1Matrine attenuates oxidative stress and cardiomyocyte apoptosis in doxorubicin-induced cardiotoxicity via maintaining AMPKα/UCP2 pathway显示文摘Oxidative stress and cardiomyocyte apoptosis are involved in the pathogenesis of doxorubicin(DOX)-induced cardiotoxicity. Matrine is well-known for its powerful anti-oxidant and anti-apoptotic capacities. Our present study aimed to investigate the effect of matrine on DOX-induced cardiotoxicity and try to unearth the underlying mechanisms. Mice were exposed with DOX to generate DOX-induced cardiotoxicity or normal saline as control. H9 C2 cells were used to verify the effect of matrine in vitro.DOX injection triggered increased generation of reactive oxygen species(ROS) and excessive cardiomyocyte apoptosis, which were significantly mitigated by matrine. Mechanistically, we found that matrine ameliorated DOX-induced uncoupling protein 2(UCP2) downregulation, and UCP2 inhibition bygenipin could blunt the protective effect of matrine on DOX-induced oxidative stress and cardiomyocyte apoptosis. Besides, 50-AMP-activated protein kinase α2(Ampkα2) deficiency inhibited matrine-mediated UCP2 preservation and abolished the beneficial effect of matrine in mice. Besides, we observed that matrine incubation alleviated DOX-induced H9 C2 cells apoptosis and oxidative stress level via activating AMPKα/UCP2, which were blunted by either AMPKα or UCP2 inhibition with genetic or pharmacological methods. 3Polysaccharide extracts of Astragalus membranaceus and Atractylodes macrocephala promote intestinal epithelial cell migration by activating the polyamine-mediated K^+ channel显示文摘Astragalus membranaceus(Radix Astragali, RA) and Atractylodes macrocephala(Rhizoma Atractylodis Macrocephalae, RAM) are often used to treat gastrointestinal diseases. In the present study, we determined the effects of polysaccharides extracts from these two herbs on IEC-6 cell migration and explored the potential underlying mechanisms. A migration model with IEC-6 cells was induced using a single-edged razor blade along the diameter of cell layers in six-well polystyrene plates. The cells were grown in control--media or media containing spermidine(5 μmol·L^(-1), SPD), alpha-difluoromethylornithine(2.5 mmol·L^(-1), DFMO), 4-Aminopyridine-(40 μ-mol·L^(-1), 4-AP), the polysaccharide extracts of RA or RAM(50, 100, or 200 mg·L^(-1)), DFMO plus SPD, or DFMO plus polysaccharide extracts of RA or RAM for 12 or 24 h. Next, cytosolic free Ca^(2+)([Ca^(2+)]cyt) was measured using laser confocal microscopy, and cellular polyamine content was quantified with HPLC. Kv1.1 mRNA expression was assessed using RT-qPCR and Kv1.1 and RhoA protein expressions were measured with Western blotting analysis. A cell migration assay was carried out using Image-Pro Plus software. In addition, GC-MS was introduced to analyze the monosaccharide composition of both polysaccharide extracts. The resutls showed that treatment with polysaccharide extracts of RA or RAM significantly increased cellular polyamine content, elevated [Ca^(2+)]cyt and accelerated migration of IEC-6 cells, compared with the controls(P < 0.01). Polysaccharide extracts not only reversed the inhibitory effects of DFMO on cellular polyamine content and [Ca^(2+)]cyt, but also restored IEC-6 cell migration to control level(P < 0.01 or < 0.05). Kv1.1 mRNA and protein expressions were increased(P < 0.05) after polysaccharide extract treatment in polyamine-deficient IEC-6 cells and RhoA protein expression was increased. Molar ratios of D-ribose, D-arabinose, L-rhamnose, D-mannose, D-glucose, and D-galactose was 1.0:14.1:0.3:19.9:181.3:6.3 in RA and 1.0:4.3:0.1:5.7:2.8:2.2 in RAM. 5Dynamic changes of behaviors, dentate gyrus neurogenesis and hippocampal miR-124 expression in rats with depression induced by chronic unpredictable mild stress显示文摘The depression-like behavior phenotype,neurogenesis in the dentate gyrus and miR-124 expression in the hippocampus are the focus of current research on the pathogenesis of depression and antidepressant therapy.The present study aimed to clarify the dynamic changes of depression-like behavior,dentate gyrus neurogenesis and hippocampal miR-124 expression during depression induced by chronic stress to reveal pathological features at different stages of depression and to further provide insight into depression treatment.Chronic unpredictable mild stress depression models were established by exposing Sprague-Dawley rats to various mild stressors,including white noise,thermal swimming,stroboscopic illumination,soiled cages,pairing with three other stressed animals,cold swimming,tail pinch,restraint and water and food deprivation.Chronic unpredictable mild stress model rats underwent dynamic observation from 1 to 8 weeks and were compared with a control group(normal feeding without any stressors).To observe changes in the depression-like behavior phenotype during chronic unpredictable mild stress-induced depression,a sucrose preference test was used to evaluate the degree of anhedonia.An open-field test was used to evaluate locomotor activity and anxiety status.Compared with the control group,chronic unpredictable mild stress rats lost weight but did not have a depression-like behavioral phenotype at 1-4 weeks.Chronic unpredictable mild stress rats presented decreased sucrose preference and locomotor activity at 5-8 weeks.In addition,chronic unpredictable mild stress rats did not have significant anxiety-like behavior during 1-8 weeks of modeling.To observe neurogenesis dysfunctions and changes in neuronal number in the dentate gyrus during chronic unpredictable mild stress-induced depression,markers(DCX and DCX/BrdU)of neural proliferation and differentiation and the neuronal marker NeuN were assessed by immunofluorescence.Compared with the control group,neurogenesis and the neuronal number in the dentate gyrus did not change from 2 to 6 weeks;however,neural proliferation and differentiation in the dentate gyrus decreased,and the number of neurons decreased until the eighth week in the chronic unpredictable mild stress group.Real-time quantitative reverse transcription polymerase chain reaction assays and fluorescence in situ hybridization were used to measure the expression of hippocampal miR-124 during chronic unpredictable mild stress-induced depression.The results showed that the expression of hippocampal miR-124 was unchanged during the first 4 weeks but increased from 5 to 6 weeks and decreased from 7 to 8 weeks compared with the control group.These findings indicate that during chronic unpredictable mild stress-induced depression,the behavioral phenotype,miR-124 expression in the hippocampus,neurogenesis in the dentate gyrus and neuronal numbers showed dynamic changes,which suggested that various pathological changes occur at different stages of depression.All experimental procedures and protocols were approved by the Experimental Animal Ethics Committee of Guangzhou University of Chinese Medicine of China in March 2015.Yun-Ling Huang Ning-Xi Zeng Jie Chen Jie Niu Wu-Long Luo Ping Liu Can Yan Li-Li Wu 2020Neural Regeneration Research2020,15,6:10
6Chemical characteristics of haze particles in Xi'an during Chinese Spring Festival: Impact of fireworks burning显示文摘Fireworks burning releases massive fine particles and gaseous pollutants, significantly deteriorating air quality during Chinese Lunar New Year(LNY) period. To investigate the impact of the fireworks burning on the atmospheric aerosol chemistry, 1-hr time resolution of PM_(2.5) samples in Xi'an during the winter of 2016 including the LNY were collected and detected for inorganic ions, acidity and liquid water content(LWC) of the fine aerosols. PM_(2.5) during the LNY was 167 ± 87 μg/m^3, two times higher than the China National Ambient Air Quality Standard(75 μg/m^3). K^+(28 wt.% of the total ion mass) was the most abundant ion in the LNY period, followed by SO_4^(2-)(25 wt.%) and Cl-(18 wt.%). In contrast, NO_3^-(34 wt.%) was the most abundant species in the haze periods(hourly PM32-2.5> 75 μg/m), followed by SO_4(29.2 wt.%) and NH_4^+(16.3 wt.%), while SO_4^(2-)(35 wt.%) was the most abundant species in the clean periods(hourly PM_(2.5)< 75 μg/m^3), followed by NO_3^-(23.1 wt.%) and NH_4^+(11 wt.%). Being different from the acidic nature in the non-LNY periods, aerosol in the LNY period presented an alkaline nature with a pH value of 7.8 ± 1.3. LWC during the LNY period showed a robust linear correlation with K_2SO_4 and KCl, suggesting that aerosol hygroscopicity was dominated by inorganic salts derived from fireworks burning. 7Monthly variation in litterfall and the amount of nutrients in an Aleurites montana plantation显示文摘In this study,the dynamics of monthly variation in litterfall and the amount of nutrients,i.e.,organic C,N,P and K,in an Aleurites montana plantation were analyzed,based on a field study and experiments over one year.The results show that the litterfall mass of A.montana collected generally presents an ascending trend with maximum defoliation occurring in the autumn and winter (October-December),accounting for 75.67% of the total amount of annual litterfall.The sequence in the amount of nutrients in A.montana litter was as follows:organic C > N > K > P;their monthly amounts show various dynamic curves.Similar to the dynamics of the mass of monthly litterfall,the monthly returns of C,N,P and K generally show an ascending trend with their peak values all occurring in December.The mass of A.montana litterfall and the dynamics of its monthly nutrient return provide,to a certain degree,a scientific reference for planting and fertilizing A.montana.LIN Han HONG Tao WU Cheng-zhen CHEN Hui CHEN Can LI Jian LIN Yong-ming FAN Hai-lan 2012Forestry Studies in China2012,14,1:10
8Exogenous pancreatic kininogenase protects against renal fibrosis in rat model of unilateral ureteral obstruction显示文摘Tissue kallikrein has protective function against various types of injury.In this study,we investigated whether exogenous pancreatic kininogenase(PK)conferred renoprotection in a rat model of unilateral ureteral obstruction(UUO)and H2O2-treated HK-2 cells in vitro.SD rats were subjected to UUO surgery,then PK(7.2 U/g per day,ip)was administered for 7 or 14 days.After the treatment,rats were euthanized;the obstructed kidneys were harvested for further examination.We found that PK administration significantly attenuated interstitial inflammation and fibrosis,and downregulated the expression of proinflammatory(MCP-1,TLR-2,and OPN)and profibrotic(TGF-β1 and CTGF)cytokines in obstructed kidney.UUO-induced oxidative stress,closely associated with excessive apoptotic cell death and autophagy via PI3K/AKT/FoxO1a signaling,which were abolished by PK administration.We further showed that PK administration increased the expression of bradykinin receptors 1 and 2(B1R and B2R)mRNA and the production of NO and cAMP in kidney tissues.Coadministration with either B1R antagonist(des-Arg9-[Leu8]-bradykinin)or B2R antagonist(icatibant)abrogated the renoprotective effects of PK,and reduced the levels of NO and cAMP in obstructed kidney.In H2O2-treated HK-2 cells,addition of PK(6 pg/mL)significantly decreased ROS production,regulated the expression of oxidant and antioxidant enzymes,suppressed the expression of TGF-β1 and MCP-1,and inhibited cell apoptosis.Our data demonstrate that PK treatment protects against the progression of renal fibrosis in obstructed kidneys.Li-zhe Jin Hui-ying Li Jian Jin Shang-guo Piao Xiong-hu Shen Yan-ling Wu Jia-chong Xu Long-ye Zhang Yu-ji Jiang Hai-lan Zheng Ying-shun Jin Sheng Cui Kang Luo Yi Quan Can Li 2020Acta Pharmacologica Sinica2020,41,12:9
9Matrine attenuates pathological cardiac fibrosis via RPS5/p38 in mice显示文摘Pathological cardiac fibrosis is a common feature in multiple cardiovascular diseases that contributes to the occurrence of heart failure and life-threatening arrhythmias.Our previous study demonstrated that matrine could attenuate doxorubicin-induced oxidative stress and cardiomyocyte apoptosis.In this study,we investigated the effect of matrine on cardiac fibrosis.Mice received aortic banding(AB)operation or continuous injection of isoprenaline(ISO)to generate pathological cardiac fibrosis and then were exposed to matrine lavage(200 mg·kg^(−1)·d^(−1))or an equal volume of vehicle as the control.We found that matrine lavage significantly attenuated AB or ISO-induced fibrotic remodeling and cardiac dysfunction.We also showed that matrine(200μmol/L)significantly inhibited the proliferation,migration,collagen production,and phenotypic transdifferentiation of cardiac fibroblasts.Mechanistically,matrine suppressed p38 activation in vivo and in vitro,and overexpression of constitutively active p38 completely abolished the protective effects of matrine.We also demonstrated that ribosomal protein S5(RPS5)upregulation was responsible for matrine-mediated inhibition on p38 and fibrogenesis.More importantly,matrine was capable of ameliorating preexisting cardiac fibrosis in mice.In conclusion,matrine treatment attenuates cardiac fibrosis by regulating RPS5/p38 signaling in mice,and it might be a promising therapeutic agent for treating pathological cardiac fibrosis.Xin Zhang Can Hu Ning Zhang Wen-ying Wei Ling-li Li Hai-ming Wu Zhen-guo Ma Qi-zhu Tang 2021Acta Pharmacologica Sinica2021,42,4:8

11Jiaweisinisan facilitates neurogenesis in the hippocampus after stress damage显示文摘The traditional Chinese medicine Jiaweisinisan has antidepressant effects, and can inhibit hypothalamus-pituitary-adrenal gland axis hyperactivity in stress-induced depression. In this study, rat hippocampal neural precursor cells were cultured in serum-free medium in vitro and a stress damage model was established with 120 μM corticosterone. Cells were treated with 10% (v/v) Jiaweisinisan drug-containing serum and the corticosterone antagonist RU38486. Results of the 3-(4,5-dimethylthiazol-2-yl)-3,5-di-phenytetrazoliumromide assay showed that both Jiaweisinisan drug-containing serum and RU38486 promoted the proliferation of neural precursor cells after corticosterone exposure. Immunofluorescence detection showed that after Jiaweisinisan drug-containing serum and RU38486 treatment, the 5-bromo-2-deoxyuridine/terminal deoxynucleotidyl transferase dUTP nick end labeling ratio in hippocampal neural precursor cells significantly increased, and the apoptotic rates of glial cells reduced, and neuron-like cell differentiation from neural precursor cells significantly increased. 12Distributed Brillouin frequency shift extraction via a convolutional neural network显示文摘Distributed optical fiber Brillouin sensors detect the temperature and strain along a fiber according to the local Brillouin frequency shift(BFS),which is usually calculated by the measured Brillouin spectrum using Lorentzian curve fitting.In addition,cross-correlation,principal component analysis,and machine learning methods have been proposed for the more efficient extraction of BFS.However,existing methods only process the Brillouin spectrum individually,ignoring the correlation in the time domain,indicating that there is still room for improvement.Here,we propose and experimentally demonstrate a BFS extraction convolutional neural network(BFSCNN)to retrieve the distributed BFS directly from the measured two-dimensional data.Simulated ideal Brillouin spectra with various parameters are used to train the BFSCNN.Both the simulation and experimental results show that the extraction accuracy of the BFSCNN is better than that of the traditional curve fitting algorithm with a much shorter processing time.The BFSCNN has good universality and robustness and can effectively improve the performances of existing Brillouin sensors.YIQING CHANG HAO WU CAN ZHAO LI SHEN SONGNIAN FU MING TANG 2020Photonics Research2020,8,5:6

14Hic74,a novel alanine and glycine rich matrix protein related to nacreous layer formation in the mollusc Hyriopsis cumingii显示文摘Organic matrix proteins play an important role in the biomineralization of mollusc shells.We have identified and characterized a novel matrix protein,namely hic74,isolated from the mantle of the freshwater mussel Hyriopsis cumingii.The hic74 gene encodes a 850 amino acid protein that is rich in alanine(ala,30.8%),glycine(gly,25.8%)and serine(ser,10.6%),with ala mainly existing in poly-ala forms within the ala/gly-rich regions.Quantitative PCR expression analysis demonstrated that hic74 was specifically amplified from the mantle and in situ hybridization showed a strong signal in the epithelial cells in the pallial region of the mantle.Lower levels of expression were detected during the early stages of pearl sac formation but increased and remained constant during nacreous layer formation suggesting that hic74 might be involved in this process.Xiaojun Liu Can Jin Leiming Wu Shaojian Dong Shimei Zeng Jiale Li 2017Aquaculture and Fisheries2017,2,3:6
15Epigallocatechin-3-gallate induces apoptosis, inhibits proliferation and decreases invasion of glioma cell显示文摘Epigallocatechin-3-gallate (EGCG), a major polyphenol in green tea, has been considered a potential therapeutic and chemopreventive agent for cancer. Glioma is a malignant tumor with high mortality but effective therapy has not yet been developed. In this study, we found that EGCG induced apoptosis in U251 glioma cells via the laminin receptor (molecular weight 67kDa) in a time- and dose-dependent manner, decreased their invasiveness and inhibited their proliferation. The mitogen-activated protein kinase pathway was shown to be involved in glioma cell apoptosis and proliferation. Furthermore, the mRNA levels of matrix metalloproteinase (MMP)-2 and MMP-9 were reduced after EGCG treatment. 16Pulmonary Toxicity in Rats Caused by Exposure to Intratracheal Instillation of SiO2 Nanoparticles显示文摘Objective The effect of the silica nanoparticles(SNs) on lungs injury in rats was investigated to evaluate the toxicity and possible mechanisms for SNs.Methods Male Wistar rats were instilled intratracheally with 1 mL of saline containing 6.25,12.5,and 25.0 mg of SNs or 25.0 mg of microscale SiO_2 particles suspensions for 30 d,were then sacrificed.Histopathological and ultrastructural change in lungs,and chemical components in the urine excretions were investigated by light microscope,TEM and EDS.MDA,NO and hydroxyproline(Hyp) in lung homogenates were quantified by spectrophotometry.Contents of TNF-α,TGF-β1,IL-1β,and MMP-2 in lung tissue were determined by immunohistochemistry staining.Results There is massive excretion of Si substance in urine.The SNs lead pulmonary lesions of rise in lung/body coefficients,lung inflammation,damaged alveoli,granuloma nodules formation,and collagen metabolized perturbation,and lung tissue damage is milder than those of microscale SiO_2 particles.The SNs also cause increase lipid peroxidation and high expression of cytokines.Conclusion The SNs result into pulmonary fibrosis by means of increase lipid peroxidation and high expression of cytokines.Milder effect of the SNs on pulmonary fibrosis comparing to microscale SiO_2 particles is contributed to its elimination from urine due to their ultrafine particle size.YANG Hong WU Qiu Yun LI Ming Yue LAO Can Shah ZHANG Ying Jian 2017Biomedical and Environmental Sciences2017,30,4:5

19Exploration of the mechanism by which icariin modulates hippocampal neurogenesis in a rat model of depression显示文摘Icariin(ICA) has a significant capacity to protect against depression and hippocampal injury,but it cannot effectively cross the bloodbrain barrier and accumulate in the brain.Therefore,the mechanism by which ICA protects against hippocampal injury in depression remains unclear.In this study,we performed proteomics analysis of cerebrospinal fluid to investigate the mechanism by which ICA prevents dysfunctional hippocampal neurogenesis in depression.A rat model of depression was established through exposure to chronic unpredictable mild stress for 6 weeks,after which 120 mg/kg ICA was administered subcutaneously every day.The results showed that ICA alleviated depressive symptoms,learning and memory dysfunction,dysfunctional neurogenesis,and neuronal loss in the dentate gyrus of rats with depression.Neural stem cells from rat embryonic hippocampi were cultured in media containing 20% cerebrospinal fluid from each group of rats and then treated with 100 μM corticosterone.The addition of cerebrospinal fluid from rats treated with ICA largely prevented the corticosterone-mediated inhibition of neuronal proliferation and differentiation.Fifty-two differentially expressed proteins regulated by chronic unpredictable mild stress and ICA were identified through proteomics analysis of cerebrospinal fluid.These proteins were mainly involved in the ribosome,PI3 K-Akt signaling,and interleukin-17 signaling pathways.Parallel reaction monitoring mass spectrometry showed that Rps4 x,Rps12,Rps14,Rps19,Hsp90 b1,and Hsp90 aa1 were up-regulated by chronic unpredictable mild stress and down-regulated by ICA.In contrast,Htr A1 was down-regulated by chronic unpredictable mild stress and up-regulated by ICA.These findings suggest that ICA can prevent depression and dysfunctional hippocampal neurogenesis through regulating the expression of certain proteins found in the cerebrospinal fluid.The study was approved by the Experimental Animal Ethics Committee of Guangzhou University of Chinese Medicine of China in March 2017.Ning-Xi Zeng Hui-Zhen Li Han-Zhang Wang Kai-Ge Liu Xia-Yu Gong Wu-Long Luo Can Yan Li-Li Wu 2022Neural Regeneration Research2022,17,3:4
20Mullite Oxidation Resistant Coating for SiC-coated Carbon/Carbon Composites by Supersonic Plasma Spraying显示文摘To improve the oxidation resistance of carbon/carbon (C/C) composites, mullite coating was prepared on the surface of SiC-coated C/C composites by supersonic plasma spraying. Phases and microstructures of mullite coating were characterized by X-ray diffraction (XRD) and scanning electron microscopy (SEM). The coating primarily consists of a single phase of mullite (3Al2O3-2SiO2). The SEM results show that mullite coating was continuous and well bonded with the SiC inner layer without penetrating crack. Mullite coating exhibited good oxidation resistance.
