| 1 | SHP2 promotes proliferation of breast cancer cells through regulating Cyclin D1 stabilityviathe PI3K/AKT/GSK3β signaling pathway显示文摘Objective:The tyrosine phosphatase SHP2 has a dual role in cancer initiation and progression in a tissue type-dependent manner.Several studies have linked SHP2 to the aggressive behavior of breast cancer cells and poorer outcomes in people with cancer.Nevertheless,the mechanistic details of how SHP2 promotes breast cancer progression remain largely undefined.Methods:The relationship between SHP2 expression and the prognosis of patients with breast cancer was investigated by using the TCGA and GEO databases.The expression of SHP2 in breast cancer tissues was analyzed by immunohistochemistry.CRISPR/Cas9 technology was used to generate SHP2-knockout breast cancer cells.Cell-counting kit-8,colony formation,cell cycle,and EdU incorporation assays,as well as a tumor xenograft model were used to examine the function of SHP2 in breast cancer proliferation.Quantitative RT-PCR,western blotting,immunofluorescence staining,and ubiquitination assays were used to explore the molecular mechanism through which SHP2 regulates breast cancer proliferation.Results:High SHP2 expression is correlated with poor prognosis in patients with breast cancer.SHP2 is required for the proliferation of breast cancer cellsin vitro and tumor growthin vivo through regulation of Cyclin D1 abundance,thereby accelerating cell cycle progression.Notably,SHP2 modulates the ubiquitin–proteasome-dependent degradation of Cyclin D1viathe PI3K/AKT/GSK3βsignaling pathway.SHP2 knockout attenuates the activation of PI3K/AKT signaling and causes the dephosphorylation and resultant activation of GSK3β.GSK3βthen mediates phosphorylation of Cyclin D1 at threonine 286,thereby promoting the translocation of Cyclin D1 from the nucleus to the cytoplasm and facilitating Cyclin D1 degradation through the ubiquitin–proteasome system.Conclusions:Our study uncovered the mechanism through which SHP2 regulates breast cancer proliferation.SHP2 may therefore potentially serve as a therapeutic target for breast cancer. | Yue Yuan Yanling Fan Zicong Gao Xuan Sun He Zhang Zhiyong Wang Yanfen Cui Weijie Song Zhaosong Wang Fei Zhang Ruifang Niu | 2020 | Cancer Biology & Medicine2020,17,3: | 11 |
| 2 | Sirt1-ROS-TRAF6 Signaling-Induced Pyroptosis Contributes to Early Injury in Ischemic Mice显示文摘Stroke is an acute cerebro-vascular disease with high incidence and poor prognosis,most commonly ischemic in nature.In recent years,increasing attention has been paid to inflammatory reactions as symptoms of a stroke.However,the role of inflammation in stroke and its underlying mechanisms require exploration.In this study,we evaluated the inflammatory reactions induced by acute ischemia and found that pyroptosis occurred after acute ischemia both in vivo and in vitro,as determined by interleukin-1β,apoptosis-associated speck-like protein,and caspase-1.The early inflammation resulted in irreversible ischemic injury,indicating that it deserves thorough investigation.Meanwhile,acute ischemia decreased the Sirtuin 1(Sirtl)protein levels,and increased the TRAF6(TNF receptor associated factor 6)protein and reactive oxygen species(ROS)levels.In further exploration,both Sirtl suppression and TRAF6 activation were found to contribute to this pyroptosis.Reduced Sirtl levels were responsible for the production of ROS and increased TRAF6 protein levels after ischemic exposure.Moreover,N-acetyl-L-cysteine,an ROS scavenger,suppressed the TRAF6 accumulation induced by oxygen-glucose deprivation via suppression of ROS bursts.These phenomena indicate that Sirtl is upstream of ROS,and ROS bursts result in increased TRAF6 levels.Further,the activation of Sirtl during the period of ischemia reduced ischemiainduced injury after 72 h of reperfusion in mice with middle cerebral artery occlusion.In sum,these results indicate that pyroptosis-dependent machinery contributes to the neural injury during acute ischemia via the Sirt1-ROS-TRAF6 signaling pathway.We propose that inflammatory reactions occur soon after oxidative stress and are detrimental to neuronal survival;this provides a promising therapeutic target against ischemic injuries such as a stroke. | Weijie Yan Wei Sun Jiahui Fan Haiqing Wang Song Han Junfa Li Yanling Yin | 2020 | Neuroscience Bulletin2020,36,8: | 9 |
| 3 | An economic assessment of the health effects and crop yield losses caused by air pollution in China's Mainland显示文摘Air pollution is severe in China, and pollutants such as PM_(2.5) and surface O_3 may cause major damage to human health and crops, respectively. Few studies have considered the health effects of PM_(2.5) or the loss of crop yields due to surface O_3 using model-simulated air pollution data in China. We used gridded outputs from the WRF-Chem model, high resolution population data, and crop yield data to evaluate the effects on human health and crop yield in China's Mainland. Our results showed that outdoor PM_(2.5) pollution was responsible for 1.70–1.99 million cases of all-cause mortality in 2006. The economic costs of these health effects were estimated to be 151.1–176.9 billion USD, of which 90% were attributed to mortality. The estimated crop yield losses for wheat, rice, maize, and soybean were approximately 9, 4.6, 0.44, and 0.34 million tons, respectively, resulting in economic losses of 3.4 billion USD. The total economic losses due to ambient air pollution were estimated to be 154.5–180.3 billion USD, accounting for approximately 5.7%–6.6% of the total GDP of China in 2006. Our results show that both population health and staple crop yields in China have been significantly affected by exposure to air pollution. | 6 | Efficacy of platinum in advanced triple-negative breast cancer with germline BRCA mutation determined by next generation sequencing显示文摘Objective:To compare the efficacy of platinum-and non-platinum-based regimens as first-line treatment for advanced triple-negative breast cancer(TNBC)and analyze the relationship between their efficacy and BRCA gene status.Methods:Retrospectively analyze clinical data of 220 patients diagnosed pathologically with advanced TNBC and treated at the Department of Breast Oncology,Peking University Cancer Hospital from 2013 to 2018 and evaluate the efficacy of chemotherapy.A total of 114 patients had BRCA1/2 gene tested by next generation sequencing(NGS)using peripheral blood,and we analyzed the correlation between their efficacy and BRCA1/2 gene status.Results:Non-platinum-based chemotherapy(NPCT)was administered to 129 and platinum-based chemotherapy(PBCT)to 91 study patients.The clinical benefit rate(CBR)and median progression-free survival(PFS)were not statistically different between NPCT and PBCT groups.The median overall survival(OS)was 30.0 and 22.5 months for PBCT and NPCT group,respectively[P=0.090,hazard ratios(HR)=0.703].BRCA status was assessed in 114 patients,14 of whom had deleterious germline BRCA1/2(g BRCA)mutations(seven in each group).In PBCT group,the CBR was 85.7%and 35.1%for patients with and without deleterious g BRCA mutations,respectively(P=0.039).The median PFS were 14.9 and 5.3 months and median OS were 26.5 and 15.5 months for patients with and without deleterious g BRCA mutations,respectively(P=0.001,P=0.161,respectively).Patients in PBCT group had significantly greater rates of grade 3-4 anemia(5.5%vs.0%)and thrombocytopenia(8.8%vs.0%),whereas palmar-plantar erythrodysesthesia(12.4%vs.0%)and peripheral neuropathy(8.6%vs.1.1%)occurred more frequently in NPCT group.Conclusions:Platinum-based regimens are more effective in patients with deleterious g BRCA mutations,but no difference in patients without BRCA gene mutations,so non-platinum is an option in patients without BRCA gene mutations considering the toxicity and side effect.And we recommend that patients with advanced TNBC should have BRCA gene test. | Nan Wang Kun Li Wenfa Huang Weiyao Kong Xiaoran Liu Weijie Shi Feng Xie Hanfang Jiang Guohong Song Lijun Di Quanren Wang Jianjun Yu Huiping Li | 2020 | Chinese Journal of Cancer Research2020,32,2: | 7 |
| 7 | Controlled release of hydrogen by implantation of magnesium induces P53-mediated tumor cells apoptosis显示文摘Hydrogen has been used to suppress tumor growth with considerable efficacy.Inhalation of hydrogen gas and oral ingestion of hydrogen-rich saline are two common systemic routes of hydrogen administration.We have developed a topical delivery method of hydrogen at targeted sites through the degradation of magnesium-based biomaterials.However,the underlying mechanism of hydrogen’s role in cancer treatment remains ambiguous.Here,we investigate the mechanism of tumor cell apoptosis triggered by the hydrogen released from magnesium-based biomaterials.We find that the localized release of hydrogen increases the expression level of P53 tumor suppressor proteins,as demonstrated by the in vitro RNA sequencing and protein expression analysis.Then,the P53 proteins disrupt the membrane potential of mitochondria,activate autophagy,suppress the reactive oxygen species in cancer cells,and finally result in tumor suppression.The anti-tumor efficacy of magnesium-based biomaterials is further validated in vivo by inserting magnesium wire into the subcutaneous tumor in a mouse.We also discovered that the minimal hydrogen concentration from magnesium wires to trigger substantial tumor apoptosis is 91.2μL/mm^(3)per day,which is much lower than that required for hydrogen inhalation.Taken together,these findings reveal the release of H2 from magnesium-based biomaterial exerts its anti-tumoral activity by activating the P53-mediated lysosome-mitochondria apoptosis signaling pathway,which strengthens the therapeutic potential of this biomaterial as localized anti-tumor treatment. | Rui Zan Hao Wang Weijie Cai Jiahua Ni Berengere J.C.Luthringer-Feyerabend Wenhui Wang Hongzhou Peng Weiping Ji Jun Yan Jiazeng Xia Yang Song Xiaonong Zhang | 2022 | Bioactive Materials2022,7,3: | 3 |

| 9 | Using SSR Marker to Trace Chinese Shrimp Fenneropenaeus chinensis Released in Natural Sea-A Feasible Strategy for Assessment of Release Effect in Natural Resources Recovery Program显示文摘Enhancement release has been proven effective in natural resources recovery of Chinese shrimp Fenneropenaeus chinensis in the last several decades in China, however, to assess the effectiveness of enhancement release, we still need to develop a high-efficient and easy-operational method to replace those physical-tagging release method with labor intensive, size-and number-limited. In the present study, single(with maternal known) parentage identification using eight simple sequence repeat(SSR) markers genotype fingerprint was used to trace Chinese shrimp released in Bohai Bay in 2013. A total of 884 shrimp spawners were collected from two hatcheries in Tianjin City respectively after enhancement release of shrimp larvae in May 2013. A total of 844 shrimp samples were recaptured around the release location approximately 4 months after the shrimp larvae were released into the natural sea. Genotype data of 8 SSR loci of the 1,726 samples were used for maternal-offspring parentage identification using CEervus 3.0 software. The allele number in each locus ranged from 8 to 68 with an average value of 33.25, which produced the cumulative exclusion probability with one parent known of all these sight loci up to 99%. Among the 844 recaptured shrimp samples, 448(♂:♀=212:232, gender information was lost for 4 samples) were successfully traced to their 337 maternal parents using a logarithm of odds(LOD) > 3.0 threshold. Among these 337 maternal parents, 253 had a single offspring, 62 had two offspring, 18 had three offspring, 3 had four offspring, and 1 had five offspring. For the first time, a large number of released shrimp were identified from recapture samples, and this study showed that it is possible to trace all released Chinese shrimp without using any type of physical tag in enhancement release activities. | 14 | Preparation of AZO Nanoparticles, Ceramic Targets and Thin Films by a Co-precipitaition Method显示文摘We comprehensively study the co-precipitation preparation of aluminum doped zinc oxide(AZO) nanoparticles, ceramic target and thin fi lm deposition. The nanoparticles calcined below 700 ℃ possessed pure wurtzite structure of ZnO. When the calcination temperature exceeded 700 ℃, ZnAl_2O_4 phase appeared. The resistivity and relative density of the AZO target pressed from nanoparticles were 3×10^(-3) Ω?cm and 99.1%, respectively. The minimum resistivity of AZO thin films prepared by DC sputtering of the ceramic target reached 4.1×10^(–4) Ω?cm with the mobility of 33 cm^2/v?s and the carrier concentration of 4.5 ×10^(20) cm^(-3). | 16 | Effective Surface Treatment for High‑Performance Inverted CsPbI2Br Perovskite Solar Cells with Efficiency of 15.92%显示文摘Developing high-efficiency and stable inverted CsPbI2Br perovskite solar cells is vitally urgent for their unique advantages of removing adverse dopants and compatible process with tandem cells in comparison with the regular.However,relatively low opening circuit voltage(Voc)and limited moisture stability have lagged their progress far from the regular.Here,we propose an effective surface treatment strategy with high-temperature FABr treatment to address these issues.The induced ions exchange can not only adjust energy level,but also gift effective passivation.Meanwhile,the gradient distribution of FA+can accelerate the carriers transport to further suppress bulk recombination.Besides,the Br-rich surface and FA+substitution can isolate moisture erosions.As a result,the optimized devices show champion efficiency of 15.92%with Voc of 1.223 V.In addition,the tolerance of humidity and operation get significant promotion:maintaining 91.7%efficiency after aged at RH 20%ambient condition for 1300 h and 81.8%via maximum power point tracking at 45°C for 500 h in N2.Furthermore,the unpackaged devices realize the rare reported air operational stability and,respectively,remain almost efficiency(98.9%)after operated under RH 35%for 600 min and 91.2%under RH 50%for 300 min. | Sheng Fu Xiaodong Li Li Wan Wenxiao Zhang Weijie Song Junfeng Fang | 2020 | Nano-Micro Letters2020,12,12: | 2 |

| 18 | Relationships between body weight and other morphological traits in young sea cucumbers Apostichopus japonicas显示文摘Correlation, multiple regression, and path analyses were used to investigate the relationships between body weight and three other morphological traits in juvenile Japanese sea cucumbers Apostichopus japonicus . We measured live body weight (BW), body length (BL), numbers of papillae (NP), and numbers of tube feet (NF) at 60, 80, 100, and 130 days post-hatching (dph). We calculated path correlation coefficients, correlation indices ( R 2 ), and coefficients of determination with BW as the dependent variable and the other morphological traits as independent variables. The coefficient of variation for BW was high across all age groups, and all measured morphological traits were significantly correlated ( P < 0.01). BL had the greatest direct eff ect on BW across all age groups (60 dph, 0.526;80 dph, 0.404;100 dph, 0.620;and 130 dph, 0.681), while NF had the greatest indirect eff ect on BW across all age groups (60 dph, 0.528;80 dph, 0.452;100 dph, 0.666;and 130 dph, 0.603). Regression analyses between morphological traits and BW indicated that R 2 was greater than 0.85 only in the 100-dph specimens. The indirect eff ects of the other measured morphological traits on BW were age-dependent. The optimal regression equations, as determined with stepwise regression, were, for 60-dph specimens: BW 60 =10 (-3.04+0.092BL+0.014NP+0.014NF)( R 2 =0.632);for 80-dph specimens: BW 80 =10 (- 3.035+0.056BL+0.017NP+0.02NF)( R 2 =0.686);for 100-dph specimens: BW 100 =10 (- 3.742+0.069BL+0. 633 · lg (NP)+0. 464 · lg (NF))( R 2 =0.893);and for 130-dph specimens: BW 130 =10 (- 2.472+0.065BL+0.012NP)( R 2 =0.774). | 20 | Tracking the reactivity of ozonation towards effluent organic matters from WWTP using two-dimensional correlation spectra显示文摘The characteristics of effluent organic matter(EfOM) from a wastewater treatment plant(WWTP) during ozonation were investigated using excitation and emission matrix(EEM)spectra, Fourier transform infrared spectroscopy(FT-IR) and high-performance size exclusion chromatography(HPSEC) at different ozone dosages. The selectivity of ozonation towards different constituents and functional groups was analysed using two-dimensional correlation spectra(2D-COS) probed by FT-IR, synchronous fluorescence spectra and HPSEC.The results indicated that ozonation can destroy aromatic structures of EfOM and change its molecular weight distribution(MWD). According to 2D-COS analysis, microbial humiclike substances were preferentially removed, and then the protein-like fractions. Terrestrial humic-like components exhibited inactivity towards ozonation compared with the above two fractions. Protein-like substances with small molecular weight were preferentially reacted during ozonation based on 2D-COS probed by HPSEC. In addition, the selectivity of ozone towards different functional groups of EfOM exhibited the following sequence:phenolic and alcoholic C\O groups > aromatic structures containing C_C double bonds >aliphatic C\H.
