4Detection of ALK translocation in non-small cell lung carcinoma(NSCLC) and its clinicopathological significance using the Ventana immunohistochemical staining method: a single-center large-scale investigation of 1,504 Chinese Han patients显示文摘Objective: The novel fully automated immunohistochemistry(IHC) assay-Ventana anaplastic lymphoma kinase(ALK)-D5F3 for screening ALK rearrangements has been approved by China's Food and Drug Administration in 2013, our previous study disclosed a highly specificity and sensitivity nearly 100%, and its efficacy needs to be evaluated in a large cohort of primary lung adenocarcinoma patients, and to compare clinicopathological features with ALK(+) and ALK(-) lung adenocarcinoma.Methods: A total of 1,504 consecutive surgical lung adenocarcinoma cases of Chinese Han population were collected and re-diagnosed according to the 2011 multidisciplinary classification of lung adenocarcinoma.Fully automated Ventana ALK-D5F3 IHC staining with a binary scoring was adopted to evaluate staining and correlated with clinicopathological characters, including age, sex, differentiation degree, histological subtype, lymph node metastasis, and clinical staging.ALK(+) patients were followed-up, and targeted therapy of ALKinhibitors was adopted and observed in patients with stage Ⅳ according to the NCCN guideline.Results: ALK positive adenocarcinomas were identified in 6.6% of the surgically resected 1,504 NSCLCs, and significantly younger than the negative group(P<0.05).Mucinous adenocarcinoma(28.2%) was determined to be predominant in ALK(+) cases, followed by the solid type(11.7%), specific type(6.8%), papillary type(5.6%), acinar type(5.5%), and lepidic type(3.1%), and the differences were statistically significant(χ~2=42.011, P<0.05).ALK(+) adenocarcinoma with lymph node metastasis(10.8%) were significantly higher than that without lymph node metastasis(4.5%)(χ~2=19.809, P<0.05); and ALK(+) in phase Ⅳ(20%) was significantly higher than phase Ⅲ(12.9%), phase Ⅱ(4.2%), phase Ⅰ(4.5%), and phase 0(0)(χ~2=36.068, P<0.05).Multivariate logistic regression disclosed that patient age, AJCC staging, and histological mucinous subtype were correlated with ALK positive staining(OR=0.959, 1.578, 5.036, respectively).Sixty eight patients had followed-up results, five patients out of which primarily diagnosed or progressed into Stage Ⅳ benefited well from targeted therapy with Crizotinib.Conclusions: The ALK fusion protein was seen in 6.6% Chinese NSCLC patients, and mostly seen in younger, clinically higher staging, mucinous and solid predominant adenocarcinoma.Clinical trials in patients of Stage Ⅳ confirmed that ALK-D5F3 Ventana IHC is serviceable in screening ALK-positive candidates for molecular targeted therapy.Lin Yang Yun Ling Lei Guo Di Ma Xuemin Xue Bingning Wang Junling Li Jianming Ying 2016Chinese Journal of Cancer Research2016,28,5:8
5Velvet antler polypeptide is able to induce differentiation of neural stem cells towards neurons in vitro显示文摘OBJECTIVE: To investigate the neural differentiation capacity of water extraction of velvet antler.METHODS: Velvet antler(Cervus Nippon Temminck) polypeptide(VAP) was used to differentiate neural stem cells(NSCs) towards neurons in the study. Firstly, we obtain the polypeptides of VAP by water extraction. Secondly, we observed the morphology, assayed the factors in the media by enzyme-linked immunosorbent assay, and detected the special neural molecules by immuno fl uorescence staining. NSCs were cultured on the cell climbing film. 6Bamboo Leaf and Pollen Fossils from the Late Miocene of Eastern Zhejiang,China and their Phytogeological Significance显示文摘Fossil bamboo leaves and pollen from Upper Miocene deposits of the Shengxian Formation in Tiantai and Ninghai counties,eastern Zhejiang,China represent a rare record in Asia.The distinctive pseudopetiole and parallel venation of the leaf blades and the clearly thickened annulus of the pollen aperture place them in the subfamily Bambusoideae.Morphological analysis supports the determination of these fossil leaves as belonging to the genus Bambusium and two new species are described.Bambusium latipseudopetiolus Q.J.Wang et B.N.Sun sp.nov.has a distinctly wide pseudopetiole of 0.23 cm in width and 0.40 cm in length,several vascular bundles on parallel veins,and 5–8 lateral veins on both sides of the midrib.Bambusium longipseudopetiolus Q.J.Wang et B.N.Sun sp.nov.has a distinctly long pseudopetiole of 0.60 cm in length and 0.10 cm in width,several vascular bundles on the pseudopetiole,and 6 lateral veins on both sides of the midrib.Pollen grains from the same layer provide additional evidence of bamboos.They are characterized by 32.0–51.0 μm in diameter,a round pore 3.0–4.0 μm in diameter with a broad thickened annulus 2.5–3.5 μm around,and several conspicuous secondary folds on the exine surface.The morphological analysis leads to their assignment to Graminidites bambusoides Stuchlik.These fossils are important for the study of bamboo phytogeography in China.They demonstrate that there were bamboos growing in southeastern China during the Late Miocene and that bamboos in Zhejiang begin to diversify no later than the Late Miocene.In combination with bamboo fossils from other places,it seems that bamboos had a wide distribution across southern China during the Miocene,ranging from southwestern Yun'nan to southeastern Zhejiang.WANG Qiujun MA Fujun YANG Yi DONG Junling WANG Haofei LI Ruiyun XU Xiaohui SUN Bainian 2014Acta Geologica Sinica(English Edition)2014,88,4:7

8Sequential therapy according to distinct disease progression patterns in advanced ALK-positive non-small-cell lung cancer after crizotinib treatment显示文摘Objective: Crizotinib is recommended as the first-line therapy for advanced anaplastic lymphoma kinase(ALK)-positive non-small-cell lung cancer(NSCLC). Despite its initial efficacy, patients ultimately acquire resistance to crizotinib within 1 year. In such patients, the optimal sequential therapy after crizotinib treatment remains unknown. This study explored which sequential therapy option confers the greatest benefit.Methods: A total of 138 patients with advanced ALK-positive NSCLC resistant to crizotinib were studied. Based on patterns of disease progression of metastases, patients were divided into 3 groups: brain progression, non-liver progression, and liver progression. Sequential therapies included crizotinib continuation plus local therapy, nextgeneration ALK inhibitors(ALKi's), and chemotherapy. The primary endpoint was overall survival(OS) from the time of crizotinib resistance to death or last follow-up.Results: The 138 patients included 64 cases with progression in brain, 57 cases in non-liver sites and 17 cases in liver. A significant difference in OS was observed among the distinct progression pattern(median OS, 25.4 months in brain, 15.8 months in non-liver, and 10.8 months in liver, respectively, P=0.020). The difference in OS among sequential therapies was statistically significant in the non-liver progression group(median OS, 27.6 months with next-generation ALKi's, 13.3 months with crizotinib continuation, and 10.8 months with chemotherapy,respectively, P=0.019). However, crizotinib continuation plus local therapy seems to provide non-inferior median OS compared with next-generation ALKi's for patients with brain progression(median OS, 28.9 months vs.32.8 months, P=0.204). And no significant differences in OS were found in patients with progression in liver(P=0.061).Conclusions: Crizotinib continuation together with local therapy might be a feasible strategy for patients with progression in brain beyond crizotinib resistance, as well as next-generation ALKi's. 10Helicobacter pylori antibody responses in association with eradication outcome and recurrence: a population-based intervention trial with 7.3-year follow-up in China显示文摘Objective:To identify serum biomarkers that may predict the short or long term outcomes of anti-Helicobacter pylori(H.pylori)treatment,a follow-up study was performed based on an intervention trial in Linqu County,China.Methods:A total of 529 subjects were selected randomly from 1,803 participants to evaluate total anti-H.pylori immunoglobulin G(Ig G)and 10 specific antibody levels before and after treatment at 1-,2-and 7.3-year.The outcomes of anti-H.pylori treatment were also parallelly assessed by 13C-urea breath test at 45-d after treatment and 7.3-year at the end of follow-up.Results:We found the medians of anti-H.pylori Ig G titers were consistently below cut-off value through 7.3years in eradicated group,however,the medians declined in recurrence group to 1.2 at 1-year after treatment and slightly increased to 2.0 at 7.3-year.While the medians were significantly higher(>3.0 at 2-and 7.3-year)among subjects who failed the eradication or received placebo.For specific antibody responses,baseline seropositivities of Fli D and Hpa A were reversely associated with eradication failure[for Fli D,odds ratio(OR)=0.44,95%confidence interval(95%CI):0.27–0.73;for Hpa A,OR=0.32,95%CI:0.17–0.60].The subjects with multiple positive specific antibodies at baseline were more likely to be successfully eradicated in a linear fashion(Ptrend=0.006).Conclusions:Our study suggested that total anti-H.pylori Ig G level may serve as a potential monitor of longterm impact on anti-H.pylori treatment,and priority for H.pylori treatment may be endowed to the subjects with multiple seropositive antibodies at baseline,especially for Fli D and Hap A.Tianyi Wang Yang Zhang Huijuan Su Zhexuan Li Lian Zhang Junling Ma Weidong Liu Tong Zhou Weicheng You Kaifeng Pan 2017Chinese Journal of Cancer Research2017,29,2:3
11Is there an optimal time to initiate adjuvant chemotherapy to predict benefit of survival in non-small cell lung cancer?显示文摘Objective: Adjuvant chemotherapy(AC) after curative resection is known to improve the survival of patients with non-small cell lung cancer(NSCLC); however, few studies have reported the correlation between the time to initiation of AC(TTAC) and survival in NSCLC patients.Methods: The clinical data of 925 NSCLC patients who received curative resection and post-operative AC at the Cancer Hospital of Chinese Academy of Medical Sciences between 2003 and 2013 were retrospectively analyzed.TTAC was measured from the date of surgery to the initiation of AC. Disease-free survival(DFS) was defined as the duration from surgery to the time of tumor recurrence or last follow-up evaluation. The optimal cut-off value of TTAC was determined by maximally selected log-rank statistics. The DFS curve was estimated using the Kaplan-Meier method, and the Cox proportional hazards regression model was used to identify risk factors independently associated with DFS. Propensity score matching(PSM) was performed for survival analysis using the match data.Results: The optimal discriminating cut-off value of TTAC was set at d 35 after curative resection based on which the patients were assigned into two groups: group A(≤35 d) and group B(>35 d). There was no significant difference in the DFS between the two groups(P=0.246), indicating that the TTAC is not an independent prognostic factor for DFS. A further comparison continued to show no significant difference in the DFS among 258 PSM pairs(P=0.283).Conclusions: There was no significant correlation between the TTAC and DFS in NSCLC patients. 12Identification and characterization of the cellular subclones that contribute to the pathogenesis of mantle cell lymphoma显示文摘Mantle cell lymphoma(MCL)is a B-cell malignancy with poor clinical outcome and undefined pathogenesis.Development of clinically relevant cellular models for MCL research is an urgent need.Our preliminary observations lead the development of two novel hypotheses that we tested in this study:1.multicellular spheroid might be a unique growth mode of earlystage cells in MCL;2.MCL might be a polyclonal tumor.We made the following original observations that have not been reported:First,we have provided a new experiment method for enriching MCL early-stage cells and characterized the spheroid mode of growth as a unique feature of early-stage MCL cells in cell line as well as in clinical samples.Second,we have established a clinically relevant cellular model of MCL,the JeKo-1-spheroid cell line,that was highly enriched in early-stage sub-clones.JeKo-1-spheroid cells and the spheroid growing cells enriched from MCL patients exhibited comparably enhanced tumorigenic abilities and similar biological features.Third,Immunophenotypic analysis has revealed that MCL may be derived from precursor-B(pre-B),immature-B and mature-B cells,not only the mature-B cells as WHO classified in 2016.Fourth,MCL may be a polyclonal disease composed of CD19e/IgMe,CD19 e/IgMt,CD19t/IgMt three sub-clones,of which the CD19e/IgMt sub-clone might be the dominant sub-clone with the strongest tumorigenic ability.Fifth,CD19t/IgMe that differentiates MCL and normal B cells may represent a new marker for MCL early detection,minor residual disease monitoring after therapies and prognosis.Junling Tang Li Zhang Tiejun Zhou Zhiwei Sun Liangsheng Kong Li Jing Hongyun Xing Hongyan Wu Yongli Liu Shixia Zhou Jingyuan Li Mei Chen Fang Xu Jirui Tang Tao Ma Min Hu Dan Liu Jing Guo Xiaofeng Zhu Yan Chen Ting Ye Jianyu Wang Xiaoming Li H.Rosie Xing 2019Genes & Diseases2019,6,4:2

14Impact of crizotinib on long-term survival of ALK-positive advanced non-small-cell lung cancer: A Chinese multicenter cohort study显示文摘Objective: Crizotinib has demonstrated promising efficacy in patients with anaplastic lymphoma kinase(ALK)-positive non-small-cell lung cancer(NSCLC) in clinical trials. We conducted this retrospective multicenter study to assess the outcomes of crizotinib therapy in, to our knowledge, a large sample cohort of patients with ALKpositive advanced NSCLC.Methods: We reviewed the medical records of 484 unselected ALK-positive NSCLC patients treated with crizotinib at 5 cancer centers in China from January 2013 to November 2017. Clinical data were collected from the initiation of crizotinib therapy to Response Evaluation Criteria in Solid Tumors(RECIST)-defined progressive disease(PD).Results: A total of 428 eligible ALK-positive NSCLC patients were enrolled, 273(63.8%) of whom received crizotinib as first-line treatment. The median progression-free survival(PFS) and overall survival(OS) from the initiation of crizotinib treatment were 14.4 [95% confidence interval(95% CI), 12.4-16.4] months and 53.4(95%CI, 33.7-73.1) months, respectively. In subgroup analyses, patients who received crizotinib as first-line treatment showed a higher disease control rate(DCR) and a longer median OS compared with second-/later-line crizotinib treatment(94.8% and OS not reached vs. 89.0% and 40.5 months, respectively). For 261 patients with RECISTdefined PD, multivariate Cox analysis revealed that in patients who received first-line crizotinib therapy, continued crizotinib beyond progressive disease(CBPD) and next-generation ALK inhibitors after crizotinib failure were associated with improved survival.Conclusions: This study has demonstrated the clinically meaningful benefit of crizotinib treatment in a large cohort of Chinese ALK-positive NSCLC patients. 15Buxus Leaves from the Oligocene of Guangxi, China and Their Biogeographical Significance显示文摘The present paper reports two new species of Buxus from the Oligocene Ningming Formation of Guangxi, South China, on the basis of several compressed fossil leaves.Buxus ningmingensis sp.nov.is characterized by an elliptic to ovate shape; pinnate venation with marginal secondary, long hairs on the adaxial side of the midvein base; and amphistomatic, anomocytic stomata with a prominent outer ring.All of these characteristics suggest a closest affinity to the living Buxus microphylla subsp.sinica.Another new species identified as Buxus preaustro-yunnanensis sp.nov.is characterized by the diagnostic formation of its admedially branched tertiaries, the trunks of which are often strongly thickened by sclereides.This species is most similar to the extant Buxus austroyunnanensis.The co-occurrence of Buxus ningmingensis sp.nov.and Buxus preaustro-yunnanensis sp.nov.suggests that these two Buxus lived during the Oligocene under a warm, tropical to subtropical climate, similar to the current climate of Ningming.Our findings provide the earliest fossil evidence of Buxus leaves with detailed illustrations of leaf architectural and cuticular features.The occurrence of these two species indicates that Buxus began to diversify in Guangxi no later than the Oligocene.MA Fujun WANG Qiujun DONG Junling YANG Yi WANG Wenjia YAN Defei SUN Bainian 2015Acta Geologica Sinica(English Edition)2015,89,5:2
