| 1 | Cropland redistribution to marginal lands undermines environmental sustainability显示文摘Cropland redistribution to marginal land has been reported worldwide;however,the resulting impacts on environmental sustainability have not been investigated sufficiently.Here we investigated the environmental impacts of cropland redistribution in China.As a result of urbanization-induced loss of high-quality croplands in south China(~8.5 t ha^(-1)),croplands expanded to marginal lands in northeast(~4.5 t ha^(-1)) and northwest China(~2.9 t ha^(-1)) during 1990-2015 to pursue food security.However,the reclamation in these low-yield and ecologically vulnerable zones considerably undermined local environmental sustainability,for example increasing wind erosion(+3.47%),irrigation water consumption(+34.42%),fertilizer use(+20.02%) and decreasing natural habitats(-3.11%).Forecasts show that further reclamation in marginal lands per current policies would exacerbate environmental costs by 2050.The future cropland security risk will be remarkably intensified because of the conflict between food production and environmental sustainability.Our research suggests that globally emerging reclamation of marginal lands should be restricted and crop yield boost should be encouraged for both food security and environmental benefits. | Wenhui Kuang Jiyuan Liu Hanqin Tian Hao Shi Jinwei Dong Changqing Song Xiaoyong Li Guoming Du Yali Hou Dengsheng Lu Wenfeng Chi Tao Pan Shuwen Zhang Rafiq Hamdi Zherui Yin Huimin Yan Changzhen Yan Shixin Wu Rendong Li Jiuchun Yang Yinyin Dou Wenbin Wu Liqiao Liang Bao Xiang Shiqi Yang | 2022 | National Science Review2022,9,1: | 13 |
| 2 | Expression of human cytomegalovirus components in the brain tissues of patients with Rasmussen's encephalitis显示文摘Rasmussen's encephalitis(RE) is a rare and severe progressive epileptic syndrome with unknown etiology. Infection by viruses, including human cytomegalovirus(HCMV), has been speculated to be a potential trigger for RE. However, no viral antigens have been detected in the brains of patients with RE; thus, a possible clinical linkage between viral infections and RE has not been firmly established. In this study, we evaluated the expression of HCMV pp65 antigen in brain sections from 26 patients with RE and 20 non-RE patients by immunohistochemistry and in situ hybridization, and assessed the associations between HCMV infection and clinical parameters.Elevated expression of HCMV pp65 protein and DNA was observed in 88.5%(23/26) and 69.2%(18/26) of RE cases, respectively. In the non-RE group, HCMV pp65 antigen was detected only in two cases(10%), both of which were negative for DNA staining. Additionally, the intensity of HCMV pp65 staining was correlated with a shorter duration of the prodromal stage, younger age of seizure onset, and more severe unilateral cortical atrophy. Elevated expression of HCMV pp65 was observed in RE brain tissue and was correlated with the clinical features of RE disease. In summary, our results suggested that HCMV infection may be involved in the occurrence and progression of RE disease. | 3 | Application and future perspective of CRISPR/Cas9 genome editing in fruit crops显示文摘Fruit crops, including apple, orange, grape,banana, strawberry, watermelon, kiwifruit and tomato, not only provide essential nutrients for human life but also contribute to the major agricultural output and economic growth of many countries and regions in the world. Recent advancements in genome editing provides an unprecedented opportunity for the genetic improvement of these agronomically important fruit crops. Here, we summarize recent reports of applying CRISPR/Cas9 to fruit crops,including efforts to reduce disease susceptibility, change plant architecture or flower morphology, improve fruit quality traits, and increase fruit yield. We discuss challenges facing fruit crops as well as new improvements and platforms that could be used to facilitate genome editing in fruit crops, including d Cas9-base-editing to introduce desirable alleles and heat treatment to increase editing efficiency. In addition, we highlight what we see as potentially revolutionary development ranging from transgene-free genome editing to de novo domestication of wild relatives. Without doubt, we now see only the beginning of what will eventually be possible with the use of the CRISPR/Cas9 toolkit. | 4 | Direct Reduction of Solid Fe_2O_3 in Molten CaCl_2 by Potentially Green Process显示文摘Sintered (300°C) porous pellets of Fe2O3 were electrolyzed to Fe in molten CaCl2 (800–900°C) under argon at 1.8–3.2 V for 2–20 h. The laboratory scale experiments show that it was a potentially direct green method to produce Fe powder. At lower electrolysis voltage (<2.2 V), higher current efficiency (>90%) and smaller energy consumption (~3.0 kWh/kg) can be obtained. When the electrolysis voltage was above 2.4 V, the deposition of metal Ca from the salt lowered the current efficiency and increased the energy consumption. The electrolysis voltage also had effects on the micrographs of the reduced powder. The cubic particles can be seen in the products at the voltage lower than 2.2 V; when the voltage was higher than 2.2 V, it was nodular. The reduction proceeds at the cathode in two steps, i.e., from Fe2O3 to FeO and then to Fe. The oxygen emits at the anode. | 6 | Detection of EBV and HHV6 in the Brain Tissue of Patients with Rasmussen's Encephalitis显示文摘Rasmussen's encephalitis (RE) is a rare pediatric neurological disorder, and the exact etiology is not clear. Viral infection may be involved in the pathogenesis of RE, but conflicting results have reported. In this study, we evaluated the expression of both Epstein-Barr virus (EBV) and human herpes virus (HHV) 6 antigens in brain sections from 30 patients with RE and16 control individuals by immunohistochemistry. In the RE group, EBV and HHV6 antigens were detected in 56.7% (17/30) and 50% (15/30) of individuals, respectively. In contrast, no detectable EBV and HHV6 antigen expression was found in brain tissues of the control group. The co-expression of EBV and HHV6 was detected in 20.0% (6/30) of individuals. In particular, a 4-year-old boy had a typical clinical course, including a medical history of viral encephalitis,intractable epilepsy, and hemispheric atrophy. The co-expression of EBV and HHV6 was detected in neurons and astrocytes in the brain tissue, accompanied by a high frequency of CD8?T cells. | 9 | Laminar Distribution of Neurochemically-Identified Interneurons and Cellular Co-expression of Molecular Markers in Epileptic Human Cortex显示文摘Inhibitory GABAergic interneurons are fundamental elements of cortical circuits and play critical roles in shaping network activity. Dysfunction of interneurons can lead to various brain disorders, including epilepsy,schizophrenia, and anxiety. Based on the electrophysiological properties, cell morphology, and molecular identity,interneurons could be classified into various subgroups. In this study, we investigated the density and laminar distribution of different interneuron types and the coexpression of molecular markers in epileptic human cortex.We found that parvalbumin(PV) and somatostatin(SST)neurons were distributed in all cortical layers except layer I, while tyrosine hydroxylase(TH) and neuropeptide Y(NPY) were abundant in the deep layers and white matter.Cholecystokinin(CCK) neurons showed a high density in layers IV and VI. Neurons with these markers constituted*7.2%(PV), 2.6%(SST), 0.5%(TH), 0.5%(NPY), and4.4%(CCK) of the gray-matter neuron population. Doubleand triple-labeling revealed that NPY neurons were also SST-immunoreactive(97.7%), and TH neurons were more likely to express SST(34.2%) than PV(14.6%). A subpopulation of CCK neurons(28.0%) also expressed PV, but none contained SST. | 10 | Elevated expression of EBV and TLRs in the brain is associated with Rasmussen's encephalitis显示文摘Rasmussen's encephalitis(RE) is a rare pediatric neurological disorder, the etiology of which remains unclear. It has been speculated that the immunopathogenesis of RE involves damage to neurons, which eventually leads to the occurrence of RE. Viral infection may be a critical factor in triggering RE immunopathogenesis. In this study, we analyzed the expression of Epstein-Barr virus(EBV) antigens as well as of Toll-like receptor 3(TLR3), TLR9, and downstream adapter TIRdomain-containing adapter-inducing interferon-β(TRIF) in the brain tissues of 26 patients with RE and 16 control individuals using immunohistochemistry(IHC). In the RE group, EBV antigens were detected in 53% of individuals at various expression levels. In contrast, there was no detectable EBV antigen expression in control brain tissues. Moreover, we found marked increases in the expression of TLR3, TLR9, and TRIF in the brain tissues of RE patients compared with levels in the control group. Furthermore, among RE cases, EBV expression and high TLR3 expression were associated with more severe brain atrophy. | 11 | Transforming the Loess Plateau of China显示文摘This paper aims to show the importance of land consolidation in transforming the Loess Plateau of China. The paper comprehensively analyzes how over recent decades the Grain for Green Project and Gully Land Consolidation Project jointly transformed the ecology and landscape of the Loess Plateau and the livelihood of its residents. The findings show that these two projects have achieved a balance between green protection, new land creation, and improved food security and livelihood of local people in the hilly areas of China. The paper points out that the successful transformation of the Plateau lies in a holistic approach incorporating various components of the human and natural systems. | 12 | Circular BANP knockdown inhibits the malignant progression of residual hepatocellular carcinoma after insufficient radiofrequency ablation显示文摘Background:Circular RNAs(circRNAs)are endogenous non-coding RNAs,some of which have pathological roles.The current study aimed to explore the role of circRNA BTG3-associated nuclear protein(circ-BANP)binding with let-7f-5p and its regulation of the toll-like receptor 4(TLR4)/signal transducer and activator of transcription 3(STAT3)signaling pathway in residual hepatocellular carcinoma(HCC)after insufficient radiofrequency ablation(RFA).Methods:Circ-BANP,let-7f-5p,and TLR4 expressions in HCC samples were assessed using reverse transcription-quantitative polymerase chain reaction(RT-qPCR)and Western blotting.Bioinformatics prediction,RNA pull-down assay,and dual luciferase reporter gene assay were used to analyze the relationships among circ-BANP,let-7f-5p,and TLR4.Huh7 cells were used to generate an in vitro model of residual HCC,defined as Huh7-H cells,which were transfected with either a plasmid or the sequence of circ-BANP,let-7f-5p,or TLR4.Expression of circ-BANP,let-7f-5p,and TLR4 mRNA was determined by RT-qPCR.TLR4,STAT3,p-STAT3,vascular endothelial growth factor A,vascular endothelial growth factor receptor-2,and epithelialmesenchymal transformation(EMT)-related factors proteins were determined by Western blotting.Cell proliferation was determined by cell counting kit-8 and 5-Ethynyl-2’-deoxyuridine(EdU)assay and cell migration and invasion by Transwell assay.Animal studies were performed by inducing xenograft tumors in nude mice.Results:Circ-BANP and TLR4 mRNAs were upregulated in HCC tissues(the fold change for circ-BANP was 1.958 and that for TLR4 was 1.736 relative to para-tumors)and expression further increased following insufficient RFA(fold change for circBANP was 2.407 and that of TLR4 was 2.224 relative to para-tumors).Expression of let-7f-5p showed an opposite tendency(fold change for let-7f-5p in HCC tissues was 0.491 and that in tumors after insufficient RFA was 0.300 relative to paratumors).Competitive binding of circ-BANP to let-7f-5p was demonstrated and TLR4 was identified as a target of let-7f-5p(P<0.01).Knockdown of circ-BANP or elevation of let-7f-5p expression inhibited the TLR4/STAT3 signaling pathway,proliferation,invasion,migration,angiogenesis,and EMT in Huh7 and Huh7-H cells(P<0.01).The effects induced by circBANP knockdown were reversed by let-7f-5p inhibition.Overexpression of TLR4 reversed the impact of let-7f-5p upregulation on the cells(P<0.01).Silencing of circ-BANP inhibited the in vivo growth of residual HCC cells after insufficient RFA(P<0.01).Conclusions:Knockdown of circ-BANP upregulated let-7f-5p to inhibit proliferation,migration,and EMT formation in residual HCC remaining after insufficient RFA.Effects occur via regulation of the TLR4/STAT3 signaling pathway. | Guoming Li Jian Kong Shuying Dong Haigang Niu Shilun Wu Wenbing Sun | 2022 | Chinese Medical Journal2022,,13: | 2 |

| 16 | Capacitive Imaging Technique for the Inspection of Composite Sucker Rod显示文摘Composite sucker rod has been extensively used due to its high strength, light weight and corrosion resistive nature. However, such composite sucker rod is diffcult for conventional non-destructive evaluation(NDE) techniques to inspect because of its complex material and/or structure. It is thus useful to embark research on developing novel NDE technique to comply the inspection requirement. This work demonstrates the feasibility of using the capacitive imaging(CI) technique for the inspection of composite sucker rod. Finite element(FE) models were constructed in COMSOL to simulate the detection of defects in the glass-fiber layer and on the carbon core surface. An FE Model based inversion method is proposed to obtain the profile of the carbon core. Preliminary CI experimental results are then presented, including the detection of surface wearing defect in the glass-fiber layer, and obtaining the profile of the carbon core. A set of accelerated aging experiments were also carried out and the results indicate that the CI technique is potentially useful in evaluating the ageing status of such composite sucker rod. | 17 | Review of flash sintering with strong electric field显示文摘Flash sintering is a novel field‐assisted sintering technology for ceramics that allows a dramatic reduction in processing time and temperature.Since 2010,when flash sintering was first reported,it has been a focus of research interest in the field of materials science.Recent study results have confirmed that the sintering temperature decreases with the strength of the applied electric field;for some ceramics,the sintering temperature can be lowered even to room temperature.This represents an innovative breakthrough for ceramic sintering at ultra‐low temperatures.However,once the electric field strength in flash sintering is increased,new questions and challenges arise,such as whether or not partial discharge occurs in the green body if under electro‐thermal coupling stress,how to quantitatively analyse the impact of discharge on sintering,and,moreover,whether the sintering mechanism under a strong electric field is consistent with that under a weak electric field.These research questions require knowledge of partial discharge detection,dielectric theory,and other topics that are beyond the scope of materials science.To address this need,this review summarises the work carried out in flash sintering with a strong electric field from the perspective of high voltage and insulation technology.First,the flash sintering process and mechanism are briefly introduced.Then,the published literature on flash sintering with a strong electric field for various ceramic materials is summarised in depth.In addition,experimental phenomena that are observed in flash sintering with a strong electric field,such as flash and blackening,are discussed.Finally,some suggestions for future work are presented.It is anticipated that the knowledge gap between different areas of study can be filled by this review. | Hongyang Zhou Xiang Li Yuchen Zhu Jieming Liu Angxuan Wu Guoming Ma Xilin Wang Zhidong Jia Liming Wang | 2022 | High Voltage2022,7,1: | 2 |
| 18 | An oral second-generation proteasome inhibitor oprozomib significantly inhibits lung cancer in a p53 independent manner in vitro显示文摘The destruction of proteins via the ubiquitin-proteasome system is amulti-step,complex process involving polyubiquitination of substrate proteins,follow ed by proteolytic degradation by the m acrom olecular 26S proteasome complex.Inhibitors of the proteasome prom ote the accum ulation of proteins that are deleterious to cell survival and are prom ising anticancer agents.Oprozomib(OPZ),an oral second-generation proteasome inhibitor,has been shown to inhibit the grow th of several cancers in preclinical and clinical trials,including m ultiple m yelom a and head and neck cancers,but its effects on lung cancer has not yet been determ ined.In this study,we evaluated the inhibitory effects of OPZ on lung cancer cell lines in vitro.The results showed that OPZ significantly suppressed cell proliferation and strongly induced apoptosis in both tested lung cancer cells independent of p53 expression.OPZ was able to cause obvious caspase 3 and PARP cleavages and stabilize p53 and its transcriptional targets p21,PUMA,and Noxa.Moreover,OPZ was capable of sensitizing lung cancer cells to the conventional chem otherapeutic drug cisplatin.Our study provides preclinical data and sheds light on the potential applications of proteasome inhibitor OPZ in lung cancer treatment. | Hongge Zhu Tianhai Wang Zhou Xin Yiyi Zhan Guoming Gu Xiaoqin Li Xiuli Wang Shune Yang Chunling Liu | 2019 | Acta Biochimica et Biophysica Sinica2019,51,10: | 2 |
