| 1 | Effect of N and P addition on soil organic C potential mineralization in forest soils in South China显示文摘Atmospheric nitrogen deposition is at a high level in some forests of South China. The effects of addition of exogenous N and P on soil organic carbon mineralization were studied to address: (1) if the atmospheric N deposition promotes soil C storage through decreasing mineralization; (2) if the soil available P is a limitation to organic carbon mineralization. Soils (0-10 cm) was sampled from monsoon evergreen broad-leaved forest (MEBF), coniferous and broad-leaved mixed forest (CBMF), and Pinus massoniana forest (PMF) in Dinghushan Biosphere Reserve (located in Guangdong Province, China). The soils were incubated at 25°C for 45 weeks, with addition of N (NH4NO3 solution) or P (KH2PO4 solution). CO2-C emission and the inorganic N (NH4+-N and NO3?-N) of the soils were determined during the incubation. The results showed that CO2-C emission decreased with the N addition. The addition of P led to a short-term sharp increase in CO2 emission after P application, and the responses of CO2-C evolution to P addition in the later period of incubation related to forest types. Strong P inhibition to CO2 emission occurred in both PMF and CBMF soils in the later incubation. The two-pool kinetic model was fitted well to the data for C turnover in this experiment. The model analysis demonstrated that the addition of N and P changed the distribution of soil organic C between the labile and recalcitrant pool, as well as their mineralization rates. In our experiment, soil pH can not completely explain the negative effect of N addition on CO2-C emission. The changes of soil inorganic N during incubation seemed to support the hypothesis that the polymerization of added nitrogen with soil organic compound by abiotic reactions during incubation made the added nitrogen retard the soil organic carbon mineralization. | 3 | Nitrogen Deposition and Its Spatial Pattern in Main Forest Ecosystems along North-South Transect of Eastern China显示文摘A continuous three-year observation(from May 2008 to April 2011)was conducted to characterize the spatial variation of dissolved inorganic nitrogen(DIN)deposition at eight main forest ecosystems along the north-south transect of eastern China(NSTEC).The results show that both throughfall DIN deposition and bulk DIN deposition increase from north to south along the NSTEC.Throughfall DIN deposition varies greatly from 2.7 kg N/(ha·yr)to 33.0 kg N/(ha·yr),with an average of 10.6 kg N/(ha·yr),and bulk DIN deposition ranges from 4.1 kg N/(ha·yr)to 25.4 kg N/(ha·yr),with an average of 9.8 kg N/(ha·yr).NH4+-N is the dominant form of DIN deposition at most sampling sites.Additionally,the spatial variation of DIN deposition is controlled mainly by precipitation.Moreover,in the northern part of the NSTEC,bulk DIN deposition is 17%higher than throughfall DIN deposition,whereas the trend is opposite in the southern part of the NSTEC.The results demonstrate that DIN deposition would likely threaten the forest ecosystems along the NSTEC,compared with the critical loads(CL)of N deposition,and DIN deposition in this region is mostly controlled by agricultural activities rather than industrial activities or transportation. | ZHAN Xiaoyun YU Guirui HE Nianpeng FANG Huajun JIA Bingrui ZHOU Mei WANG Chuankuan ZHANG Junhui ZHAO Guangdong WANG Silong LIU Yunfen YAN Junhua | 2014 | Chinese Geographical Science2014,24,2: | 10 |
| 4 | Application and future perspective of CRISPR/Cas9 genome editing in fruit crops显示文摘Fruit crops, including apple, orange, grape,banana, strawberry, watermelon, kiwifruit and tomato, not only provide essential nutrients for human life but also contribute to the major agricultural output and economic growth of many countries and regions in the world. Recent advancements in genome editing provides an unprecedented opportunity for the genetic improvement of these agronomically important fruit crops. Here, we summarize recent reports of applying CRISPR/Cas9 to fruit crops,including efforts to reduce disease susceptibility, change plant architecture or flower morphology, improve fruit quality traits, and increase fruit yield. We discuss challenges facing fruit crops as well as new improvements and platforms that could be used to facilitate genome editing in fruit crops, including d Cas9-base-editing to introduce desirable alleles and heat treatment to increase editing efficiency. In addition, we highlight what we see as potentially revolutionary development ranging from transgene-free genome editing to de novo domestication of wild relatives. Without doubt, we now see only the beginning of what will eventually be possible with the use of the CRISPR/Cas9 toolkit. | 5 | A Short Text Classification Algorithm Based on Semantic Extension显示文摘A semantic-extension-based algorithm for short texts is proposed, by involving the Word2vec and the LDA model, to improve the performance of classification,which is frequently deteriorated by semantic dependencies and scarcity of features. For every keyword within a short text, weighted synonyms and related words can be generated by the Word2Vec and LDA model, respectively,and subsequently be inserted to extend the short text to a reasonable length. We not only have established a criterion by means of similarity estimation to determine whether a sentence should be extended, we designed a scheme to choose the number of extended words. The extended text will be classified. | 9 | Self-assembled PEG-b-PDPA-b-PGEM copolymer nanoparticles as protein antigen delivery vehicles to dendritic cells:preparation,characterization and cellular uptake显示文摘Antigen uptake by dendritic cells(DCs)is a key step for initiating antigen-specific T cell immunity.In the present study,novel synthetic polymeric nanoparticles were prepared as antigen delivery vehicles to improve the antigen uptake by DCs.Well-defined cationic and acid-responsive copolymers,monomethoxy poly(ethylene glycol)-block-poly(2-(diisopropyl amino)ethyl methacrylate)-block-poly(2-(guanidyl)ethyl methacrylate)(mPEG-b-PDPA-b-PGEM,PEDG)were synthesized by reversible addition-fragmentation chain transfer polymerization of 2-(diisopropylamino)ethyl methacrylate and N-(tert-butoxycarbonyl)amino ethyl methacrylate monomers,followed by deprotection of tert-butyl protective groups and guanidinylation of obtained primary amines.1H NMR,13C NMR and GPC results indicated the successful synthesis of well-defined PEDG copolymers.PEDG copolymers could self-assemble into nanoparticles in aqueous solution,which were of cationic surface charges and showed acid-triggered disassembly contributed by PGEM and PDPA moieties,respectively.Significantly,PEDG nanoparticles could effectively condense with negatively charged model antigen ovalbumin(OVA)to form OVA/PEDG nanoparticle formulations with no influence on its secondary and tertiary structures demonstrating by far-UV circular dichroism and UV-vis spectra.In vitro antigen cellular uptake by bone marrow DCs(BMDCs)indicated using PEDG nanoparticles as antigen delivery vehicles could significantly improve the antigen uptake efficiency of OVA compared with free OVA or the commercialized Alum adjuvant.Moreover,as the surface cationic charges of OVA/PEDG nanoparticle formulations reduced,the uptake efficiency decreased correspondingly.Collectively,our work suggests that guanidinylated,cationic and acid-responsive PEDG nanoparticles represent a new kind of promising antigen delivery vehicle to DCs and hold great potential to serve as immunoadjuvants in the development of vaccines. | Pan Li Junhui Zhou Pingsheng Huang Chuangnian Zhang Weiwei Wang Chen Lia Deling Kong | 2017 | Regenerative Biomaterials2017,4,1: | 2 |
| 10 | Generation of Nb-enriched mafic rocks and associated adakitic rocks from the southeastern Central Asian Orogenic Belt:Evidence of crust-mantle interaction显示文摘Melting of subducting oceanic lithosphere and associated melt-mantle interactions in convergent plate margins require specific geodynamic environment that allows the oceanic slab to be abnormally heated.Here we focus on the Early Mesozoic mafic rocks and granite porphyry,which provide insights into slab melting processes associated with final closure of the Paleo-Asian Ocean.The granite porphyry samples are calc-alkaline and distinguished by high Sr contents,strong depletion of heavy rare earth elements,resulting in high(La/Yb);and Sr/Y ratios,and negligible Eu anomalies.Based on their high Na_(2)O and Mg O,low K_(2)O contents,positiveε_(Hf)(t)andε_(Nd)(t)and low(^(87)Sr/^(86)Sr)ivalues,we propose that the granite porphyry was likely derived from partial melting of subducting Paleo-Asian oceanic crust.The Nb-enriched mafic rocks are enriched in Rb,Th,U,Pb and K,and depleted in Nb,Ta,Ba,P and Ti,corroborating a subduction-related origin.Their heterogeneous Sr-Nd-Hf-O isotopic compositions and other geochemical features suggest that they were likely derived from partial melting of peridotitic mantle wedge interacted with oceanic slab-derived adakitic melts.Trace element and isotope modeling results and elevated zirconδ^(18)O values suggest variable subducting sediments input into the mantle wedge,dominated by terrigenous sediments.Synthesizing the widely-developed bimodal rock associations,conjugated dikes,thermal metamorphism,tectonic characteristics,paleomagnetic constraints,and paleogeographical evidence along the Solonke-Changchun suture zone,we identify a slab window triggered by slab break-off,which accounts for slab melting and formation of the Nb-enriched mafic rocks and associated adakitic granite porphyry in southeastern Central Asian Orogenic Belt. | Yan Jing Wenchun Ge M.Santosh Yu Dong Hao Yang Zheng Ji Junhui Bi Hongying Zhou Dehe Xing | 2022 | Geoscience Frontiers2022,13,2: | 2 |

| 13 | Study on Plasticity Modification for Measuring High Residual Stress by Hole-Drilling Method显示文摘Hole-drilling method is a commonly used method for measuring residual stress. The calibration coefficients in ASTM E837-13 a would cause large errors due to the plasticity deformation of materials. In the study, calibration coefficients were modified in the plasticity deformation stage based on the distortion energy theory. The calibration experiment of calibration coefficients was simulated by the finite element model, and the plasticity modification formulas of 7075 aluminum alloy were obtained. From the results of uniaxial tensile loading test, the measuring errors of high residual stress are significantly reduced from-4.071%~53.440% to-5.140% ~ 0.609% after the plasticity modification. | 16 | Ferroelastic domain identification and toughening mechanism for yttrium tantalate–zirconium oxide显示文摘Yttrium tantalate(YTaO_(4))is the next generation of higher service temperature thermal barrier coatings(TBCs)materials due to its smaller volume effect in phase change,lower thermal conductivity and unique ferroelastic domain structure.However,the low fracture toughness limits its application.We first characterized the diffraction patterns of variants,and two variants(M_(1)and M_(2))observed in transmission electron microscopy(TEM)results were determined from four possible variants by mechanical derivation.The role of Zr^(4+)doping in ferroelastic toughening was explained in detail.With the increase of Zr^(4+)doping concentration,the monoclinic angle β and the domain rotation angleαdecrease,respectively.The spontaneous strain component and the principal strain in the main space also have a similar decreasing trend.The decrease of the ferroelastic domain inversion energy barrier is beneficial to the improvement of fracture toughness.Combining the results of Vickers indentation,we found that Zr^(4+)could be enriched at the domain boundary to inhibit the generation of cracks.An appropriate amount of Zr^(4+)is conducive to the improvement of fracture toughness,and the excessive Zr^(4+)will reduce the fracture toughness due to the generation of by-product t-ZrO_(2).So,the optimal composition is Y_(0.44)Ta_(0.44)Zr_(0.12)O_(2) and the best fracture toughness(2.9–3.8MPa m^(1/2))is equivalent to the commercial 8YSZ.This result will promote the application of a new generation of TBCs. | Cheng Luo Cong Li Ke Cao Junbao Li Junhui Luo Qinghua Zhang QianQian Zhou Fan Zhang Lin Gu Li Yang Yichun Zhou | 2022 | Journal of Materials Science & Technology2022,,32: | 1 |
| 17 | Junction plakoglobin regulates and destabilizes HIF2α to inhibit tumorigenesis of renal cell carcinoma显示文摘Background:Increased hypoxia-inducible factor 2α(HIF2α)activation is a common event in clear cell renal cell carcinoma(ccRCC)progression.However,the function and underlying mechanism of HIF2α in ccRCC remains uninvestigated.We conducted this study to access the potential link between junction plakoglobin(JUP)and HIF2αin ccRCC.Methods:Affinity purification and mass spectrometry(AP-MS)screening,glutathione-s-transferase(GST)pull-down and co-immunoprecipitation(Co-IP)assays were performed to detect the interacting proteins of HIF2α.Quantitative PCR(qPCR)and Western blotting were used to detect the expression of JUP in human ccRCC samples.Luciferase reporter assays,chromatin immunoprecipitation(ChIP),cycloheximide chase assays,and ubiquitination assays were conducted to explore the regulation of JUP on the activity of HIF2α.Cell Counting Kit-8(CCK-8)assays,colony formation assays,transwell assays,and xenograft tumor assays were performed to investigate the effect of JUP knockdown or overexpression on the tumorigenicity of renal cancer cells.Results:We identified JUP as a novel HIF2α-binding partner and revealed an important role of JUP in recruiting von Hippel-Lindau(VHL)and histone deacetylases 1/2(HDAC1/2)to HIF2α to regulate its stability and transactivation.JUP knockdown promoted and overexpression suppressed the tumorigenicity of renal cell carcinoma in vitro and in vivo.Importantly,the low expression of JUP was found in clinical ccRCC samples and correlated with enhanced hypoxia scores and poor treatment outcomes.Conclusion:Taken together,these data support a role of JUP in modulating HIF2α signaling during ccRCC progression and identify JUP as a potential therapeutic target. | Ke Chen Jin Zeng Yi Sun Wei Ouyang Gan Yu Hui Zhou Yangjun Zhang Weimin Yao Wei Xiao Junhui Hu Jinchun Xing Kefeng Xiao Lily Wu Zhiqiang Chen Zhangqun Ye Hua Xu | 2021 | Cancer Communications2021,41,4: | 1 |
