| 5 | Adsorption of Phosphate onto Ferrihydrite and Ferrihydrite-Humic Acid Complexes显示文摘The adsorption of phosphate onto ferrihydrite(FH) and two FH-humic acid(HA) complexes, obtained by co-precipitating FH with low(FH-HA1) and relatively high amounts of humic acid(FH-HA2), was studied through kinetics and isotherm experiments to determine the differences in phosphate adsorption between FH-HA complexes and FH and to reveal the mechanisms of phosphate adsorption onto two soil compositions. The isoelectric point(IEP) and the specific surface area(SSA) of the mineral decreased as the particle porosity of the mineral increased, which corresponded to an increase in the amount of organic carbon. The adsorption capacity of phosphate was higher on FH than on FH-HA1 and FH-HA2 at the scale of micromoles per kilogram. The initial adsorption rate and adsorption affinity of phosphate decreased with an increase in the amount of HA in the mineral. The sensitivity of phosphate adsorption to the change in the pH was greater for FH than for FH-HA complexes. | 6 | Cordycepin inhibits pancreatic cancer cell growth in vitro and in vivo via targeting FGFR2 and blocking ERK signaling显示文摘Cordycepin(3′-deoxyadenosine) from Cordyceps militaris has been reported to have anti-tumor effects. However, the molecular target and mechanism underlying cordycepin impeding pancreatic cancer cell growth in vitro and in vivo remain vague. In this study, we reported functional target molecule of cordycepin which inhibited pancreatic cancer cells growth in vitro and in vivo.Cordycepin was confirmed to induce apoptosis by activating caspase-3, caspase-9 and cytochrome c. Further studies suggested that MAPK pathway was blocked by cordycepin via inhibiting the expression of Ras and the phosphorylation of Erk. Moreover, cordycepin caused S-phase arrest and DNA damage associated with activating Chk2(checkpoint kinase 2) pathway and downregulating cyclin A2 and CDK2 phosphorylation. Very interestingly, we showed that cordycepin could bind to FGFR2(KD = 7.77 × 10-9) very potently to inhibit pancreatic cancer cells growth by blocking Ras/Er K pathway. | 8 | Epigallocatechin-3-gallate induces apoptosis, inhibits proliferation and decreases invasion of glioma cell显示文摘Epigallocatechin-3-gallate (EGCG), a major polyphenol in green tea, has been considered a potential therapeutic and chemopreventive agent for cancer. Glioma is a malignant tumor with high mortality but effective therapy has not yet been developed. In this study, we found that EGCG induced apoptosis in U251 glioma cells via the laminin receptor (molecular weight 67kDa) in a time- and dose-dependent manner, decreased their invasiveness and inhibited their proliferation. The mitogen-activated protein kinase pathway was shown to be involved in glioma cell apoptosis and proliferation. Furthermore, the mRNA levels of matrix metalloproteinase (MMP)-2 and MMP-9 were reduced after EGCG treatment. | 11 | Pulmonary Toxicity in Rats Caused by Exposure to Intratracheal Instillation of SiO2 Nanoparticles显示文摘Objective The effect of the silica nanoparticles(SNs) on lungs injury in rats was investigated to evaluate the toxicity and possible mechanisms for SNs.Methods Male Wistar rats were instilled intratracheally with 1 mL of saline containing 6.25,12.5,and 25.0 mg of SNs or 25.0 mg of microscale SiO_2 particles suspensions for 30 d,were then sacrificed.Histopathological and ultrastructural change in lungs,and chemical components in the urine excretions were investigated by light microscope,TEM and EDS.MDA,NO and hydroxyproline(Hyp) in lung homogenates were quantified by spectrophotometry.Contents of TNF-α,TGF-β1,IL-1β,and MMP-2 in lung tissue were determined by immunohistochemistry staining.Results There is massive excretion of Si substance in urine.The SNs lead pulmonary lesions of rise in lung/body coefficients,lung inflammation,damaged alveoli,granuloma nodules formation,and collagen metabolized perturbation,and lung tissue damage is milder than those of microscale SiO_2 particles.The SNs also cause increase lipid peroxidation and high expression of cytokines.Conclusion The SNs result into pulmonary fibrosis by means of increase lipid peroxidation and high expression of cytokines.Milder effect of the SNs on pulmonary fibrosis comparing to microscale SiO_2 particles is contributed to its elimination from urine due to their ultrafine particle size. | YANG Hong WU Qiu Yun LI Ming Yue LAO Can Shah ZHANG Ying Jian | 2017 | Biomedical and Environmental Sciences2017,30,4: | 5 |

| 14 | Seasonal variation and size distribution of biogenic secondary organic aerosols at urban and continental background sites of China显示文摘Size-resolved biogenic secondary organic aerosols(BSOA) derived from isoprene and monoterpene photooxidation in Qinghai Lake, Tibetan Plateau(a continental background site) and five cities of China were measured using gas chromatography/mass spectrometry(GC/MS). Concentrations of the determined BSOA are higher in the cities than in the background and are also higher in summer than in winter. Moreover, strong positive correlations(R2= 0.44–0.90) between BSOA and sulfate were found at the six sites,suggesting that anthropogenic pollution(i.e., sulfate) could enhance SOA formation,because sulfate provides a surface favorable for acid-catalyzed formation of BSOA. Size distribution measurements showed that most of the determined SOA tracers are enriched in the fine mode(< 3.3 μm) except for cis-pinic and cis-pinonic acids, both presented a comparable mass in the fine and coarse(> 3.3 μm) modes, respectively. Mass ratio of oxidation products derived from isoprene to those from monoterpene in the five urban regions during summer are much less than those in Qinghai Lake region. In addition, in the five urban regions relative abundances of monoterpene oxidation products to SOA are much higher than those of isoprene. | 15 | Chinese medicine Yu-Ping-Feng-San attenuates allergic inflammation by regulating epithelial derived pro-allergic cytokines显示文摘This study aimed to investigate the mechanisms of Yu-Ping-Feng-San(YPFS) on attenuating allergic inflammation in the initial stage of atopic dermatitis(AD). AD mouse model was established with fluorescein isothiocyanate(FITC) sensitization and elicitation. Epithelial barrier structure was observed with transmission electron microscope. The populations of dendritic cells(DCs)and group 2 innate lymphoid cells(ILC2 s) were detected by flow cytometry. Human immortalized keratinocyte(Ha Ca T) cells were stimulated with Poly(I:C)/TNF-α in vitro to assess thymic stromal lymphopoietin(TSLP), interleukin(IL)-33 and nuclear factor-κB(NF-κB) levels or expressions by immunofluorescence, enzyme linked immunosorbent assay(ELISA) and western blot. In the initial stage of AD, ear swelling and infiltration of inflammatory cells in ear tissues were markedly attenuated with YPFS treatments. The damaged structures of ear epithelium and the increased levels of Th2-cytokines induced by FITC were significantly rescued in YPFS-treated mice. The production of pro-allergic cytokines, TSLP and IL-33, as well as the cell populations of their target cells DCs and ILC2 s were decreased in AD model, respectively. Likewise, the levels of TSLP and IL-33 in Poly(I:C)/TNF-α-stimulated HaCaT cells showed the same results. Lower levels of p-NF-κB were detected with YPFS treatment, and the expressions of TSLP and IL-33 could be further decreased with inhibiting of NF-κB. | 16 | THREE-DIMENSIONAL NUMERICAL MODELINGOF COHESIVE SEDIMENT TRANSPORT BY TIDAL CURRENTIN PEARL RIVER ESTUARV显示文摘A three-dimensional numerical model of suspended sediment transport taking into account the effects of cohesiveness between sediments is presented. An efficient operator splitting scheme is employed to solve the equations of mass conservation, momentum and suspended sediment transport. The splitting method partitions a time step into three fractional time steps according to physical phenomena. In the first step, advection and Coriolis force are solved by Eulerian-Lagrangian method (ELM). Horizontal diffusion is approximated by implicit finite element method (FEM) for each horizontal layer in the second step. In the last step, vertical diffusion and pressure gradient are discretized by implicit finite difference method (FDM) for each nodal column. The continuity equation is solved by implicit FEM within an entire time step. The sigma coordinate system for vertical columns and nine-node isoparameteric finite elements for horizontal planes are employed in the model. Empirical functions specifically developed for cohesive sediments in China have been incorporated into the model. The model has been applied to investigate the suspended sediment concentration and seabed evolution in the Pearl River Estuary (PRE) in China. Computed results show that the distribution of sediment concentration and seabed evolution is in agreement with field measurements. In addition, the effect of the size of time step on result is discussed. | 17 | A male-ABCD algorithm for hepatocellular carcinoma risk prediction in HBs Ag carriers显示文摘Objective: Hepatocellular carcinoma(HCC) development among hepatitis B surface antigen(HBs Ag) carriers shows gender disparity, influenced by underlying liver diseases that display variations in laboratory tests. We aimed to construct a risk-stratified HCC prediction model for HBs Ag-positive male adults.Methods: HBs Ag-positive males of 35-69 years old(N=6,153) were included from a multi-center populationbased liver cancer screening study. Randomly, three centers were set as training, the other three centers as validation. Within 2 years since initiation, we administrated at least two rounds of HCC screening using Bultrasonography and α-fetoprotein(AFP). We used logistic regression models to determine potential risk factors,built and examined the operating characteristics of a point-based algorithm for HCC risk prediction.Results: With 2 years of follow-up, 302 HCC cases were diagnosed. A male-ABCD algorithm was constructed including participant's age, blood levels of GGT(γ-glutamyl-transpeptidase), counts of platelets, white cells,concentration of DCP(des-γ-carboxy-prothrombin) and AFP, with scores ranging from 0 to 18.3. The area under receiver operating characteristic was 0.91(0.90-0.93), larger than existing models. At 1.5 points of risk score,26.10% of the participants in training cohort and 14.94% in validation cohort were recognized at low risk, with sensitivity of identifying HCC remained 100%. At 2.5 points, 46.51% of the participants in training cohort and 33.68% in validation cohort were recognized at low risk with 99.06% and 97.78% of sensitivity, respectively.
