1China National Medical Products Administration approval summary:anlotinib for the treatment of advanced non-small cell lung cancer after two lines of chemotherapy显示文摘Background:On May 8,2018,the China National Medical Products Administration(NMPA)approved anlotinib,an orally administered anti-angiogenesis inhibitor,for the treatment of patients with advanced non-small cell lung can-cer(NSCLC)who have progressed after treatment with two or more lines of prior systemic chemotherapy.Main body of the abstract:China NMPA reviewed and inspected a regional double-blinded,placebo-controlled,Phase III trial comparing the overall survival(OS)of NSCLC patients between the anlotinib and placebo arms.A total of 437 patients were randomized(2:1)to receive either anlotinib(n=294)or placebo(n=143)once daily on a 2-week on and 1-week off schedule.Patients with epidermal growth factor receptor(EGFR)or activating anaplastic lymphoma kinase(ALK)genomic tumor aberrations should have disease progression on NMPA-approved therapy.Anlotinib is the first NMPA-approved drug for patients with advanced NSCLC who have progressed on at least two lines of prior systemic chemotherapies in China.The approval was based on a statistically and clinically significant improvement in median OS with anlotinib(9.46 months)compared with placebo[6.37 months;hazard ratio(HR])=0.70,95%confidence interval(CI)=0.55-0.89;two-sided log-rank P=0.002].The confirmed objective response rate(ORR)was 9.2%in the anlotinib arm and 0.7%in the placebo arm.The median duration of response(DoR)was 4.83 months,with a 95%CI of 3.31-6.97 months.The toxicity profile of anlotinib was consistent with that of known anti-angiogenesis inhibitors.Common adverse drug reactions(ADRs)in anlotinib-treated patients included hypertension(67.4%),hand-foot syndrome(43.9%),hemoptysis(14.0%),thyroid stimulating hormone(TSH)elevation(46.6%),and corrected QT interval(QTc)prolongation(26.2%).Short conclusion:Anlotinib demonstrated a clinically significant OS prolongation as a novel therapeutic option for advanced or metastatic NSCLC following at least two lines of chemotherapy.Ming Zhou Xiaoyuan Chen Hong Zhang Lin Xia Xin Tong Limin Zou Ruimin Hao Jianhong Pan Xiao Zhao Dongmei Chen Yuanyuan Song Yueli Qi Ling Tang Zhifang Liu Rong Gao Yuankai Shi Zhimin Yang 2019Cancer Communications2019,39,1:35
2Human umbilical cord-derived mesenchymal stem cell therapy in patients with COVID-19:a phase 1 clinical trial显示文摘No effective drug treatments are available for coronavirus disease 2019(COVID-19).Host-directed therapies targeting the underlying aberrant immune responses leading to pulmonary tissue damage,death,or long-term functional disability in survivors require clinical evaluation.We performed a parallel assigned controlled,non-randomized,phase 1 clinical trial to evaluate the safety of human umbilical cord-derived mesenchymal stem cells(UC-MSCs)infusions in the treatment of patients with moderate and severe COVID-19 pulmonary disease.The study enrolled 18 hospitalized patients with COVID-19(n=9 for each group).The treatment group received three cycles of intravenous infusion of UC-MSCs(3×107 cells per infusion)on days 0,3,and 6.Both groups received standard COVID-treatment regimens.Adverse events,duration of clinical symptoms,laboratory parameters,length of hospitalization,serial chest computed tomography(CT)images,the PaO2/FiO2 ratio,dynamics of cytokines,and IgG and IgM anti-SARS-CoV-2 antibodies were analyzed.No serious UC-MSCs infusion-associated adverse events were observed.Two patients receiving UC-MSCs developed transient facial flushing and fever,and one patient developed transient hypoxia at 12 h post UC-MSCs transfusion.Mechanical ventilation was required in one patient in the treatment group compared with four in the control group.All patients recovered and were discharged.Our data show that intravenous UC-MSCs infusion in patients with moderate and severe COVID-19 is safe and well tolerated.Phase 2/3 randomized,controlled,double-blinded trials with long-term follow-up are needed to evaluate the therapeutic use of UC-MSCs to reduce deaths and improve long-term treatment outcomes in patients with serious COVID-19.Fanping Meng Ruonan Xu Siyu Wang Zhe Xu Chao Zhang Yuanyuan Li Tao Yang Lei Shi Junliang Fu Tianjun Jiang Lei Huang Peng Zhao Xin Yuan Xing Fan Ji-Yuan Zhang Jinwen Song Dawei Zhang Yanmei Jiao Limin Liu Chunbao Zhou Markus Maeurer Alimuddin Zumla Ming Shi Fu-Sheng Wang 2020Signal Transduction and Targeted Therapy2020,5,1:16
3Potassium Channels: A Potential Therapeutic Target for Parkinson's Disease显示文摘The pathogenesis of the second major neurodegenerative disorder, Parkinson's disease(PD), is closely associated with the dysfunction of potassium(K^+ ) channels. Therefore, PD is also considered to be an ion channel disease or neuronal channelopathy. Mounting evidence has shown that K^+ channels play crucial roles in the regulations of neurotransmitter release, neuronal excitability, and cell volume. Inhibition of K^+ channels enhances the spontaneous firing frequency of nigral dopamine(DA)neurons, induces a transition from tonic firing to burst discharge, and promotes the release of DA in the striatum.Recently, three K^+ channels have been identified to protect DA neurons and to improve the motor and non-motor symptoms in PD animal models: small conductance(SK)channels, A-type K^+ channels, and KV7/KCNQ channels.In this review, we summarize the physiological and pharmacological effects of the three K^+ channels. 4High-performance silicon−graphene hybrid plasmonic waveguide photodetectors beyond 1.55μm显示文摘Graphene has attracted much attention for the realization of high-speed photodetection for silicon photonics over a wide wavelength range.However,the reported fast graphene photodetectors mainly operate in the 1.55μm wavelength band.In this work,we propose and realize high-performance waveguide photodetectors based on bolometric/photoconductive effects by introducing an ultrathin wide silicon−graphene hybrid plasmonic waveguide,which enables efficient light absorption in graphene at 1.55μm and beyond.When operating at 2μm,the present photodetector has a responsivity of ~70 mA/W and a setup-limited 3 dB bandwidth of >20 GHz.When operating at 1.55μm,the present photodetector also works very well with a broad 3 dB bandwidth of >40 GHz(setup-limited)and a high responsivity of ~0.4 A/W even with a low bias voltage of−0.3 V.This work paves the way for achieving highresponsivity and high-speed silicon-graphene waveguide photodetection in the near/mid-infrared ranges,which has applications in optical communications,nonlinear photonics,and on-chip sensing.Jingshu Guo Jiang Li Chaoyue Liu Yanlong Yin Wenhui Wang Zhenhua Ni Zhilei Fu Hui Yu Yang Xu Yaocheng Shi Yungui Ma Shiming Gao Limin Tong Daoxin Dai 2020Light(Science & Applications)2020,9,1:10

6Catalytic properties of Cu-Co catalysts supported on HNO_3-pretreated CNTs for higher-alcohol synthesis显示文摘HNO 3 -pretreated CNTs were employed as supports, and a special ultrasound-assisted impregnation method was designed to prepare supported Cu-Co catalysts for higher-alcohol synthesis from syngas. The catalysts used in this work were characterized by N 2 adsorption-desorption, TEM, XRD, H 2 -TPR, CO-TPD techniques. It was found that the pre-treatment procedure of CNTs remarkably promoted the catalytic properties of the Cu-Co/CNTs catalysts. For the Cu-Co catalyst supported on CNTs pre-treated by 68 wt% HNO 3 , some active components were introduced into the CNTs channels, their dispersions and the amount of strongly adsorbed CO-species were improved. 8Catalytic performance of Zr-doped CuO-CeO2 oxides for CO selective oxidation in H2-rich stream显示文摘Zr-doped CuO-CeO2 catalysts for CO selective oxidation were designed and prepared by the hydrothermal method and coprecipitation. The experimental samples were characterized by means of N2 adsorption-desorption isotherms, powder X-ray diffraction, temperature-programmed reduction and Xray photoelectron spectroscopy. It is observed that the catalyst prepared by hydrothermal method exhibits larger specific surface area, smaller crystalline size and higher dispersion of active components compared with those of the catalyst obtained by coprecipitation. Meanwhile, redox properties of copper oxide are improved significantly and highly dispersed copper species providing CO oxidation sites are present on the surface. Furthermore, adsorptive centers of CO and active oxygen species increase on the copper-ceria interfaces. The Zr-doped CuO-CeO2 catalyst prepared by hydrothermal method possesses superior catalytic activity and selectivity for selective oxidation of CO at low temperature compared with those of the sample prepared by coprecipitation. 10Uncommon EGFR mutations in a cohort of Chinese NSCLC patients and outcomes of first-line EGFR-TKIs and platinum-based chemotherapy显示文摘Objective: Data on the clinical activity of epidermal growth factor receptor(EGFR) tyrosine kinase inhibitors(TKIs) in patients with non-small-cell lung cancer(NSCLC) and uncommon EGFR mutations remain insufficient.This study aimed to investigate the effect of first-line EGFR-TKIs or platinum-based chemotherapy in NSCLC patients with uncommon EGFR mutations.Methods: We retrospectively enrolled 504 patients with EGFR-mutant NSCLC.The clinical characteristics and treatment outcomes were collected and compared between patients with common and uncommon EGFR-mutant NSCLC.Results: Seventy patients(13.9%) harboring uncommon EGFR mutations were included.Thirty of these patients received EGFR-TKIs and 40 received platinum-based chemotherapy as first-line therapy.The objective response rate(ORR) and median progression-free survival(m PFS) of patients treated with TKIs in the uncommon mutation group was significantly inferior to that in the common mutation group(ORR: 23.3% vs.51.8%,P=0.003; m PFS:7.1 vs.10.9 months,P<0.001).In the uncommon group,m PFS was similar between first-line EGFR-TKIs treatment and platinum-based chemotherapy(7.1 vs.6.1 months,P=0.893).In patients with EGFR G719 X or L861 Q mutations,the m PFS was longer in the first-line EGFR-TKIs treatment group than in the chemotherapy group,but the difference was not statistically significant(G719 X: 8.2 vs.5.8 months,P=0.061; L861 Q: 7.6 vs.4.1 months,P=0.872).Multivariate analyses identified adenocarcinoma(P=0.003) as the independent predictive factor for PFS in patients with uncommon EGFR mutations who were treated with first-line EGFR-TKIs.Conclusions: The current study demonstrated that the effect of first-line EGFR-TKIs was similar to that of platinum-based chemotherapy in patients with uncommon EGFR-mutant NSCLC.Adenocarcinoma was the independent predictive factor for PFS in uncommon EGFR-mutant NSCLC patients treated with first-line EGFRTKIs.Jinpeng Shi Hui Yang Tao Jiang Xuefei Li Chao Zhao Limin Zhang Sha Zhao Xiaozhen Liu Yijun Jia Yan Wang Lei Xi Shijia Zhang Chunxia Su Shengxiang Ren Caicun Zhou 2017Chinese Journal of Cancer Research2017,29,6:6

Complex 3 exhibits a mononuclear structure. Complex 4 owns binuclear symmetry units, which were bridged via the dba ligands forming a 2D framework. 12超深井高速率连续波MWD信道传输系统(英文)显示文摘At present,the MWD systems continuous wave mud pulse telemetry transmit is not faster than 1or 2bits/s from deep wells containing highly attenuative mud,because positive pulsers create strong signals but large axial flow forces impede fast reciprocation while mud sirens provide high data rates without higher signal strength.A high-data-rate system providing 10bits/s and operable up to30 000 ft is described,which creates strong source signals by using downhole constructive wave interference in two novel ways.Telemetry schemes,frequencies and pulser locations in the MWD drill collar are selected for positive wave phasing,and sirens-in-series are used to create additive signals——both without incurring power and erosion penalties.The positions normally occupied by pulsers and turbines are reversed to minimize times required for constructive interference and modulation.A system design approach is undertaken,e.g.,strong source signals are augmented with new multiple-transducer surface signal processing methods to remove mudpump noise and signal reflections at both pump and desurger,and mud,bottomhole assembly and drill pipe properties,to the extent possible in practice,are controlled to reduce signal attenuation.The results of detailed acoustic modeling in realistic drilling telemetry channels are described,and short and long wind tunnels for signal strength,torque,erosion and jamming testing,and telemetry and signal processing evaluation,respectively,are introduced.Special scaling methods are developed to extrapolate cost-effective wind tunnel test results to real muds flowing at any downhole speed.New siren concept prototype hardware and also typical acoustic test results are also given.Wilson CHIN SU Yinao SHENG Limin LI Lin BIAN Hailong SHI Rong 2014测井技术2014,38,6:3

19Single-atom Sn-Zn pairs in CuO catalyst promote dimethyldichlorosilane synthesis显示文摘Single-atom catalysts are of great interest because they can maximize the atom-utilization efficiency and generate unique catalytic properties;however,much attention has been paid to single-site active components,rarely to catalyst promoters.Promoters can significantly affect the activity and selectivity of a catalyst,even at their low concentrations in catalysts.In this work,we designed and synthesized CuO catalysts with atomically dispersed co-promoters of Sn and Zn.When used as the catalyst in the Rochow reaction for the synthesis of dimethyldichlorosilane,this catalyst exhibited much-enhanced activity,selectivity and stability compared with the conventional CuO catalysts with promoters in the form of nanoparticles.Density functional theory calculations demonstrate that single-atomic Sn substitution in the CuO surface can enrich surface Cu vacancies and promote dispersion of Zn to its atomic levels.Sn and Zn single sites as the co-promoters cooperatively generate electronic interaction with the CuO support,which further facilitates the adsorption of the reactant molecules on the surface,thereby leading to the superior catalytic performance.Qi Shi Yongjun Ji Wenxin Chen Yongxia Zhu Jing Li Hezhi Liu Zhi Li Shubo Tian Ligen Wang Ziyi Zhong Limin Wang Jianmin Ma Yadong Li Fabing Su 2020National Science Review2020,7,3:2
