| 3 | Clinical applications of MARSALA for preimplantation genetic diagnosis of spinal muscular atrophy显示文摘Conventional PCR methods combined with linkage analysis based on short tandem repeats(STRs) or Karyomapping with single nucleotide polymorphism(SNP) arrays, have been applied to preimplantation genetic diagnosis(PGD) for spinal muscular atrophy(SMA), an autosome recessive disorder. However, it has limitations in SMA diagnosis by Karyomapping, and these methods are unable to distinguish wildtype embryos with carriers effectively. Mutated allele revealed by sequencing with aneuploidy and linkage analyses(MARSALA) is a new method allowing embryo selection by a one-step next-generation sequencing(NGS) procedure, which has been applied in PGD for both autosome dominant and X-linked diseases in our group previously. In this study, we carried out PGD based on MARSALA for two carrier families with SMA affected children. As a result, one of the couples has given birth to a healthy baby free of mutations in SMA-causing gene. It is the first time that MARSALA was applied to PGD for SMA, and we can distinguish the embryos with heterozygous deletion(carriers) from the wild-type(normal) ones accurately through this NGS-based method. In addition, direct mutation detection allows us to identify the affected embryos(homozygous deletion), which can be regarded as probands for linkage analysis, in case that the affected family member is absent. | 4 | Potential role of Lactobacillus plantarum in colitis induced by dextran sulfate sodium through altering gut microbiota and host metabolism in murine model显示文摘Inflammatory bowel disease(IBD) is a chronic lifelong disease characterized by inflammation of the gastrointestinal tract.Although more and more treatment options serve IBD, there is still no cure. It is important to find an effective treatment for IBD.This study aims to investigate whether Lactobacillus plantarum(L. plantarum) could alleviate colitis induced by dextran sulfate sodium(DSS). Following the DSS challenge, L. plantarum on DSS-mediated inflammatory colon lesions in mice, and L.plantarum therapy heightened the relative abundance of the colon-resident Actinobacteria. Analysis of serum metabolomics also indicated that the content of MG(18:4(6 Z, 9 Z, 12 Z, 15 Z)/0:0/0:0) was increased in response to L. plantarum therapy, and this was also the case for indolepyruvate and 1-hydroxyibuprofen. However, 13-oxooctadecadienoic acid(13-oxo ODE) and indolylacryloylglycine content fell following the DSS challenge. Based on these results, the study elucidates the mitigatory effects of L. plantarum in colitis, which depend on its regulation of the colonic microbial community and its modification of serum metabolites. The results revealed that L. plantarum mitigated inflammatory colon lesions, reprogrammed the microbial community and altered the level of serum metabolites in a murine model challenged with DSS. | 5 | The YDA-MKK4/MKK5-MPK3/MPK6 Cascade Functions Downstream of the RGF1-RGI Ligand-Receptor Pair in Regulating Mitotic Activity in Root Apical Meristem显示文摘The mitotic activity of root apical meristem(RAM)is critical to primary root growth and development.Previous studies have identified the roles of ROOT GROWTH FACTOR 1(RGF1),a peptide ligand,and its receptors,RGF1 INSENSITIVEs(RGIs),a clade of five leucine-rich-repeat receptor-like kinases,in promoting cell division in the RAM,which determines the primary root length.However,the downstream signaling components remain elusive.In this study,we identify a complete mitogen-activated protein kinase(MAPK or MPK)cascade,composed of YDA,MKK4/MKK5,and MPK3/MPK6,that functions downstream of the RGF1-RGI ligand-receptor pair.Similar to the rgi1/2/3/4/5 quintuple mutant,loss-of-function mutants of MPK3 and MPK6,MKK4 and MKK5,or YDA show a short-root phenotype,which is associated with reduced mitotic activity and lower expression of PLETHORA 1(PLT1)/PLT2 in the RAM.Furthermore,MPK3/MPK6 activation in response to exogenous RGF1 treatment is impaired in the rgi1/2/3/4/5 quintuple,yda single,and mkk4 m kk5 double mutants.Epistatic analyses demonstrated that the expression of constitutively active MKK4,MKK5,or YDA driven by the RGI2 promoter can rescue the short-root phenotype of the rgi1/2/3/4/5 mutant.Taken together,these results suggest that the YDA-MKK4/MKK5-MPK3/MPK6 cascade functions downstream of the RGF1-RGI ligand-receptor pair and upstream of PLT1/PLT2 to modulate the stem cell population and primary root growth in Arabidopsis. | Yiming Shao Xinxing Yu Xuwen Xu Yong Li Wenxin Yuan Yan Xu Chuanzao Mao Shuqun Zhang Juan Xu | 2020 | Molecular Plant2020,13,11: | 6 |
| 6 | Genetic analysis and preimplantation genetic diagnosis of Chinese Marfan syndrome patients显示文摘Marfan syndrome (MFS)(OMIM 154700) is a relatively common autosomal dominant genetic disease that causes skeletal, ocular, and cardiovascular defects and was first described by a French pediatrician in 1896 (Bitterman and Sponseller, 2017). Its prevalence rate is 1/3000—1/5000, and more than 25% of cases are sporadic (Chiu et al., 2014). Studies have shown that about 90% of MFS is caused by variants in the fibrillin-1 gene (FBN1, OMIM 134797). FBN1, located on chromosome 15q21.1, encodes a macromolecular glycoprotein-fibrin 1, which aggregates to form microfibers in the extracellular matrix and distributes in various human connective tissues, such as periosteum, vessel wall, and crystal suspensor ligament. Variants in FNB1 have been reported in 65 exons, but the relationship between genotype and phenotype remains rather unclear (Sakai et al., 2016). Studies have also shown that patients with MFS and similar diseases may have variants in other related genes such as members of the transforming growth factor beta receptor (TGFBR) family (Mizuguchi et al., 2004;Sakai et al., 2006;Bolar et al., 2012;De Cario et al., 2018). | 11 | The cost of obsessive- compulsive disorder (OCD) in China: a multi- center cross- sectional survey based on hospitals显示文摘Background Obsessive-compulsive disorder(OCD)is considered a very debilitating disorder with severe loss of quality of life and income.Aims This study estimates the quality of life and economic consequences of OCD in China.Methods The research team interviewed 639 patients with OCD in 13 hospitals in 12 cities in China.The direct method was used to get the direct cost of OCD.Indirect costs associated with OCD were estimated using the human capital approach.Linear regression analysis was conducted for quality of life and generalised linear model analysis was conducted for total cost.Sensitivity analysis was used to analyse the uncertainty of total cost.Results The mean quality of life score for OCD was 52.78(20.46).The annual total cost of OCD per capita was 24503.78(95%CI:22621.53 to 26386.03)renminbi(RMB)(US$3465.88(95%CI:US$3199.65 to US$3732.11)).The annual cost of OCD in China was estimated to be 37.74 billion(95%CI:34.95 billion to 40.53 billion)RMB(equal to US$5.34 billion(95%CI:US$4.94 billion to US$5.73 billion)).Sensitivity analysis showed that the total annual cost of OCD in China was between 23.15 billion RMB(US$3.27 billion)and 370.00 billion RMB(US$52.33 billion).Worse social function status,more psychiatric symptoms and higher Yale Brown Obsessive-Compulsive Scale(Y-BOCS)score were associated with worse quality of life.The numbers of clinic visits and hospitalisations,socioeconomic status,education,Y-BOCS scores and age were found to be significantly associated with total cost.Conclusions OCD is associated with low quality of life and high costs in China.The findings call for concerted efforts to improve services for patients with OCD.Improvements may include early detection and diagnosis,the provision of evidence-based treatments and relapse prevention strategies. | Weili Yang Zhen Tang Xijin Wang Xiancang Ma Yuqi Cheng Bin Wang Ping Sun Wenxin Tang Jia Luo Changhong Wang Ping Li Guiyun Xu Jun Yan Vlasios Brakoulias Zhen Wang | 2021 | General Psychiatry2021,34,6: | 3 |

| 14 | Prediction of a common neutralizing epitope of H5N1 avian influenza virus by in silico molecular docking显示文摘The H5N1 avian influenza virus (AIV) has widely spread in Asia, Europe and Africa, making a large amount of economic loss. Recently, our research group has screened a common neutralizing mono- clonal antibody named 8H5, which can neutralize almost all H5 subtype AIV ever isolated so far. Obvi- ously, this monoclonal antibody would benefit for research and development of the universal AIV vac- cine and design of the drug against H5N1 AIV in high mutation rate. In this study, the homology mod- eling was applied to generate the 3D structure of 8H5 Fab fragment, and 'canonical structure' method was used to define the specified loop conformation of CDR regions. The model was subjected to en- ergy minimization in cvff force field with Discovery module in Insight II program. The resulting model has correct stereochemistry as gauged from the Ramachandran plot calculation and good 3D-structure compatibility as assessed by interaction energy analysis, solvent accessible surface (SAS) analysis, and Profiles-3D approach. Furthermore, the 8H5 Fab model was subjected to docking with three H5 subtype hemagglutinin (HA) structures deposited in PDB (ID No: 1jsm, 2ibx and 2fk0) respectively. The result indicates that the three docked complexes share a common binding interface, but differ in bind- ing angle related with HA structure similarity between viral subtypes. In the light of the three HA inter- faces with structural homology analysis, the common neutralizing epitope on HA recognized by 8H5 consists of 9 incontinuous amino acid residues: Asp68, Asn72, Glu112, Lys113, Ile114, Pro118, Ser120, Tyr137, Tyr252 (numbered as for 1jsm sequence). | 18 | Quasi-Spherical Brookite TiO_2 Nanostructures Synthesized Using Solvothermal Method in the Presence of Oxalic Acid显示文摘Brookite TiO_2,the latest TiO_2 photocatalyst,promises to be an interesting candidate for photocatalytic applications because of its unique physical and chemical properties. In this study,pure-phase brookite TiO_2 with a quasi-spherical nanostructure was successfully synthesized via a solvothermal method using tetrabutyl titanate(Ti(OC_4H_9)_4,TBOT) as the Ti source in the presence of oxalic acid. NaOH was used to regulate the pH of solution. The structure and morphology of the samples were then analyzed using multiple methods,such as X-ray diff raction(XRD),Raman spectroscopy,scanning electron microscopy(SEM),transmission electron microscopy(TEM),Brunauer–Emmett–Teller(BET) measurements and ultraviolet–visible diff use spectroscopy(UV–Vis). Photocatalytic activities of the as-synthesized brookite TiO_2 were evaluated by degrading aqueous methylene blue solution under UV light irradiation. The effect of thermal treatment temperature on photocatalytic activity of the samples was also investigated. The produced brookite TiO_2 nanopowders calcined at 500 °C for 2 h showed the highest photocatalytic activity,and the corresponding degradation rate of methylene blue(10 mg/L) reached 96.7% after 90 min of illumination. In addition,the formation mechanism of pure-phase brookite TiO_2 was investigated. | 20 | Trends of SARS-CoV-2 Infection in Rural Area in Sentinel Community-Based Surveillance——China,December 2022 to January 2023显示文摘Introduction:In late 2022,a rapid transmission of Omicron variants of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)occurred throughout China.The purpose of this study was to provide the latest data and evaluate trends of SARS-CoV-2 infection in rural China among the community population.Methods:Data on SARS-CoV-2 infection among approximately 90,000 participants in rural China were collected by the National Sentinel Community-Based Surveillance(NSCS)system.Participants were tested for SARS-CoV-2 infection(defined as positive for SARS-CoV-2 nucleic acid or antigen)twice weekly from December 16,2022 to January 12,2023.The daily average of newly positive rate and its estimated daily percentage change were calculated to describe the national and regional trends of SARS-CoV-2 infection in rural China.Results:In rural China,the daily average new positive rate of SARS-CoV-2 infection peaked at 4.79%between December 20–22,2022 and then decreased to 0.57%between January 10–12,2023,with an average decrease of 29.95%per round.The peak of new SARS-CoV-2 infection was slightly earlier and lower in North China(5.28%between December 20–22,2022)than in South China(5.63%between December 23–26,2022),and then converged from December 30,2022 to January 2,2023.The peak of 6.09%occurred between December 20–22,2022 in eastern China,while the peak of 5.99%occurred later,between December 27–29,2022,in central China.Conclusions:Overall,the epidemic wave in rural China peaked between December 20–22,2022,and passed quickly following the optimization of prevention and control measures.Currently,SARSCoV-2 infection in community populations in rural China is sporadic. | Wenzhan Jing Fan Ding Yanping Zhang Guoqing Shi Jue Liu Yu Wu Fangyuan Li Wenxin Yan Chenyuan Qin Min Liu Qun Li Wannian Liang | 2023 | China CDC weekly2023,5,11: | 1 |
