| 1 | One-year clinical study of NeuroR egen scaffold implantation following scar resection in complete chronic spinal cord injury patients显示文摘The objective of this clinical study was to assess the safety and feasibility of the collagen scaffold, Neuro Regen scaffold, one year after scar tissue resection and implantation. Scar tissue is a physical and chemical barrier that prevents neural regeneration. However, identification of scar tissue is still a major challenge. In this study, the nerve electrophysiology method was used to distinguish scar tissue from normal neural tissue, and then different lengths of scars ranging from 0.5–4.5 cm were surgically resected in five complete chronic spinal cord injury(SCI) patients. The NeuroR egen scaffold along with autologous bone marrow mononuclear cells(BMMCs), which have been proven to promote neural regeneration and SCI recovery in animal models, were transplanted into the gap in the spinal cord following scar tissue resection. No obvious adverse effects related to scar resection or Neuro Regen scaffold transplantation were observed immediately after surgery or at the 12-month follow-up. In addition, patients showed partially autonomic nervous function improvement, and the recovery of somatosensory evoked potentials(SSEP) from the lower limbs was also detected. | 2 | Human placenta-derived mesenchymal stem cells loaded on linear ordered collagen scaffold improves functional recovery after completely transected spinal cord injury in canine显示文摘Traumatic spinal cord injury(SCI) is a major challenge in the clinic. In this study, we sought to examine the synergistic effects of linear ordered collagen scaffold(LOCS) and human placenta-derived mesenchymal stem cells(hPMSCs) when transplanted into completely transected beagle dogs. After 36 weeks observation, we found that LOCS+hPMSCs implants promoted better hindlimb locomotor recovery than was observed in the non-treatment(control) group and LOCS group. Histological analysis showed that the regenerated tissue after treatment was well integrated with the host tissue, and dramatically reduced the volume of cystic and chondroitin sulfate proteoglycans(CSPGs) expression. Furthermore, the LOCS+hPMSCs group also showed more neuron-specific βIII-tubulin(Tuj-1)-and NeuN-positive neurons in the lesion area, as well as axonal regeneration, remyelination and synapse formation in the lesion site. Additionally, dogs in the LOCS+hPMSCs group experienced enhanced sprouting of both ascending(CGRP-positive) sensory fibers and descending(5-HT-and TH-positive) motor fibers at the lesion area. All these data together suggested that the combined treatment had beneficial effects on neuronal regeneration and functional improvement in a canine complete transection model. | 3 | Properties of Supersulphated Phosphogysum-slag Cement (SSC) Concrete显示文摘Supersulphated phosphogysum-slag cement(SSC) is a newly developed non-burned cementitious material mainly composed of phosphogysum(PG) and ground granulated blast furnace slag(GGBFS), with small amount of steel slag(SS) and clinker(CL). SSC is a kind of environmentally-friendly cementitious material due to its energy-saving, low-carbon emission, and waste-utilization. We prepared concretes with supersulphated phosphogysum-slag cement, and studied the mechanical properties, microproperties and resistance to chloride penetration of concrete in comparison with those of portland slag cement(PSC) and ordinary portland cement(OPC) concrete. The test results show that the compressive strength of SSC concrete can reach 38.6 MPa after 28 d, close to PSC concrete and OPC concrete. Microanalyses indicate that large quantities of ettringite and C-S-H, and little amount of Ca(OH)2 are generated during the hydration of SSC. The dense cement paste structure of SSC is formed by ettringite and C-S-H, surrounded unreacted phosphogysum. | 5 | The Arabidopsis phosphatase PP2C49 negatively regulates salt tolerance through inhibition of At HKT1;1显示文摘Type 2C protein phosphatases(PP2Cs)are the largest protein phosphatase family.PP2Cs dephosphorylate substrates for signaling in Arabidopsis,but the functions of most PP2 Cs remain unknown.Here,we characterized PP2 C49(AT3G62260,a Group G PP2C),which regulates Na^(+)distribution under salt stress and is localized to the cytoplasm and nucleus.PP2C49 was highly expressed in root vascular tissues and its disruption enhanced plant tolerance to salt stress.Compared with wild type,the pp2c49 mutant contained more Na^(+)in roots but less Na^(+)in shoots and xylem sap,suggesting that PP2C49 regulates shoot Na^(+)extrusion.Reciprocal grafting revealed a root-based mechanism underlying the salt tolerance of pp2 c49.Systemic Na+distribution largely depends on AtHKT1;1 and loss of function of AtHKT1;1 in the pp2c49 background overrode the salt tolerance of pp2c49,resulting in salt sensitivity.Furthermore,compared with plants overexpressing PP2C49 in the wild-type background,plants overexpressing PP2C49 in the athtk1;1 mutant background were sensitive to salt,like the athtk1;1 mutants.Moreover,protein-protein interaction and two-voltage clamping assays demonstrated that PP2C49 physically interacts with AtHKT1;1 and inhibits the Na^(+)permeability of AtHKT1;1.This study reveals that PP2C49 negatively regulates AtHKT1;1 activity and thus determines systemic Na^(+)allocation during salt stress. | Moli Chu Pengwang Chen Sufang Meng Peng Xu Wenzhi Lan | 2021 | Journal of Integrative Plant Biology2021,63,3: | 5 |

| 7 | A preliminary investigation on genetic diversity of Sousa chinensis in the Pearl River Estuary and Xiamen of Chinese waters显示文摘In this study, the mitochondrial DNA (mtDNA) control region and the mitochondrial cytochrome b gene of stranded Indo-Pacific humpback dolphin (Sousa chinensis) samples from the Pearl River Estuary and Xiamen waters were sequenced and analyzed. The result of mtDNA control region revealed 34 variable sites and four unique haplotypes (named as A, B, C and D) identified among the total samples from these two water areas, and the most common haplotype (A) was shared by 75% of the dolphins sampled from the two water areas. The haplotypic diversity (h) was 0.455 and the nucleotide diversity (π) was 0.0088. The phylogenetic analysis showed that the hap- lotype A, C, and D were closely related, but the haplotype B (unique for XM01 from Xiamen) was far from the other three. By scanning cytochrome b fragments, two haplotypes (A and B) were identified in these two water areas, and the most common haplotype (A) was shared by 91.67% individuals, while XM01 from Xiamen as the only exception. | 8 | Antrodia cinnamomea exerts an anti-hepatoma effect by targeting PI3K/AKT-mediated cell cycle progression in vitro and in vivo显示文摘Antrodia cinnamomea is extensively used as a traditional medicine to prevention and treatment of liver cancer.However,its comprehensive chemical fingerprint is uncertain,and the mechanisms,especially the potential therapeutic target for anti-hepatocellular carcinoma(HCC)are still unclear.Using UPLC-Q-TOF/MS,139 chemical components were identified in A.cinnamomea dropping pills(ACDPs).Based on these chemical components,network pharmacology demonstrated that the targets of active components were significantly enriched in the pathways in cancer,which were closely related with cell proliferation regulation.Next,HCC data was downloaded from Gene Expression Omnibus database(GEO).The Cancer Genome Atlas(TCGA)and Dis Ge NET were analyzed by bioinformatics,and 79 biomarkers were obtained.Furtherly,nine targets of ACDP active components were revealed,and they were significantly enriched in PI3 K/AKT and cell cycle signaling pathways.The affinity between these targets and their corresponding active ingredients was predicted by molecular docking.Finally,in vivo and in vitro experiments showed that ACDPs could reduce the activity of PI3 K/AKT signaling pathway and downregulate the expression of cell cycle-related proteins,contributing to the decreased growth of liver cancer.Altogether,PI3 K/AKT-cell cycle appears as the significant central node in anti-liver cancer of A.Cinnamomea. | Yan Zhang Pin Lv Junmei Ma Ning Chen Huishan Guo Yan Chen Xiaoruo Gan Rong Wang Xuqiang Liu Sufang Fan Bin Cong Wenyi Kang | 2022 | Acta Pharmaceutica Sinica B2022,12,2: | 2 |

| 13 | In vitro Transcriptome Analysis of Two Chinese Isolates of Streptococcus suis Serotype 2显示文摘The Streptococcus suis serotype 2(S. suis 2) isolates 05ZYH33 and 98HAH33 have caused severe human infections in China. Using a strand-specific RNA-seq analysis, we compared the in vitro transcriptomes of these two Chinese isolates with that of a reference strain(P1/7). In the89 K genomic island that is specific to these Chinese isolates, a toxin–antitoxin system showed relatively high levels of transcription among the S. suis. The known virulence factors with high transcriptional activity in these two highly-pathogenic strains are mainly involved in adhesion, biofilm formation, hemolysis and the synthesis and transport of the outer membrane protein. Furthermore,our analysis of novel transcripts identified over 50 protein-coding genes with one of them encoding a toxin protein. We also predicted over 30 small RNAs(s RNAs) in each strain, and most of them are involved in riboswitches. We found that six s RNA candidates that are related to bacterial virulence, including csp A and rli38, are specific to Chinese isolates. | 20 | Rapid-acting antidepressants targeting modulation of the glutamatergic system: clinical and preclinical evidence and mechanisms显示文摘Major depressive disorder(MDD)is a devastating mental illness that affects approximately 20%of the world's population.It is a major disease that leads to disability and suicide,causing a severe burden among communities.Currently available medications for treating MDD target the monoaminergic systems.The most prescribed medications include selective serotonin reuptake inhibitors and selective norepinephrine reuptake inhibitors.However,these medications have serious drawbacks,such as a delayed onset requiring weeks ormonths to reach efficacy and drug resistance,as one-third of patients are unresponsive to the medications.Therefore,it is imperative to develop novel therapies with rapid action,high efficacy and few adverse effects.The discovery of the rapid antidepressant effect of ketamine has triggered tremendous enthusiasm for studying new antidepressants that target the glutamatergic system in the central nervous system.Many agents that directly or indirectly modulate the glutamatergic system have been shown to provide rapid and lasting antidepressant action.Among these agents,ketamine,an antagonist of metabotropic glutamate 2/3 receptors,and scopolamine,an unspecific muscarinic acetylcholine receptor antagonist,have been extensively studied.In this review,we discuss the clinical and preclinical evidence supporting theantidepressant efficacy of these agents and the current understanding of the underlying mechanisms. | Shikai Wang Sufang Tang Jintao Huang Huanxin Chen | 2022 | General Psychiatry2022,35,6: | 0 |
