transduction,transcriptional regulation,and metabolism.Eight key pathways and eight DEGs were shared both in leaves and roots.Notably,the pathway with the most core genes in leaves is the plant hormone signal transduction pathway,and in roots are the MAPK signaling pathway,reactive oxygen species metabolism and the key genes such as VcXTH3,VcPP2C51,and VcPNC1 were identified.For transcription factors,VcABR1,VcABF2,VcMYB108 and VcMYB93 are likely involved in drought response.In the metabolism category,VcCYP75A1 was likely involved in anthocyanin biosynthesis,and VcPNC1 in the monoterpenoid biosynthesis pathway.The eight DEGs markedly induced under drought exhibited differential expression in leaves and roots.Correspondingly,physiological assays showed that POD activity,ABA content,and anthocyanin biosynthesis in leaves and roots were significantly increased.Collectively,our study revealed the synergistic and distinct defense pathways and mechanisms in leaves and roots of blueberry and explored the potential regulatory network in blueberry response to drought stress.Aibin Wang Lei Wang Kui Liu Kehao Liang Shiwen Yang Yibo Cao Lingyun Zhang 2022Fruit Research2022,2,1:0

14Different sites of extranodal involvement may affect the survival of patients with relapsed or refractory non-Hodgkin lymphoma after chimeric antigen receptor T cell therapy显示文摘Factors associated with complete and durable remissions after anti-CD19 chimeric antigen receptor T(CAR-T)cell immunotherapy for relapsed or refractory non-Hodgkin lymphoma(r/r NHL)have not been well characterized.In this study,we found that the different sites of extranodal involvement may affect response,overall survival(OS),and progression-free survival(PFS)in patients with r/r NHL treated with anti-CD19 CAR-T cells.In a cohort of 32 treated patients,12(37.5%)and 8(25%)patients exhibited soft tissue lymphoma and bone marrow(BM)infiltrations,respectively,and 13(41%)patients exhibited infiltration at other sites.The factors that may affect prognosis were identified through multivariable analysis.As an independent risk factor,soft tissue infiltration was the only factor significantly correlated with adverse prognosis(P<0.05),whereas other factors did not reach statistical significance.Furthermore,the site of extranodal tumor infiltration significantly and negatively affected OS and PFS in patients with r/r NHL treated with anti-CD19 CAR-T cell therapy.PFS and OS in patients with BM involvement were not significantly different from those of patients with lymph node involvement alone.Thus,anti-CD19 CAR-T cell therapy may improve the prognosis of patients with BM infiltration.Lili Zhou Ping Li Shiguang Ye Xiaochen Tang Junbang Wang Jie Liu Aibin Liang 2020Frontiers of Medicine2020,14,6:0
15Chimeric antigen receptor T-cell therapy: a promising treatment modality for relapsed/refractory mantle cell lymphoma显示文摘Mantle cell lymphoma(MCL)is a distinct histological type of B-cell lymphoma with a poor prognosis.Several agents,such as proteasome inhibitors,immunomodulatory drugs,and inhibitors of B cell lymphoma-2 and Bruton’s tyrosine kinase have shown efficacy for relapsed or refractory(r/r)MCL but often have short-term responses.Chimeric antigen receptor(CAR)T-cell therapy has emerged as a novel treatment modality for r/r non-Hodgkin’s lymphoma.However,long-term safety and tolerability associated with CAR T-cell therapy are not defined well,especially in MCL.In this report,we described a 70-year-old patient with r/r MCL with 48-month duration of follow-up who achieved long-term remission after CAR T-cell therapy.CAR T-cell-related toxicities were also mild and tolerated well even in this elderly patient.This report suggested that CAR T-cell therapy is a promising treatment modality for patients with MCL,who are generally elderly and have comorbid conditions.Ping Li Ningxin Dong Yu Zeng Jie Liu Xiaochen Tang Junbang Wang Wenjun Zhang Shiguang Ye Lili Zhou Alex Hongsheng Chang Aibin Liang 2020Frontiers of Medicine2020,14,6:0
