| 2 | T cell–associated immunoregulation and antiviral effect of oxymatrine in hydrodynamic injection HBV mouse model显示文摘Although oxymatrine(OMT) has been shown to directly inhibit the replication of hepatitis B virus(HBV) in vitro, limited research has been done with this drug in vivo. In the present study, the antiviral effect of OMT was investigated in an immunocompetent mouse model of chronic HBV infection.The infection was achieved by tail vein injection of a large volume of DNA solution. OMT(2.2, 6.7 and20 mg/kg) was administered by daily intraperitoneal injection for 6 weeks. The efficacy of OMT was evaluated by the levels of HBV DNA, hepatitis B surface antigen(HBs Ag), hepatitis B e antigen(HBe Ag)and hepatitis B core antigen(HBc Ag). The immunoregulatory activity of OMT was evaluated by serum ELISA and flow cytometry. Results shows that OMT at 20 mg/kg inhibited HBV replication, and it was more efficient than entecavir(ETV) in the elimination of serum HBs Ag and intrahepatic HBc Ag. Inaddition, OMT accelerated the production of interferon-γ(IFN-γ) in a dose-dependent manner in CD4^+T cells. Our findings demonstrate the beneficial effects of OMT on the enhancement of immunological function and in the control of HBV antigens. | 4 | Transcriptional Regulation of miR528 by OsSPL9 Orchestrates Antiviral Response in Rice显示文摘Many microRNAs (miRNAs) are critical regulators of plant antiviral defense.However,little is known about how these miRNAs respond to virus invasion at the transcriptional level.We previously show that defense against Rice stripe virus (RSV) invasion entailed a reduction of miR528 accumulation in rice,alleviating miR528-mediated degradation of L-Ascorbate Oxidase (AO) mRNA and bolstering the antiviral activity of AO.Here we show that the miR528-A0 defense module is regulated by the transcription factor SPL9.SPL9 displayed high-affinity binding to specific motifs within the promoter region of miR528 and activated the expression of miR528 gene in vivo.Loss-of-function mutations in SPL9 caused a significant reduction in miR528 accumulation but a substantial increase of AO mRNA,resulting in enhanced plant resistance to RSV.Conversely,transgenic overexpression of SPL9 stimulated the expression of miR528 gene,hence lowering the level of AO mRNA and compromising rice defense against RSV.Importantly,gain in RSV susceptibility did not occur when SPL9 was overexpressed in mir528 loss-of-function mutants,or in transgenic rice expressing a miR528-resistant AO.Taken together,the finding of SPL9-mediated transcriptional activation of miR528 expression adds a new regulatory layer to the miR528-A0 antiviral defense pathway. | Shengze Yao Zhirui Yang Rongxin Yang Yu Huang Ge Guo Xiangyue Kong Ying Lan Tong Zhou He Wang Wenming Wang Xiaofeng Cao Jianguo Wu Yi Li | 2019 | Molecular Plant2019,12,8: | 10 |

| 6 | Ketamine induces tau hyperphosphorylation at serine 404 in the hippocampus of neonatal rats显示文摘Male Wistar 7-day-old rats were injected with 40 mg/kg ketamine intraperitoneally, followed by three additional injections of 20 mg/kg ketamine each upon restoration of the righting reflex. Neonatal rats injected with equivalent volumes of saline served as controls. Hippocampal samples were collected at 1, 7 or 14 days following administration. Electron microscopy showed that neuronal structure changed noticeably following ketamine treatment. Specifically, microtubular structure became irregular and disorganized. Quantitative real time-PCR revealed that phosphorylated tau mRNA was upregulated after ketamine. Western blot analysis demonstrated that phosphorylated tau levels at serine 396 initially decreased at 1 day after ketamine injection, and then gradually returned to control values. At 14 days after injection, levels of phosphorylated tau were higher in the ketamine group than in the control group. Tau protein phosphorylated at serine 404 significantly increased after ketamine injection, and then gradually decreased with time. However, the levels of tau protein at serine 404 were significantly greater in the ketamine group than in the control group until 14 days. | 10 | Hypoxia-induced GBE1 expression promotes tumor progression through metabolic reprogramming in lung adenocarcinoma显示文摘Hypoxia mediates a metabolic switch from oxidative phosphorylation to glycolysis and increases glycogen synthesis.We previously found that glycogen branching enzyme(GBE1)is downstream of the hypoxia-inducible factor-1(HIF1)signaling pathway in lung adenocarcinoma(LUAD)cells;however,the molecular mechanism underlying HIF1 regulation of GBE1 expression remains unknown.Herein,the effect of GBE1 on tumor progression via changes in metabolic signaling under hypoxia in vitro and in vivo was evaluated,and GBE1-related genes from human specimens and data sets were analyzed.Hypoxia induced GBE1 upregulation in LUAD cells.GBE1-knockdown A549 cells showed impaired cell proliferation,clone formation,cell migration and invasion,angiogenesis,tumor growth,and metastasis.GBE1 mediated the metabolic reprogramming of LUAD cells.The expression of gluconeogenesis pathway molecules,especially fructose-1,6-bisphosphatase(FBP1),was markedly higher in shGBE1 A549 cells than it was in the control cells.FBP1 inhibited the tumor progression of LUAD.GBE1-mediated FBP1 suppression via promoter methylation enhanced HIF1αlevels through NF-κB signaling.GBE1 may be a negative prognostic biomarker for LUAD patients.Altogether,hypoxia-induced HIF1αmediated GBE1 upregulation,suppressing FBP1 expression by promoter methylation via NF-κB signaling in LUAD cells.FBP1 blockade upregulated HIF1α,triggered the switch to anaerobic glycolysis,and enhanced glucose uptake.Therefore,targeting HIF1α/GBE1/NF-κB/FBP1 signaling may be a potential therapeutic strategy for LUAD. | Lifeng Li Li Yang Zhirui Fan Wenhua Xue Zhibo Shen Yongliang Yuan Xiangdong Sun Dan Wang Jingyao Lian Liping Wang Jie Zhao Yi Zhang | 2020 | Signal Transduction and Targeted Therapy2020,5,1: | 3 |

| 12 | Enhanced carbon acquisition and use efficiency alleviate microbial carbon relative to nitrogen limitation under soil acidification显示文摘Background:Soil microbial communities cope with an imbalanced supply of resources by adjusting their element acquisition and utilization strategies.Although soil pH has long been considered an essential driver of microbial growth and community composition,little is known about how soil acidification affects microbial acquisition and utilization of carbon(C)and nitrogen(N).To close the knowledge gap,we simulated soil acidification and created a pH gradient by adding eight levels of elemental sulfur(S)to the soil in a meadow steppe.Results:We found that S-induced soil acidification strongly enhanced the ratio of fungi to bacteria(F:B)and microbial biomass C to N(MBC:MBN)and subsequently decreased the C:N imbalance between microbial biomass and their resources.The linear decrease in the C:N imbalance with decreasing soil pH implied a conversion from N limitation to C limitation.To cope with enhanced C versus N limitation,soil microbial communities regulated the relative production of enzymes by increasing the ratio ofβ-glucosidase(BG,C-acquiring enzyme)to leucine aminopeptidase(LAP,N-acquiring enzyme),even though both enzymatic activities decreased with S addition.Structural equation modeling(SEM)suggested that higher C limitation and C:N-acquiring enzyme stimulated microbial carbon-use efficiency(CUE),which counteracted the negative effect of metal stress(i.e.,aluminum and manganese)under soil acidification.Conclusions:Overall,these results highlight the importance of stoichiometric controls in microbial adaption to soil acidification,which may help predict soil microbial responses to future acid deposition. | Tianpeng Li Ruzhen Wang Jiangping Cai Yani Meng Zhirui Wang Xue Feng Heyong Liu Ronald F.Turco Yong Jiang | 2021 | Ecological Processes2021,10,1: | 2 |

We selected 64 prepubertal children with FT4 levels in the lowest third of the normal range as the lowerFT4 group, and these children were divided randomly into two subgroups: L-thyroxine (L-T4)-treated subgroupwas treated with L-T4 (0.5-3.0 g/(kg, d)) from the beginning of the study, and the non-L-T4-treated subgroupreceived placebo. We also selected 39 ISS children with FT4 in the upper two-thirds of the normal range as thehigher FT4 group. During the first year, the lower FT4 group featured lower FT3, FT4, thyroid stimulatinghormone (TSH), and insulin-like growth factor-I standard deviation score (IGF-I SDS) and significantly lowerheight velocity (HV) compared with the higher FT4 group. However, in the lower FT4 group, the L-T4-treatedsubgroup presented higher FT4, FT3, TSH, and IGF-I SDS concentrations and significantly higher HV comparedwith children in the non-L-T4-treated subgroup. | 16 | Hyperactive PI3Kδ predisposes naive T cells to activation via aerobic glycolysis programs显示文摘Activated phosphoinositide 3-kinaseδsyndrome(APDS)is an autosomal-dominant combined immunodeficiency disorder resulting from pathogenic gain-of-function(GOF)mutations in the PIK3CD gene.Patients with APDS display abnormal T cell homeostasis.However,the mechanisms by which PIK3CD GOF contributes to this feature remain unknown.Here,with a cohort of children with PIK3CD GOF mutations from multiple regions of China and a corresponding CRISPR/Cas9 gene-edited mouse model,we reported that hyperactive PI3Kδdisrupted TNaive cell homeostasis in the periphery by intrinsically promoting the growth,proliferation,and activation of TNaive cells.Our results showed that PIK3CD GOF resulted in loss of the quiescence-associated gene expression profile in naive T cells and promoted naive T cells to overgrow,hyperproliferate and acquire an activated functional status.Naive PIK3CD GOF T cells exhibited an enhanced glycolytic capacity and reduced mitochondrial respiration in the resting or activated state.Blocking glycolysis abrogated the abnormal splenic T cell pool and reversed the overactivated phenotype induced by PIK3CD GOF in vivo and in vitro.These results suggest that enhanced aerobic glycolysis is required for PIK3CD GOF-induced overactivation of naive T cells and provide a potential therapeutic approach for targeting glycolysis to treat patients with APDS as well as other immune disorders. | Yanjun Jia Qiuyun Yang Yanping Wang Wenyan Li Xuemei Chen Tao Xu Zhirui Tian Minxuan Feng Liang Zhang Wenjing Tang Na Tian Lina Zhou Wenxia Song Xiaodong Zhao | 2021 | Cellular & Molecular Immunology2021,18,7: | 2 |
