The R2R3-type MYB transcription factor MdMYB90-like is responsible for the enhanced skin color of an apple bud sport mutant显示文摘The anthocyanin content in apple skin determines its red coloration,as seen in a Fuji apple mutant.Comparative RNA-seq analysis was performed to determine differentially expressed genes at different fruit development stages between the wild-type and the skin color mutant.A novel R2R3-MYB transcription factor,MdMYB90-like,was uncovered as the key regulatory gene for enhanced coloration in the mutant. The expression of MdMYB90-like was 21.3 times higher in the mutant.MdMYB90-like regulates anthocyanin biosynthesis directly through the activation of anthocyanin biosynthesis genes and indirectly through the activation of other transcription factors that activate anthocyanin biosynthesis.MdMYB90-like bound to the promoters of both structural genes(MdCHS and MdUFGT)and other transcription factor genes(MdMYB1 and MdbHLH3)in the yeast one-hybrid system, electrophoretic mobility shift assay, and dual-luciferase assay.Transgenic analysis showed that MdMYB90-like was localized in the nucleus, and its overexpression induced the expression of other anthocyanin-related genes, including MdCHS,MdCHI,MdANS,MdUFGT,MdbHLH3,and MdMYB1.The mutant had reduced levels of DNA methylation in two regions(−1183 to−988 and−2018 to−1778)of the MdMYB90-like gene promoter, which might explain the enhanced expression of the gene and the increased anthocyanin content in the mutant apple skin. | Chao Sun Chunming Wang Wang Zhang Shuai Liu Weiyao Wang Xinyi Yu Tao Song Maxwell Yu Weichang Yu Shenchun Qu | 2021 | Horticulture Research2021,8,1: | 6
A high-Tc dc-SQUID-based single-channel MCG and its performance显示文摘A single-channel high-Tc dc-SQUID magnetometer and gradiometer have been developed to record the magnetic field component perpendicular to the human chest generated by heart-beat. Magnetocardiogram (MCG) measurements have been carried out inside a magnetically shielded room. By sequentially adjusting a non-magnetic patient table with 5 cm pitch in X and Y directions, the field signals on a rectangle grid 5×5 over the chest area were registered in real time trace point by point with a typical dwell time over 30 cardiocycles each. Utilizing standard electrocardiogram (ECG) recordings as timing reference measured simultaneously with the MCG signals, the MCG data were then averaged and combined to form magnetic field patterns every 10 ms or so. Both the current dipole, which is parallel to MCG measuring plane and produces the vertical magnetic field, and its depth were determined as a function of time in a standard way. We have compared the MCG of healthy hearts with that of a heart with right bundle | WANG Huiwu DING Hongsheng ZHANG Lihua HUANG Xuguang CHEN Weichang WANG Ziqiang FENG Ji LIU Yiping ZHAO Shiping REN Yufeng YU Hongwei XU Fengzhi WANG Xiaoyin CHEN Genghua YANG Qiansheng | 2002 | Chinese Science Bulletin2002,47,22: | 2

Fluorescent bovine serum albumin-silver nanoclusters loaded with paclitaxel can traverse the blood-brain barrier to inhibit the migration of glioma显示文摘Objective::Glioma is the most common and aggressive primary brain tumor. Here, we aimed to establish a nano-drug carrier system to traverse the blood-brain barrier for the treatment and inhibition of glioma migration.Methods::The synthesis of bovine serum albumin protected-silver nanoclusters (BSA-AgNCs) was performed using chemical reduction. The drug paclitaxel (PTX) can be loaded into BSA-AgNCs through electrostatic and hydrophobic interactions to formulate spherical BSA-AgNC-PTX nanoparticles (BSA-AgNC-PTX NPs). A glioma mouse model was established by injecting U251-GFP-Luc cells into the mouse striatum, and all surgical procedures were approved by the Animal Ethics Committee of Nanchang University (SYXK2019-0003) on December 29, 2019.Results::The BSA-AgNC-PTX NPs were able to efficiently pass through the blood-brain barrier, both in vitro and in vivo, to deliver the drug to the tumor site. The in vivo assessment of BSA-AgNC-PTX NPs in glioblastoma multiforme-bearing mice revealed the significant inhibition of tumor growth and migration, prolonging the survival of the mice.Conclusion::These results indicated that BSA-AgNCs might represent an ideal nanocarrier for the treatment of glioma and has the potential to be used in the treatment of a variety of central nervous system diseases. | Yu Liu Yu Liu Yini Zhang Weichang Xie Yalan Yang Kaigeng Hu Hongbo Xin Ping Hu Fanrong Ai | 2020 | Journal of Bio-X Research2020,3,4: | 0
Electrode structure enabling dendrite inhibition for high cycle stability quasi-solid-state lithium metal batteries显示文摘Lithium(Li)metal batteries(LMBs)are widely regarded as the ultimate choice for the next generation of high-energy–density batteries. However, the uncontrollable growth of Li dendrites formed by inhomogeneous deposition seriously hinders its commercialization.Although many studies have achieved significant results in inhibiting the formation of Li dendrites,it is still impossible to eradicate them completely. Therefore, regulating the deposition behavior, such as the growth direction of unevenly deposited Li,is preferable to unilaterally suppressing them in some cases. Here we report a structured anode that can confine the deposited Li within holes and tune it to become vertical-up/horizontal-centripetal mixed growth mode by optimizing the electric field/Li^(+)concentration gradient. The Li^(+) adsorbed by the poly(amic acid)(PAA)insulating layer coated on the anode surface can form the Li^(+)concentration gradient pointing to the center of the hole.Combined with the special electric field formed by the hole structure,it is favorable for the Li^(+)to move into the vertically arrayed holes and simultaneously deposit on the bottom and walls.Furthermore, both in-situ and ex-situ observations confirm that the growth mode is changed and the Li deposition morphology is denser, which can greatly delay capacity fading and prolong cycle life in both liquid and quasi-solid-state LMBs.All the results show that the novel anode provides a new perspective for deep research into solid-state LMBs. | Kaiming Wang Ao Yu Zhiyi Zhou Fei Shen Manni Li Liang Zhang Weichang Guo Yifei Chen Le Shi Xiaogang Han | 2023 | Journal of Energy Chemistry2023,,4: | 0
Fabrication and Wettability of ZnO Nanorod Array显示文摘ZnO nanorod arrays were prepared in an open system by using a simple aqueous solution method. Spindle-like, wimble-like, tower-like and hexagonal rod-like ZnO rods were obtained under different conditions. ZnO nanorod arrays with different morphology and size were studied by X-ray diffraction (XRD), scanning electron microscopy (SEM), and contact angle measurement (CAM). The size of ordered zinc oxide rods can be controlled by temperature of water bath, because this temperature can influences growing speeds in different crystal directions. Some additives, such as urea and thiourea, were introduced into reaction solution to improve quality of arrays. Surface character of ZnO nanorod arrays can be changed from hydrophilic to hydrophobic, which was proved to be dependence on size of air grooves on surface. | Meng Sun Yi Du Weichang Hao Huaizhe Xu Youxing Yu Tianmin Wang | 2009 | Journal of Materials Science & Technology2009,25,1: | 0
Plant latent defense response to microbial non-pathogenic factors antagonizes compatibility显示文摘Unlike microbe-associated molecular patterns(MAMPs)that are readily targeted by host immunity, microbial non-pathogenic factors(NPFs)appear negligible as they do not elicit defense.Little is known about whether and how NPFs may be monitored by hosts to control compatibility.Herein,a forward genetic screening isolated an Arabidopsis mutant with a loss of plant-rhizobacteria mutualism, leading to the disclosure of a plant latent defense response(LDR)to NPFs.The activation of LDR in the mutant, named rol1 for regulator of LDR 1,is triggered by several non-pathogenic volatile organic compounds and antagonizes plant compatibility with the beneficial bacterium Bacillus amyloliquefaciens GB03.The activation of LDR in rol1 is mediated through the prokaryotic pathway of chloroplastic lipid biosynthesis. The rol1 root microbiome showed a reduced proportion of the Bacillaceae family. We propose that, parallel to the forefront immunity to MAMPs,LDR to certain NPFs provides a hidden layer of defense for controlling compatibility with commensal or beneficial microbes. | Yu Yang Shenglan Chen Xiaoxuan Wu Li Peng Juan I.Vílchez Richa Kaushal Xiaomin Liu Sunil K.Singh Danxia He Fengtong Yuan Suhui Lv Rafael J.L.Morcillo Wei Wang Weichang Huang Mingguang Lei Jian-Kang Zhu Paul W.Paré Huiming Zhang | 2022 | National Science Review2022,9,8: | 0
