Mesenchymal stem cell treatment improves outcome of COVID-19 patients via multiple immunomodulatory mechanisms显示文摘The infusion of coronavirus disease 2019(COVID-19)patients with mesenchymal stem cells(MSCs)potentially improves clinical symptoms, but the underlying mechanism remains unclear. We conducted a randomized,single-blind,placebo-controlled(29 patients/group)phase II clinical trial to validate previous findings and explore the potential mechanisms.Patients treated with umbilical cord-derived MSCs exhibited a shorter hospital stay(P=0.0198)and less time required for symptoms remission(P=0.0194)than those who received placebo.Based on chest images, both severe and critical patients treated with MSCs showed improvement by day 7(P=0.0099)and day 21(P=0.0084).MSC-treated patients had fewer adverse events.MSC infusion reduced the levels of C-reactive protein, proinflammatory cytokines, and neutrophil extracellular traps(NETs)and promoted the maintenance of SARS-CoV-2-specific antibodies. To explore how MSCs modulate the immune system,we employed single-cell RNA sequencing analysis on peripheral blood. Our analysis identified a novel subpopulation of VNN2+hematopoietic stem/progenitorlike(HSPC-like)cells expressing CSF3R and PTPRE that were mobilized following MSC infusion.Genes encoding chemotaxis factors—CX3CR1 and L-selectin—were upregulated in various immune cells.MSC treatment also regulated B cell subsets and increased the expression of costimulatory CD28 in T cells in vivo and in vitro. In addition,an in vivo mouse study confirmed that MSCs suppressed NET release and reduced venous thrombosis by upregulating kindlin-3 signaling.Together, our results underscore the role of MSCs in improving COVID-19 patient outcomes via maintenance of immune homeostasis. | Rongjia Zhu Tingdong Yan Yingmei Feng Yan Liu Hongcui Cao Gongxin Peng Yanlei Yang Zhen Xu Jingqi Liu Wei Hou Xiaoyue Wang Zhe Li Luchan Deng Shihua Wang Jing Li Qin Han Hongling Li Guangliang Shan Yinghao Cao Xingyan An Jianshe Yan Zhonghui Zhang Huafei Li Xuebin Qu Jiaqi Zhu Shumin Zhou Jiao Wang Fengchun Zhang Jinming Gao Ronghua Jin Dayong Xu Yan-Qing Ma Tao Huang Shuang Peng Zhi Zheng Ilia Stambler Eric Gilson Lee Wei Lim Alexey Moskalev Antonio Cano Sasanka Chakrabarti Brun Ulfhake Huanxing Su Haoying Xu Sihuan Xu Feng Wei Holly MBrown-Borg Kyung-Jin Min Georgina Ellison-Hughes Calogero Caruso Kunlin Jin Robert Chunhua Zhao | 2021 | Cell Research2021,31,12: | 5

Effect of integrated Traditional Chinese and Western Medicine on Gout显示文摘OBJECTIVE:To evaluate the curative effect of integrated Traditional Chinese and Western Medicine on gout, and to investigate the therapy timing and exact treatment options of integrated medicine.METHODS:Totally 860 patients were enrolled, including 460 patients with intermittent gout,200 patients with active Traditional Chinese Medicine(TCM)syndrome(TCM syndrome score≥6)and200 patients with stable TCM syndrome(score<6).They were randomly divided into intervention and control groups. The control group was treated according to Western Medicine guidelines. The intervention group was treated with integrated Traditional Chinese and Western Medicine. The efficacy of TCM syndrome, joint pain score, joint swelling score,ESR,C-reactive protein, serum uric acid, liver and kidney function, and the duration of remission of TCM syndrome were compared between the two groups before and after treatments.RESULTS:For the patients with stable TCM syndrome, there was no significant difference in the effective rate and inefficiency between the intervention group and the control group. For the active type, the effective rate of the intervention group is better than the control group significantly. For the stable type, there was no significant difference between the intervention group and the control group in improving the scores of joint pain and swelling, reducing the level of ESR,C-reactive protein, serum uric acid and improving liver and kidney function. For the active type, the differences between the two groups were significant. The stable stage of gout in the intervention group was longer than the control group.CONCLUSION:For the gout patients with stable TCM syndrome in the acute stage of gout,we can use TCM treatment or Western Medicine alternatively;for the patients with active TCM syndrome, the scheme of combination of Traditional Chinese and Western Medicine can be applied, with the better curative effect than any medicine alone. | LIU Wei WU Yuanhao XUE Bin JIN Yue ZHANG Shumin LI Peihao XIE Qing WANG Aihua GAO Jingyue CAI Yue ZHANG Bo LIU Xiaoya WANG Yi DUAN Ran | 2021 | Journal of Traditional Chinese Medicine2021,41,5: | 3
Rapidly developable therapeutic-grade equine immunoglobulin against the SARS-CoV-2 infection in rhesus macaques显示文摘Dear Editor,The unprecedented COVID-19 pandemic caused by SARS-CoV-2 remains ongoing, but there is a lack of fully effective treatments.Convalescent plasma-derived hyperimmune globulins have been a safe and effective treatment but restricted by the difficulties in obtaining sufficient plasma with high antibody titers from a large number of recovered patients.Heterologous antibodies, particularly equine antibodies, have been widely used for decades as the therapeutics against some viral infections or as antivenoms.1 Equine antibodies could be rapidly developed and manufactured into therapeutic antibodies in large quantities under WHO standardized guidelines. Here we explored the development of equine antibody-derived F(ab′)2 as an option to treat COVID-19 by targeting the receptor-binding domain(RBD)of the viral spike protein that is essential for the viral entry into the host cells.2 We observed excellent neutralization titers of the F(ab′)2 in vitro and high potency against SARS-CoV-2 infection in the nonhuman primate rhesus macaques. | Xiaolei Liu Yi Liu Xuemin Jin Zhanlong He Zhen Huang Shumin Sun Yuwei Gao Jingyu Li Qin Ning Zhongping Xie Ningyi Jin Mingyuan Liu | 2022 | Signal Transduction and Targeted Therapy2022,7,8: | 2

Suggestion on further strengthening ultra-low emission standards for PM emission from coal-fired power plants in China显示文摘China has established the largest clean coal-fired power generation system in the world by accomplishing the technological transformation of coal-fired power plants(CFPPs)to achieve ultra-low emission. The potential for further particulate matter(PM)emission reduction to achieve near-zero emission for CFPPs has become a hotspot issue. In this study,PM emission from some ultra-low emission CFPPs adopting advanced air pollutant control technologies in China was reviewed. The results revealed that the average filterable particulate matter(FPM)concentration, measured as the total particulate matter(TPM)according to the current national monitoring standard,was(1.67±0.86)mg/m^(3),which could fully achieve the ultra-low emission standard for key regions(5 mg/m^(3)),but that achieving the near-zero emission standard was difficult(1 mg/m^(3)).However,the condensable particulate matter(CPM),with an average concentration of(1.06±1.28)mg/m^(3),was generally ignored during monitoring, which led to about 38.7%underestimation of the TPM.Even considering both FPM and CPM,the TPM emission from current CFPPs would contribute to less than 5%of atmospheric PM_(2.5) concentrations in the key cities and regions in China. Therefore, further reduction in FPM emission proposed by the near-zero emission plan of CFPPs may have less environmental benefit than emission control of other anthropogenic sources. However,it is suggested that the management of CPM emission should be strengthened, and a national standard for CPM emission monitoring based on the indirect dilution method should be established for CFPPs.Those measurements are helpful for optimal operation of air pollutant control devices and continuously promoting further emission reduction. | Jianguo Deng Shumin Wang Jiawei Zhang Yi Zhang Jingkun Jiang Yongzheng Gu Tao Han Lei Feng Jian Gao Lei Duan | 2023 | Journal of Environmental Sciences2023,,1: | 1
The Influence of Typhoon'Hongxia'on the Intrusion of the Kuroshio Current into the South China Sea显示文摘This paper uses the Coupled Ocean-Atmosphere-Wave-Sediment Transport(COAWST)model to analyze the impact of typhoon‘Hongxia’on the velocity and position movement of the Kuroshio axis, the impact of typhoons on the Kuroshio intrusion into South China Sea(SCS),the corresponding water, heat, and salt fluxes, and the impact of Kuroshio water in the northeastern SCS.When typhoon‘Hongxia’passed, the Kuroshio intrusion into the SCS was the most significant at 21?N latitude. In the vertical direction, the Kuroshio intrusion was strongest in the subsurface layer, leading to the most significant changes in temperature and salinity in the northeastern part of the SCS in the subsurface layer.Under the influence of the southeastern monsoon in summer,a large amount of low-salinity water accumulates at the surface of the northeastern part of the SCS,and Kuroshio intrusive water remains in the bottom and middle portions of the subsurface layer. The westward deviation of the Kuroshio axis caused by the typhoon displays a certain lag compared with the hot and salty water intrusion into the SCS approximately 7 d later. The impact of the typhoon on the Kuroshio intrusion into the SCS lasts for 20 d.The typhoon caused increases in the water, heat, and salt fluxes associated with the Kuroshio intrusion into the SCS,and the contribution of the typhoon to these fluxes was as high as 40%.Under typhoon conditions, the maximum Kuroshio intrusion flux reached more than twice that before the typhoon. | GAO Shumin HAN Shuzong WANG Shicheng WU Dexing WANG Mingjie WU Kejian LIU Lili | 2023 | Journal of Ocean University of China2023,22,2: | 1

Quick photofabrication of functional nanospheres from de novo designed peptides for NIR fluorescence and MR imaging显示文摘Combining the noncovalent and covalent interactions,a series of peptide amphiphiles were designed de novo and synthesized to architect functional assemblies by means of photochemistry. The strand of peptide sequence was structurally capped with photoactive tyrosine-tyrosine(YY)motifs at both termini, and the spacing was filled by alternating of hydrophilic D(L-aspartate)and hydrophobic X(ε-aminocaproic acid)structure.Upon visible-light irradiation, these de novo designed peptides underwent rapid photocrosslinking within merely 10 min.Interestingly, the modulation of alternating D-X pairs in occupying spacer would adjust molecular amphiphilicity, regulate charge distribution, and control particle size and loading capacity of peptide nanospheres(PNS)in aqueous media. With entirely peptide-based matrix, this PNS system could host cationic indicators of fluorescent rhodamine and magnetic GdIII for exemplar near infrared(NIR)fluorescence and magnetic resonance(MR)imaging, which paves a pathway to biomaterial and biomedical applications using de novo designed peptides. | Jingyi Zhao Chen Li Xue-Wang Gao Ke Feng Hao Liu Sijie He Wenhua Zhao Shumin Yang Jianqun Shao Ling Ye Bin Chen Nan Xie Chen-Ho Tung Li-Zhu Wu | 2023 | Nano Research2023,16,3: | 0
Prediction of Newhall navel orange internal quality based on digital microscopy显示文摘The traditional technology for internal quality detection of citrus fruit is believed to be destructive, inefficient, and error-prone.To address these issues,a novel method has been developed to evaluate nondestructively the sugar-acid ratio(SAR)of soluble solids by using the oil glands density(OGD).In this study,a total of 584 samples with different widths were collected. The sample data were correlated that the SAR and OGD decreased with time during storage. The relevance with time was significant between SAR and OGD of citrus positively correlated in the calibration group(R2=0.82),which indicated that OGD could be used to predict the internal quality of citrus. It indicated that a new generation of digital microscopy could provide an alternative to predict nondestructively the internal quality of citrus fruit,as well as some theoretical insight for further studies of online quality detection in the future. | Shumin Gao Cong Cao Dingyi Hu Rangwei Xu Yunjiang Cheng Hong Chen Ming Zhu Shanjun Li | 2021 | International Journal of Agricultural and Biological Engineering2021,14,6: | 0
FTO is a transcriptional repressor to auto-regulate its own gene and potentially associated with homeostasis of body weight显示文摘Fat mass and obesity-associated (FTO) protein is a ferrous ion (Fe^2+)/2-oxoglutarate (2-OG)-dependent demethylase preferentially catalyzing m^6A sites in RNA. The FTO gene is highly expressed in the hypothalamus with fluctuation in response to various nutritional conditions, which is believed to be involved in the control of whole body metabolism. However, the underlying mechanism in response to different nutritional cues remains poorly understood. Here we show that ketogenic diet-derived ketone body β-hydroxybutyrate (BHB) transiently increases FTO expression in both mouse hypothalamus and cultured cells. Interestingly, the FTO protein represses Fto promoter activity, which can be offset by BHB. We then demonstrate that FTO binds to its own gene promoter, and Fe^2+, but not 2-OG, impedes this binding and increases FTO expression. The BHB-induced occupancy of the promoter by FTO influences the assembly of the basal transcriptional machinery. Importantly, a loss-of-function FTO mutant (I367F), which induces a lean phenotype in FTO^I367F mice, exhibits augmented binding and elevated potency to repress the promoter. Furthermore, FTO fails to bind to its own promoter that promotes FTO expression in the hypothalamus of high-fat diet-induced obese and 48-h fasting mice, suggesting a disruption of the stable expression of this gene. Taken together, this study uncovers a new function of FTO as a Fe^(2+)-sensitive transcriptional repressor dictating its own gene switch to form an auto-regulatory loop that may link with the hypothalamic control of body weight. | Shu-Jing Liu Hui-Ling Tang Qian He Ping Lu Tao Fu Xu-Ling Xu Tao Su Mei-Mei Gao Shumin Duan Yan Luo Yue-Sheng Long | 2019 | Journal of Molecular Cell Biology2019,11,2: | 0
cFos-ANAB:A cFos-based Web Tool for Exploring Activated Neurons and Associated Behaviors显示文摘cFos is one of the most widely-studied genes in the field of neuroscience.Currently, there is no systematic database focusing on cFos in neuroscience. We developed a curated database-cFos-ANAB-a cFos-based web tool for exploring activated neurons and associated behaviors in rats and mice, comprising 398 brain nuclei and sub-nuclei,and five associated behaviors:pain, fear, feeding, aggression, and sexual behavior.Direct relationships among behaviors and nuclei(even cell types)under specific stimulating conditions were constructed based on cFos expression profiles extracted from original publications. Moreover, overlapping nuclei and sub-nuclei with potentially complex functions among different associated behaviors were emphasized, leading to results serving as important clues to the development of valid hypotheses for exploring as yet unknown circuits.Using the analysis function of cFos-ANAB,multi-layered pictures of networks and their relationships can quickly be explored depending on users’purposes. These features provide a useful tool and good reference for early exploration in neuroscience. The cFos-ANAB database is available at www.cfos-db.net. | Fan Wang Wenjie Sun Lei Chang Kefang Sun Leying Hou Linna Qian Chaoyin Jin Jiandong Chen Jiali Pu Panmeng Ye Shuang Qiu Jianhong Luo Shumin Duan Baorong Zhang Zhihua Gao Xiaojun Hu | 2021 | Neuroscience Bulletin2021,37,10: | 0
Adult tiger beetles Cicindela gemmata modify their foraging strategy in different hunting contexts显示文摘Predator–prey interaction has long been an interesting item in the research of animal behaviors.Given that live prey can damage their predators, predators must trade foraging efficiency for safety while hunting, but the extent of this trade-off is not yet clear.Tiger beetles display diversity in their diets and hunting strategies, and hence, they become an ideal system to address how self-security affects foraging efficiency. We addressed this question in captive adult tiger beetles Cicindela gemmata.By offering several types of arthropod and plant foods,we confirmed that C.gemmata is carnivorous. We found that C.gemmata hunt by either ambushing or chasing their prey, and that they switch between strategies based on differences in the number of prey, the prey status and encounter rate, and the number of predators.Ambushing success increased with the number of prey but decreased with prey encounter rate.Chasing success decreased as prey body size and encounter rate increased.Foraging Cicindela gemmata often gave up an attack when it was nonfatal. This active giving up of hunting may be a consequence of a trade-off between foraging efficiency and self-security.Therefore,it is an adaptive response to the risk of injury when hunting for larger live prey. | Shumin Wang Panshuai Fei Zifeng Wu Zeyi Luo Yichen Wu Ningning Sun Yujie Wang Shumei Zi Lifang Gao Bo Du | 2023 | Insect Science2023,30,6: | 0
Engineering Janus gold nanorod–titania heterostructures with enhanced photocatalytic antibacterial activity against multidrugresistant bacterial infection显示文摘Photocatalytic antibacterial approach shows great potential in treating multidrug-resistant bacterial infections. However, the bactericidal efficiency heavily depends on the photocatalytic activity of semiconductor materials, which is limited by the fast recombination of photogenerated electron–hole pairs.Janus nano-heterostructures with spatial control growth of TiO_(2)nanoparticles(NPs)at one end of gold nanorods(Au NRs)are designed via surface ligand regulation for photocatalytic sterilization and infected wound healing. The asymmetric nanostructure of Janus gold nanorod-titanium dioxide nanoparticles(Janus AuNR-TiO_(2) NPs)promotes the directional migration of charge carriers and is more conducive to the spatial separation of electron–hole pairs. Moreover, the injection of hot electrons and enhancement of plasmon near-fields from the surface plasmon resonance(SPR)effect further improve the photocatalytic efficiency of Janus AuNR-TiO_(2) NPs.Under simulated sunlight irradiation, large amounts of reactive oxygen species(ROS)are generated for photocatalytic antibacterial activity.Enhanced bactericidal efficiency up to 99.99%against methicillin-resistant Staphylococcus aureus(MRSA)is achieved in vitro.Furthermore,Janus AuNR-TiO_(2) NPs exhibit superior biocompatibility, structural stability, and also remarkably accelerate MRSA-infected wound healing.Taking the above all into consideration,Janus AuNR-TiO_(2) NPs,as an efficient antibacterial photocatalyst, offers a promising strategy for MRSA infectious therapy. | Shuqin Li Hongqi Huo Xing Gao Luntao Liu Shumin Wang Jiamin Ye Jing Mu Jibin Song | 2023 | Nano Research2023,16,2: | 0

| 19 | Sentinel Lymph Node Identification in Endometrial Cancer显示文摘OBJECTIVE To evaluate the feasibility of intra-operativedetection of sentinel lymph nodes (SLN) in the patient withendometrial cancer (EC).METHODS Thirty-one patients with Stage Ⅰ and Ⅱ endometrialcancer, who underwent a hysterectomy and a lymphadenectomy, were enrolled in the study. At laparotomy, methylene blue dyetracer was injected into the subserosal myometrium of corpusuteri at multiple sites, and dye uptake into the lymphatic channelswas observed. The blue nodes which were identified as SLNs weretraced and excised. The other nodes were then removed. All of theexcised nodes were submitted for pathological hematoxylin andeosin (H&E) staining examination.RESULTS Failure of dye uptake occurred in 4 of the 31 cases(12.9%) because of spillage, and no lymphatic coloration wasobserved there. Lymphatic staining was clearly observable as bluedye diffused to the lymphatic channels of the uterine surface andthe infundibulopelvic ligaments in 27 (87.1%) cases. Concurrentcoloration in the pelvic lymphatic vessels was also observed in 22of the 27 patients. The SLNs were identified in 23 of the 27 (85.2%)cases with a lymphatic staining, with a total number of 90 SLNs,and a mean of 3.9 in each case (range, 1-10). Besides one SLN (1.1%)in the para-aortic area, the other 89 (98.9%) were in the nodes ofthe pelvis. The most dense locations of SLNs included obturator in38 (42.2%) and interiliac in 19 (21.1%) cases. In our group, pelviclymphadenectomy was conducted in 27 (87.1%) patients andpelvic nodal sampling in 4 (12.9%). Of the 31 cases, a concurrentabdominal para-aortic lymph node sampling was conducted in7. A total of 926 nodes were harvested, with an average of 39.8 ineach case (range, 14-55). Nodal metastases occurred in 3 patients(9.7%), 2 of them with SLN involvement and the other withoutSLN involvement. Adverse reactions or injury related to the studywas not found.CONCLUSION Application of methylene blue dye is feasible inan intra-operative SLN identification of endometrial cancer.
