2A Pilot Study of the Therapeutic Efficacy and Mechanism of Artesunate in the MRL/lpr Murine Model of Systemic Lupus Erythematosus显示文摘Recent evidence indicates that artesunate has immunomodulatory properties that might be useful for treating autoimmune disease.In this study,we conducted a pilot study and explored the effect and mechanism of artesunate on the treatment of systemic lupus erythematosus using an MRL/lpr murine model.MRL/lpr mice were divided into control,cyclophosphamide(CTX)and artesunate treatment groups.Blood was collected to measure serum levels of creatinine,antinuclear antibody(ANA)and anti-double-stranded DNA(anti-dsDNA)antibody.Twenty-four-hour urine was collected to measure levels of proteinuria.The concentration of monocyte chemotactic protein-1(MCP-1)in serum and urine was measured.The expression of MCP-1 in kidney was detected by Western blot and immunohistochemistry assay,respectively.The expression of B cell activating factor(BAFF)in spleen was determined by real time-PCR and immunoblotting.We found that artesunate significantly increased the survival rate,body weight and blood leukocyte counts,and reduced the serum levels of ANA and anti-dsDNA antibody titer,24 h urinary protein,and serum creatinine.Our results indicated that artesunate could decrease MCP-1,major pro-inflammation cytokine,in serum,urine and kidney.We also found that the level of BAFF,the major B cell activation factor,was decreased in artesunate treated MRL/lpr mice.Its efficacy was comparable with that of CTX in this study.Taken together,we have demonstrated that artesunate can inhibit the progression of disease and reverse the pathologic lesion of lupus nephritis.Ouyang Jin Huayong Zhang Zhifeng Gu Shengnan Zhao Ting Xu Kangxing Zhou Bo Jiang Jie Wang Xiaofeng Zeng Lingyun Sun 2009Cellular & Molecular Immunology2009,6,6:20

4N6-Methyladenine DNA Methylation in Japonica and Indica Rice Genomes and Its Association with Gene Expression,Plant Development, and Stress Responses显示文摘N6-Methyladenine (6mA)DNA methylation has recently been implicated as a potential new epigenetic marker in eukaryotes,including the dioot modelArabidopsis thaliana.However,the conservation and divergence of 6mA distribution patterns and functions in plants remain elusive.Here we report high-quality 6mA methylomes at single-nucleotide resolution in rice based on substantially improved genome sequences of two rice cultivars,Nipponbare (Nip;Japonica)and 93-11 (Indica).Analysis of 6mA genomic distribution and its association with transcription suggest that 6mA distribution and function is rather conserved between rice and Arabidopsis.We found that 6mA levels are positively correlated with the expression of key stressrelated genes,which may be responsible for the difference in stress tolerance between Nip and 93-11. Moreover,we showed that mutations in DDM1 cause defects in plant growth and decreased 6mA level. 5Zircon SHRIMP U-Pb dating for olivine gabbro at Wangmuguan in the Beihuaiyang zone and its geological significance显示文摘Zircon SHRIMP U-Pb dating was per- formed for olivine gabbro at Wangmuguan in the Beihuaiyang zone of the Dabie orogen and its coun- try rock (garnet-bearing epidote-mica-quartz schist). The results show that the gabbro was crystallized at 635 ± 5 Ma, in the late Neoproterozoic rather than in the late Paleozoic as previously suggested; its coun- try rocks formed at 464±7 Ma, younger than the en- closed gabbro. The U-Pb age for the gabbro is in good agreement with ages for tuff interbedded with sediments from the Doushantuo Formation in the South China Block and late-Neoproterozoic basic dyke swarms distributed on a large scale over areas of Suizhou to Zaoyang of Hubei Province in the northern margin of the South China Block. This sug- gests a large-scale magmatic activity occurred at the late Neoproterozoic in the South China Block, so that the gabbro at Wangmuguan in the western segment of the Beihuaiyang zone is geotectonic affinity to the northern margin of the South China Block. Since the olivine gabbro occurs within the schist of Ordovician protolith with tectonic contact between them but forming in different tectonic settings, it is concluded that the late-Neoproterozoic gabbro was detached from the Precambrian basement of the South China Block during the Triassic subduction of the South China Block, and tectonically thrusted over the metamorphosed rocks in the southern margin of the North China Block.LIU Yican LI Shuguang GU Xiaofeng HOU Zhenhui 2006Chinese Science Bulletin2006,51,20:16

7Genome Characteristics and Evolution of Pseudorabies Virus Strains in Eastern China from 2017 to 2019显示文摘Since late 2011,outbreaks of pseudorabies virus(PRV)have occurred in southern China causing major economic losses to the pig industry.We previously reported that variant PRV forms and recombination in China could be the source of continued epidemics.Here,we analyzed samples from intensive pig farms in eastern China between 2017 and 2019,and sequenced the main glycoproteins(gB,gC,gD,and gE)to study the evolution characteristics of PRV.Based on the gC gene,we found that PRV variants belong to clade 2 and detected a founder effect during by the PRV epidemic.In addition,we detected inter-and intra-clade recombination;in particular,inter-clade recombination in the gB genes of strains FJ-ZXF and FJ-W2,which were recombinant with clade 1 strains.We also found specific amino-acid changes and positively selected sites,possibly associated with functional changes.This analysis of the emergence of PRV in China illustrates the need for continuous monitoring and the development of vaccines against specific variants of PRV.Xiaofeng Zhai Wen Zhao Kemang Li Cheng Zhang Congcong Wang Shuo Su Jiyong Zhou Jing Lei Gang Xing Haifeng Sun Zhiyu Shi Jinyan Gu 2019Virologica Sinica2019,34,6:12

10Occurrence of Neoproterozoic low-grade metagranite in the western Beihuaiyang zone,the Dabie orogen显示文摘Based on detailed field investigations and petrographic observations,we discovered Neoproterozoic-emplaced granite from the metavolcanics of the Dingyuan Formation in the western Beihuaiyang zone,the Dabie orogen. This study reports the results of zircon U-Pb dating and preliminary petrographic observations on two metagranite samples. The studied rocks experienced epidote-amphibolite facies metamorphism and strong structural deformation. Their U-Pb ages are 726 ± 6 and 758 ± 12 Ma,respectively,similar to those for the Luzhenguan complex in the eastern segment of the Beihuaiyang zone. In combination with previously determined 635 ± 5 Ma low-grade metagabbro,this study suggests the occurrence of at least two types of Neoproterozoic intrusive rocks in the Beihuaiyang zone,the northern margin of the South China Block (SCB):726-758 Ma metagranite and 635 Ma metagabbro. These rocks occur within the metamorphosed Ordovician volcanic zone (originally named the Dingyuan Formation) and are in tectonic contact to each other,but they formed in different tectonic settings. The protolith ages for the Neoproterozoic low-grade metaigneous rocks are in good agreement not only with ages for two episodes of mid-and late-Neoproterozoic mafic and felsic magmatism in the Suizhou and Zaoyang area,Hubei Province,but also agree with protolith ages of ultrahigh-pressure metaigneous rocks in the Dabie-Sulu orogenic belt. 12Transcriptional landscape of rice roots at the single-cell resolution显示文摘There are two main types of root systems in flowering plants,namely taproot systems of dicots and fibrous root systems found in monocots.Despite this fundamental split,our current knowledge of cellular and molecular mechanism driving root development is mainly based on studies of the dicot model Arabidopsis.However,the world major crops are monocots and little is known about the transcriptional programs underlying cell-type specification in this clade.Here,we report the transcriptomes of more than 20000 single cells derived from root tips of two agronomically important rice cultivars.Using combined computational and experimental analyses we were able to robustly identify most of the major cell types and define novel cell-type-specific marker genes for both cultivars.Importantly,we found divergent cell types associated with specific regulatory programs,including phytohormone biosynthesis,signaling,and response,which were well conserved between the two rice cultivars.In addition,we detected substantial differences between the cell-type transcript profiles of Arabidopsis and rice.These species-specific features emphasize the importance of analyzing tissues across diverse model species,including rice.Taken together,our study provides insight into the transcriptomic landscape of major cell types of rice root tip at single-cell resolution and opens new avenues to study cell-type specification,function,and evolution in plants.Qing Liu Zhe Liang Dan Feng Sanjie Jiang Yifan Wang Zhuoying Du Ruoxi Li Guihua Hu Pingxian Zhang Yanfei Ma Jan ULohmann Xiaofeng Gu 2021Molecular Plant2021,14,3:9

14Reversal effect of bufalin on multidrug resistance in K562/VCR vincristine-resistant leukemia cell line显示文摘OBJECTIVE: To probe insights into the reversal effect of bufalin on vincristine-acquired multidrug resistance(MDR) in human leukemia cell line K562/VCR.METHODS: Proliferative inhibition rate and the reversal index(RI) of bufalin were determined by Methyl thiazolyl tetrazolium assay. The uptake of Adriamycin(ADM) in K562/VCR cells, cell cycle and apoptosis rate were determined by flow cytometry(FCM). Cell morphologic changes were observed with Wright-Giemsa staining. The expression of P-glycoprotein(P-gp), multidrug-associated protein-1(MRP1), Bcl-x L and Bax protein were measured by immunocytochemistry.RESULTS: The human leukemia multidrug resistant K562/VCR cells showed no cross-resistance to bufalin. The RIs of bufalin at concentrations of 0.0002,0.001 and 0.005 μmol/L were 4.85, 6.94 and 14.77,respectively. Preincubation of 0.001 μmol/L bufalin for 2 h could increase intracellular ADM fluorescence intensity to 28.07%(P<0.05) and down-regulate MRP1 expression simultaneously, but no remarkable effect was found on P-gp protein. Cell cycle analysis indicated increased apoptosis rate and apparent decreased G2/M phase proportion after treatment with bufalin. When exposed to 0.01μmol/L bufalin, typical morphological changes of apoptosis could be observed. 16Optical observations of LIGO source GW 170817 by the Antarctic Survey Telescopes at Dome A,Antarctica显示文摘The LIGO detection of gravitational waves(GW) from merging black holes in 2015 marked the beginning of a new era in observational astronomy. The detection of an electromagnetic signal from a GW source is the critical next step to explore in detail the physics involved. The Antarctic Survey Telescopes(AST3),located at Dome A, Antarctica, is uniquely situated for rapid response time-domain astronomy with its continuous night-time coverage during the austral winter. We report optical observations of the GW source(GW 170817) in the nearby galaxy NGC 4993 using AST3. The data show a rapidly fading transient at around 1 day after the GW trigger, with the i-band magnitude declining from 17:23 ± 0:13 magnitude to 17:72 ± 0:09 magnitude in ~1:8 h. The brightness and time evolution of the optical transient associated with GW 170817 are broadly consistent with the predictions of models involving merging binary neutron stars. 19A simple model of radial nerve injury in the rhesus monkey to evaluate peripheral nerve repair显示文摘Current research on bone marrow stem cell transplantation and autologous or xenogenic nerve transplantation for peripheral nerve regeneration has mainly focused on the repair of peripheral nerve defects in rodents.In this study,we established a standardized experimental model of radial nerve defects in primates and evaluated the effect of repair on peripheral nerve injury.We repaired 2.5-cm lesions in the radial nerve of rhesus monkeys by transplantation of autografts,acellular allografts,or acellular allografts seeded with autologous bone marrow stem cells.Five months after surgery,regenerated nerve tissue was assessed for function,electrophysiology,and histomorphometry.Postoperative functional recovery was evaluated by the wrist-extension test.Compared with the simple autografts,the acellular allografts and allografts seeded with bone marrow stem cells facilitated remarkable recovery of the wrist-extension functions in the rhesus monkeys.This functional improvement was coupled with radial nerve distal axon growth,a higher percentage of neuron survival,increased nerve fiber density and diameter,increased myelin sheath thickness,and increased nerve conduction velocities and peak amplitudes of compound motor action potentials.Furthermore,the quality of nerve regeneration in the bone marrow stem cells-laden allografts group was comparable to that achieved with autografts.The wrist-extension test is a simple behavioral method for objective quantification of peripheral nerve regeneration.Dong Wang Xijun Huang Guo Fu Liqiang Gu Xiaolin Liu Honggang Wang Jun Hu Jianhua Yi Xiaofeng Niu Qingtang Zhu 2014Neural Regeneration Research2014,9,10:3
20Self-organized phase-transition lithography for all-inorganic photonic textures显示文摘Realizing general processing applicable to various materials by one basic tool has long been considered a distant dream.Fortunately,ultrafast laser-matter interaction has emerged as a highly universal platform with unprecedented optical phenomena and provided implementation paths for advanced manufacturing with novel functionalities.Here,we report the establishment of a three-dimensional(3D)focal-area interference field actively induced by a single ultrafast laser in transparent dielectrics.Relying on this,we demonstrate a radically new approach of self-organized phase-transition lithography(SOPTL)to achieve super-resolution construction of embedded all-inorganic photonic textures with extremely high efficiency.The generated textures exhibit a tunable photonic bandgap(PBG)in a wide range from〜1.3 to〜2 pm.More complicated interlaced textures with adjustable structural features can be fabricated within a few seconds,which is not attainable with any other conventional techniques.Evidenee suggests that the SOPTL is extendable to more than one material system.This study augments light-matter interaction physics,offers a promising approach for constructing robust photonic devices,and opens up a new research direction in advaneed lithography.Bo Zhang Dezhi Tan Zhuo Wang Xiaofeng Liu Beibei Xu Min Gu Limin Tong Jianrong Qiu 2021Light(Science & Applications)2021,10,6:3
