1A multicenter,retrospective.epidemiologic survey of the clinical features and management of bone metastatic disease in China显示文摘Background:Bone metastases are common in patients with advanced cancer.Bisphosphonates(BPs) could prevent or delay the development of skeleton-related events(SREs).The present study aimed to identify the clinical features of and treatment strategies for Chinese patients with bone metastases.Methods:Consecutive cancer patients who had bone metastases and received BP treatment were enrolled.A questionnaire was developed to collect the patients' clinical data,as well as information on the diagnosis and management of bone metastases.Physicians' awareness of the guidelines and knowledge of the application of BP were also assessed.Results:A total of 3223 patients with lung cancer(36.5%),breast cancer(30.9%),prostate cancer(8.5%),and gastrointestinal cancer(5.7%) were included in this study.The sites of bone metastases were the thoracic spine(56.0%),lumbar spine(47.1%),ribs(32.6%),and pelvis(23.2%).The SRE frequency was the highest in patients with multiple myeloma(36.6%),followed by those with lung cancer(25.9%),breast cancer(20.2%),prostate cancer(18.2%),and gastrointestinal cancer(17.3%).Irradiation to the bone was the most frequent SRE(58%in lung cancer patients,45%in breast cancer patients,and 48%in prostate cancer patients).Our survey also showed that 45.5%of patients received BP within 3 months after their diagnosis of bone metastases,whereas the remaining 54.5%of patients did not receive BP treatment until at least 3 months after their diagnosis of bone metastases.The SRE frequency in the former group was significantly lower than that in the latter group(4.0%vs.42.3%,P < 0.05).In patients with more than 6 months of continuous BP treatment,the mean time to the first SRE was significantly longer than that in patients with less than 6 months of continuous BP treatment(7.2 vs.3.4 months,P < 0.05).In addition,12.2%of the physicians were not aware of the efficacy of BP in preventing and delaying SRE.Only half(52.3%) of the physicians agreed that the BP treatment should persist for at least 6 months unless it was intolerable.Conclusions:Our study suggested that prompt and persistent BP treatment was associated with a reduced risk of SREs.However,our survey also revealed that the proper application of BP was not as common as expected in China.Yunpeng Yang Yuxiang Ma Jin Sheng Yan Huang Yuanyuan Zhao Wenfeng Fang Shaodong Hong Ying Tian Cong Xue Li Zhang 2016Chinese Journal of Cancer2016,35,5:13
2Role of pre-vertical compression in deformation behavior of Mg alloy AZ31B during super-high reduction hot rolling process显示文摘Mg alloy AZ31 B plates were processed by hot rolling with different thickness reductions per pass and prevertical compression followed by super-high reduction hot rolling(PVCR), respectively. Microstructure evolution, rolling formability variation and mechanical responses were investigated. As reduction per pass increased, the number of shear bands deflecting toward rolling direction increased, resulting in easy crack initiation in and around the bands. With increasing reduction per pass up to 80%, twinning and twinning-induced dynamic recrystallization(DRX) dominated the deformation of the edge material at350?C, resulting in local recrystallization with coarse grains and further largest edge-crack degree. Preinduced {10ī12} tensile twins by pre-vertical compression(PVC) increased number density of nucleation sites for dynamic recrystallization during the subsequent severe rolling, which enhanced the dominant role of continuous dynamic recrystallization. Designed PVCR-b was proved to be a relatively effective method to improve rolling formability of rolled Mg alloy AZ31 B plates. With this method, mean grain size of AZ31 B plate was significantly refined from ~600 mm to ~14.1 mm and more homogeneous grain size distribution along transverse direction(TD) was achieved. In addition, basal texture intensity was greatly weakened. 5The ACTIVE study protocol:apatinib or placebo plus gefitinib as first-line treatment for patients with EGFR-mutant advanced non-small cell lung cancer(CTONG1706)显示文摘Background:Gefitinib,as the first epidermal growth factor receptor tyrosine kinase inhibitors(EGFR-TKI)approved for the treatment of advanced non-small cell lung cancer(NSCLC),has been proved to significantly improve the progression-free survival(PFS)in the first-line setting but suffers from resistance 7-10 months after treatment initia-tion.Apatinib(YN968D1),a potent vascular endothelial growth factor receptor(VEGFR)2-TKI,specifically binds to VEGFR2 and leads to anti-angiogenetic and anti-neoplastic effect.Concurrent inhibition of VEGFR and EGFR path-ways represents a rational approach to improve treatment responses and delay the onset of treatment resistance in EGFR-mutant NSCLC.This ACTIVE study aims to assess the combination of apatinib and gefitinib as a new treatment approach for EGFR-mutant NSCLC as a first-line setting.Methods:This multicenter,randomized,double-blind,placebo-controlled phase III study(NCT02824458)has been designed to assess the efficacy and safety of apatinib or placebo combined with gefitinib as a first-line treatment for patients with EGFR-mutant advanced NSCLC.A total of 310 patients with EGFR-mutation(19del or 21L858R),pathological stage IIIB to IV non-squamous NSCLC were to be enrolled.The primary endpoint is investigator assessment of PFS,and the secondary endpoints include independent radiological central(IRC)-confirmed PFS,overall survival(OS),objective response rate(ORR),disease control rate(DCR),time to progressive disease(TTPD),duration of response(DoR),quality of life(QoL)and safety.The patients are randomized in a 1:1 ratio to receive gefitinib(250 mg,p.o.q.d.)plus apatinib(500 mg,p.o.q.d.)or gefitinib plus placebo,given until disease progression or intolerable adverse events.Exploratory biomarker analysis will be performed.This study is being conducted across China and comprises of 30 participating centers.Enrollment commenced in August 2017 and finished in December 2018,most of the patients are in the follow-up period.Anticipated outcomes and significance:The present study will be the first to evaluate the efficacy and safety profile of the combination of apatinib plus gefitinib as a first-line therapy for patients with EGFR-positive advanced non-squamous NSCLC.Importantly,this trial will provide comprehensive evidence on the treatment of EGFR-TKIs combined with antiangiogenic therapy.Zhonghan Zhang Fan Luo Yang Zhang Yuxiang Ma Shaodong Hong Yunpeng Yang Wenfeng Fang Yan Huang Li Zhang Hongyun Zhao 2019Cancer Communications2019,39,1:5
6Effect of electroacpuncture of Neiguan(PC 6)and Tianquan(PC 2)on skin temperature,blood perfusion,and adrenoceptor response in rats with acute myocardial ischemia显示文摘OBJECTIVE: To investigate the effect of electroacupuncture(EA) of acupoints Neiguan(PC 6) and Tianquan(PC 2) on the skin temperature, blood perfusion, and alpha1-and beta2-adrenoreceptor(α1-AR and β2-AR) protein and m RNA level in rats with acute myocardial ischemia.METHODS: Thirty male adult Wistar rats [(230 ±20) g] were randomly assigned into five groups(n = 6 each): a control group, sham operation group, model group, model group treated with EA at low frequency(L-EA, 2 Hz, 1 m A) and model group treated with EA at high frequency(H-EA, 100 Hz, 1 m A). The rat model was prepared by ligating the left anterior descending coronary artery. Electroacupuncture was performed at the left Neiguan(PC 6) for 20 min daily for 3 d. After the 3 rd time of the treatment, measurements of skin blood perfusion and temperature in Neiguan(PC 6) and Tianquan(PC 2) in all groups were made by laser speckle contrast imaging and infrared thermal image instrument. Real-time PCR and ELISA were used to measure m RNA level and protein level of α1-AR andβ2-AR in the skin tissues of Neiguan(PC 6) and Tianquan(PC 2), respectively. Serum levels of c Tn T and electrocardiogram were used to identify the state of myocardium.RESULTS: In the group receiving electroacupunture at Neiguan(PC 6), compared with control group,the skin temperature, blood perfusion, and β2-AR m RNA and protein level of model group significantly decreased(P < 0.05), α1-AR m RNA and protein level of model group significantly increased(P < 0.05);compared with model group, the skin temperature,blood perfusion, and β2-AR m RNA and protein level of L-EA significantly increased(P < 0.05), α1-AR m RNA and protein level of L-EA and H-EA significantly decreased(P < 0.05). 7Tranexamic acid for acute intracerebral haemorrhage growth based on imaging assessment (TRAIGE): a multicentre, randomised, placebo- controlled trial显示文摘Background Studies show tranexamic acid can reduce the risk of death and early neurological deterioration after intracranial haemorrhage.We aimed to assess whether tranexamic acid reduces haematoma expansion and improves outcome in intracerebral haemorrhage patients susceptible to haemorrhage expansion.Methods We did a prospective,double-blind,randomised,placebo-controlled trial at 10 stroke centres in China.Acute supratentorial intracerebral haemorrhage patients were eligible if they had indication of haemorrhage expansion on admission imaging(eg,spot sign,black hole sign or blend sign),and were treatable within 8 hours of symptom onset.Patients were randomly assigned(1:1)to receive either tranexamic acid or a matching placebo.The primary outcome was intracerebral haematoma growth(>33% relative or>6 mL absolute)at 24 hours.Clinical outcomes were assessed at 90 days.Results Of the 171 included patients,124(72.5%)were male,and the mean age was 55.9±11.6 years.89 patients received tranexamic acid and 82 received placebo.The primary outcome did not differ significantly between the groups:36(40.4%)patients in the tranexamic acid group and 34(41.5%)patients in the placebo group had intracranial haemorrhage growth(OR 0.96,95% CI 0.52 to 1.77,p=0.89).The proportion of death was lower in the tranexamic acid treatment group than placebo group(8.1%vs 10.0%),but there were no significant differences in secondary outcomes including absolute intracranial haemorrhage growth,death and dependency.Conclusions Among patients susceptible to haemorrhage expansion treated within 8 hours of stroke onset,tranexamic acid did not significantly prevent intracerebral haemorrhage growth.Larger studies are needed to assess safety and efficacy of tranexamic acid in intracerebral haemorrhage patients.Jingyi Liu Ximing Nie Hongqiu Gu Qi Zhou Haixin Sun Ying Tan Dacheng Liu Lina Zheng Jiahui Zhao Yan Wang Yibin Cao Haomeng Zhu Yunpeng Zhang Lijin Yi Yuehua Pu Miao Wen Zhonghua Yang Shengjun Sun Wenzhi Wang Xingquan Zhao Liping Liu Yongjun Wang 2021Stroke & Vascular Neurology2021,6,2:3
8Review on strategies of space-based optical space situational awareness显示文摘Space-based optical(SBO)space surveillance has attracted widespread interest in the last two decades due to its considerable value in space situation awareness(SSA).SBO observation strategy,which is related to the performance of space surveillance,is the top-level design in SSA missions reviewed.The recognized real programs about SBO SAA proposed by the institutions in the U.S.,Canada,Europe,etc.,are summarized firstly,from which an insight of the development trend of SBO SAA can be obtained.According to the aim of the SBO SSA,the missions can be divided into general surveillance and space object tracking.Thus,there are two major categories for SBO SSA strategies.Existing general surveillance strategies for observing low earth orbit(LEO)objects and beyond-LEO objects are summarized and compared in terms of coverage rate,revisit time,visibility period,and image processing.Then,the SBO space object tracking strategies,which has experienced from tracking an object with a single satellite to tracking an object with multiple satellites cooperatively,are also summarized.Finally,this paper looks into the development trend in the future and points out several problems that challenges the SBO SSA.HU Yunpeng LI Kebo LIANG Yan’gang CHEN Lei 2021Journal of Systems Engineering and Electronics2021,32,5:3

10Radiation-hardened property of single-walled carbon nanotube film-based field-effect transistors under low-energy proton irradiation显示文摘Strong C-C bonds,nanoscale cross-section and low atomic number make single-walled carbon nanotubes(SWCNTs)a potential candidate material for integrated circuits(ICs)applied in outer space.However,very little work combines the simulation calculations with the electrical measurements of SWCNT field-effect transistors(FETs),which limits further understanding on the mechanisms of radiation effects.Here,SWCNT film-based FETs were fabricated to explore the total ionizing dose(TID)and displacement damage effect on the electrical performance under low-energy proton irradiation with different fluences up to 1×1015 p/cm2.Large negative shift of the threshold voltage and obvious decrease of the on-state current verified the TID effect caused in the oxide layer.The stability of the subthreshold swing and the off-state current reveals that the displacement damage caused in the CNT layer is not serious,which proves that the CNT film is radiation-hardened.Specially,according to the simulation,we found the displacement damage caused by protons is different in the source/drain contact area and channel area,leading to varying degrees of change for the contact resistance and sheet resistance.Having analyzed the simulation results and electrical measurements,we explained the low-energy proton irradiation mechanism of the CNT FETs,which is essential for the construction of radiation-hardened CNT film-based ICs for aircrafts.Xiaorui Zhang Huiping Zhu Song’ang Peng Guodong Xiong Chaoyi Zhu Xinnan Huang Shurui Cao Junjun Zhang Yunpeng Yan Yao Yao Dayong Zhang Jingyuan Shi Lei Wang Bo Li Zhi Jin 2021Journal of Semiconductors2021,42,11:2
11SARS-CoV-2 impairs the disassembly of stress granules and promotes ALS-associated amyloid aggregation显示文摘The nucleocapsid(N)protein of SARS-CoV-2 has been reported to have a high ability of liquid-liquid phase separation,which enables its incorporation into stress granules(SGs)of host cells.However,whether SG invasion by N protein occurs in the scenario of SARS-CoV-2 infection is unknow,neither do we know its con-sequence.Here,we used SARS-CoV-2 to infect mam-malian cells and observed the incorporation of N protein into SGs,which resulted in markedly impaired self-dis-assembly but stimulated cell cellular clearance of SGs.NMR experiments further showed that N protein binds to the SG-related amyloid proteins via non-specific tran-sient interactions,which not only expedites the phase transition of these proteins to aberrant amyloid aggre-gation in vitro,but also promotes the aggregation of FUS with ALS-associated P525L mutation in cells.In addition,we found that ACE2 is not necessary for the infection of SARS-CoV-2 to mammalian cells.Our work indicates that SARS-CoV-2 infection can impair the dis-assembly of host SGs and promote the aggregation of SG-related amyloid proteins,which may lead to an increased risk of neurodegeneration.Yichen Li Shuaiyao Lu Jinge Gu Wencheng Xia Shengnan Zhang Shenqing Zhang Yan Wang Chong Zhang Yunpeng Sun Jian Lei Cong Liu Zhaoming Su Juntao Yang Xiaozhong Peng Dan Li 2022Protein & Cell2022,13,8:1

16Compressible and Lightweight MXene/Carbon Nanofiber Aerogel with “Layer‑Strut” Bracing Microscopic Architecture for Efficient Energy Storage显示文摘Two-dimensional MXene has recently captured widespread research attention in energy storage and conversion fields due to its high conductivity,large specific surface area,and remarkable electro-activity.However,its performance is still hindered by severe self-restacking of MXene flakes.Herein,conductive Ti_(3)C_(2)T_(x)/carbon nanofiber(CNF)composite aerogel with typical“layer-strut”bracing 3D microscopic architecture has been fabricated via synergistic assembly and freeze-drying process.In virtu of the strong interfacial interaction between polymeric precursor nanofibers and MXene mono-layers,gelation capability and 3D formability of Ti_(3)C_(2)T_(x) is greatly reinforced,as resulted Ti_(3)C_(2)T_(x)/CNF aerogels possess a highly ordered microporous structure with interlayered CNF penetrating between large size MXene lamellae.This special configuration guarantees the stability and pliability of the composite aerogels.Furthermore,the 3D form interconnected conductive network and the parallell alignment of the pores allow free electrical carriers motion and ion migration.As a result,the prepared Ti_(3)C_(2)T_(x)/CNF aerogel-based electrode exhibits an exceptional gravimetric specific capacitance of 268 F g^(−1) at a current density of 0.5 A g^(−1) and an excellent cycling stability of 8000 cylcles,and the assembled symmetric supercapacitor,delivers a high energy density of 3.425 W h kg^(−1) at 6000 W kg^(−1).This work offers a new route for the rational construction of 3D MXene assembly for advanced energy storage materials.Yongpeng Liu Dan Wang Chao Zhang Yan Zhao Piming Ma Weifu Dong Yunpeng Huang Tianxi Liu 2022Advanced Fiber Materials2022,4,4:1

19MdWRKY11 improves copper tolerance by directly promoting the expression of the copper transporter gene MdHMA5显示文摘Overuse of fungicides and fertilizers has resulted in copper(Cu)contamination of soils and toxic levels of Cu in apple fruits.To breed Cu-resistant apple(Malus domestica)cultivars,the underlying molecular mechanisms and key genes involved in Cu resistance must be identified.Here,we show that MdWRKY11 increases Cu tolerance by directly promoting the transcription of MdHMA5.MdHMA5 is a Cu transporter that may function in the storage of excess Cu in root cell walls and stems for Cu tolerance in apple.The transcription factor MdWRKY11 is highly induced by excess Cu.MdWRKY11 overexpression in transgenic apple enhanced Cu tolerance and decreased Cu accumulation.Apple calli transformed with an MdWRKY11-RNAi construct exhibited the opposite phenotype.Both an in vivo chromatin immunoprecipitation assay and an in vitro electrophoretic mobility shift assay indicated that MdWRKY11 binds to the promoter of MdHMA5.Furthermore,MdWRKY11 promoted MdHMA5 expression in transgenic apple plants,as revealed by quantitative PCR.Moreover,inhibition of MdWRKY11 expression by RNA interference led to a significant decrease in MdHMA5 transcription.Thus,MdWRKY11 directly regulates MdHMA5 transcription.Our work resulted in the identification of a novel MdWRKY11-MdHMA5 pathway that mediates Cu resistance in apple.Kun Shi Xuan Liu Yunpeng Zhu Yixue Bai Dongqian Shan Xiaodong Zheng Lin Wang Haixia Zhang Chanyu Wang Tianci Yan Fangfang Zhou Zehui Hu Yanzhao Sun Yan Guo Jin Kong 2020Horticulture Research2020,7,1:1
