The vicK gene of Streptococcus mutans mediates its cariogenicity via exopolysaccharides metabolism显示文摘Streptococcus mutans(S.mutans)is generally regarded as a major contributor to dental caries because of its ability to synthesize extracellular polysaccharides(EPS)that aid in the formation of plaque biofilm. The VicRKX system of S.mutans plays an important role in biofilm formation. The aim of this study was to investigate the effects of vicK gene on specific characteristics of EPS in S.mutans biofilm. We constructed single-species biofilms formed by different mutants of vicK gene.Production and distribution of EPS were detected through atomic force microscopy, scanning electron microscopy and confocal laser scanning microscopy.Microcosmic structures of EPS were analyzed by gel permeation chromatography and gas chromatography-mass spectrometry.Cariogenicity of the vicK mutant was assessed in a specific pathogen-free rat model.Transcriptional levels of cariogenicityassociated genes were confirmed by quantitative real-time polymerase chain reaction. The results showed that deletion of vicK gene suppressed biofilm formation as well as EPS production, and EPS were synthesized mostly around the cells.Molecular weight and monosaccharide components underwent evident alterations.Biofilms formed in vivo were sparse and contributed a decreased degree of caries. Moreover, expressional levels of genes related to EPS synthesis were down-regulated,except for gtfB.Our report demonstrates that vicK gene enhances biofilm formation and subsequent caries development. And this may due to its regulations on EPS metabolism, like synthesis or microcosmic features of EPS.This study suggests that vicK gene and EPS can be considered as promising targets to modulate dental caries. | Yalan Deng Yingming Yang Bin Zhang Hong Chen Yangyu Lu Shirui Ren Lei Lei Tao Hu | 2021 | International Journal of Oral Science2021,13,4: | 2
Potential chronic liver toxicity in rats orally administered an ethanol extract of Huangqin(Radix Scutellariae Baicalensis)显示文摘OBJECTIVE: To investigate the potential chronic liver toxicity of oral administration of ethanol extract of Huangqin(Radix Scutellariae Baicalensis)(SBE) in Wistar rats.METHODS: SBE was administered to rats by gavage for 26 weeks, at doses of 300, 1250, or 2500 mg·kg-1·d-1 respectively. The rats were euthanized at the end of 13 and 26 weeks daily oral dosing and following 4 weeks of recovery time. The changes of hematology, urinary, blood biochemistry and histomorphology were examined at each time point and focus on liver function and histological changes.RESULTS: When SBE at a dose of up to 2500 mgkg-1 d-1 was fed to male and female rats for 2··6 weeks, the liver tissue showed some inflammatory change that predominated by leukocyte infiltrationbut returned to normal after withdrawal. In addition, high-dose SBE treatment of 26 weeks in rats, glucose, electrolyte and lipid levels also have some changes. In addition, there are no other functional or organic lesions related to SBE treatment.CONCLUSIONS: Long-term and high-dose SBE may cause liver damage, however, the structural damage of the liver can be restored after the ethanol extract stopping. SBE will be well-tolerated for long-term use as a drug or health food, but in order to ensure drug safety, liver function, and serum glucose, electrolyte and lipid levels should be monitored when using SBE long term. | Yi Yan Zhao Yong Li Chunying Zhang Yushi Bin Yang Yuan Yalan Pan Chen Wang Lianmei Liang Aihua | 2018 | Journal of Traditional Chinese Medicine2018,38,2: | 1
Structure-design and theoretical-calculation for ultrasmall Co_(3)O_(4) anchored into ionic liquid modified graphene as anode of flexible lithium-ion batteries显示文摘Cobalt oxide(Co_(3)O_(4))is currently suitable in energy storage applications because of its high capacity based on the conversion reaction mechanism. However, unmodified Co_(3)O_(4)suffers from distinctly inferior rate capability and poor cycling stability. On the basis of the aforementioned considerations and density functional theory(DFT)simulations, the three-dimensional hierarchical porous structure(HPS)ultrasmall Co_(3)O_(4)anchored into ionic liquid(IL)modified graphene oxide(GO)has been successfully prepared(ultrasmall/Co_(3)O_(4)-GA-IL).The ultrasmall/Co_(3)O_(4)-GA-IL consists of Co_(3)O_(4)co-assembled with IL modified GO to generate the HPS which can facilitate ion transfer channels through reduction of the electron and ion transportation path and transmission impedance. In addition,N-doping graphene can enhance the inherent electrical conductivity of Co_(3)O_(4),which is proved by the DFT calculations.By virtue of the novel superstructure, the ultrasmall/Co_(3)O_(4)-GA-IL electrode demonstrates a high reversible capacity of 1,304 mAh·g^(−1),an enhanced high-rate capability(715 mAh·g^(−1)at 5 C),and a capacity retention of 98.4%even after 500 cycles at 5 C rate, which corresponds to 0.0003%capacity loss per cycle.Pouch cells based on the cathode are further fabricated and demonstrate excellent mechanical and electrochemical properties under bent and folded state, highlighting the practical application of our deliberately designed electrode in wearable electronics. | Longda Cong Shichao Zhang Hengyao Zhu Weixin Chen Xueyan Huang Yalan Xing Jun Xia Puheng Yang Xia Lu | 2022 | Nano Research2022,15,3: | 1
Multiscale analysis of Asian Monsoon over the past 640 ka显示文摘The empirical mode decomposition(EMD) method is used to re-analyse the high-resolution and precisely-dated stalagmite record from Chinese caves over the past 640 ka. Results show that(1) the variation in the Asian Monsoon can be completely decomposed into ten quasiperiod oscillations, among which the precession and semiprecession band oscillations are the most prominent periodicities, with contribution rates of 31.1% and 30.7%, respectively;(2) the cross-spectrum analysis of the semiprecession component and bi-hemisphere insolation(BHI) are strongly correlated, indicating an amplified response of precipitation and temperature variability to the interhemispheric insolation in the low-latitude regions, thus further affecting the intensity of the Asian Monsoon;(3) on millennial timescales, obvious oscillations at the 5 ka and 1–2 ka bands roughly correspond to the classical Bond and Dansgaard-Oeschger(DO) cycles. Additionally, a strong correlation is found between the detrended stalagmite δ18 O records and Ca/Sr sequence from the North Atlantic(especially at the 5 ka band). This result means that the 5 ka cycle is characteristic of the glacial-interglacial cycle since the middle and late Pleistocene and may imply that climate change on the millennial timescale is the result of an interaction between global ice volume and insolation. | Yalan ZENG Shitao CHEN Shaohua YANG Yijia LIANG Yongjin WANG | 2019 | Science China Earth Sciences2019,62,5: | 1
Insights into Ti doping for stabilizing the Na_(2/3)Fe_(1/3)Mn_(2/3)O_(2)cathode in sodium ion battery显示文摘Iron-and manganese-based layered metal oxides,as cathodes for sodium ion batteries, have received widespread attention because of the low cost and high specific capacity. However, the Jahn-teller effect of Mn^(3+)ions and the resulted unstable structure usually lead to continuously capacity decay.Herein,Titanium(Ti)has been successfully doped into Na_(2/3)Fe_(2/3)Mn_(2/3)O_(2)to suppress the Jahn-Teller distortion and improve both cycling and rate performance of sodium ion batteries. In situ high-energy synchrotron X-ray diffraction study shows that Ti-doped compound(Na_(2/3)Fe_(1/3)Mn_(0.57)Ti_(0.1)O_(2))can maintain the single P2 phase without any phase transition during the whole charging/discharging process.Various electrochemical characterizations are also applied to explore the better kinetics of sodium ions transfer in the Na_(2/3)Fe_(1/3)Mn_(0.5)7 Ti_(0.1)O_(2).This work provides a comprehensive insight into the Ti-doping effects on the performance from both structural and electro kinetic perspectives. | Tingting Yang Yalan Huang Jian Zhang He Zhu Jincan Ren Tianyi Li Leighanne C.Gallington Si Lan Ligao Yang Qi Liu | 2022 | Journal of Energy Chemistry2022,31,10: | 1
Beneficial Effects of Celastrol on Immune Balance by Modulating Gut Microbiota in Experimental Ulcerative Colitis Mice显示文摘Ulcerative colitis(UC)is a chronic inflammatory bowel disease caused by many factors including colonic inflammation and microbiota dysbiosis.Previous studies have indicated that celastrol(CSR)has strong anti-inflammatory and immune-inhibitory effects. Here,we investigated the effects of CSR on colonic inflammation and mucosal immunity in an experimental colitis model, and addressed the mechanism by which CSR exerts the protective effects. We characterized the therapeutic effects and the potential mechanism of CSR on treating UC using histological staining, intestinal permeability assay, cytokine assay, flow cytometry, fecal microbiota transplantation(FMT),16S rRNA sequencing, untargeted metabolomics, and cell differentiation.CSR administration significantly ameliorated the dextran sodium sulfate(DSS)-induced colitis in mice, which was evidenced by the recovered body weight and colon length as well as the decreased disease activity index(DAI)score and intestinal permeability.Meanwhile,CSR down-regulated the production of pro-inflammatory cytokines and up-regulated the amount of anti-inflammatory mediators at both mRNA and protein levels, and improved the balances of Treg/Th1 and Treg/Th17 to maintain the colonic immune homeostasis.Notably, all the therapeutic effects were exerted in a gut microbiota-dependent manner.Furthermore,CSR treatment increased the gut microbiota diversity and changed the compositions of the gut microbiota and metabolites, which is probably associated with the gut microbiota-mediated protective effects. In conclusion, this study provides the strong evidence that CSR may be a promising therapeutic drug for UC. | Mingyue Li Weina Guo Yalan Dong Wenzhu Wang Chunxia Tian Zili Zhang Ting Yu Haifeng Zhou Yang Gui Kaming Xue Junyi Li Feng Jiang Alexey Sarapultsev Huafang Wang Ge Zhang Shanshan Luo Heng Fan Desheng Hu | 2022 | Genomics, Proteomics & Bioinformatics2022,20,2: | 1
Developmental stage-specific A-to-I editing pattern in the postnatal pineal gland of pigs(Sus scrofa)显示文摘Background: RNA editing is a widespread post-transcriptional modification mechanism in mammalian genomes.Although many editing sites have been identified in domestic pigs(Sus scrofa), little is known about the characteristics and dynamic regulation of RNA editing in the pineal gland(PG), a small neuroendocrine gland that synthesizes and secretes melatonin, which is primarily responsible to modulate sleep patterns. Results: This study analyzed the expression of adenosine-to-inosine(A-to-I) editing regulators and profiled the first dynamic A-to-I RNA editome during postnatal PG development. The results identified ADAR1 as the most abundantly expressed ADAR enzyme, which was down-regulated during postnatal PG development. Furthermore,47,284 high-confidence RNA editing sites were identified, the majority of which(93.6%) were of the canonical A-to-I editing type, followed by C-to-T editing. Analysis of its characteristics showed that the A-to-I editing sites mostly localized in SINE retrotransposons PRE-1/Pre0_SS. Moreover, a strong deficiency and preference for guanine nucleotides at positions of one base upstream or downstream were found, respectively. The overall editing level at the puberty stage was higher than at both infancy and adulthood stages. Additionally, genome-wide RNA editing was found to exhibit a dynamic stage-specific fashion(postnatally). Genes that underwent developmental changes in RNA editing were associated with catabolic processes as well as protein localization and transport functions, implying that RNA editing might be responsible for the molecular machineries of the postnatal developing PG.Remarkably, RNA editing in 3′-UTRs might regulate gene expression by influencing miRNA binding during PG development. Conclusions: This study profiles the first comprehensive developmental RNA editome in the pig PG, which contributes to the understanding of the importance of post-transcriptionally mediated regulation during mammalian postnatal PG development. Moreover, this study widely extends RNA editome resources in mammals. | Rong Zhou Wenye Yao Chundi Xie Leixia Zhang Yangli Pei Hua Li Zheng Feng Yalan Yang Kui Li | 2021 | Journal of Animal Science and Biotechnology2021,12,1: | 1
A reporter gene assay for determining the biological activity of therapeutic antibodies targeting TIGIT显示文摘T cell immunoglobulin and ITIM domain(TIGIT)is a novel immune checkpoint that has been considered as a target in cancer immunotherapy.Current available bioassays for measuring the biological activity of therapeutic antibodies targeting TIGIT are restricted to mechanistic investigations because donor primary T cells are highly variable. Here,we designed a reporter gene assay comprising two cell lines,namely,CHO-CD112-CD3 scFv,which stably expresses CD 112(PVRL2,nectin-2)and a membranebound anti-CD3 single-chain fragment variable(scFv)as the target cell, and Jurkat-NFAT-TIGIT,which stably expresses TIGIT as well as the nuclear factor of activated T-cells(NFAT)response element-controlled luciferase gene,as the effector cell. The anti-CD3 scFv situated on the target cells activates Jurkat-NFATTIGIT cells through binding and crosslinking CD3 molecules of the effector cell, whereas interactions between CD 112 and TIGIT prevent activation. The presence of anti-TIGIT mAbs disrupts their interaction, which in turn reverses the inactivation and luciferase expression.Optimization and validation studies have demonstrated that this assay is superior in terms of specificity, accuracy, linearity, and precision. In summary, this reliable and effective reporter gene assay may potentially be utilized in lot release control, stability assays, screening, and development of novel TIGIT-targeted therapeutic antibodies. | Zhihao Fu Hongchuan Liu Lan Wang Chuanfei Yu Yalan Yang Meiqing Feng Junzhi Wang | 2021 | Acta Pharmaceutica Sinica B2021,11,12: | 1
Regeneration of immunocompetent B lymphopoiesis from pluripotent stem cells guided by transcription factors显示文摘Regeneration of functional B lymphopoiesis from pluripotent stem cells(PSCs)is challenging, and reliable methods have not been developed. Here,we unveiled the guiding role of three essential factors,Lhx2,Hoxa9,and Runx1,the simultaneous expression of which preferentially drives B lineage fate commitment and in vivo B lymphopoiesis using PSCs as a cell source. In the presence of Lhx2,Hoxa9,and Runx1 expression,PSC-derived induced hematopoietic progenitors(iHPCs)immediately gave rise to pro/pre-B cells in recipient bone marrow, which were able to further differentiate into entire B cell lineages, including innate B-1a,B-1b,and marginal zone B cells,as well as adaptive follicular B cells. In particular, the regenerative B cells produced adaptive humoral immune responses, sustained antigen-specific antibody production, and formed immune memory in response to antigen challenges. The regenerative B cells showed natural B cell development patterns of immunoglobulin chain switching and hypermutation via cross-talk with host T follicular helper cells, which eventually formed T cell-dependent humoral responses. This study exhibits de novo evidence that B lymphopoiesis can be regenerated from PSCs via an HSC-independent approach, which provides insights into treating B cell-related deficiencies using PSCs as an unlimited cell resource. | Qi Zhang Bingyan Wu Qitong Weng Fangxiao Hu Yunqing Lin Chengxiang Xia Huan Peng Yao Wang Xiaofei Liu Lijuan Liu Jiapin Xiong Yang Geng Yalan Zhao Mengyun Zhang Juan Du Jinyong Wang | 2022 | Cellular & Molecular Immunology2022,19,4: | 0
Towards extreme fast charging of 4.6 V LiCoO_(2) via mitigating high-voltage kinetic hindrance显示文摘High-voltage LiCoO_(2)(LCO) is an attractive cathode for ultra-high energy density lithium-ion batteries(LIBs) in the 3 C markets. However, the sluggish lithium-ion diffusion at high voltage significantly hampers its rate capability.Herein, combining experiments with density functional theory(DFT) calculations,we demonstrate that the kinetic limitations can be mitigated by a facial Mg^(2+)+Gd^(3+)co-doping method. The as-prepared LCO shows significantly enhanced Li-ion diffusion mobility at high voltage, making more homogenous Li-ion de/intercalation at a high-rate charge/discharge process. The homogeneity enables the structural stability of LCO at a high-rate current density, inhibiting stress accumulation and irreversible phase transition. When used in combination with a Li metal anode, the doped LCO shows an extreme fast charging(XFC) capability, with a superior high capacity of 193.1 mAh g^(-1)even at the current density of 20 C and high-rate capacity retention of 91.3% after 100 cycles at 5 C.This work provides a new insight to prepare XFC high-voltage LCO cathode materials. | Yu Tang Jun Zhao He Zhu Jincan Ren Wei Wang Yongjin Fang Zhiyong Huang Zijia Yin Yalan Huang Binghao Zhang Tingting Yang Tianyi Li Leighanne CGallington Si Lan Yang Ren Qi Liu | 2023 | Journal of Energy Chemistry2023,,3: | 0
Effectiveness and safety of different amifostine regimens:Preliminary results of a phase Ⅱ multicenter randomized controlled trial显示文摘Objective:The radioprotective effects of amifostine remain uncertain in patients with nasopharyngeal carcinoma(NPC),and adverse effects and cost limit generalization of its classical everyday regimen. This phase II multicenter randomized controlled trial aimed to explore whether amifostine could ameliorate the toxicities of NPC patients in the era of intensity-modulated radiotherapy(IMRT),and to compare different regimens of amifostine on effectiveness and safety. Methods:Patients with stage I–IVB NPC were involved prospectively from January 1st,2013.All patients received radical treatment based on IMRT.After a randomization stratified by their stage, these patients were allocated into 3 groups:the group treated without amifostine, the group treated with the everyday regimen of amifostine, and the group treated with the every-other-day regimen. The 3 groups of patients were compared on radiotherapy-related acute toxicities, treatment effects of NPC,and amifostine-related complications. This trial was registered on the clinicaltrials.gov(ID:NCT01762514).Results:Until August 31st,2017,totally 187 patients completed experimental intervention.Only amifostine of everyday regimen appeared to reduce the patient proportion of mucositis(79.1%vs.96.8%,P=0.002).Hypocalcemia was less common in patients treated without amifostine than in those treated with amifostine(22.6%vs.53.4%vs.41.8%,P=0.002).Neither complete remission rates nor the survivals were affected by amifostine. Conclusions:Amifostine of everyday regimen could reduce mucositis in NPC patients who received IMRT,though it also had the possibility to cause more hypocalcemia. | Hui Chang Wei Yi Xiaohui Wang Yalan Tao Xin Yang Chen Chen Wenwen Zhang Shu Zhou Songran Liu Xiaohui Li Shirong Ding Jing Li Gong Li Xunfan Shao Yimin Liu Weishu Song Yunfei Xia | 2018 | Chinese Journal of Cancer Research2018,30,3: | 0
Intrinsic lithiophilic carbon host derived from bacterial cellulose nanofiber for dendrite-free and long-life lithium metal anode显示文摘Although lithium metal is considered a promising anode for advanced Li-S and Li-air batteries, the uncontrolled dendrite growth and infinite volume change impede its practical application.Herein,we report an ideal framework composed of carbonized bacterial cellulose(CBC)nanofibers, which shows intrinsic lithiophilicity to molten lithium without any lithiophilic surface modification. The wetting behavior of molten lithium can be significantly improved because its surface functional groups provide thermodynamical driving force, and the high surface roughness derived from nanocracks leads to rapid infusion in kinetics. The hybrid anode exhibits long cycle life up to 2000 h and excellent deep stripping-platting capacity up to 20 mAh·cm^(-2).When the anode is assembled with LiFePO_(4) cathode, the full cell delivers a good cycling stability up to 700 cycles. This is attributed to the intrinsic lithiophilic scaffold, which can not only lower the nucleation barrier of Li and provide uniform nucleation sites for stable Li stripping/plating,but also offer interspace to accommodate volume fluctuation of lithium during long cycling. This work provides a new manner to achieve a series of intrinsic lithiophilic carbon skeletons based on the large family of biomass materials and organic materials. | Gangyi Xiong Jiayu Yu Yalan Xing Puheng Yang Shichao Zhang | 2024 | Nano Research2024,17,5: | 0

Here, we aimed to establish a nano-drug carrier system to traverse the blood-brain barrier for the treatment and inhibition of glioma migration.Methods::The synthesis of bovine serum albumin protected-silver nanoclusters (BSA-AgNCs) was performed using chemical reduction. The drug paclitaxel (PTX) can be loaded into BSA-AgNCs through electrostatic and hydrophobic interactions to formulate spherical BSA-AgNC-PTX nanoparticles (BSA-AgNC-PTX NPs). A glioma mouse model was established by injecting U251-GFP-Luc cells into the mouse striatum, and all surgical procedures were approved by the Animal Ethics Committee of Nanchang University (SYXK2019-0003) on December 29, 2019.Results::The BSA-AgNC-PTX NPs were able to efficiently pass through the blood-brain barrier, both in vitro and in vivo, to deliver the drug to the tumor site. The in vivo assessment of BSA-AgNC-PTX NPs in glioblastoma multiforme-bearing mice revealed the significant inhibition of tumor growth and migration, prolonging the survival of the mice.Conclusion::These results indicated that BSA-AgNCs might represent an ideal nanocarrier for the treatment of glioma and has the potential to be used in the treatment of a variety of central nervous system diseases. | Yu Liu Yu Liu Yini Zhang Weichang Xie Yalan Yang Kaigeng Hu Hongbo Xin Ping Hu Fanrong Ai | 2020 | Journal of Bio-X Research2020,3,4: | 0 |
Relationship between groundwater cadmium and vicinity resident urine cadmium levels in the non-ferrous metal smelting area,China显示文摘Cadmium(Cd)has received widespread attention owing to its persistent toxicity and non-degradability.Cd in the human body is mainly absorbed from the external environment and is usually assessed using urinary Cd.Hunan Province is the heartland of the Chinese non-ferrous mining area, where several serious Cd pollution events have occurred, including high levels of Cd in the urine of residents. However, the environmental factors influencing high urinary Cd levels(UCLs)in nearby residents remain unclear.Therefore,211 nearby residents’UCLs and the corresponding sociological characteristics from nine groundwater samples in this area were analyzed using statistical analysis models.Groundwater Cd concentration ranged from 0.02 to 1.15μg/L,aligning with class III of the national standard;the range of UCL of nearby residents was 0.37–36.60μg/L,exceeding the national guideline of 0–2.5μg/L.Groundwater Cd levels were positively correlated with the UCL(P<0.001,correlation coefficient 95%CI=9.68,R^(2)=0.06).In addition, sociological characteristics, such as smoking status and education level, also affect UCL.All results indicate that local governments should strengthen the prevention and abatement of groundwater Cd pollution. This study is the first to systematically evaluate the relationship between groundwater Cd and UCL using internal and external environmental exposure data. These findings provide essential bases for relevant departments to reduce Cd exposure in regions where the heavy metal industry is globally prevalent. | Yujie Pan Yalan Li Hongxia Peng Yiping Yang Min Zeng Yang Xie Yao Lu Hong Yuan | 2023 | Frontiers of Environmental Science & Engineering2023,17,5: | 0
Development of a dual-fluorescence reporter system for high-throughput screening of L-aspartate-a-decarboxylase显示文摘β-Alanine (3-am inopropionic acid)holds great potential in industrial application. It can be obtained through a chemical synthesis route, which is hazardous to the environm ent.It is w ell known that L-aspartate-a-decarboxylase(ADC)can convert L-aspartate to β-alanine in bacteria. However, due to the low activity of ADC,industrial production of β-alanine through the green biological route remains unclear. Thus,im proving the activity of ADC is critical to reduce the cost of p-alanine production. In this study,we established a dual-fluorescence high-throughput system fo r efficient ADC screening.By measuring the am ount of β-alanine and the expression level of ADC using two different fluorescence markers,we can rapidly quantify the relative activity of ADC variants. From a mutagenesis library containing 2000 ADC variants,we obtained am utant w ith 33%increased activity.Further analysis revealed that m utations o f K43R and P103Q in ADC significantly improved the yield of β-alanine produced by the whole-cell biocatalysis.Compared with the previous singlefluorescence method, our system can not only quantify the amount of β-alanine but also measure the expression level of ADC with different fluorescence, making it able to effectively screen out ADC variants with im proved relative activity. The dual-fluorescence high-throughput system for rapid screening of ADC provides a good strategy fo r industrial production of β-alanine via the biological conversion route in the future. | Mingyue Fei Xudan Mao Yiyang Chen Yalan Lu Lin Wang Jie Yang Juanping Qiu Dongchang Sun | 2020 | Acta Biochimica et Biophysica Sinica2020,52,12: | 0
