The temporal dynamics of antimicrobial-resistant Salmonella enterica and predominant serovars in China显示文摘Salmonella enterica is one of the most common bacterial pathogens in humans and animals.Systematic studies on the trends and geographical distribution of antimicrobial-resistant Salmonella and dominant serovars have been well studied in European and American countries while not in China. Here, taking the One-Health strategy,we used>35000 Salmonella enterica isolates to explore the temporal and spatial dynamics of dominant serovars in China. We found that Salmonella Typhimurium was the dominant serovar causing human infection in China, which was consistent with Australia but inconsistent with North American and European countries. The proportion of Salmonella serovars Typhimurium,London,Rissen,Corvallis,Meleagridis,Kentucky, and Goldcoast showed an increasing trend during 2006-2019.We randomly selected 1962 isolates for comparative genomics and antimicrobial resistance studies and found that the number of antibiotic resistance genes(ARGs)per isolate increased 1.84 and 2.69 times of human and non-human origins, respectively, spanning 14 years. The proportion of antimicrobial-resistant Salmonella isolates had an increasing trend during 2006-2019,especially beta-lactam,quinolone,tetracycline,and rifampicin resistance. Moreover,we found that higher diversity of sequence types(STs)in S.Typhimurium than in other serovars,ST34 from pig and ST 19 from chicken origin, were mainly associated with isolates causing child and adult gastro-infection,respectively.Our results fill in the data gap on the trends of dominant serovars and antimicrobial resistance of Salmonella enterica in China. These data provide useful information for public health decision-makers prioritizing interventions for foodborne diseases and food safety. | Yanan Wang Yue Liu Na Lyu Zhiyuan Li Sufang Ma Demin Cao Yuanlong Pan Yongfei Hu Hua Huang George F.Gao Xuebin Xu Baoli Zhu | 2023 | National Science Review2023,10,3: | 4
CD38 deficiency alleviates Ang II-induced vascular remodeling by inhibiting small extracellular vesicle-mediated vascular smooth muscle cell senescence in mice显示文摘CD38 is the main enzyme for nicotinamide adenine dinucleotide(NAD)degradation in mammalian cells.Decreased NAD levels are closely related to metabolic syndromes and aging-related diseases. Our study showed that CD38 deficiency significantly alleviated angiotensin II(Ang ll)-induced vascular remodeling in mice,as shown by decreased blood pressures;reduced vascular media thickness,media-to-lumen ratio, and collagen deposition;and restored elastin expression. However, our bone marrow transplantation assay showed that CD38 deficiency in lymphocytes led to lack of protection against Ang ll-induced vascular remodeling, suggesting that the effects of CD38 on Ang ll-induced vascular remodeling might rely primarily on vascular smooth muscle cells(VSMCs),not lymphocytes. In addition,we observed that CD38 deficiency or NAD supplementation remarkably mitigated Ang ll-induced vascular senescence by suppressing the biogenesis, secretion, and internalization of senescence-associated small extracellular vesicles(SA-sEVs),which facilitated the senescence of neighboring non-damaged VSMCs.Furthermore,we found that the protective effects of CD38 deficiency on VSMC senescence were related to restoration of lysosome dysfunction, particularly with respect to the maintenance of sirtuin-mediated mitochondrial homeostasis and activation of the mitochondria-lysosomal axis in VSMCs.In conclusion, our findings demonstrated that CD38 and its associated intracellular NAD decline are critical for Ang ll-induced VSMC senescence and vascular remodeling. | Lu Gan Demin Liu Jing Liu Erya Chen Chan Chen Lian Liu Hang Hu Xiaohui Guan Wen Ma Yanzi Zhang Yarong He Bofu Liu Songling Tang Wei Jiang Jianxin Xue Hongbo Xin | 2021 | Signal Transduction and Targeted Therapy2021,6,7: | 2

Electron-ion collider in China显示文摘Lepton scattering is an established ideal tool for studying inner structure of small particles such as nucleons as well as nuclei. As a future high energy nuclear physics project,an Electron-ion collider in China(EicC)has been proposed. It will be constructed based on an upgraded heavy-ion accelerator,High Intensity heavy-ion Accelerator Facility(HIAF)which is currently under construction, together with a new electron ring. The proposed collider will provide highly polarized electrons(with a po-larization of 80%)and protons(with a polarization of 70%)with variable center of mass energies from 15 to 20 GeV and the luminosity of(2–3)×1033 cm^(−2)·s^(−1).Polarized deuterons and Helium-3,as well as unpolarized ion beams from Carbon to Uranium, will be also available at the EicC.The main foci of the EicC will be precision measurements of the structure of the nucleon in the sea quark region, including 3D tomography of nucleon;the partonic structure of nuclei and the parton interaction with the nuclear environment;the exotic states, especially those with heavy flavor quark contents. In addition, issues fundamental to understanding the origin of mass could be addressed by measurements of heavy quarkonia near-threshold production at the EicC.In order to achieve the above-mentioned physics goals,a hermetical detector system will be constructed with cutting-edge technologies. This document is the result of collective contributions and valuable inputs from experts across the globe. The EicC physics program complements the ongoing scientific programs at the Jefferson Laboratory and the future EIC project in the United States. The success of this project will also advance both nuclear and particle physics as well as accelerator and detector technology in China. | Daniele PAnderle Valerio Bertone Xu Cao Lei Chang Ningbo Chang Gu Chen Xurong Chen Zhuojun Chen Zhufang Cui Lingyun Dai Weitian Deng Minghui Ding Xu Feng Chang Gong Longcheng Gui Feng-Kun Guo Chengdong Han Jun He Tie-Jiun Hou Hongxia Huang Yin Huang KrešImir KumeričKi LPKaptari Demin Li Hengne Li Minxiang Li Xueqian Li Yutie Liang Zuotang Liang Chen Liu Chuan Liu Guoming Liu Jie Liu Liuming Liu Xiang Liu Tianbo Liu Xiaofeng Luo Zhun Lyu Boqiang Ma Fu Ma Jianping Ma Yugang Ma Lijun Mao Cédric Mezrag HervéMoutarde Jialun Ping Sixue Qin Hang Ren Craig DRoberts Juan Rojo Guodong Shen Chao Shi Qintao Song Hao Sun PawełSznajder Enke Wang Fan Wang Qian Wang Rong Wang Ruiru Wang Taofeng Wang Wei Wang Xiaoyu Wang Xiaoyun Wang Jiajun Wu Xinggang Wu Lei Xia Bowen Xiao Guoqing Xiao Ju-Jun Xie Yaping Xie Hongxi Xing Hushan Xu Nu Xu Shusheng Xu Mengshi Yan Wenbiao Yan Wencheng Yan Xinhu Yan Jiancheng Yang Yi-Bo Yang Zhi Yang Deliang Yao Zhihong Ye Peilin Yin C-PYuan Wenlong Zhan Jianhui Zhang Jinlong Zhang Pengming Zhang Yifei Zhang Chao-Hsi Chang Zhenyu Zhang Hongwei Zhao Kuang-Ta Chao Qiang Zhao Yuxiang Zhao Zhengguo Zhao Liang Zheng Jian Zhou Xiang Zhou Xiaorong Zhou Bingsong Zou Liping Zou | 2021 | Frontiers of physics2021,16,6: | 1
Late Jurassic Intracontinental Extension and Related Mineralisation in Southwestern Fujian Province of SE China:Insights from Deformation and Syn-Tectonic Granites显示文摘Late Mesozoic igneous intrusions and extensional structures in Carboniferous to Permian sequences in the SW Fujian region acted as important controls on the localisation of Fe-polymetallic de-posits.Here we document the identification of extensional deformation at shallow crustal levels and syn-tectonic granites related to normal faults.Based on spatial distribution and structural features, the extensional deformation can be divided into cover-only and basement-intersecting styles.A series of syn-tectonic plutons were emplaced into the footwall of normal faults.Representative samples of the Tangquan Granite have high SiO2(66.4 wt.%-73.9 wt.%)assays and Mg#values(37-59).The samples also have relatively homogenous initial 87Sr/86Sr(0.7083-0.7089)andεNd(-9.2--10.2)values.Geochemical and isotopic evidences indicate that the Tangquan granite originates from a hybrid source including lower crustal-derived felsic and lithospheric mantle-derived mafic magmas.Zircon U-Pb dating indicates that the granodiorite phase from the pluton crystallised at 161±4 Ma and the monzogranite phase crystallised at 159±1 Ma.Combined with the granitic rocks in a wider region of SE China, the widespread granitic magmatism and polymetallic mineralisation have been synchronous during the Late Mesozoic, probably resulting from extensional tectonics related to the lithospheric thinning. | Rui Cao Xinghua Ma Leon Bagas Yongbao Gao Demin Liu Zailai Mou | 2021 | Journal of Earth Science2021,32,1: | 1
Macrophages in Ischemic Heart Failure:Yesterday,Today, and Tomorrow显示文摘With continually improving reperfusion strategies and patient care, the overall mortality of acute myocardial infarction(AMI)has been significantly reduced during the past two decades. However, this success is a double-edged sword,as many patients surviving an AMI will progress towards ischemic heart failure(HF)over time. The pathologic causes of ischemic HF are undoubtedly multifactorial. However, the inflammatory response is considered one of the most important causes of pathological remodeling because it spans the whole process of HF development. The macrophage-mediated inflammatory response was once considered a purely harmful factor leading to pathological remodeling and HF.However, growing evidence demonstrates that multiple subgroups of macrophage exist and contribute differently to ischemic HF development.Understanding macrophage populations and how they contribute to post-MI remodeling and consequent ischemic HF is,therefore, critical to understanding and treating the disease. This review focuses on different macrophage populations that regulate post-MI cardiac injury and how immunoregulation therapy may benefit patients with ischemic HF. | Demin Liu Wenjun Yan Jingwen Huang Jianli Zhao Houston Kilby Theodore AChristopher Bernard Lopez Ling Tao Xinliang Ma Guoqiang Gu Yajing Wang | 2021 | Cardiology Discovery2021,1,2: | 0
circPVT1 promotes silica-induced epithelial-mesenchymal transition by modulating the miR-497-5p/TCF3 axis显示文摘Epithelial-mesenchymal transition(EMT)is a vital pathological feature of silica-induced pulmonary fibrosis. However, whether circRNA is involved in the process remains unclear. The present study aimed to investigate the role of circPVT1 in the silica-induced EMT and the underlying mechanisms. We found that an elevated expression of circPVT1 promoted EMT and enhanced the migratory capacity of silica-treated epithelial cells. The isolation of cytoplasmic and nuclear separation assay showed that circPVT1 was predominantly expressed in the cytoplasm.RNA immunoprecipitation assay and RNA pull-down experiment indicated that cytoplasmic-localized circPVT1 was capable of binding to miR-497-5p.Furthermore,we found that miR-497-5p attenuated the silica-induced EMT process by targeting transcription factor 3(TCF3),an E-cadherin transcriptional repressor,in the silica-treated epithelial cells.Collectively, these results reveal a novel role of the circPVT1/miR-497-5p/TCF3 axis in the silica-induced EMT process in lung epithelial cells.Once validated, this finding may provide a potential theoretical basis for the development of interventions and treatments for pulmonary fibrosis. | Siyun Zhou Yan Li Wenqing Sun Dongyu Ma Yi Liu Demin Cheng Guanru Li Chunhui Ni | 2024 | The Journal of Biomedical Research2024,38,2: | 0
Proximity-enabled covalent binding of IL-2 to IL-2Rα selectively activates regulatory T cells and suppresses autoimmunity显示文摘Interleukin-2(IL-2)is a pleiotropic cytokine that orchestrates bidirectional immune responses via regulatory T cells(Tregs)and effector cells, leading to paradoxical consequences. Here,we report a strategy that exploited genetic code expansion-guided incorporation of the latent bioreactive artificial amino acid fluorosulfate-L-tyrosine(FSY)into IL-2 for proximity-enabled covalent binding to IL-2Rαto selectively promote Treg activation. We found that FSY-bearing IL-2 variants, such as L72-FSY,covalently bound to IL-2Rαvia sulfur-fluoride exchange when in proximity, resulting in persistent recycling of IL-2 and selectively promoting the expansion of Tregs but not effector cells.Further assessment of L72-FSY-expanded Tregs demonstrated that L72-FSY maintained Tregs in a central memory phenotype without driving terminal differentiation,as demonstrated by simultaneously attenuated expression of lymphocyte activation gene-3(LAG-3)and enhanced expression of programmed cell death protein-1(PD-1).Subcutaneous administration of L72-FSY in murine models of pristane-induced lupus and graft-versus-host disease(GvHD)resulted in enhanced and sustained therapeutic efficacy compared with wild-type IL-2 treatment. The efficacy of L72-FSY was further improved by N-terminal PEGylation, which increased its circulatory retention for preferential and sustained effects. This proximity-enabled covalent binding strategy may accelerate the development of pleiotropic cytokines as a new class of immunomodulatory therapies. | Bo Zhang Jiaqi Sun Yeshuang Yuan Dezhong Ji Yeting Sun Yudong Liu Shengjie Li Xingxing Zhu Xunyao Wu Jin Hu Qiu Xie Ling Wu Lulu Liu Boyang Cheng Yuanjie Zhang Lingjuan Jiang Lidan Zhao Fei Yu Wei Song Min Wang Yue Xu Shiliang Ma Yunyun Fei Lihe Zhang Demin Zhou Xuan Zhang | 2023 | Signal Transduction and Targeted Therapy2023,8,2: | 0
| 10 | Feasibility and physics potential of detecting ^(8)B solar neutrinos at JUNO显示文摘The Jiangmen Underground Neutrino Observatory(JUNO)features a 20 kt multi-purpose underground liquid scintillator sphere as its main detector. Some of JUNO's features make it an excellent location for^8B solar neutrino measurements, such as its low-energy threshold, high energy resolution compared with water Cherenkov detectors, and much larger target mass compared with previous liquid scintillator detectors. In this paper,we present a comprehensive assessment of JUNO's potential for detecting^8B solar neutrinos via the neutrino-electron elastic scattering process.A reduced 2 MeV threshold for the recoil electron energy is found to be achievable, assuming that the intrinsic radioactive background^(238)U and^(232)Th in the liquid scintillator can be controlled to 10^(-17)g/g.With ten years of data acquisition, approximately 60,000 signal and 30,000 background events are expected. This large sample will enable an examination of the distortion of the recoil electron spectrum that is dominated by the neutrino flavor transformation in the dense solar matter, which will shed new light on the inconsistency between the measured electron spectra and the predictions of the standard three-flavor neutrino oscillation framework.IfDelta m^(2)_(21)=4.8times10^(-5);(7.5times10^(-5))eV^(2),JUNO can provide evidence of neutrino oscillation in the Earth at approximately the 3sigma(2sigma)level by measuring the non-zero signal rate variation with respect to the solar zenith angle. Moreover,JUNO can simultaneously measureDelta m^2_(21)using^8B solar neutrinos to a precision of 20% or better, depending on the central value, and to sub-percent precision using reactor antineutrinos.A comparison of these two measurements from the same detector will help understand the current mild inconsistency between the value of Delta m^2_(21)reported by solar neutrino experiments and the KamLAND experiment.
