1Characterization of the receptor-binding domain(RBD)of 2019 novel coronavirus:implication for development of RBD protein as a viral attachment inhibitor and vaccine显示文摘The outbreak of Coronavirus Disease 2019(COVID-19)has posed a serious threat to global public health,calling for the development of safe and effective prophylactics and therapeutics against infection of its causative agent,severe acute respiratory syndrome coronavirus 2(SARS-CoV-2),also known as 2019 novel coronavirus(2019-nCoV).The CoV spike(S)protein plays the most important roles in viral attachment,fusion and entry,and serves as a target for development of antibodies,entry inhibitors and vaccines.Here,we identified the receptor-binding domain(RBD)in SARS-CoV-2 S protein and found that the RBD protein bound strongly to human and bat angiotensin-converting enzyme 2(ACE2)receptors.SARS-CoV-2 RBD exhibited significantly higher binding affinity to ACE2 receptor than SARS-CoV RBD and could block the binding and,hence,attachment of SARS-CoV-2 RBD and SARS-CoV RBD to ACE2-expressing cells,thus inhibiting their infection to host cells.SARS-CoV RBD-specific antibodies could crossreact with SARS-CoV-2 RBD protein,and SARS-CoV RBD-induced antisera could cross-neutralize SARS-CoV-2,suggesting the potential to develop SARS-CoV RBD-based vaccines for prevention of SARS-CoV-2 and SARS-CoV infection.Wanbo Tai Lei He Xiujuan Zhang Jing Pu Denis Voronin Shibo Jiang Yusen Zhou Lanying Du 2020Cellular & Molecular Immunology2020,17,6:35
2Long-term changes in the fishery ecosystem structure of Laizhou Bay,China显示文摘Laizhou Bay provides a critical spawning and nursery habitat for many fishery species,including commercially important species,such as Fenneropenaeus chinensis and Larimichthys polyactis.The bay is severely stressed due to high fishing pressure and environmental changes.Based on the long-term ecosystem surveys in Laizhou Bay during the main spawning period(May) of most fishery species from 1959 to 2008,the responses of the Laizhou Bay fishery ecosystem were analyzed here,including regime shifts in species composition,biomass,species diversity,zooplankton,phytoplankton,and environmental variables.The dominant species of large-size and high economic value(e.g.Trichiurus haumela,L.polyactis) have been replaced by the short-lived,low-trophic-level planktivorous pelagic species(e.g.Setipinna taty,Engraulis japonicus) since the 1980s,and it is probable that the small-sized pelagic fishes have been recently replaced by invertebrates(e.g.Oratosquilla oratoria,Crangon affinis).The biomass of fishery resources declined continuously from 423.6 kg haul?1 h?1 in 1959 to 164.6 kg haul?1 h?1 in 1982,37.7 kg haul?1 h?1 in 1993,and less than 8 kg haul?1 h?1 in 1998-2008.Moreover,the biomass of zooplankton showed an increasing trend during 1959-2006,but showed a slight decline in 2008.The abundance of phytoplankton increased from 1959 through 1982,decreased substantially in 1993,and increased again until 2004.More recently,however,the phytoplankton abundance was very low.The sea surface temperature(SST) and sea bottom temperature(SBT) in May increased by 0.23°C a?1 and 0.16°C a?1,respectively,during 1982-2008.The salinity in May showed large fluctuations and reached its lowest values in 2004 and 2006.The ratio of dissolved inorganic nitrogen(DIN) to dissolved inorganic phosphate(DIP) increased.However,the dissolved silicon(DSi):DIP and DSi:DIN ratios decreased to a low level during 1959-2008.These changes seriously impacted primary production,and cascade effects then changed the structure and function of the fishery ecosystem.Further analysis indicated that multiple stresses caused the alterations in the structure of the Laizhou Bay fishery ecosystem.The top-down effect was identified as the main influence on the fishery species(at the top of the food chain) over the past five decades due to the increasing fishing pressure,whereas the bottom-up effect increased over the past three decades due to the strong variations in the environment.JIN XianShi SHAN XiuJuan LI XianSen WANG Jun CUI Yi ZUO Tao 2013Science China Earth Sciences2013,56,3:31
3Genome analysis of Taraxacum kok-saghyz Rodin provides new insights into rubber biosynthesis显示文摘The Russian dandelion Taraxacum kok-saghyz Rodin(TKS), a member of the Composite family and a potential alternative source of natural rubber(NR) and inulin, is an ideal model system for studying rubber biosynthesis. Here we present the draft genome of TKS, the first assembled NR-producing weed plant. The draft TKS genome assembly has a length of 1.29 Gb, containing 46 731 predicted protein-coding genes and68.56% repeats, in which the LTR-RT elements predominantly contribute to the genome enlargement. We analyzed the heterozygous regions/genes, suggesting its possible involvement in inbreeding depression.Through comparative studies between rubber-producing and non-rubber-producing plants, we found that enzymes of the mevalonate(MVA) pathway and rubber elongation might be critical for rubber biosynthesis, and several key isoforms have been isolated and shown to be predominantly expressed in the latex, indicating their crucial functions in rubber biosynthesis. Moreover, for two important families in rubber elongation, the CPT/CPTL and REF/SRPP families, diverse evolutionary tracks have been revealed. 4Disentangling the effects of topography and space on the distributions of dominant species in a subtropical forest显示文摘Topography and space are two important factors determining plant species assemblages in forest communities.Quantification of the contribution of these two factors in determining species distribution helps us to evaluate their relative importance in determining species assemblages.This study aims to disentangle the effect of topography and space on the distributions of 14 dominant species in a subtropical mixed forest.Spearman correlation analysis and the torustranslation test were used to test the species–habitat associations.Variation partitioning was used to quantify the relative contributions of topography and space at three sampling scales and three life stages.Correlation analyses and torus-translation tests showed species abundance was mostly correlated with topographic wetness index,vertical distance from the channel network and convexity.Variation partitioning showed that pure topography,pure space and spatially structured topography explained about 2.1 %,41.2 % and 13.8 %of the variation in species distributions,respectively.For nine species,total topography fractions peaked in 20 m quadrats.For ten species,the pure space fractions peaked in 50 m quadrats.For many species,the total topography fraction andthe pure space fraction were larger for the most abundant life stages,which reflected the importance of sampling effect.However,some cases did not follow this trend suggesting that the effects of ecological processes such as habitat filtering,density dependence or dispersal limitation may exceed the sampling effects.In conclusion,we found that spatially structured topography and pure space primarily shaped the distribution of dominant tree species.Furthermore,their effects were both scale- and life stage-dependent.Qinggang Wang Yaozhan Xu Zhijun Lu Dachuan Bao Yili Guo Junmeng Lu Kuihan Zhang Haibo Liu Hongjie Meng Xiujuan Qiao Handong Huang Mingxi Jiang 2014Chinese Science Bulletin2014,59,35:16
5Density dependence and habitat preference shape seedling survival in a subtropical forest in central China显示文摘Aims seedlings are vulnerable to many kinds of fatal abiotic and biotic agents,and examining the causes of seedling dynamics can help understand mechanisms of species coexistence.To disentangle the relative importance of neighborhood densities,habitat factors and phylogenetic relatedness on focal seedling survival,we monitored the survival of 5306 seedlings of 104 species>15 months.We address the following questions:(i)How do neighborhood densities,habitat variables and phylogenetic relatedness affect seedling survival?What is the relative importance of conspecific densities,habitat variables and phylogenetic relatedness to seedling survival?(ii)Does the importance of the neighborhood densities,habitat variables and phylogenetic relatedness vary among growth forms,leaf habits or dispersal modes?specially,does the conspecific negative density dependence inhibit tree and decidu-ous seedlings more compared with shrub and evergreen species?Does density dependence affect the wind and animal-dispersed species equally?Methods We established 135 census stations to monitor seedling dynamics in a 25-ha subtropical forest plot in central China.Conspecific and heterospecific seedling density in the 1-m2 seedling plot and adult basal area within a 20-m radius provided neighborhood density var-iables.mean elevation,convexity and aspect of every 5-×5-m grid with seedling plots were used to quantify habitat characteristics.We calculated the relative average phylodiversity between focal seed-ling and heterospecific neighbors to quantify the species related-ness in the neighborhood.Eight candidate generalized linear mixed models with binominal error distribution were used to compare the relative importance of these variables to seedling survival.akaike’s information criteria were used to identify the most parsimonious models.Important Findingsat the community level,both the neighborhood densities and phylogenetic relatedness were important to seedling survival.We found negative effects of increasing conspecific seedlings,which suggested the existence of species-specific density-dependent mortality.Phylodiversity of heterospecific neighbors was negatively related to survival of focal seedlings,indicating similar habitat preference shared among phylogenetically closely related species may drive seedling survival.The relative importance of neighborhood densities,habitat variables and phylogenetic relatedness varied among ecological guilds.Conspecific densities had significant negative effect for deciduous and wind-dispersed species,and marginally significant for tree seedlings>10 cm tall and animal-dispersed species.Habitat variables had limited effects on seedling survival,and only elevation was related to the sur-vival of evergreen species in the best-fit model.We conclude that both negative density-dependent mortality and habitat preference reflected by the phylogenetic relatedness shape the species coex-istence at seedling stage in this forest.Junmeng Lu Daniel J.Johnson Xiujuan Qiao Zhijun Lu Qinggang Wang Mingxi Jiang 2015Journal of Plant Ecology2015,8,6:12
6Characteristics of ozone vertical profile observed in the boundary layer around Beijing in autumn显示文摘In the autumn of 2008,the vertical profiles of ozone and meteorological parameters in the low troposphere (0-1000 m) were observed at two sites around Beijing,specifically urban Nanjiao and rural Shangdianzi.At night and early morning,the lower troposphere divided into two stratified layers due to temperature inversion.Ozone in the lower layer showed a large gradient due to the titration of NO.Air flow from the southwest brought ozone-rich air to Beijing,and the ozone profiles were marked by a continuous increase in the residual layer at night.The accumulated ozone in the upper layer played an important role in the next day's surface peak ozone concentration,and caused a rapid increase in surface ozone in the morning.Wind direction shear and wind speed shear exhibited different influences on ozone profiles and resulted in different surface ozone concentrations in Beijing.Zhiqiang Ma Xiaoling Zhang Jing Xu xiujuan Zhao Wei Meng 2011Journal of Environmental Sciences2011,23,8:11
7Biological coupling anti-wear properties of three typical molluscan shells—Scapharca subcrenata, Rapana venosa and Acanthochiton rubrolineatus显示文摘Molluscan shells are fascinating examples of highly ordered hierarchical structure and complex organic-inorganic biocomposite material. However, their anti-wear properties were rarely studied especially in the perspective of biological coupling. So in the current study three typical shells, Scapharca subcrenata, Rapana venosa and Acanthochiton rubrolineatus, were selected as coupling models to further study their anti-wear properties. Stereomicroscope and scanning electron microscopic observations showed that all these three shells had specific surface morphologies and complicated section microstructures. Importantly, a special structure, pore canal tubules, was discovered in the shells of Scapharca subcrenata and Acanthochiton rubrolineatus, which probably contributed most to their anti-wear properties. X-ray diffraction and micro-Vikers hardness tester were further adopted to analyze the phase compositions and micro-hardness of the shells. The measured results demonstrated that aragonite was the most extensive phase present in the shell, and possesed a relatively high micro-hardness. In this paper, the shells were described in details in morphology, structure and material with emphasis on the relationship with anti-wear property. 10Detecting protein complexes from DPINs by density based clustering with Pigeon-Inspired Optimization Algorithm显示文摘Detecting protein complexes is crucial to understand principles of cellular organization. Plenty evidences have indicated that sub-graphs with high density in protein-protein interaction(PPI) network, especially dynamic PPI network(DPIN), usually correspond to protein complexes. As a well-known density-based clustering algorithm, Density-Based Spatial Clustering of Applications with Noise(DBSCAN) has been used in many areas due to its simplicity and the ability to detect clusters of different sizes and shapes. However, one of its limitations is that the performance of DBSCAN depends on two specified parameters ε and Min Pts, where εrepresents the maximum radius of a neighborhood from an observing point while Min Pts means the minimum number of data points contained in such a neighborhood. In this article, we develop a new method named as P-DBSCAN to detect protein complexes in DPIN by using Pigeon-Inspired Optimization(PIO) Algorithm to optimize the parameters ε and Min Pts in DBSCAN. 13Sequential use of ultraviolet light and chlorine for reclaimed water disinfection显示文摘Several disinfection processes of ultraviolet (UV),chlorine or UV followed by chlorine were investigated in municipal wastewater according to the inactivation of Escherichia coli,Shigella dysenteriae and toxicity formation.The UV inactivation of the tested pathogenic bacteria was not affected by the quality of water.It was found that the inactivated bacteria were obviously reactivated after one day in dark.Fluorescent light irradiation increased the bacteria repair.The increase of UV dosage could cause more damage to bacteria to inhibit bacteria self-repair.No photoreactivation was detected when the UV dose was up to 80 mJ/cm2 for E.coli DH5α,and 23 mJ/cm2 for S.dysenteriae.Nevertheless,sequential use of 8 mJ/cm2 of UV and low concentration of chlorine (1.5 mg/L) could effectively inhibit the photoreactivation and inactivate E.coli below the detection limits within seven days.Compared to chlorination alone,the sequential disinfection decreased the genotoxicity of treated wastewater,especially for the sample with high NH 3-N concentration.Xiujuan Wang Xuexiang Hu Chun Hu Dongbin Wei 2011Journal of Environmental Sciences2011,23,10:8
14Differentiation potential of bone marrow mesenchymal stem cells in duck显示文摘The bone marrow mesenchymal stem cells(MSCs) are multipotent stem cells which can differentiate into mesenchymal cells in vitro.In this study,MSCs in duck were isolated from bone marrow by density gradient centrifuge separation,purified and expanded in the me-dium.The primary MSCs were expanded for 11 passages.The different-passage MSCs were induced to differentiate into osteoblasts and neuron-like cells.Karyotype analysis indicated that MSCs kept diploid condition and the hereditary feature was stable.The different-passage MSCs expressed CD44,ICAM-1 and SSEA-4,but not CD34,CD45 and SSEA-1 when detected by immunofluorescence staining.There was no significant difference among the positive rates of passages 2,6 and 8(P > 0.05),but a significant difference existed among those of passages 2,6,8 and 11(P < 0.05).After the osteogenic inducement was added,the induced different-passage MSCs expressed high-level alkaline phosphatase(ALP),and are positive for tetracycline staining,Alizarin Red staining and Von Kossa staining.After the neural inducement was added,about 70% cells exhibited typical neuron-like phenotype,the induced different-passage MSCs expressed Nestin,neuron-specific enolase(NSE) and glial fibrillary acidic protein(GFAP) when detected by immunofluorescence staining.There was no significant difference among the positive rates of passages 3,4 and 6(P>0.05),but a significant difference existed among those of passages 3,4,6 and 8(P<0.05).These results suggest that MSCs in duck were capable of differentiating into osteoblasts and neuron-like cells in vitro.Linfeng Li Xiujuan Bai Xuelian Gong Hongkun Liu Lina Chen Weijun Guan Yuehui Ma 2009Journal of Genetics and Genomics2009,36,3:8

16Geochronological and geochemical constraints on the Cuonadong leucogranite, eastern Himalaya显示文摘First comprehensive investigations of the Cuonadong leucogranite exposed in North Himalayan gneiss dome of southern Tibet are presented in this study.The SIMS U–Pb ages of oscillatory zircon rims scatter in a wide range from 34.1 to 16.0 Ma, and the Cuonadong leucogranite probably emplaced at 16.0 Ma. High-precision ^(40)Ar/^(39)Ar dating on a muscovite sample yields an essentially flat age spectrum with consistent plateau and isochron ages, indicating that the Cuonadong leucogranite cooled below 450 °C at 14 Ma. Based on the youngest zircon U–Pb age and muscovite^(40)Ar/^(39)Ar age, the Cuonadong leucogranite experienced rapid cooling with a rate of 119 °C/Myr from 16 to 14 Ma. The geochronological data of this undeformed leucogranite also suggest that the ductile extension of the South Tibetan Detachment System in the eastern Himalaya ceased by ca. 14 Ma. Furthermore,the initial Sr–Nd isotopic compositions and Nd model ages demonstrate that the leucogranite was derived from metapelitic source within the Greater Himalayan Crystalline Complex. 17Highly stable lead-free Cs3Bi2I9 perovskite nanoplates for photodetection applications显示文摘Lead halide perovskites have received tremendous attentions recently for their excellent properties such as high light absorption coefficient and long charge carrier diffusion length. However, the stability issues and the existence of toxic lead cations have largely limited their applications in optoelectronic area. Herein, we report the synthesis and investigation of highly stable and lead-free Cs3Bi2I9 perovskite nanoplates for visible light photodetection applications. The Cs3Bi2I9 nanoplates were synthesized through a facile solution-processed method, which is also applicable to various substrates. The achieved nanoplates present very good crystal quality and exhibit excellent long-term stability even exposed in moist air for several months. Photodetectors were constructed based on these high-quality perovskite nanoplates for the first time, and display a maximum photoresponsivity of 33.1 mA/W under the illumination of 450 nm laser, which is six times higher than the solution-synthesized CH3NH3PbI3 nanowire photodetectors. The specific detectivity of these devices can reach up to 10^10 Jones. Additionally, the devices exhibit fast rise and decay time of 10.2 and 37.2 ms, respectively, and highly stable photoswitching behavior with their photoresponse well retaining under alternating light and darkness. 18Study on fire retardant mechanism of nano-LDHs in intumescent system显示文摘This paper investigated the fire-retardant mechanism of the nano-LDHs in the intumescent system by the temperature programmed oxidation (TPO). The results indicated that the nano-LDHs species were responsible for the catalytic oxidation of the rich-carbon compound in oxygen atmosphere. 20Molecular epidemiology demonstrates that imported and local strains circulated during the 2014 dengue outbreak in Guangzhou, China显示文摘The dengue virus(DENV) is a vital global public health issue. The 2014 dengue epidemic in Guangzhou, China, caused approximately 40,000 cases of infection and five deaths. We carried out a comprehensive investigation aimed at identifying the transmission sources in this dengue epidemic. To analyze the phylogenetics of the 2014 dengue strains, the envelope(E) gene sequences from 17 viral strains isolated from 168 dengue patient serum samples were sequenced and a phylogenetic tree was reconstructed. All 17 strains were serotype Ⅰ strains, including 8genotype Ⅰ and 9 genotype V strains. Additionally, 6 genotype Ⅰ strains that were probably introduced to China from Thailand before 2009 were widely transmitted in the 2013 and 2014 epidemics, and they continued to circulate until 2015, with one affinis strain being found in Singapore. The other 2 genotype Ⅰ strains were introduced from the Malaya Peninsula in 2014. The transmission source of the 9 genotype Ⅴ strains was from Malaysia in 2014. DENVs of different serotypes and genotypes co-circulated in the 2014 dengue outbreak in Guangzhou.
