2The Reference Genome of Tea Plant and Resequencing of 81 Diverse Accessions Provide Insights into Its Genome Evolution and Adaptation显示文摘Tea plant is an important economic crop,which is used to produce the world's oldest and most widely consumed tea beverages.Here,we present a high-quality reference genome assembly of the tea plant(Camellia sinensis var.sinensis)consisting of 15 pseudo-chromosomes.LTR retrotransposons(LTR-RTs)account for 70.38%of the genome,and we present evidence that LTR-RTS play critical roles in genome size expansion and the transcriptional diversification of tea plant genes through preferential insertion in promoter regions and introns.Genes,particularly those coding for terpene biosynthesis pro-teins,associated with tea aroma and stress resistance were significantly amplified through recent tandem duplications and exist as gene clusters in tea plant genome.Phylogenetic analysis of the sequences of 81 tea plant accessions with diverse origins revealed three well-differentiated tea plant populations,support-ing the proposition for the southwest origin of the Chinese cultivated tea plant and its later spread to western Asia through introduction.Domestication and modern breeding left significant signatures on hundreds of genes in the tea plant genome,particularly those associated with tea quality and stress resis-tance.The genomic sequences of the reported reference and resequenced tea plant accessions provide valuable resources for future functional genomics study and molecular breeding of improved cul-tivars of tea plants.Enhua Xia Wei Tong Yan Hou Yanlin An Linbo Chen Qiong Wu Yunlong Liu Jie Yu Fangdong Li Ruopei Li Penghui Li Huijuan Zhao Ruoheng Ge Jin Huang Ali Inayat Mallano Yanrui Zhang Shengrui Liu Weiwei Deng Chuankui Song Zhaoliang Zhang Jian Zhao Shu Wei Zhengzhu Zhang Tao Xia Chaoling Wei Xiaochun Wan 2020Molecular Plant2020,13,7:51
3Performance value of high risk factors in colorectal cancer screening in China显示文摘AIM:To analyze the performance value of high risk factors in population-based colorectal cancer(CRC) screening in China.METHODS:We compared the performance value of the immunochemical fecal occult blood test(iFOBT) and other high risk factors questionnaire in a population sample of 13 214 community residents who completed both the iFOBT and questionnaire investigation.Patients with either a positive iFOBT and/or questionnaire were regarded as a high risk population and those eligible were asked to undergo colonoscopy.RESULTS:The iFOBT had the highest positive predictive value and negative predictive value in screening for advanced neoplasia.The iFOBT had the highest sensitivity,lowest number of extra false positive results associated with the detection of one extra abnormality for screening advanced neoplasias and adenomas.A history of chronic cholecystitis or cholecystectomy,chronic appendicitis or appendectomy,and chronic diarrhea also had a higher sensitivity than a history of adenomatous polyps in screening for advanced neoplasias and adenomas.The sensitivity of a history of chronic cholecystitis or cholecystectomy was highest among the 10 high risk factors in screening for nonadenomatous polyps.A history of chronic appendicitis or appendectomy,chronic constipation,chronic diarrhea,mucous and bloody stool,CRC in first degree relatives,malignant tumor and a positive iFOBT also had higher sensitivities than a history of adenomas polyps in screening for non-adenomatous polyps.Except for a history of malignant tumor in screening for non-adenomatous polyps,the gain in sensitivity was associated with an increase in extra false positive results associated with the detection of one extra abnormality.CONCLUSION:The iFOBT may be the best marker for screening for advanced neoplasias and adenomas.Some unique high risk factors may play an important role in CRC screening in China.Wen Meng Shan-Rong Cai Lun Zhou Qi Dong Shu Zheng Su-Zhan Zhang 2009World Journal of Gastroenterology2009,15,48:41
4Prevalence of Atrial Fibrillation in China and Its Risk Factors显示文摘Objective To study the prevalence of atrial fibrillation(AF)and the relation with its risk factors in China.Methods A total of 19 363 participants(8635 males and 10 728 females) aged ≥35 years in geographically dispersed urban and rural regions of China were included in this cross-sectional survey.All participants received questionnaire,physical and blood examination.Echocardiography were performed for AF patients found in the survey.Results Of the 19 363 participants,199 were diagnosed with AF.The estimated age-standardized prevalence of AF was 0.78% in men and 0.76% in women.The prevalence of AF in participants aged <60 years was 0.41% in men and 0.43% in women,and was 1.83% in both men and women aged ≥60 years.About 19.0% of males and 30.9% of females with AF were diagnosed with valve disease.Age-and sex-adjusted multivariable logistic regression analysis revealed that myocardial infarction,left ventricular hypertrophy(LVH),obesity,and alcohol consumption were associated with a increased risk of AF(P<0.05).Conclusion The age standardized prevalence of AF is 0.77% in the participants enrolled in the present study.The number of AF cases aged ≥35 years is 5.26 million according to 2010 Chinese Census.Most risk factors for AF,identified mainly in Western countries,are also detected in China.LI Ying WU Yang Feng CHEN Ke Ping LI Xian ZHANG Xing XIE Gao Qiang WANG Fang Zheng ZHANG Shu 2013Biomedical and Environmental Sciences2013,26,9:48

6Comparison of microRNA profiles of human periodontal diseased and healthy gingival tissues显示文摘MicroRNAs (miRNAs) have been demonstrated to play an important role in regulation of the immuno- inflammatory response; however, the function of miRNAs in periodontal inflammation has not been investigated. The objective of this study was to explore the properties of miRNAs in periodontal inflammation by comparing miRNA profiles of inflamed and healthy gingival tissues. Gingival tissues were obtained from 10 periodontitis patients and 10 healthy subjects. After RNA extraction, miRNA profiles were analyzed by microarray, and expression levels of selected miRNAs were confirmed by real-time quantitative reverse transcription polymerase chain reaction (RT-PCR). Analyses using two computational methods, Targetscan and MicroRNA.org, were combined to identify common targets of these miRNAs. Finally, the individual miRNA expression levels of three toll-like receptor (TLR)-related miRNAs from inflamed and healthy gingival tissues were evaluated by RT-PCR. Ninety-one miRNAs were found to be upregulated and thirty-four downregulated over two-fold in inflamed gingival tissue compared with those in healthy gingival tissue. Twelve selected inflammatory-related miRNAs, hsa-miR-126*, hsa-miR-20a, hsa-miR-142-3p, hsa-miR-19a, hsa-let-7f, hsa-miR-203, hsa-miR-17, hsa-miR-223, hsa-miR-146b, hsa-miR-146a, hsa-miR-155, and hsa-miR-205 showed comparable expression levels by microarray and real-time quantitative RT-PCR analyses. In addition, the putative inflammation targets of these miRNAs were predicted, and three that were tested (hsa-miRNA-146a, hsa-miRNA-146b, and hsa-miRNA-155), showed significant differences between inflamed and healthy gingiva. This remarkable difference in miRNA profiles between periodontal diseased and healthy gingiva implicates a probable close relationship between miRNAs and periodontal inflammation. 7Reduction of the incidence and mortality of rectal cancer bypolypectomy:a prospective cohort study in Haining County显示文摘AIM: To reduce the incidence and morlality of rectal cancerand address the hypothesis that colorectal cancer often arisefrom precursor lesion (s), either adenomas or non-alenomatous polyps, by conducting a population-basedmass screening for colorectal cancer in Haining County,Zhejiang, PRCMETHODS: From 1977 to 1980, physicians screened thepopulation of Haining County using 15 cm rigid endoscopy.Of over 240 000 participants, 4076 of them were diagnoselwith Precursor Iesions, eitler alenomas or non-adenomatous polyps, which were then removedl surgically.All individuals with precursor lesionswere followed up andreexamined by endoscopy every two to five years upto 1998.RESULTS: After the initial screening, 953 metactronousadenomas and 417 non-adenomatous polyps ware detected andremoved from the members of this cohort. Further, 27 cases ofcolorectal cancer were detected and treated. Log-rank tuestsshowed that the survival time among those cancer patients whounderwent mass screening increased significantly comparedtothat of other colorectal cancer patients (P < 0.0001). 9Application of serum protein fingerprinting coupled with artificial neural network model in diagnosis of hepatocellular carcinoma显示文摘Background Hepatocellular carcinoma tends to present at a late clinical stage with poor prognosis. Therefore, it is urgent to explore and develop a simple, rapid diagnostic method, which has high sensitivity and specificity for hepatocellular carcinoma at an early stage. In this study, the serum proteins in patients with hepatocellular carcinoma or liver cirrhosis and in normal controls were analysed. Surface enhanced laser desorption/ionization time-of-flight mass (SELDI-TOF-MS) spectrometry was used to fingerprint serum protein using the protein chip technique and explore the value of the fingerprint, coupled with artificial neural network, to diagnose hepatocellular carcinoma.Methods Of the 106 serum samples obtained, 52 were from patients with hepatocellular carcinoma, 22 from patients with liver cirrhosis and 32 from healthy volunteers. The samples were randomly assigned into a training group ( n =70, 35 patients with hepatocellular carcinoma, 14 with liver cirrhosis, and 21 normal controls) and a testing group ( n =36, 17 patients with hepatocellular carcinoma, 8 with liver cirrhosis, and 11 normal controls). An artificial neural network was trained on data from 70 individuals in the training group to develop an artificial neural network diagnostic model and this model was tested. The 36 sera in the testing group were analysed with blind prediction by using the same flowchart and procedure of data collection. The 36 serum protein spectra were clustered with the preset clustering method and the same mass/charge ( M/Z ) peak values as those in the training group. Matrix transfer was performed after data were output. Then the data were input into the previously built artificial neural network model to get the prediction value. The M/Z peaks of the samples with more than 2000 M/Z were normalized with biomarker wizard of ProteinChip Software version 3.1 for noise filtering. The first threshold for noise filtering was set at 5, and the second was set at 2. The 10% was the minimum threshold for clustering. The statistical analysis of the data of serum protein mass spectrum was performed in the groups (normal vs. hepatocellular carcinoma, and liver cirrhosis vs. hepatocellular carcinoma) with the t test. Results Comparison between the groups of hepatocellular carcinoma and normal control: The mass spectra from 56 samples (hepatocellular carcinoma and normal controls) in the training group were analysed and 241 peaks were obtained. In addition, 21 peaks from them were used for comparison between the groups of hepatocellular carcinoma and normal controls ( P <0.01). Only 2 peaks at 3015 M/Z and 5900 M/Z were selected with significant difference ( P <10 -9). A model was developed based on these two proteins with different M/Z . It was confirmed that this artificial neural network model can be used for comparison between the groups of hepatocellular carcinoma and normal controls. The sensitivity was 100% (17/17), and the specificity was 100% (11/11). Comparison between the groups of hepatocellular carcinoma and liver cirrhosis: The mass spectra from 49 samples in the training group (including patients with hepatocellular carcinoma and liver cirrhosis) were analysed and 208 peaks were obtained. In addition, 21 peaks from them were used for comparison between the groups of hepatocellular carcinoma and liver cirrhosis ( P <0.01). Only 2 peaks at 7759 M/Z , 13134 M/Z were selected with significant difference ( P <10 -9). A model was developed based on these two proteins with dfferent M/Z . It was confirmed that this artificial neural network model can be used for comparison between the groups of hepatocellular carcinoma and liver cirrhosis. 10A novel m6A reader Prrc2a controls oligodendroglial specification and myelination显示文摘While N6-methyladenosine (m6A), the most abundant internal modification in eukaryotic mRNA, is linked to cell differentiation and tissue development, the biological significance of m6A modification in mammalian glial development remains unknown. Here, we identify a novel m6A reader, Prrc2a (Proline rich coiled-coil 2A), which controls oligodendrocyte specification and myelination. Nestin-Cre-mediated knockout of Prrc2a induces significant hypomyelination, decreased lifespan, as well as locomotive and cognitive defects in a mouse model. Further analyses reveal that Prrc2a is involved in oligodendrocyte progenitor cells (OPCs) proliferation and oligodendrocyte fate determination. Accordingly, oligodendroglial-lineage specific deletion of Prrc2a causes a similar phenotype of Nestin-Cre-mediated deletion. Combining transcriptome-wide RNA-seq, m6A-RIP-seq and Prrc2a RIP-seq analysis, we find that Olig2 is a critical downstream target gene of Prrc2a in oligodendrocyte development. Furthermore, Prrc2a stabilizes Olig2 mRNA through binding to a consensus GGACU motif in the Olig2 CDS (coding sequence) in an m6A-dependent manner. Interestingly, we also find that the m6A demethylase, Fto, erases the m6A modification of Olig2 mRNA and promotes its degradation. Together, our results indicate that Prrc2a plays an important role in oligodendrocyte specification through functioning as a novel m6A reader. 16Geological characteristics and mineralization setting of the Zhuxi tungsten(copper) polymetallic deposit in the Eastern Jiangnan Orogen显示文摘The Zhuxi ore deposit is a super-large scheelite(copper) polymetallic deposit discovered in recent years. It grew above copper/tungsten-rich Neoproterozoic argilloarenaceous basement rocks and was formed in the contact zone between Yanshanian granites and Carboniferous-Permian limestone. Granites related to this mineralization mainly include equigranular, middle- to coarse-grained granites and granitic porphyries. There are two mineralization types: skarn scheelite(copper) and granite scheelite mineralization. The former is large scale and has a high content of scheelite, whereas the latter is small scale and has a low content of scheelite. In the Taqian-Fuchun Basin, its NW boundary is a thrust fault, and the SE boundary is an angular unconformity with Proterozoic basement. In Carboniferous-Permian rock assemblages, the tungsten and copper contents in the limestone are both very high. The contents of major elements in granitoids do not differ largely between the periphery and the inside of the Zhuxi ore deposit. In both areas, the values of the aluminum saturation index are A/CNK>1.1, and the rocks are classified as potassium-rich strongly peraluminous granites. In terms of trace elements, compared to granites on the periphery of the Zhuxi ore deposit, the granites inside the Zhuxi ore deposit have smaller d Eu values, exhibit a significantly more negative Eu anomaly, are richer in Rb, U, Ta, Pb and Hf, and are more depleted in Ba, Ce, Sr, La and Ti, which indicates that they are highly differentiated S-type granites with a high degree of evolution. Under the influence of fluids, mineralization of sulfides is evident within massive rock formations inside the Zhuxi ore deposit, and the mean SO_3 content is 0.2%. Compared to peripheral rocks, the d Eu and total rare earth element(REE) content of granites inside the Zhuxi ore deposit are both lower, indicating a certain evolutionary inheritance relationship between the granites on the periphery and the granites inside the Zhuxi ore deposit. For peripheral and ore district plutons, U-Pb zircon dating shows an age range of 152–148 Ma. In situ Lu-Hf isotope analysis of zircon in the granites reveals that the calculated e_(Hf)(t) values are all negative, and the majority range from -6 to -9. The T_(DM2) values are concentrated in the range of 1.50–1.88 Ga(peak at 1.75 Ga), suggesting that the granitic magmas are derived from partial melting of ancient crust. This paper also discusses the metallogenic conditions and ore-controlling conditions of the ore district from the perspectives of mineral contents, hydrothermal alteration, and ore-controlling structures in the strata and the ore-bearing rocks. It is proposed that the Zhuxi ore deposit went through a multistage evolution, including oblique intrusion of granitic magmas, skarn mineralization, cooling and alteration, and precipitation of metal sulfides. 17Research on the maceral characteristics of Shenhua coal and efficient and directional direct coal liquefaction technology显示文摘In this research,molecular structure models were developed respectively for Shenhua coal vitrinite concentrates(SDV)and inertinite concentrates(SDI),on the basis of information on constitutional unit of Shenhau coal and elemental analysis results obtained from^(13)C-NMR analysis characterization,FTIR analysis characterization,X-ray diffraction XRD and XPS analysis characterization.It can be observed from characterization data and molecular structure models that the structure of SDV and SDI is dominated by aromatic hydrocarbon,with aromaticity of SDI higher than that of SDV;SDV mainly consists of small molecule basic structure unit,while SDI is largely made from macromolecular structure unit.Based on bond-level parameters of the molecular model,the research found through the autoclave experiment that vitrinite liquefaction process goes under thermodynamics control and inertinite liquefaction process under dynamics control.The research developed an efficient directional direct coal liquefaction technology based on the maceral characteristics of Shenhua coal,which can effectively improve oil yield and lower gas yield.Geping Shu Yuzhuo Zhang 2014International Journal of Coal Science & Technology2014,1,1:24
18Geochemistry, geochronology, and petro-genesis of the early Paleozoic granitic plutons in the central-southern Jiangxi Province,China显示文摘This paper reports the systematic study on petrology, geochemistry, LA ICPMS zircons U-Pb dating, and in situ Hf isotope geology of the four plutons in the central-southern Jiangxi Province, an important part of the South China Block. In the outcrops, rocks are gradually changed from wall rock (slate or schist) to pluton (gneissic granite); some residual blocks of sandy rock occur in the margin of pluton, and the foliations of residual blocks are parallel to those of both wall rock and gneissic granite. The thin-section observations show that the four plutons contain peraluminous minerals such as muscovite and sillimanite. The flattened and elongated feldspar and quartz grains are often visible in the gneissic granite, parallel to direction of lineation, suggesting that the granitic rock were subjected to a strong ductile shearing. Geochemically, the A/CNK values from 13 granitic samples are between 1.03 and 1.37 with an average of 1.16, indicating that the granites are of strongly peraluminous plutons. The REE compositions of the 13 samples are similar, showing higher REE contents, with enrichment in LREEs, depletion in Eu and REE patterns with relative LREE-enrichment and negligible Eu anomalies. They show enrichment in Rb, Th, U and depletion in Ba, Sr, Nb, Ti, belonging to a low Ba-Sr type of granite. Thus, the four bodies should be derived from the same magmatic source. Zircons used as U-Pb dating mostly exhibit euhedral shape and high Th/U values from 0.52 to 1.54 with an average of 1.08, suggesting that most zircons are of magmatic genesis. The zircons from four plutons yielded rather similar 206 Pb/ 238 U vs. 207 Pb/ 235 U concordia ages: 436.1±5.7 Ma for the Tangwan granite, 440.6±4 Ma for the Jiekou gneissic granite, 435.9±6.2 Ma for the Dongbao gneissic granite, and 441.9±3.1 Ma for the Jinxi K-granite, respectively, corresponding to Silurian Llandovery. Several xenocrysts yielded U-Pb ages around 700 Ma, implying that a breakup event took place during Neoproterozoic in the South China Block. In situ Lu-Hf isotopic analysis shows that all Hf (t) values of zircons are negative and have two-stage Hf model ages (TDM2) from 1.4 to 3.6 Ga, indicating that the Silurian granitic magma came from the recycle of Meso-Paleoproterozoic basement and even partly Archean rocks, and had not been effected by mantle magma. Researches on regional geology suggest that an intracontinental tectono-magmatic event took place during the early Paleozoic in the study areas, which is characterized by folding and thrusting, leading to crustal shortening and thickening, up to 20 km thickness. The high geothermal temperature from thickening crust and accumulation of producing high-heat radioactive elements gradually softened crustal rocks and caused a partial melting, forming peraluminous granitic magma. 19Dihydromyricetin inhibits migration and invasion of hepatoma cells through regulation of MMP-9 expression显示文摘AIM:To investigate the effects of dihydromyricetin(DHM)on the migration and invasion of human hepatic cancer cells.METHODS:The hepatoma cell lines SK-Hep-1 and MHCC97L were used in this study.The cells were cultured in RPIM-1640 medium supplemented with 10%fetal bovine serum at 37℃in a humidified 5%CO2incubator.DHM was dissolved in dimethyl sulfoxide and diluted to various concentrations in medium before applying to cells.MTT assays were performed to measure the viability of the cells after DHM treatment.Wound healing and Boyden transwell assays were used to assess cancer cell motility.The invasive capacity of cancer cells was measured using Matrigel-coated transwell chambers.Matrix metalloproteinase(MMP)-2/9 activity was examined by fluorescence analysis.Western blot was carried out to analyze the expression of MMP-2,MMP-9,p-38,JNK,ERK1/2 and PKC-δproteins.All data were analyzed by Student’s t tests in GraphPad prism 5.0software and are presented as mean±SD.RESULTS:DHM was found to strongly inhibit the migration of the hepatoma cell lines SK-Hep-1(without DHM,24 h:120±8μmol/L vs 100μmol/L DHM,24h:65±10μmol/L,P<0.001)and MHCC97L(without DHM,24 h:126±7μmol/L vs 100μmol/L DHM,24h:74±6μmol/L,P<0.001).The invasive capacity of the cells was reduced by DHM treatment(SK-Hep-1cells without DHM,24 h:67±4μmol/L vs 100μmol/L DHM,24 h:9±3μmol/L,P<0.001;MHCC97L cells without DHM,24 h:117±8μmol/L vs 100μmol/L DHM,24 h:45±2μmol/L,P<0.001).MMP2/9 activity was also inhibited by DHM exposure(SK-Hep-1 cells without DHM,24 h:600±26μmol/L vs 100μmol/L DHM,24 h:100±6μmol/L,P<0.001;MHCC97L cells without DHM,24 h:504±32μmol/L vs 100μmol/L DHM 24 h:156±10μmol/L,P<0.001).Western blot analysis showed that DHM decreased the expression level of MMP-9 but had little effect on MMP-2.Further investigation indicated that DHM markedly reduced the phosphorylation levels of p38,ERK1/2 and JNK in a concentration-dependent manner but had no impact on the total protein levels.In addition,PKC-δprotein,a key protein in the regulation of MMP family protein expression,was up-regulated with DHM treatment.CONCLUSION:These findings demonstrate that DHM inhibits the migration and invasion of hepatoma cells and may serve as a potential candidate agent for the prevention of HCC metastasis.Qing-Yu Zhang Ran Li Guo-Fang Zeng Bin Liu Jie Liu Yang Shu Zhong-Kao Liu Zhi-Dong Qiu Dong-Jun Wang Hui-Lai Miao Ming-Yi Li Run-Zhi Zhu 2014World Journal of Gastroenterology2014,20,29:23
20Left posterior fascicular block: a new endpoint of ablation for verapamil-sensitive idiopathic ventricular tachycardia显示文摘Background Verapamil-sensitive, idiopathic left ventricular tachycardia (ILVT) with right bundle branch block configuration and left-axis deviation is known to be due to re-entry mechanism but the exact nature of reentrant circuit in ILVT is not fully elucidated. Radiofrequency (RF) ablation was applied during ventricular tachycardia (VT) and termination of the VT or abolishing the inducibility of the tachycardia was used as an endpoint for successful RF. In this study, the left posterior fascicular block in surface electrocardiogram (ECG) was used as a new endpoint of ablation to cure ILVT. Methods Electrophysiological studies and radiofrequency ablation were performed in 39 consecutive patients [30 men, 9 women; age ranging from 10 to 64 years, mean (29±16) years] with verapamil-sensitive ILVT and structurally normal hearts. VT could be terminated by the intravenous administration of verapamil in all patients. The target site was the midseptum of LV where the earliest Purkinje potentials were recorded during VT. RF current was applied to the target site with or without late diastolic potential (LDP) during sinus rhythm in 37 patients and during VT in 2 patients to meet the ablation endpoint: the left posterior fascicular block in the surface ECG. Results Thirty-seven patients with ILVT had been treated by RF ablation during sinus rhythm and two had been treated during VT. All of them met the endpoint of the left posterior fascicular block. Thirty-eight cases were symptom-free without medications during the follow-up period (range from 3 to 95 months, median 17 months). One patient developed a clinical recurrence and the left posterior fascicular block in surface ECG disappeared. The patient received another treatment. The endpoint was met and the procedure was successful. Conclusions The left posterior fascicular block in surface ECG used as an endpoint of RF ablation to treat ILVT is effective. It is important especially in those patients whose VT can not be induced or the inducible condition is unstable.
