| 1 | Solid-pseudopapillary tumor of the pancreas: Clinical experience and literature review显示文摘AIM: To evaluate the clinical presentations of solidpseudopapillary tumor of the pancreas (SPT) and examine the diagnosis, treatment, low grade malignant potential of this rare disease.METHODS: We retrospectively reviewed a series of seven patients with SPT managed in our hospital between July 1990 and October 2003. Six females and one male with mean age of 31 years (range 13 to 50 years) were diagnosed with SPT at our institution.RESULTS: Clinical presentation included a palpable abdominal mass in two patients and vague abdominal discomfort in another two. Two patients were asymptomatic;their tumors were found incidentally on abdominal sonographic examination for other reasons. The final patient was admitted with hemoperitoneum secondary to tumor rupture. The mean diameter of the tumors in the seven patients was 10.5 cm (range 5 to 20 cm). The lesions were located in the body and tail in five cases and in the head of the pancreas in two. Surgical procedures included distal pancreatectomy (3), distal pancreatectomy with splenectomy (2), pancreaticoduodenectomy (1) and a pylorus-preserving Whipple procedure (1). There were gross adhesions or histological evidence of infiltration to the adjacent pancreas and/or splenic capsule in four cases. None of the patients received adjuvant therapy.The mean follow up was 7 years (range 0.5 to 14 years).One patient developed multiple liver metastases after 14 years of follow up.CONCLUSION: SPT is a rare tumor that behaves less aggressively than other pancreatic tumor. | 3 | SlMYB75,an MYB-type transcription factor,promotes anthocyanin accumulation and enhances volatile aroma production in tomato fruits显示文摘Genetic manipulation of genes to upregulate specific branches of metabolic pathways is a method that is commonly used to improve fruit quality.However,the use of a single gene to impact several metabolic pathways is difficult.Here,we show that overexpression of the single gene SlMYB75(SlMYB75-OE)is effective at improving multiple fruit quality traits.In these engineered fruits,the anthocyanin content reached 1.86mg g−1 fresh weight at the red-ripe stage,and these SlMYB75-OE tomatoes displayed a series of physiological changes,including delayed ripening and increased ethylene production.In addition to anthocyanin,the total contents of phenolics,flavonoids and soluble solids in SlMYB75-OE fruits were enhanced by 2.6,4,and 1.2 times,respectively,compared to those of wild-type(WT)fruits.Interestingly,a number of aroma volatiles,such as aldehyde,phenylpropanoid-derived and terpene volatiles,were significantly increased in SlMYB75-OE fruits,with some terpene volatiles showing more than 10 times higher levels than those in WT fruits.Consistent with the metabolic assessment,transcriptomic profiling indicated that the genes involved in the ethylene signaling,phenylpropanoid and isoprenoid pathways were greatly upregulated in SlMYB75-OE fruits.Yeast one-hybrid and transactivation assays revealed that SlMYB75 is able to directly bind to the MYBPLANT and MYBPZM cis-regulatory elements and to activate the promoters of the LOXC,AADC2 and TPS genes.The identification of SlMYB75 as a key regulator of fruit quality attributes through the transcriptional regulation of downstream genes involved in several metabolic pathways opens new avenues towards engineering fruits with a higher sensory and nutritional quality. | Wei Jian Haohao Cao Shu Yuan Yudong Liu Juanfang Lu Wang Lu Ning Li Jianhui Wang Jian Zou Ning Tang Chan Xu Yulin Cheng Yanqiang Gao Wanpeng Xi Mondher Bouzayen Zhengguo Li | 2019 | Horticulture Research2019,6,1: | 39 |

| 12 | Chemical screen identifies a geroprotective role of quercetin in premature aging显示文摘Aging increases the risk of various diseases. The main goal of aging research is to find therapies that attenuate aging and alleviate aging-related diseases. In this study, we screened a natural product library for geroprotective compounds using Werner syndrome (WS) human mesenchymal stem cells (hMSCs), a premature aging model that we recently established. Ten candidate compounds were identified and quercetin was investigated in detail due to its leading effects. Mechanistic studies revealed that quercetin alleviated senescence via the enhancement of cell proliferation and restoration of heterochromatin architecture in WS hMSCs. RNA-sequencing analysis revealed the transcriptional commonalities and differences in the geroprotective effects by quercetin and Vitamin C. Besides WS hMSCs, quercetin also attenuated cellular senescence in Hutchinson-Gilford progeria syndrome (HGPS) and physiological-aging hMSCs. | 14 | Application of optical coherence tomography angiography in assessment of posterior scleral reinforcement for pathologic myopia显示文摘AIM: To investigate the effect of posterior scleral reinforcement(PSR) on circulation of pathologic myopia eyes with posterior staphyloma by optical coherence tomography angiography(OCTA).·METHODS: The study included 30 pathologic myopia eyes with posterior staphyloma which underwent PSR(PSR group) for 6 to 18 mo ago, and 30 age and myopia matched eyes without PSR surgery as control group.Macular, choriocapillaris and radial peripapillary capillary(RPC) flow density were measured by OCTA, and the measurements were compared between groups.·RESULTS: OCTA found no significant differences in macular flow density between PSR and control groups.For the superficial flow, whole enface flow density(WED), fovea density(FD), and parafoveal density(PD)were 46. 55 % ± 5. 19 % vs 47. 29 % ± 4. 12 %( P = 0. 542),31.45%±6.35% vs 31.17%±4.48%(P =0.841), and 48.82%±5.66% vs 49.21% ±4.15%(P =0.756) in PSR and control groups, respectively. For the deep flow, WED, FD, and PD were 52.07% ±5.78% vs 53.95% ±4.62%(P =0.168),29.62%±6.55% vs 29.50%±6.38%(P=0.940), and 56.93%±6.17% vs 58.15% ±5.13%(P =0.407) in PSR and control groups, respectively. The choriocapillary flow density was 61.18 ±3.25% in PSR group vs 60.88% ±2.56% in control group(P=0.692). Also, OCTA found no significant differences in RPCs flow density between PSR and control groups. The optic disc WED, inside disc flow density and peripapillary flow density were 48.47%±4.77%vs 48.11%±4.57%(P =0.813), 45.47%±11.44% vs 46.68%±9.02%(P=0.709), 54.32%±5.29% vs 52.47%±6.62%(P =0.349) in PSR and control groups, respectively.·CONCLUSION: OCTA provides a non-invasive and quantitative approach for monitoring macular and papillary blood flow in pathologic myopia. Ganoderma lucidum (GL) has emerged as a novel age nt that protects neurons from oxidative stress. However, the detailed mechanisms underlying GL-induced neuroprotection have not been documented. In this study, we investigated the neuroprotective effects of GL extract (GLE) and the underlying mechanisms in the classic MPTP(1 -methyl-4-phenyl-1,2,3,6- tetrahydropyridine)-induced mouse model of PD. Mice were injected with MPTP to in duce parkins on ism. The n the mice were administered GLE (400 mg kg^-1 d^-1,ig) for 4 weeks. We observed that GLE administration significantly improved locomotor performanee and increased tyrosine hydroxylase expression in the substantia nigra pars compact (SNpc) of MPTP-treated mice. In in vitro study, treatment of neuroblastoma neuro-2a cells with 1 -methyl-4-phenyIpyridinium (MPP^+, 1 mmol/L) caused mitochondrial membrane potential collapse, radical oxygen species accumulation, and ATP depletion. Application of GLE (800 pg/mL) protected n euroblastoma neu ro-2a cells agai nst MPP in suit. Application of GLE also improved mitochondrial movement dysfunction in cultured primary mesencephalic neurons. In addition, GLE counteracted the decline in NIX (also called BNIP3L) expression and increase in the LC3-II/LC3-I ratio evoked by MPP I Moreover, GLE reactivated MPP^+-inhibited AMPK, mTOR, and ULK1. Similarly, GLE was sufficient to counteract MPP1 -induced inhibition of PINK1 and Parkin expression. GLE suppressed MPP^+-induced cytochrome C release and activation of caspase-3 and caspase-9. | 19 | Constraint-induced movement therapy enhances AMPA receptor-dependent synaptic plasticity in the ipsilateral hemisphere following ischemic stroke显示文摘Constraint-induced movement therapy(CIMT)can promote the recovery of motor function in injured upper limbs following stroke,which may be associated with upregulation ofα-amino-3-hydroxy-5-methyl-4-isoxazole propionic acid receptor(AMPAR)at synapses in the ipsilateral sensorimotor cortex in our previous study.However,AMPAR distribution is tightly regulated,and only AMPARs on the postsynaptic membrane can mediate synaptic transmission.We speculated that synaptic remodeling induced by movement-associated synaptic activity can promote functional recovery from stroke.To test this hypothesis,we compared AMPAR expression on the postsynaptic membrane surface in a rat model of ischemic stroke induced by middle cerebral artery occlusion(MCAO)with versus without CIMT,which consisted of daily running wheel training for 2 weeks starting on day 7 after MCAO.The results showed that CIMT increased the number of glutamate receptor(Glu R)2-containing functional synapses in the ipsilateral sensorimotor cortex,and reduced non-Glu R2 AMPARs in the ipsilateral sensorimotor cortex and hippocampal CA3 region.In addition,CIMT enhanced AMPAR expression on the surface of post-synaptic membrane in the ipsilateral sensorimotor cortex and hippocampus.Thus,CIMT promotes the recovery of motor function of injured upper limbs following stroke by enhancing AMPAR-mediated synaptic transmission in the ischemic hemisphere.These findings provide supporting evidence for the clinical value of CIMT for restoring limb movement in stroke patients.All experimental procedures and protocols were approved by the Department of Laboratory Animal Science of Fudan University,China(approval No.201802173 S)on March 3,2018. | Jian Hu Pei-Le Liu Yan Hua Bei-Yao Gao Yu-Yuan Wang Yu-Long Bai Chan Chen | 2021 | Neural Regeneration Research2021,16,2: | 9 |
