| 1 | SHP2 promotes proliferation of breast cancer cells through regulating Cyclin D1 stabilityviathe PI3K/AKT/GSK3β signaling pathway显示文摘Objective:The tyrosine phosphatase SHP2 has a dual role in cancer initiation and progression in a tissue type-dependent manner.Several studies have linked SHP2 to the aggressive behavior of breast cancer cells and poorer outcomes in people with cancer.Nevertheless,the mechanistic details of how SHP2 promotes breast cancer progression remain largely undefined.Methods:The relationship between SHP2 expression and the prognosis of patients with breast cancer was investigated by using the TCGA and GEO databases.The expression of SHP2 in breast cancer tissues was analyzed by immunohistochemistry.CRISPR/Cas9 technology was used to generate SHP2-knockout breast cancer cells.Cell-counting kit-8,colony formation,cell cycle,and EdU incorporation assays,as well as a tumor xenograft model were used to examine the function of SHP2 in breast cancer proliferation.Quantitative RT-PCR,western blotting,immunofluorescence staining,and ubiquitination assays were used to explore the molecular mechanism through which SHP2 regulates breast cancer proliferation.Results:High SHP2 expression is correlated with poor prognosis in patients with breast cancer.SHP2 is required for the proliferation of breast cancer cellsin vitro and tumor growthin vivo through regulation of Cyclin D1 abundance,thereby accelerating cell cycle progression.Notably,SHP2 modulates the ubiquitin–proteasome-dependent degradation of Cyclin D1viathe PI3K/AKT/GSK3βsignaling pathway.SHP2 knockout attenuates the activation of PI3K/AKT signaling and causes the dephosphorylation and resultant activation of GSK3β.GSK3βthen mediates phosphorylation of Cyclin D1 at threonine 286,thereby promoting the translocation of Cyclin D1 from the nucleus to the cytoplasm and facilitating Cyclin D1 degradation through the ubiquitin–proteasome system.Conclusions:Our study uncovered the mechanism through which SHP2 regulates breast cancer proliferation.SHP2 may therefore potentially serve as a therapeutic target for breast cancer. | Yue Yuan Yanling Fan Zicong Gao Xuan Sun He Zhang Zhiyong Wang Yanfen Cui Weijie Song Zhaosong Wang Fei Zhang Ruifang Niu | 2020 | Cancer Biology & Medicine2020,17,3: | 11 |
| 2 | PKM2 promotes reductive glutamine metabolism显示文摘Objective:Pyruvate kinases M(PKM),including the PKM1 and PKM2 isoforms,are critical factors in glucose metabolism.PKM2promotes aerobic glycolysis,a phenomenon known as'the Warburg effect'.The purpose of this study was to identify the roles of PKM2 in regulating cellular metabolism.Methods:The CRISPR/Cas9 system was used to generate the PKM-knockout cell model to evaluate the role of PKM in cellular metabolism.Lactate levels were measured by the Vitros LAC slide method on an autoanalyzer and glucose levels were measured by the autoanalyzer AU5800.The metabolism of ^(13)C_6-glucose or ^(13)C_5-glutamine was evaluated by liquid chromatography/mass spectrometry analyses.The effects of PKM on tumor growth were detected in vivo in a tumor-bearing mouse model.Results:We found that both PKM1 and PKM2 enabled aerobic glycolysis,but PKM2 converted glucose to lactate much more efficiently than PKM1.As a result,PKM2 reduced glucose levels reserved for intracellular utilization,particularly for the production of citrate,and thus increased theα-ketoglutarate/citrate ratio to promote the generation of glutamine-derived acetylcoenzyme A through the reductive pathway.Furthermore,reductive glutamine metabolism facilitated cell proliferation under hypoxia conditions,which supports in vivo tumor growth.In addition,PKM-deletion induced a reverse Warburg effect in tumorassociated stromal cells.Conclusions:PKM2 plays a critical role in promoting reductive glutamine metabolism and maintaining proton homeostasis.This study is helpful to increase the understanding of the physiological role of PKM2 in cancer cells. | Miao Liu Yuanyuan Wang Yuxia Ruan Changsen Bai Li Qiu Yanfen Cui Guoguang Ying Binghui Li | 2018 | Cancer Biology & Medicine2018,15,4: | 6 |
| 3 | Changes in soil microbial community response to precipitation events in a semi-arid steppe of the Xilin River Basin, China显示文摘In the context of climate change, precipitation is predicted to become more intense at the global scale. Such change may alter soil microbial communities and the microbially mediated carbon and nitrogen dynamics. In this study, we experimentally repackaged precipitation patterns during the growing season(from June to September) of 2012 in a semi-arid temperate steppe of the Xilin River Basin in Inner Mongolia of China, based on the 60-year growing season precipitation data. Specifically, a total amount of 240 mm simulated precipitation was assigned to experimental plots by taking the following treatments:(1) P6(6 extreme precipitation events, near the 1^(st) percentile);(2) P10(10 extreme precipitation events, near the 5^(th) percentile);(3) P16(16 moderate precipitation events, near the 50^(th) percentile); and(4) P24(24 events, 60-year average precipitation, near the 50^(th) percentile). At the end of the growing season, we analyzed soil microbial community structure and biomass, bacterial abundance, fungal abundance and bacterial composition, by using phospholipid fatty acid(PLFA), real-time quantitative polymerase chain reaction(RT-qPCR) and 16S rRNA gene clone library methods. The extreme precipitation events did not change soil microbial community structure(represented by the ratio of PLFA concentration in fungi to PLFA concentration in bacteria, and the ratio of PLFA concentration in gram-positive bacterial biomass to PLFA concentration in gram-negative bacterial biomass). However, the extreme precipitation events significantly increased soil microbial activity(represented by soil microbial biomass nitrogen and soil bacterial 16S rRNA gene copy numbers). Soil fungal community showed no significant response to precipitation events. According to the redundancy analysis, both soil microbial biomass nitrogen and soil ammonium nitrogen(NH_4-N) were found to be significant in shaping soil microbial community. Acidobacteria, Actinobacteria and Proteobacteria were the dominant phyla in soil bacterial composition, and responded differently to the extreme precipitation events. | 12 | Risk factors for intracerebral hemorrhage and coronary heart diseases over a 3-year period in patients admitted to some hospitals of Tongliao city of Nei Monggol Autonomous Region Retrospective analysis on case data显示文摘BACKGROUND: Intracerebral hemorrhage(ICH) and coronary heart disease (CHD) have the same pathological base, atherosclerosis, and the similar risk factors,such as smoking ,drinking, hypertension, hyperlipemia, diabetes mellitus, etc; but the distributions of two diseases are very different in the populations. This may be related to the exposure of risk factors and different effects of risk factors on two diseases. OBJECTIVE: To analyze the distribution difference of risk factors for ICH and CHD in the populations of Tongliao city of Nei Monggol Autonomous Region. DESIGN: Retrospective analysis. SETTING: School of Radiation Medicine and Public Health, Soochow University; Tongliao Hospital, Nei Monggol Autonomous Region. PARTICIPANTS: Random sampling was used to select 6 hospitals from 10 hospitals affiliated to Tongliao City of Nei Monggol Autonomous Region. Totally 1 672 medical records of patients with ICH and 2 195 medical records of patients with CHD admitted to Department of Neurology and Department of Cardiovascular Internal Medicine of above-mentioned 6 hospitals between January 2003 and December 2005 were collected according to the investigation need. METHODS: The subjects, whose medical records were involved, were performed retrospective analysis with pre-prepared questionnaire 'Stroke and Coronary Heart Disease Epidemiologic Questionnaire'. The main contents included: ①Social demography condition: The distributions of gender, age, nationality, etc. ②Previous history of disease: hypertension, diabetes mellitus, etc. ③Related risk factors: systolic blood pressure, diastolic blood pressure, total cholesterol, triglyceride, high-density lipoprotein cholesterol, low-density lipoprotein cholesterol, smoking, drinking and glucose (GLU). The database of Epidata was transformed to SPSS database. Single-and multiple-factor non-conditional Logistic regression analysis were performed on the data, and OR value and 95% CI were calculated. The distribution differences of risk factors for two diseases were compared. MAIN OUTCOME MEASURES: Single- and multi-factor non-conditional Logistic regression analysis results of each factor of patients. RESULTS: Single-factor non-conditional Logistic regression analysis showed that statistical significance existed in gender, age, nationality, smoking, drinking, history of hypertension, history of diabetes mellitus, hypertension, triglyceride (TG), and GLU ten factors(OR =0.199, OR 95% CI 0.142-0.280 to OR =7.484, OR 95% CI 6.186-9.054, P < 0.01). ②The results of multiple-factor non-conditional Logistic regression analysis showed 8 factors including age, gender, smoking, hypertension, history of hypertension, history of diabetes mellitus, GLU and TG(OR =0.203, OR 95% CI 0.114-0.361 to OR =8.262,OR 95% CI 5.466- 12.491, P < 0.01). CONCLUSION: ICH and CHD are the diseases induced by various risk factors. | 15 | Synthesis of carbon dots with a tunable photoluminescence and their applications for the detection of acetone and hydrogen peroxide显示文摘Carbon dots(CDs) with multi-color emissive properties and a high photoluminescent quantum yield(PLQY) have attracted great attention recently due to their potential applications in chemical,environmental,biological and photo-electronic fields.Solvent-dependent effect in photoluminescence provides a facial and effective approach to tune the emission of CDs.In this study,green emissive nitrogen-doped carbon dots(N-CDs) are synthesized from p-hydroquinone and ethylenediamine through a simple hydrothermal method.The as-prepared N-CDs possess a robust excitation-independent green luminescence and a high PLQY of up to 15.9%.Further spectroscopic characterization indicates that the high PLQY is achieved by the balance of nitrogen doping states and the surface passivation extent in CDs.The N-CDs also exhibit solvent-dependent multi-color emissive property and distinct PLQY in different solvents(the maximum can reach up to 25.3%).Furthermore,the as-prepared N-CDs are applied as fluorescence probes to detect acetone and H2O2 in water.This method has exhibited a low detection limit of acetone(less than 0.1 %) and a quick and linear response to the H_2O_2 with the concentration from 0 to 120 μmol/L.This work broadens the knowledge of applying CDs as probes in the bio and chemical sensing fields. | Shitong Cui Yanfen Wu Yao Liu Qingwen Guan Yida Zhang Yilin Zhang Song Luo Meng Xu Juncheng Wang | 2020 | Chinese Chemical Letters2020,31,2: | 1 |

| 18 | Water resource conservation promotes synergy between economy and environment in China’s northern drylands显示文摘Water resource availability is the major limiting factor for sustainable development in drylands.Climate change intensifies the conflicting water demands between people and the environment and highlights the importance of effective water resource management for achieving a balance between economic development and environmental protection.In 2008,Inner Mongolia,typical dryland in northern China,proposed strict regulations on water exploitation and utilization aimed at achieving sustainable development.Our study is the first to investigate the effectiveness and performance of these long-standing water conservation regulations.Our analyses found that the regulations drove industrial transformation,evidenced by the decreasing proportion of environmentally harmful industries such as coal and steel,and the increasing proportion of tertiary industries(especially tourism).Following industrial transformation,economic development decoupled from industrial water consumption and subsequently led to reduced negative environmental impacts.Based on these results,adaptive strategies were developed for 12 cities by revealing and integrating their development pathways and relative status in achieving sustainable development.Integration and cooperation between cities were proposed,e.g.,a water trade agreement between eastern Inner Mongolia(an economically underdeveloped region with relatively abundant water resources)and central Inner Mongolia(an economically developed region with high water stress).Such an agreement may enable the holistic achievement of sustainable development across regions.By integrating the findings of our research,our study presents a reproducible framework for water-management-based sustainable development strategies in drylands. | Yali Liu Jianqing Du Boyang Ding Yuexian Liu Wenjun Liu Anquan Xia Ran Huo Qinwei Ran Yanbin Hao Xiaoyong Cui Yanfen Wang | 2022 | Frontiers of Environmental Science & Engineering2022,16,3: | 0 |
