| 2 | HPLC Fingerprint - Antioxidant Properties Study of Buckwheat显示文摘For quality control of buckwheat, fingerprint-efficacy study of buckwheat was carried out in this work. 2,2′-Bipyridyl,2,2- diphenyl-1-picrylhydrazyl radical (DPPH) scavenging activity of the buckwheat was measured using UV spectrometer. Chemical fingerprints of tartary buckwheat from 29 different sources were determined by HPLC-UV and investigated by similarity analysis and hierarchical clustering analysis. The fingerprint-efficacy relationship between chemical fingerprint and DPPH radical scavenging activity of tartary buckwheat were established by multi-linear regression analysis. The results showed that the sources of buckwheat had some important influence on the chemical fingerprints and DPPH radical scavenging activity. | 4 | Grape Seed Proanthocyanidin Extract Alleviates Arsenic-induced Oxidative Reproductive Toxicity in Male Mice显示文摘Objective To determine the ability of grape seed proanthocyanidin extract(GSPE) in alleviating arsenic-induced reproductive toxicity. Methods Sixty male Kunming mice received the following treatments by gavage: normal saline solution(control); arsenic trioxide(ATO; 4 mg/kg); GSPE(400 mg/kg); ATO+GSPE(100 mg/kg); ATO+GSPE(200 mg/kg) and ATO+GSPE(400 mg/kg). Thereafter, the mice were sacrificed and weighed, and the testis was examined for pathological changes. Nuclear factor(erythroid-derived 2)-like 2(Nrf2), heme oxygenase 1(HO1), glutathione S-transferase(GST), NAD(P)H dehydrogenase, and quinone 1(NQO1) expression in the testis was detected by real-time PCR. Superoxide dismutase(SOD), glutathione(GSH), total antioxidative capability(T-AOC), malondialdehyde(MDA), 8-hydroxydeoxyguanosine(8-OHd G), and reproductive indexes were analyzed. Results ATO-treated mice showed a significantly decreased sperm count and testis somatic index and activity levels of SOD, GSH, and T-AOC than control group. Compared to the ATO-treated group, ATO +GSPE group showed recovery of the measured parameters. Mice treated with ATO+high-dose GSPE showed the highest level of m RNA expression of Nrf2, HO, NQO1, and GST. | 5 | The Predictive Value of Baseline HBs Ag Level and Early Response for HBs Ag Loss in Patients with HBe Ag-positive Chronic Hepatitis B during Pegylated Interferon Alpha-2a Treatment显示文摘Objective To explore the predictive value of baseline HBsAg level and early response for HBsAg loss in patients with HBeAg-positive chronic hepatitis B during pegylated interferon alpha-2a treatment. Methods A total of 121 patients with HBeAg-positive chronic hepatitis B who achieved HBsAg loss were enrolled; all patients were treated with PEG-IFNα-2a 180 μg/week. Serum HBV DNA and serological indicators(HBsAg, anti-HBs, HBeAg, and anti-HBe) were determined before and every 3 months during treatment. Results The median treatment time for HBsAg loss was 84 weeks(7-273 weeks), and 74.38%(90 cases) of the patients needed extended treatment(> 48 weeks). The correlation between baseline HBsAg levels and the treatment time of HBsAg loss was significant(B = 14.465, t = 2.342, P = 0.021). Baseline HBsAg levels together with the decline range of HBsAg at 24 weeks significantly correlated with the treatment time of HBsAg loss(B = 29.862, t = 4.890, P = 0.000 and B = 27.993, t = 27.993, P = 0.005). Conclusion Baseline HBsAg levels and extended therapy are critical steps toward HBsAg loss. | 7 | Robotic gastrectomy with transvaginal specimen extraction for female gastric cancer patients显示文摘AIM: To describe the application of complete robotic gastrectomy with transvaginal specimen extraction(TVSE) for gastric cancer patients.METHODS: Between July and November 2014, eight female patients who were diagnosed with gastric adenocarcinoma underwent a TVSE following a full robot-sewn gastrectomy. According to the tumor location, the patients were allocated to two different groups; two patients received robotic total gastrectomy with TVSE and the other six received robotic distal gastrectomy with TVSE.RESULTS: Surgical procedures were successfully performed in all eight cases without conversion. The mean age was 55.3(range, 42-69) years, and the mean body mass index was 23.2(range, 21.6-26.0) kg/m2. The mean total operative time and blood loss were 224(range, 200-298) min and 62.5(range, 50-150) m L, respectively. The mean postoperative hospital stay was 3.6(range, 3-5) d. The mean number of lymph nodes resected was 23.6(range, 17-27). None was readmitted within 30 d of postoperation. | 9 | Insight-HXMT observations of the first binary neutron star merger GW170817显示文摘Finding the electromagnetic(EM) counterpart of binary compact star merger, especially the binary neutron star(BNS) merger,is critically important for gravitational wave(GW) astronomy, cosmology and fundamental physics. On Aug. 17, 2017,Advanced LIGO and Fermi/GBM independently triggered the first BNS merger, GW170817, and its high energy EM counterpart,GRB 170817 A, respectively, resulting in a global observation campaign covering gamma-ray, X-ray, UV, optical, IR, radio as well as neutrinos. The High Energy X-ray telescope(HE) onboard Insight-HXMT(Hard X-ray Modulation Telescope) is the unique high-energy gamma-ray telescope that monitored the entire GW localization area and especially the optical counterpart(SSS17 a/AT2017 gfo) with very large collection area(~1000 cm^2) and microsecond time resolution in 0.2-5 MeV. In addition,Insight-HXMT quickly implemented a Target of Opportunity(ToO) observation to scan the GW localization area for potential X-ray emission from the GW source. Although Insight-HXMT did not detect any significant high energy(0.2-5 MeV) radiation from GW170817, its observation helped to confirm the unexpected weak and soft nature of GRB 170817 A. Meanwhile,Insight-HXMT/HE provides one of the most stringent constraints(~10^(-7) to 10^(-6) erg/cm^2/s) for both GRB170817 A and any other possible precursor or extended emissions in 0.2-5 MeV, which help us to better understand the properties of EM radiation from this BNS merger. The mission aims at determining the equation of state of matter at supra-nuclear density, measuring effects of QED, and understanding the dynamics of matter in strong-field gravity. In addition to investigating fundamental physics, eXTP will be a very powerful observatory for astrophysics that will provide observations of unprecedented quality on a variety of galactic and extragalactic objects. | 13 | Protective Effects of Quercetin on Cadmium-induced Cytotoxicity in Primary Cultures of Rat Proximal Tubular Cells显示文摘Objective To investigate the protective effects of quercetin on cadmium-induced cytotoxicity in primary cultures of rat proximal tubular(rPT)cells.Methods Primary cultures of rPT cells undergoing exponential growth were incubated with 1.0 μg/mL quercetin and/or cadmium(2.5,5.0 μmol/L),in a serum-free medium at 37 ℃ at different time intervals.Commercial kits were used and flow cytometric analyses were performed on rPT cell cultures to assay apoptosis and oxidative stress.Results Exposure of rPT cells to cadmium acetate(2.5,5.0 μmol/L)induced a decrease in cell viability,caused an increase in apoptotic rate and apoptotic morphological changes.Simultaneously,elevation of intracellular reactive oxygen species,malondialdehyde and calcium levels,depletion of mitochondrial membrane potential and intracellular glutathione,and inhibition of Na +,K +-ATPase,Ca 2+-ATPase,glutathione peroxidase(GSH-Px),catalase(CAT),and superoxide dismutase(SOD)activities were revealed during the cadmium exposure of rPT cells.However,simultaneous supplementation with 1 μg/mL quercetin protected rPT cells against cadmium-induced cytotoxicity through inhibiting apoptosis,attenuating lipid peroxidation,renewing mitochondrial function and elevating the intracellular antioxidants(non-enzymatic and enzymic)levels.Conclusion The present study has suggested that quercetin,as a widely distributed dietary antioxidant,contributes potentially to prevent cadmium-induced cytotoxicity in rPT cells. | WANG Lin LIN Shu Qian HE Yuan Long LIU Gang WANG Zhen Yong | 2013 | Biomedical and Environmental Sciences2013,26,4: | 8 |
| 14 | Microstructures and mechanical properties of Nb-alloyed CoCrCuFeNi high-entropy alloys显示文摘Nb has a positive effect on improving the mechanical properties of metal materials, and it is expected to strengthen CoCrCuFeNi high-entropy alloys(HEAs) with outstanding ductility and relatively weak strength. In this paper, the alloying effects of Nb on the microstructural evolution and the mechanical properties of the(CoCrCuFeNi)_(100-x)Nb_xHEA were investigated systematically. The result shows that Nb promotes the phase transition from FCC(face-centered cubic) to Laves phase, and the volume fractions of Laves phase increase from 0% to 58.2% as the Nb content increases. Compressive testing shows that the addition of Nb has a positive effect on improving the strength of CoCrCuFeNi HEA. The compressive yield strength of(CoCrCuFeNi)_(100-x)Nb_xHEAs increases from 338 MPa to 1322 MPa and the fracture strain gradually reduces from 60.0%(no fracture) to 8.1% as the Nb content increases from 0 to 16 at.%. | 15 | Influence of chronic HBV infection on superimposed acute hepatitis E显示文摘AIM:To investigate the influence of chronic hepatitis B virus(HBV)infection[based on the status of hepatitis B e antigen(HBeAg),HBV DNA,and cirrhosis]on superimposed acute hepatitis E.METHODS:A total of 294 patients were recruited from the Department of Infectious Diseases of the Third Affiliated Hospital,Sun Yat-sen University,from January 2003 to January 2012.The patients were classified into two groups:an HBV+hepatitis E virus(HEV)group(a group with chronic HBV infection that was superinfected with acute hepatitis E,n=118)and an HEV group(a group with acute hepatitis E,n=176).We retrospectively analyzed and compared the clinical features of the two groups.Statistical analyses were performed using theχ2test or Fisher’s exact test for categorical variables and the Student’s t test forcontinuous variables.A P value<0.05 was considered statistically significant.RESULTS:The peak values of prothrombin time,serum total bilirubin,and Model for End-Stage Liver Disease scores were significantly higher in the HBV+HEV group.More patients in the HBV+HEV group had complications(39.8%vs 16.5%,P=0.000)and developed liver failure(35.6%vs 8.5%,P=0.000).Additionally,the mortality of the HBV+HEV group was significantly higher(20.3%vs 7.4%,P=0.002).Further analysis of the HBV+HEV group showed that there were no significant differences in complication occurrence,liver failure incidence,or mortality between patients with different HBeAg and HBV DNA statuses.However,in patients with underlying cirrhosis,complication occurrence and liver failure incidence significantly increased.In total,12.7%of the patients in the HBV+HEV group received anti-HBV treatment,but this therapy failed to reduce mortality in patients who developed liver failure.CONCLUSION:The presence of underlying cirrhosis in chronic HBV infection results in more severe clinical outcomes with superimposed acute hepatitis E.AntiHBV treatment cannot improve the prognosis of liver failure caused by HBV-HEV superinfection. | Si-Hong Cheng Li Mai Feng-Qin Zhu Xing-Fei Pan Hai-Xia Sun Hong Cao Xin Shu Wei-Min Ke Gang Li Qi-Huan Xu | 2013 | World Journal of Gastroenterology2013,19,35: | 8 |

Population Using cyp17a1-Deficient Neomale Carp显示文摘Due to sexual dimorphism in the growth of certain cultured fish species,the production of monosex fishes is desirable for the aquaculture industry.Nowadays,the most widely practiced technique available for the mass production of monosex fish populations is sex steroid-induced sex reversal.Here,a novel strategy for the successful production of all-female(AF)common carp(Cyprinus carpio L.),to take advantage of the sexual dimorphism in growth documented in this species,has been developed using genetic engineering via single gene-targeting manipulation without any exogenous hormone treatments.Male and female heterozygous cyp17a1-deficient common carp were first obtained using the clustered regularly interspaced short palindromic repeats/CRISPR-associated endonuclease 9(CRISPR/Cas9)technique.An all-male phenotype for homozygous cyp17a1-deficient carp,regardless of the individuals’sexdetermination genotypes(XY or XX),has been observed.A male-specific DNA marker newly identified in our laboratory was used to screen the neomale carp population with the XX genotype from the cyp17a1-deficient carp.These neomale carp develop a normal testis structure with normal spermatogenesis and sperm capacity.The neomale common carp were then mated with wild-type(WT)females(cyp17a1^(+/+)XX genotype)using artificial fertilization.All the AF offspring sample fish from the neomale-WT female mating were confirmed as having the cyp17a^(1+/-)XX genotype,and normal development of gonads to ovaries was observed in 100.00%of this group at eight months post-fertilization(mpf).A total of 1000 carp fingerlings,500 from the WT male and female and 500 from the neomale and WT female mating,were mixed and reared in the same pond.The average body weight of cyp17a1^(+/-)XX females was higher by 6.60%(8 mpf)and 32.66%(12 mpf)than that of the control common carp.Our study demonstrates the first successful production of a monosex teleost population with the advantages of sexual dimorphism in growth using genetic manipulation targeting a single locus. | Gang Zhai Tingting Shu Kuangxin Chen Qiyong Lou Jingyi Jia Jianfei Huang Chuang Shi Xia Jin Jiangyan He Donghuo Jiang Xueqiao Qian Wei Hu Zhan Yin | 2022 | Engineering2022,8,1: | 7 |

It has been suggested that androgen signaling regulates critical genes for the differentiation and development of the testis. To elucidate the exact regulatory mechanisms involved in amh transcription mediated by androgen signaling, androgen signaling was manipulated in zebrafish by cytochrome P45017a1(cyp17a1) knockout and Flutamide treatment. In cyp17a1-deficient and Flutamide-treated testes,up-regulated sry-box9a(sox9a) and down-regulated amh were observed. Moreover, a physical association of the zebrafish androgen receptor(AR) and SOX9 A was found. The interaction between AR and SOX9 A was mediated via the DNA binding domain(DBD) of AR and the transactivation domain(TA) of SOX9 A,and was further enhanced by 5-alpha dihydrotestosterone(DHT), one of the most potent androgens.Intriguingly, together with SOX9 A, androgen signaling synergistically promoted amh transcription,mainly through the proximal 1 kb of the amh promoter region. | 20 | Sedimentary Evolution and Transgressions of the Western Subei Basin in Eastern China since the Late Pliocene显示文摘Sedimentary sequences in the Subei Basin are complex and have been affected by interactions between the ocean and rivers since the Late Pliocene, including the Yellow River, Huaihe River, and the Yangtze River. This sedimentary evolution, in particular the timing of Pleistocene transgressions, has long been a matter of controversy owing to the lack of precise chronological evidence. The aim of this study is to explore the evolution of the sedimentary environment throughout the past 3.00 Ma in this region on the basis of a comprehensive analysis of particle size and foraminifera and ostracods collected in the TZK9 core from the Subei Basin combined with geochronological studies of magnetostratigraphy, AMS^(14)C and optically stimulated luminescence(OSL). The results show that fluvial facies in the sedimentary environment from 3.00 to 1.01 Ma. There were fluvial facies and reflects six sea-level high stands from 1.01 to 0.25 Ma. The study area was affected by four large-scale transgressions since 0.25 Ma. The four marine sedimentary layers known as DU7(buried at 48–52 m), DU5(buried at 35–41 m), DU3(buried at 16–23 m), and DU1(buried at 2–4 m) are recorded in the MIS7(210–250 ka), MIS5, MIS3, and Holocene, respectively. The magnitude of the DU5 transgression was identical to that of the DU3 transgression, both were larger than the DU7 transgression, and the DU1 transgression was the weakest. The variation of transgression strength reflects the influence of global changes in sea level, tectonic subsidence, shell ridges, and sand dams. In the TZK9 core, we found evidence of seven sea-level high stands from the Early–Middle Pleistocene, and the first one caused by regional rapid subsidence and could be traced back to 0.83-0.84 Ma. The sea-level high stands and the age of the first one recorded above was different from other cores in eastern China, this was caused by the lack of absolute age control and the differences in paleotopography during this period. This study reconstructs sedimentary evolution, determines the transgression and its age, establishes the chronology since the Late Pliocene, and provides a scientific framework for further paleoenvironmental and tectonic studies.
