1Identification of a novel coronavirus causing severe pneumonia in human:a descriptive study显示文摘Background:Human infections with zoonotic coronaviruses(CoVs),including severe acute respiratory syndrome(SARS)-CoV and Middle East respiratory syndrome(MERS)-CoV,have raised great public health concern globally.Here,we report a novel batorigin CoV causing severe and fatal pneumonia in humans.Methods:We collected clinical data and bronchoalveolar lavage(BAL)specimens from five patients with severe pneumonia from Wuhan Jinyintan Hospital,Hubei province,China.Nucleic acids of the BAL were extracted and subjected to next-generation sequencing.Virus isolation was carried out,and maximum-likelihood phylogenetic trees were constructed.Results:Five patients hospitalized from December 18 to December 29,2019 presented with fever,cough,and dyspnea accompanied by complications of acute respiratory distress syndrome.Chest radiography revealed diffuse opacities and consolidation.One of these patients died.Sequence results revealed the presence of a previously unknownβ-CoV strain in all five patients,with 99.8%to 99.9%nucleotide identities among the isolates.These isolates showed 79.0%nucleotide identity with the sequence of SARS-CoV(GenBank NC_004718)and 51.8%identity with the sequence of MERS-CoV(GenBank NC_019843).The virus is phylogenetically closest to a bat SARS-like CoV(SL-ZC45,GenBank MG772933)with 87.6%to 87.7%nucleotide identity,but is in a separate clade.Moreover,these viruses have a single intact open reading frame gene 8,as a further indicator of bat-origin CoVs.However,the amino acid sequence of the tentative receptor-binding domain resembles that of SARS-CoV,indicating that these viruses might use the same receptor.Conclusion:A novel bat-borne CoV was identified that is associated with severe and fatal respiratory disease in humans.Li-Li Ren Ye-Ming Wang Zhi-Qiang Wu Zi-Chun Xiang Li Guo Teng Xu Yong-Zhong Jiang Yan Xiong Yong-Jun Li Xing-Wang Li Hui Li Guo-Hui Fan Xiao-Ying Gu Yan Xiao Hong Gao Jiu-Yang Xu Fan Yang Xin-Ming Wang Chao Wu Lan Chen Yi-Wei Liu Bo Liu Jian Yang Xiao-Rui Wang Jie Dong Li Li Chao-Lin Huang Jian-Ping Zhao Yi Hu Zhen-Shun Cheng Un-Lin Liu Zhao-Hui Qian Chuan Qin Qi Jin Bin Cao Jian-Wei Wang 2020Chinese Medical Journal2020,,9:99

5Base-Editing-Mediated Artificial Evolution of OsALS1 In Planta to Develop Novel Herbicide-Tolerant Rice Germplasms显示文摘Recently developed CRISPR-mediated base editors,which enable the generation of num erous nucleotide changes in target genomic regions,have been widely adopted for gene correction and generation of crop germ plasms containing im portant gain-of-function genetic variations.How ever,to engineer target genes with unknown functional SNPs remains challenging.To address this issue,we present here abase-e diting-mediated gene evolution(BEMGE)m ethod,employing both Cas9n-based cytosine and adenine base editors as well as a single-guide RNA(sgRNA)library tiling the full-length coding region,for developing novel rice germ plasm swith mutations in any endogenous gene.To this end,OsALS1 was artificially evolved in rice cells using BEMGE through both Agrobacterium-mediated and particle-bom bardment-mediated transform ation.Four different types of amino acid substitutions in the evolved OsALS1,derived from two sites that have never been targeted by natural or human selection during rice dom estication,were identified,conferring varying levels of tolerance to the herbicide bispyribac-sodium.Furtherm ore,the P171F substitution identified in a strong OsALS1 allele was quickly introduced into the commercial rice cultivar Nangeng 46 through precise base editing w ith the corresponding base editor and sgRNA.Collectively,these data indicate great potential of BEMGE in creating important genetic variants of target genes for crop improvement.Yongjie Kuang Shaofang Li Bin Ren Fang Yan Carl Spetz Xiangju Li Xueping Zhou Huanbin Zhou 2020Molecular Plant2020,13,4:37

7Morphological study on colonic pathology in ulcerative colitis treated by moxibustion显示文摘AIM To observe the therapeutic effect ofmoxibustion on ulcerative colitis and itsinfluence on the colonic mucosal morphology.METHODS Forty-six patients with ulcerativecolitis were randomly divided into themoxibustion with herbal medicine underneathgroup and the western medicine group.Thirtypatients were treated with the abovemoxibustion and 16 patients with Salicylayefapyridine(SASP).The colonic mucosa of 13patients in the moxibustion group was observedby colonoscopy before and after the treatment.Mucin was also analyzed by H.E and AB-PASstaining.RESULTS Seventeen patients were clinicallycured,12 were improved and 1 unchanged in themoxibustion group.In the control group,5patients were clinically cured,7 improved and 4unchanged.Thirteen patients with active UCwere taken as the subjects for histopathologicanalysis in this study.The colonic mucosallesions were remarkably improved and thecharacteristic of the mucin also changed.Inmost sections,the chronic inflammation ofmucosa was geatly ameliorated(P<0.01).Theinflammatory cell infiltratation much decreased and neutrophils,disapeared in most sections(P<0.001).The goblet cells significantlyincreased(P<0.001);crypt paracrypt abscessor mucosal ulceration was seen(P<0.001).CONCLUSION The rate of cure of ulcerative colitis by moxibustion with herbal medicine beneath is superior to that by SASP. 13Cas9-NG Greatly Expands the Targeting Scope of the Genome-Editing Toolkit by Recognizing NG and Other Atypical PAMs in Rice显示文摘CRISPR technologies enabling precise genome manipulation are valuable for gene function studies and molecular crop breeding. However, the requirement of a protospacer adjacent motif (PAM)y such as NGG and TTN, for Cas protein recognition restricts the selection of targetable genomic loci in practical applications of CRISPR technologies. Recently Cas9-NG, which recognizes a minimal NG PAM, was reported to expand the targeting space of genome editing in human cells, but it remains unclear whether this Cas9 variant can be used in plants. In this study, we evaluated the nuclease activity of Cas9-NG toward various NGN PAMs by targeting endogenous genes in transgenic rice. We found that Cas9-NG edits all NGG, NGA, NGT, and NGC sites with impaired activity, while the gene-edited plants were dominated by monoallelic mutations. Cas9-NG-engineered base editors were then developed and used to generate O s B Z R I gainof- function plants that can not be created by other available Cas9-engineered base editors. Moreover, we showed that a Cas9-NG-based transcriptional activator efficiently upregulated the expression of endogenous target genes in rice. In addition, we discovered that Cas9-NG recognizes NAC, NTG, NTT, and NCG apart from NG PAM. 14Evaluation of high-resolution satellite precipitation products with surface rain gauge observations from Laohahe Basin in northern China显示文摘Three high-resolution satellite precipitation products,the Tropical Rainfall Measuring Mission (TRMM) standard precipitation products 3B42V6 and 3B42RT and the Climate Precipitation Center's (CPC) morphing technique precipitation product (CMORPH),were evaluated against surface rain gauge observations from the Laohahe Basin in northern China.Widely used statistical validation indices and categorical statistics were adopted.The evaluations were performed at multiple time scales,ranging from daily to yearly,for the years from 2003 to 2008.The results show that all three satellite precipitation products perform very well in detecting the occurrence of precipitation events,but there are some different biases in the amount of precipitation.3B42V6,which has a bias of 21%,fits best with the surface rain gauge observations at both daily and monthly scales,while the biases of 3B42RT and CMORPH,with values of 81% and 67%,respectively,are much higher than a normal receivable threshold.The quality of the satellite precipitation products also shows monthly and yearly variation:3B42RT has a large positive bias in the cold season from September to April,while CMORPH has a large positive bias in the warm season from May to August,and they all attained their best values in 2006 (with 10%,50%,and-5% biases for 3B42V6,3B42RT,and CMORPH,respectively).Our evaluation shows that,for the Laohahe Basin,3B42V6 has the best correspondence with the surface observations,and CMORPH performs much better than 3B42RT.The large errors of 3B42RT and CMORPH remind us of the need for new improvements to satellite precipitation retrieval algorithms or feasible bias adjusting methods.Shan-hu JIANG Li-liang REN Bin YONG Xiao-li YANG Lin SHI 2010Water Science and Engineering2010,3,4:17
15Small molecule inhibitors reveal allosteric regulation of USP14 via steric blockade显示文摘The ubiquitin system is important for drug discovery,and the discovery of selective small-molecule inhibitors of deubiquitinating enzymes (DUBs) remains an active yet extremely challenging task. With a few exceptions,previously developed inhibitors have been found to bind the evolutionarily conserved catalytic centers of DUBs,resulting in poor selectivity. The small molecule IU1 was the first-ever specific inhibitor identified and exhibited surprisingly excellent selectivity for USP14 over other DUBs. However,the molecular mechanism for this selectivity was elusive. Herein,we report the high-resolution co-crystal structures of the catalytic domain of USP14 bound to IU1 and three IU1 derivatives. All the structures of these complexes indicate that IU1 and its analogs bind to a previously unknown steric binding site in USP14,thus blocking the access of the C-terminus of ubiquitin to the active site of USP14 and abrogating USP14 activity. Importantly,this steric site in USP14 is very unique,as suggested by structural alignments of USP14 with several known DUB X-ray structures. These results,in conjunction with biochemical characterization,Indicate a coherent steric blockade mechanism for U5P14 inhibition by compounds of the IU series. 17Comparative analysis of GF-1,HJ-1,and Landsat-8 data for estimating the leaf area index of winter wheat显示文摘Using simultaneously collected remote sensing data and field measurements,this study firstly assessed the consistency and applicability of China high-resolution earth observation system satellite 1(GF-1) wide field of view(WFV) camera,environment and disaster monitoring and forecasting satellite(HJ-1) charge coupled device(CCD),and Landsat-8 operational land imager(OLI) data for estimating the leaf area index(LAI) of winter wheat via reflectance and vegetation indices(VIs). The accuracies of these LAI estimates were then assessed through comparison with an empirical model and the PROSAIL radiative transfer model. The effects of radiation calibration,spectral response functions,and spatial resolution on discrepancies in the LAI estimates between the different sensors were also analyzed. The results yielded the following observations:(1) The correlation between reflectance from different sensors is relative good,with the adjusted coefficients of determination(R2) between 0.375 to 0.818. The differences in reflectance are ranging from 0.002 to 0.054. The correlation between VIs from different sensors is high with the R2 between 0.729 and 0.933. The differences in the VIs are ranging from 0.07 to 0.156. These results show the three sensors' images can all be used for cross calibration of the reflectance and VIs.(2) The four VIs from the three sensors are all demonstrated to be highly correlated with LAI(R2 between 0.703 and 0.849). The linear models associated with the 2-band enhanced vegetation index(EVI2),which feature the highest R2(higher than 0.746) and the lowest root mean square errors(RMSE)(less than 0.21),were selected to estimate the winter wheat LAI. The accuracy of the estimated LAI from Landsat-8 was the highest,with the relative errors(RE) of 2.18% and an RMSE of 0.13,while the HJ-1 was the lowest,with the RE of 2.43% and the RMSE of 0.15.(3) The inversion errors in the different sensors' LAI estimates using the PROSAIL model are small. 18SIS accuracy and service performance of the BDS-3 basic system显示文摘On December 27,2018,the basic system of the third-generation BeiDou navigation satellite system(BDS-3)completed the deployment of its constellation of 18 MEO networking satellites as well as the construction of the operation control system(OCS)and began to provide basic navigation services to users worldwide.Compared with BDS-2,BDS-3 aims to offer users better navigation signals and higher precision with a series of new technologies.For example,the spaceborne atomic clock of BDS-3 is upgraded for higher performance,the Ka-band inter-satellite link is adopted for inter-satellite ranging and communication,and new B1C and B2a signals are broadcast in addition to B1I and B3I signals(compatible with BDS-2).In addition,a 9-parameter model based on a spherical harmonic function is employed for ionospheric delay corrections.Using the observation data from 18 satellites of the basic system,this paper conducts a comprehensive evaluation of the pseudorange measurement characteristics,signal-in-space(SIS)accuracy of navigation messages and global service capability of BDS-3.The results indicate that the pseudorange measurement multipath effect and observation noise of BDS-3 satellites are better than those of BDS-2;additionally,with the support of inter-satellite links,the user range error(URE)of the BDS-3 satellite broadcast ephemeris is better than 10 cm,the precision of the broadcast clock parameter is better than 1.5 ns,and the SIS accuracy is better than 0.6 m overall.Different from the traditional Klobuchar model,the BeiDou global broadcast ionospheric delay correction model(BDGIM)can provide ionospheric delay corrections better than 70%for worldwide single-frequency users.The service capability evaluation of the basic system consists mainly of the accuracy improvement of the B1I and B3I signals according to BDS-2 as well as the global positioning accuracy of the new signals.These results prove that the BDS-3 basic system has achieved the design goal;that is,both the horizontal and the vertical global positioning accuracies are better than 10 m(95%).In the future,6 MEO satellites as well as 3 GEO satellites and 3 IGSO satellites for regional enhancement purposes will be deployed for full operation;consequently,BDS-3 will definitely provide a higher SIS accuracy and better service capability.JinPing Chen XiaoGong Hu ChengPan Tang ShanShi Zhou YuFei Yang JunYang Pan Hui Ren YueXin Ma QiuNing Tian Bin Wu Yang Yu 2020Science China(Physics,Mechanics & Astronomy)2020,63,6:14
19Photoperiodism dynamics during the domestication and improvement of soybean显示文摘Soybean(Glycine max) is a facultative short-day plant with a sensitive photoperiod perception and reaction system, which allows it to adjust its physiological state and gene regulatory networks to seasonal and diurnal changes in environmental conditions. In the past few decades, soybean cultivation has spread from East Asia to areas throughout the world. Biologists and breeders must now confront the challenge of understanding the molecular mechanism of soybean photoperiodism and improving agronomic traits to enable this important crop to adapt to geographical and environmental changes. In this review, we summarize the genetic regulatory network underlying photoperiodic responses in soybean. Genomic and genetic studies have revealed that the circadian clock, in conjunction with the light perception pathways, regulates photoperiodic flowering. Here, we provide an annotated list of 844 candidate flowering genes in soybean, with their putative biological functions. Many photoperiod-related genes have been intensively selected during domestication and crop improvement. 20Long-term phosphorus accumulation and agronomic and environmtal critical phosphorus levels in Haplic Luvisol soil, northern China显示文摘Sufficient soil phosphorus(P) content is essential for achieving optimal crop yields,but accumulation of P in the soil due to excessive P applications can cause a risk of P loss and contribute to eutrophication of surface waters.Determination of a critical soil P value is fundamental for making appropriate P fertilization recommendations to ensure safety of both environment and crop production.In this study,agronomic and environmental critical P levels were determined by using linear-linear and linear-plateau models,and two segment linear model,for a maize(Zea mays L.)-winter wheat(Triticum aestivum L.) rotation system based on a 22-yr field experiment on a Haplic Luvisol soil in northern China.This study included six treatments:control(unfertilized),no P(NoP),application of mineral P fertilizer(MinP),MinP plus return of maize straw(MinP+StrP),MinP plus low rate of farmyard swine manure(MinP+L.Man) and MinP plus high rate of manure(MinP+H.Man).Based on the two models,the mean agronomic critical levels of soil Olsen-P for optimal maize and wheat yields were 12.3 and 12.8 mg kg^(-1),respectively.The environmental critical P value as an indicator for P leaching was 30.6 mg Olsen-P kg^(-1),which was 2.4 times higher than the agronomic critical P value(on average 12.5 mg P kg^(-1)).It was calculated that soil Olsen-P content would reach the environmental critical P value in 41 years in the MinP treatment,but in only 5-6years in the two manure treatments.Application of manure could significantly raise soil Olsen-P content and cause an obvious risk of P leaching.In conclusion,the threshold range of soil Olsen-P is from 12.5 to 30.6 mg P kg^(-1) to optimize crop yields and meanwhile maintain relatively low risk of P leaching in Haplic Luvisol soil,northern China.XI Bin ZHAI Li-mei LIU Jian LIU Shen WANG Hong-yuan LUO Chun-yan REN Tian-zhi LIU Hong-bin 2016Journal of Integrative Agriculture2016,15,1:13
