1N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. 3Effects of electroacupuncture at Taichong(LR 3) and Baihui(DU 20)on cardiac hypertrophy in rats with spontaneous hypertension显示文摘OBJECTIVE: To investigate the effects of electroacupuncture(EA) at Taichong(LR 3) and Baihui(DU 20)on myocardial hypertrophy in spontaneously hypertensive rats(SHRs).METHODS: Thirty-six SHRs were randomly assigned to model, EA, and Losartan groups, with twelve rats per group. Twelve Wistar Kyoto rats were selected as the normal control group. Systolic blood pressure(SBP) and cardiac function were measured in all rats.Expression levels of factors associated with the phosphoinositide 3-kinase/protein kinase B/mammalian target of rapamycin(PI3K/Akt/mTOR) pathway were evaluated by Western blotting and real-time PCR.Pathological changes of the heart tissue were observed by hematoxylin-eosin staining.RESULTS: After treatment, enhanced SBP was significantly decreased in the EA and Losartan groups compared with the model group(P < 0.01). Echocardiographic and morphological analyses revealed that enhanced end-diastolic interventricular septal thickness and left ventricular posterior wall thickness, as well as ratio of left ventricular weight to body weight were markedly diminished in the EA and Losartan groups(P < 0.01 or P < 0.05), while reduced left ventricular end-diastolic dimension and left ventricular ejection fraction were significantly ameliorated(P < 0.01). 4Effects of different acupuncture manipulations on protein expression in the parietal cortex of spontaneously hypertensive rats显示文摘Objective: To analyze the effects of twirling reinforcing and reducing manipulations on protein expression in parietal cortex of spontaneously hypertensive rats(SHRs), and elucidate the main mechanisms and differences between two manipulations in hypertension treatment.Methods: Rats were randomly divided into the control, model, twirling reinforcing manipulation(TRFM),and twirling reducing manipulation(TRDM) groups. The control and model groups received catch and fixation stimulations once a day for 14 days. The TRFM and TRDM groups were intervened once a day for 20 min for 14 days. On days 0, 2, 6, 10, and 14 after acupuncture, rat systolic blood pressures(SBPs) were measured. Differential protein(DP) expression in the rat parietal cortices was detected. Thereafter, GO functional significance and KEGG pathway enrichment analyses were performed.Results: Compared with the model group, SBP of rats in the TRDM and TRFM groups decreased on days 6 and 10 of acupuncture, respectively(P=.009;P <.001). Moreover, SBP of the TRDM group was significantly lower than that of the TRFM group on days 10 and 14 of acupuncture(P=.015;P=.013).Compared with control group, 601 and 1040 DPs were up-and downregulated, respectively, in the model group. Compared with model group, 44 and 28 up-and downregulated DPs were expressed, respectively,in the TRFM group. Compared with model group, expression of 616 and 427 up-and downregulated DPs,respectively, was found in the TRDM group. 7Boosting practical high voltage lithium metal batteries by butyronitrile in ether electrolytes via coordination, hydrolysis of C≡N and relatively mild concentration strategy显示文摘Currently ether solvents have been regarded as the most compatible organic solvents with lithium metal in electrolytes of lithium batteries.However,ether solvents are unstable under high voltage (>4.0 V),and prone to side reactions with nickel-rich high-voltage cathode materials.In this work,a novel dual-solvent electrolyte in ethylene glycol dimethyl ether (DME) and butyronitrile (BN) mixed solvent was designed and fabricated for Li/Li Ni_(0.5)Mn_(0.3)Co_(0.2)O_(2)-based lithium metal batteries.When charged to high voltage4.3 V,the battery cycled in this optimal electrolyte can maintain the capacity at 133.7 m Ah g^(-1) with a retention of 88.84%after 150 cycles at 0.2 C and-10℃.During long-term cycling,the battery also exhibits excellent cycling performance with capacity maintained at about 112.0 m Ah g^(-1) after 500 cycles at 1C and-10℃.BN has strong oxidation resistance and high conductivity,which can inhibit the decomposition of ether solvents under high voltage and improve the low temperature performance of battery effectively.Additionally,the cyano (–C≡N) group in BN molecular has a strong coordination ability with the high-valent metal ions and can mask the active ions on the cathode,correspondingly reducing the corrosion of cathode material by the electrolyte.Moreover,cyano group can participate in the hydrolysis to remove trace amounts of water and acidic by-products such as HF in the electrolyte.Therefore,the boosting effect of butyronitrile for ether solvents can provide a promising strategy for enhancing the performance of high voltage lithium metal batteries for practical industrialization.Jingrong Ning Kaijia Duan Kai Wang Jianwen Liu Shiquan Wang Jiujun Zhang 2022Journal of Energy Chemistry2022,31,4:2
8Contribution of bacterivorous nematodes to soil resistance and resilience under copper or heat stress显示文摘The functional performance of soil ecosystems following disturbance determines ecosystem stability,and although contributions of bacterivorous nematodes to soil ecosystems are recognized,their roles in functional stability have received little attention.The objective of this study was to evaluate the roles of bacterivorous nematodes in functional stability following stress.In a factorial laboratory experiment,soil microcosms were prepared with two levels of nematode abundance,either an enriched abundance of bacterivores(Nema soil)or background abundance of nematodes(CK soil),and three levels of stress,copper,heat,or an unstressed control.The resistance and resilience of nematode abundance,as well as soil microbial function by determining decomposition of plant residues and microbial substrate utilization pattern using a BIOLOG microplate,were followed post stress.The relative changes of two dominant bacterivores,Acrobeloides and Protorhabditis,responded differently to stresses.The resistance and resilience of Protorhabditis were greater than that of Acrobeloides to copper stress during the whole incubation period,while both bacterivores only showed higher resilience under heat stress at the end of incubation.The enrichment of bacterivores had no significant effects on the soil microbial resistance but significantly increased its resilience to copper stress.Under heat stress,the positive effect of bacterivores on soil resilience was only evident from 28 days to the end of incubation.The differences in the responses of soil function to stress with or without bacterivores suggested that soil nematodes could be conducive to ecosystem stability,highlighting the soil fauna should be taken into account in soil sustainable management.Xiaoyun Chen Wenfeng Xue Jingrong Xue Bryan S.Griffiths Manqiang Liu 2020Soil Ecology Letters2020,2,3:1

14High-Throughput Drug Screening on Borrelia garinii and Borrelia afzelii Identified Hypocrellin A as an Active Drug Candidate Against Borrelia Species显示文摘Lyme disease(LD)is a tick-transmitted infection caused by Borrelia burgdorferi sensu lato species,which include B.burgdorferi,Borrelia afzelii and Borrelia garinii.The majority of patients with early LD can be cured by the standard treatment,yet some still suffer from posttreatment LD syndrome.The presence of Borrelia persisters has been proposed as a contributing factor,because they cannot be completely eradicated by the currently used antibiotics for LD.Finding new pharmaceuticals targeting Borrelia persisters is crucial for developing more effective treatments.Here,we first confirmed the existence of persisters in B.garinii and B.afzelii cultures and then conducted a high-throughput screening of a customdrug library against persister-rich stationary-phase B.garinii and B.afzelii cultures.Among 2427 compounds screened,hypocrellin A(HA),anthracycline class of drugs and topical antibiotics along with some other natural compounds were identified to have strong potential for killing persisters of B.garinii and B.afzelii.HA was the most active anti-Borrelia compound,capable of eradicating stationary-phase Borrelia persisters,in particular when combined with doxycycline and/or ceftriaxone.Liposoluble antioxidant vitamin E was found to antagonize the activity of HA,indicating HA’s target is the cell membrane where HA triggers the generation of reactive oxygen species in the presence of light.HA was found to have distinct bactericidal activity against Borrelia species but had poor or no activity against gram-positive and gram-negative bacteria.Identification of the abovementioned drug candidates may help develop more effective therapies for LD.Tingting Li Yuxian Xin Dongxia Liu Jingrong Sun Jingwei Li Ying Zhang Jie Feng 2023Infectious Microbes & Diseases2023,5,2:0
15Overexpressed NEDD8 as a potential therapeutic target in esophageal squamous cell carcinoma显示文摘Objective:The hyperactivated neddylation pathway plays an important role in tumorigenesis and is emerging as a promising anticancer target.We aimed to study whether NEDD8(neural precursor cell expressed,developmentally down-regulated 8)might serve as a therapeutic target in esophageal squamous cell carcinoma(ESCC).Methods:The clinical relevance of NEDD8 expression was evaluated by using The Cancer Genome Atlas(TCGA)database and tissue arrays.NEDD8-knockdown ESCC cells generated with the CRISPR/Cas9 system were used to explore the anticancer effects and mechanisms.Quantitative proteomic analysis was used to examine the variations in NEDD8 knockdown-induced biological pathways.The cell cycle and apoptosis were assessed with fluorescence activated cell sorting.A subcutaneous-transplantation mouse tumor model was established to investigate the anticancer potential of NEDD8 silencing in vivo.Results:NEDD8 was upregulated at both the mRNA and protein expression levels in ESCC,and NEDD8 overexpression was associated with poorer overall patient survival(mRNA level:P=0.028,protein level:P=0.026,log-rank test).Downregulation of NEDD8 significantly suppressed tumor growth both in vitro and in vivo.Quantitative proteomic analysis revealed that downregulation of NEDD8 induced cell cycle arrest,DNA damage,and apoptosis in ESCC cells.Mechanistic studies demonstrated that NEDD8 knockdown led to the accumulation of cullin-RING E3 ubiquitin ligases(CRLs)substrates through inactivation of CRLs,thus suppressing the malignant phenotype by inducing cell cycle arrest and apoptosis in ESCC.Rescue experiments demonstrated that the induction of apoptosis after NEDD8 silencing was attenuated by DR5 knockdown.Conclusions:Our study elucidated the anti-ESCC effects and underlying mechanisms of NEDD8 knockdown,and validated NEDD8 as a potential target for ESCC therapy.Jingrong Xian Shiwen Wang Yanyu Jiang Lihui Li Lili Cai Ping Chen Yue Liu Xiaofei Zeng Guoan Chen Chen Ding Robert MHoffman Lijun Jia Hu Zhao Yanmei Zhang 2022Cancer Biology & Medicine2022,19,4:0
16A hippocampal anti-hypertensive mechanism induced by twirling reinforcing-reducing manipulation in rats显示文摘Objective:To investigate a hippocampal anti-hypertensive mechanism induced by twirling reinforcingreducing manipulation(TRRM)using proteomics in rats.Methods:Forty-two male spontaneously hypertensive rats were randomly divided into 3 groups,and 14 Wistar-Kyoto rats were served as control group.In the twirling reinforcing(TRF)group,needles were directly inserted into the Taichong(LR 3)point,then thumbs were moved heavily forward and lightly backward for 3 min,while needles remained inserted for 17 min.In the twirling reduction(TRD)group,the same treatment was applied as in the TRF group except that the thumb moved lightly forward and heavily backward.In the model and control groups,only the corresponding grasping and fixation were applied.All interventions were conducted for 14 days.The blood pressure(BP)of all rats was measured one day before intervention and every other day after.Then hematoxylin-eosin(H&E)staining,label-free and parallel reaction monitoring proteomic techniques were used to assess hippocampal samples from each group.Results:Systolic BPs showed a significant decrease in the TRF and TRD groups compared with the model group(P<.01).In the model group,H&E staining showed obvious pathological changes in the hippocampus,while in the TRF and TRD groups,the hippocampal morphology was only slightly altered.Labelfree proteomic analysis revealed 1163 differential protein expressions between groups.Gene Ontology enrichment analysis confirmed that the differentially expressed proteins were enriched in different biological processes,cellular components,and molecular functions.Kyoto Encyclopedia of Genes and Genomes enrichment analysis showed that TRRM proteins were expressed in the serotonergic synapse pathway,renin-angiotensin system,the mitogen-activated protein kinase signaling pathway,and the peroxisome pathway,which were all also related to BP regulation.Conclusion:TRRM can significantly lower the BP of SHRs.The mode of action may be through the activation of various protein pathways in the hippocampus that are related to BP regulation.Jiao Sun Liu Yan Yunhong Zhu Jingrong Liang Yingying Zang Tianxiao Zeng Xiaomin Hao Pui Yee Wong Qingguo Liu 2023Journal of Traditional Chinese Medical Sciences2023,10,1:0

18Allocation of grassland, livestock and arable based on the spatial and temporal analysis for food demand in China显示文摘To explore the distribution of food demand and the projected trend in future food demand in China, this paper analyzed the change in current(1998–2012) percapita demand for grain, grain-consuming and herbivorous livestock products, and predicted the food demand in 2020 The results indicated that in 1998–2012, the national percapita consumption of grain ration declined by about36.66%, and the per-capita consumption of grain-consuming and herbivorous livestock products increased by about 48% and 34.09%, respectively. The grain-consuming livestock products have become the primary source of both calories and protein for consumers. The proportion of herbivorous livestock products in consumer diets has increased steadily and there has been huge potential in substituting beef and mutton for pork in this dynamic market. The demand for food in different regions of China is highly variable, which is important for planning grassland agriculture development and ensuring food safety. The demand for grain, and grain-consuming and herbivorous livestock products will increase by about3.3%, 20% and 14% respectively by 2020. 19NEDD8-conjugating enzyme UBC12 as a novel therapeutic target in esophageal squamous cell carcinoma显示文摘Dear Editor,Esophageal squamous cell carcinoma(ESCC)is the major histologic subtype of esophageal cancer with high incidence and mortality.1 However,few achievements have been made in the development of targeted drugs.2 Neddylation,a reversible post-translational modification,attaches ubiquitin-like molecule NEDD8 to substrates in a three-step enzymatic reaction catalyzed by NEDD8-activating enzyme E1(NAE,NAE1 and UBA3 heterodimer),NEDD8-conjugating enzyme E2s(UBE2M/UBC12 or UBE2F)and substrate-specific NEDD8-E3 ligases.3 The best-characterized substrates of neddylation are cullin family proteins,the essential components of multiunit Cullin-RING ubiquitin ligases(CRLs).4 Currently,the inhibition of cullin neddylation by targeting overactivated neddylation pathway has emerged as an attractive approach for anticancer therapy.5,6 Our previous study reported that MLN4924,a specific inhibitor of NAE,significantly inhibited the tumor growth of ESCC by blocking cullin neddylation and inactivating CRLs activity.7 However,recent studies found that MLN4924 treatment-emergent NAE mutations would confer the drug resistance.8,9 Therefore,it is urgent to identify other neddylation enzymes(E2s or E3s)as alternative anticancer targets and develop novel anti-ESCC strategies.Shiwen Wang Jingrong Xian Lihui Li Yanyu Jiang Yue Liu Lili Cai Robert M.Hoffman Lijun Jia Hu Zhao Yanmei Zhang 2020Signal Transduction and Targeted Therapy2020,5,1:0
