| 1 | N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. | 2 | Effect of acupuncture at Renying(ST 9) on gene expression profile of hypothalamus in spontaneously hypertensive rats显示文摘OBJECTIVE: To investigate changes in gene expression profiles in the hypothalamus related to the effects of acupuncture at the Renying(ST 9) acupoint in spontaneously hypertensive(SH) rats.METHODS: We randomly divided 18 SH rats into Renying(ST 9) group and model control group, 9 body weight-matched Wistar-Kyoto rats were used as blank controls. Acupuncture was performed manually for 20-min daily over 28 d in the Renying(ST 9) group. Rat Gene 2.0 array technology was used for the determination of gene expression profiles and the screened key genes were verified by real-time quantitative polymerase chain reaction(RT-PCR) analyses.RESULTS: The different groups exhibited differential gene expression: compared with the blank control group, 48 genes were up-regulated and 91 genes were down-regulated in the model group;compared with the model group, 79 genes were up-regulated and 80 genes were down-regulated in Renying(ST 9) group. The RT-PCR results of the key genes including Chi3 l1, Ephx2, Klk1, 5-HT1 A and Cbs were consistent with that of gene chip analysis.CONCLUTION: Acupuncture at Renying(ST 9)could significantly lower the blood pressure of SH rats and affect their hypothalamic gene expression profile. | 4 | CS1003, a novel human and mouse cross-reactive PD-1 monoclonal antibody for cancer therapy显示文摘The programmed cell death protein 1(PD-1)is an immune-checkpoint that negatively regulates the immune system and a key mechanism that tumors utilize to escape from immune surveillance.PD-1 antibodies can block the interaction of PD-1 with its ligands(PD-L1 and PD-L2),restore T cells activation,and elicit antitumor activity.In this paper,we reported a novel PD-1 monoclonal antibody(mAb)CS1003,which is a humanized IgG4 PD-1 mAb generated by conventional hybridoma technology,and currently being developed in multiple clinical trials as monotherapy or in combination with other anticancer agents.We showed that CS1003 bound to recombinant human,cynomolgus monkey,and mouse PD-1 with EC50 values of 0.1757,0.2459,and 0.3664 nM,respectively.CS1003 blocked PD-1 interaction with its ligands,dose-dependently enhanced T cell proliferation and secretion of cytokines(IL-2 and IFN-γ)to the levels comparable to the reference antibody pembrolizumab.Intraperitoneal administration of CS1003(0.1,0.5,2.5 mg/kg,once every 3 days)dose-dependently suppressed the growth of MC38-hPD-L1 colon cancer in hPD-1 knock-in mice.Pharmacokinetics(PK)study revealed a linear PK profile within the dose range of 2–18 mg/kg following single intravenous administration in cynomolgus monkey.These data provide a comprehensive preclinical characterization of CS1003 that supports its clinical development for cancer immunotherapy. | Fu Li Jingrong Li Ke Yin Juan Zhang Zhen-hu Li Liang Lu Yuan-wu Bao Zhen Qin Yong Zheng Bao-tian Yang Jing Li Xinzhong Wang | 2021 | Acta Pharmacologica Sinica2021,42,1: | 2 |
