5Super enhancers targeting ZBTB16 in osteogenesis protect against osteoporosis显示文摘As the major cell precursors in osteogenesis, mesenchymal stem cells(MSCs) are indispensable for bone homeostasis and development. However, the primary mechanisms regulating osteogenic differentiation are controversial. Composed of multiple constituent enhancers, super enhancers(SEs) are powerful cis-regulatory elements that identify genes that ensure sequential differentiation. The present study demonstrated that SEs were indispensable for MSC osteogenesis and involved in osteoporosis development. Through integrated analysis, we identified the most common SE-targeted and osteoporosis-related osteogenic gene,ZBTB16. ZBTB16, positively regulated by SEs, promoted MSC osteogenesis but was expressed at lower levels in osteoporosis.Mechanistically, SEs recruited bromodomain containing 4(BRD4) at the site of ZBTB16, which then bound to RNA polymerase IIassociated protein 2(RPAP2) that transported RNA polymerase Ⅱ(POL Ⅱ) into the nucleus. The subsequent synergistic regulation of POL Ⅱ carboxyterminal domain(CTD) phosphorylation by BRD4 and RPAP2 initiated ZBTB16 transcriptional elongation, which facilitated MSC osteogenesis via the key osteogenic transcription factor SP7. 7Targeting HECTD3-IKKα axis inhibits inflammation-related metastasis显示文摘Metastasis is the leading cause of cancer-related death.The interactions between circulating tumor cells and endothelial adhesion molecules in distant organs is a key step during extravasation in hematogenous metastasis.Surgery is a common intervention for most primary solid tumors.However,surgical trauma-related systemic inflammation facilitates distant tumor metastasis by increasing the spread and adhesion of tumor cells to vascular endothelial cells(ECs).Currently,there are no effective interventions to prevent distant metastasis.Here,we show that HECTD3 deficiency in ECs significantly reduces tumor metastasis in multiple mouse models.HECTD3 depletion downregulates expression of adhesion molecules,such as VCAM-1,ICAM-1 and E-selectin,in mouse primary ECs and HUVECs stimulated by inflammatory factors and inhibits adhesion of tumor cells to ECs both in vitro and in vivo.We demonstrate that HECTD3 promotes stabilization,nuclear localization and kinase activity of IKKa by ubiquitinating IKKa with K27-and K63-linked polyubiquitin chains at K296,increasing phosphorylation of histone H3 to promote NF-kB target gene transcription.Knockout of HECTD3 in endothelium significantly inhibits tumor cells lung colonization,while conditional knockin promotes that.IKKa kinase inhibitors prevented LPS-induced pulmonary metastasis.These findings reveal the promotional role of the HECTD3-IKKa axis in tumor hematogenous metastasis and providea potential strategy for tumormetastasis prevention.Fubing Li Huichun Liang Hua You Ji Xiao Houjun Xia Xi Chen Maobo Huang Zhuo Cheng Chuanyu Yang Wenjing Liu Hailin Zhang Li Zeng Yingying Wu Fei Ge Zhen Li Wenhui Zhou Yi Wen Zhongmei Zhou Rong Liu Dewei Jiang Ni Xie Bin Liang Zhenzhen Liu Yanjie Kong Ceshi Chen 2022Signal Transduction and Targeted Therapy2022,7,9:1
8Efficacy and safety of S-1 based adjuvant chemoradiotherapy for resected pancreatic ductal adenocarcinoma with high-risk pathological feature: a prospective, single-arm, interventional study显示文摘Objective: It remains unclear whether adjuvant chemoradiotherapy (CRT) improves survival outcome of pancreatic ductal adenocarcinoma (PDAC) patients after surgery. This study aimed to investigate the efficacy and safety of tegafur/gimeracil/oteracil (S-1)-based adjuvant concurrent chemoradiotherapy in resected PDAC patients with defined high-risk pathological features.Methods: We conducted a single-arm, prospective, and interventional study at Zhongshan Hospital Fudan University from December 2012 to December 2019 and the last follow-up was conducted in December 2021. This study was approved by the Ethics Committee of Zhongshan Hospital Fudan University on December 27, 2012 (approval No. B2012-139). Resected PDAC patients with high-risk pathological features, including positive resection margin, pathological T3-4N1-2M0 disease, peripancreatic fat invasion, microvascular invasion, and perineural invasion, were recruited. Primary endpoint was overall survival and secondary endpoints were disease-free survival, treatment toxicity, and 2-, 5-year survival rates.Results: A total of 54 patients were recruited. Mean age was 63.6 years old (±7.2). The distribution of T and N stages were 24.1% for T1, 46.3% for T2, 27.8% for T3, 1.9% for T4, 33.3% for N1, and 11.1% for N2. Seven patients had R1 resection. The median overall survival and disease-free survival were 27.1 and 13.7 months, respectively, while no fatal adverse events were recorded. Subgroup analyses showed differences in survival outcomes among patients with microvascular invasion, different N stages, and preoperative CA 19-9 levels. 10Context-Aware Recommendation in Mobile Environments:An Approach Based on Interest Resonance显示文摘The recommendation system can efficiently solve the information overload in mobile Internet. Thus, how to effectively utilize context information to improve the accuracy of recommendation becomes the research focus in the field. This article puts forward a novel approach to realize the context-aware recommendation in mobile environments. It first gets users' interest resonance with a hash-based interest resonance mining algorithm. Then, it calculates the association degree between the user and the item and then predicts the user's rating on the item. Finally, it comprehensively figures out the recommending index. Moreover, this article also designs a personal recommendation model for the users and provides relevant decision-making coefficients. 13An activatable ratiometric near-infrared fluorescent probe for hydrogen sulfide imaging in vivo显示文摘Ratiometric fluorescent probes hold great promise for in vivo imaging;however,stimuli-activatable ratiometric probes with fluorescence emissions in near-infrared(NIR)region are still very few.Herein,we report a hydrogen sulfide(H_2S)-activatable ratiometric NIR fluorescent probe(1-SPN)by integrating a H_2S-responsive NIR fluorescent probe 1 into a H_2S-inert poly[2,6-(4,4-bis-(2-ethylhexyl)-4 H-cyclopenta[2,1-b;3,4-b′]dithiophene)-alt-4,7(2,1,3-benzothiadiazole)](PCPDTBT)-based NIR semiconducting polymer nanoparticle(SPN).1-SPN shows'always on'PCPDTBT fluorescence at 830 nm and weak probe 1 fluorescence at 725 nm under excitation at 680 nm.The ratio of NIR fluorescence intensities between 725 and 830 nm(I_(725)/I_(830))is small.Upon interaction with H_2S,the fluorescence at 725 nm is rapidly switched on,resulting in a large enhancement of I_(725)/I_(830),which is allowed for sensitive visualization and quantification of H_2S concentrations in living cells.Taking advantage of enhanced tissue penetration depth of NIR fluorescence,1-SPN is also applied for real-time ratiometric fluorescence imaging of hepatic and tumor H_2S in living mice.This study demonstrates that activatable ratiometric NIR fluorescent probes hold great potential for in vivo imaging.Luyan Wu Wenhui Zeng Liandong Feng Yuxuan Hu Yidan Sun Yingxiao Yan Hong-Yuan Chen Deju Ye 2020Science China Chemistry2020,63,5:0

17Neutralization against SARS-CoV-2 Delta/Omicron variants and B cell response after inactivated vaccination among COVID-19 convalescents显示文摘Emerging SARS-CoV-2 variants have made COVID-19 convalescents susceptible to re-infection and have raised concern about the efficacy of inactivated vaccination in neutralization against emerging variants and antigen-specific B cell response.To this end,a study on a long-term cohort of 208 participants who have recovered from COVID-19 was conducted,and the participants were followed up at 3.3(Visit 1),9.2(Visit 2),and 18.5(Visit 3)months after SARS-CoV-2 infection.They were classified into three groups(no-vaccination(n=54),one-dose(n=62),and two-dose(n=92)groups)on the basis of the administration of inactivated vaccination.The neutralizing antibody(NAb)titers against the wild-type virus continued to decrease in the no-vaccination group,but they rose significantly in the one-dose and two-dose groups,with the highest NAb titers being observed in the two-dose group at Visit 3.The NAb titers against the Delta variant for the no-vaccination,one-dose,and two-dose groups decreased by 3.3,1.9,and 2.3 folds relative to the wild-type virus,respectively,and those against the Omicron variant decreased by 7.0,4.0,and 3.8 folds,respectively.Similarly,the responses of SARS-CoV-2 RBD-specific B cells and memory B cells were boosted by the second vaccine dose.Results showed that the convalescents benefited from the administration of the inactivated vaccine(one or two doses),which enhanced neutralization against highly mutated SARS-CoV-2 variants and memory B cell responses.Two doses of inactivated vaccine among COVID-19 convalescents are therefore recommended for the prevention of the COVID-19 pandemic,and vaccination guidelines and policies need to be updated.Hao Wang Yu Yuan Bihao Wu Mingzhong Xiao Zhen Wang Tingyue Diao Rui Zeng Li Chen Yanshou Lei Pinpin Long Yi Guo Xuefeng Lai Yuying Wen Wenhui Li Hao Cai Lulu Song Wei Ni Youyun Zhao Kani Ouyang Jingzhi Wang Qi Wang Li Liu Chaolong Wang An Pan Xiaodong Li Rui Gong Tangchun Wu 2023Frontiers of Medicine2023,17,4:0
18The Dendritic Cells’ Immunological Behaviors Modulated by the Spatial Confinements of Deposited Fibrin Matrix显示文摘Implant materials,as foreign objects to host,can cause various degrees of inflammation in most cases.The inflammation is triggered by a series of immune responses and directly impacts the tissue regeneration process,which determines the outcome of tissue repair.The immune responses are complex process involving numerous immune cells and can be divide into innate immune and adaptive immune responses.Once materials are implanted,innate immune responses are activated under the mediation of several immune cells(e.g.neutrophils and macrophages),meanwhile immature dendritic cells(imDCs)are recruited to the implant sites to recognize,internalize and process antigens.Upon antigen uptake,imDCs gradually differentiate into mature dendritic cells(mDCs)and migrate to secondary lymph nodes.In the lymph nodes,mDCs present processed antigen peptides to naive T lymphocytes and activate their antigen specific proliferation,resulting in initiation of adaptive immune responses.Due to their key position in the immune system,serving to bridge innate and adaptive immunity,DCs are crucial to guiding and modulating the immune responses caused by implanted materials.Therefore,figuring out the response of DCs to implanted materials and the exact role of DCs in tissue healing processes will provide deeper insight for the rational design of biomaterials.Previous studies on the effects of implants on immune functions of DCs are mainly focused on physical and chemical properties of the materials(e.g.released chemical composition,surface chemistry,substrate stiffness and surface topography).All these factors will change the microenvironment of the tissue around implant materials,which affect the immune functions of DCs.However,the change of microenvironment not only directly derives from the physical and chemical properties of the material(intrinsic),but also indirectly results from the remodeled extracellular matrix(ECM)caused by implanted materials.When blood or tissue fluid contact with materials after implantation,proteins(e.g.fibrin and collagen)will absorb and deposit on the surface of implants,leading to a provisionally stable matrix with microporous fibrous-liked network structure.It means that the remodeled ECM can provide adhesion sites for recruited DCs and form spatial confinement.DCs,as a kind of cells that are extremely sensitive to mechanical stimuli,theoretically,can response to the mechanical stimuli coming from spatial confinement of remodeled ECM,which may lead to a series of modulations in their cell morphologies and immune functions.Then,the remodeled ECM is a non-negligible mechanical cue.However,to the best of our knowledge,there is a lack of a simple and effective model to establish the relationship between the immune functions of DCs and remodeled ECM.Most studies on the responses of DCs to implanted materials are still based on suspension culture model,which is the normal status of DCs in vitro culture systems.In addition,the processes by which DC exerts immune functions(both endocytosis and antigen presentation)are dynamically physical interaction.It means that the changes of DCs’immune functions are highly correlated with the changes of their biomechanical characteristics caused by remodeled ECM.In this work,we have found that the ECM was remodeled by a large amount of fibrin matrix deposited on the surface of implants in the early stage of the inflammations following implantation.Thus,we used non-toxic salmon fibrin hydrogels with microporous fibrous-liked network structure to mimic the deposited fibrin matrix.Then,human monocyte-derived DCs were cultured on the surface and inside of the fibrin hydrogels to mimic the different spatial confinement states of fibrin matrix.Our results indicated that cell morphologies and cytoskeleton structures of DCs were regulated by the spatial confinement of fibrin hydrogels,resulting in generating mechanical stimuli for DCs.Furthermore,we have found that the biomechanical characteristics and the immune functions of both imDCs and mDC were also modulated.Considering the changes in surface markers,secreted cytokines and biomechanical characteristics of DCs,it indicates that the tendency and magnitude of modulations were highly associated with the spatial confinement of fibrin hydrogels.This model demonstrated that mechanical stimuli deriving from spatial confinement of deposited fibrin matrix is an important factor for regulating the biomechanical characteristics and immune functions of DCs.Wenhui Hu Yun Wang Jin Chen Yonggang Song Jinhua Long Zhu Zeng 2019医用生物力学2019,34,A01:0
19Diets enriched with finely ground wheat bran alter digesta passage rate and composition of the gut microbiome in sows显示文摘We investigated the effects of finely ground wheat bran on the nutrient digestibility, digesta passage rate,and gut microbiota structure in sows. A 3 × 3 Latin square design with 3 test periods and 3 experimental diets was used. Six non-pregnant sows(parity: 5 to 7) were randomly assigned to 3 experimental diets with 2 replicates per treatment in each period. Each period lasted 19 d(12 d for adaptation and 7 d for experiment). The experimental diets included(a) a basal corn and soybean meal diet(CON),(b) a basal diet with 20% coarse wheat bran(CWB;particle size: 605 μm), and(c) a basal diet with 20% fine wheat bran(FWB;particle size: 438 μm). The results demonstrated that the apparent total tract digestibility of neutral detergent fiber, acid detergent fiber and energy were reduced(P < 0.05) in the FWB and CWB groups compared with those in the CON group. Viscosity of digesta increased(P < 0.001) in FWB-fed sows. The passage rate of digesta from the mouth to the ileum decreased(P < 0.001) in FWB-fed sows. Peptide YY(PYY) concentration increased(P = 0.01) in FWB-fed sows after 30 min of feeding.In the FWB group, the relative abundance of Lactobacillaceae at the family level increased(P < 0.05) in the ileal digesta. At the class level, the relative abundance of Clostridia in feces decreased(P < 0.05) in FWB-fed sows. FWB enhanced the concentration of butyrate in feces compared with CON and CWB(P = 0.04). These results suggest that dietary supplementation with finely ground wheat bran reduces the passage rate of digesta, increases the abundance of beneficial microorganisms, and elevates the concentration of short-chain fatty acids and PYY in sows. 20Cyanidin-3-O-glucoside alleviates trimethyltin chloride-induced neurodegeneration by maintaining glutamate homeostasis through modulation of the gut microbiota显示文摘Trimethyltin chloride(TMT)is a potent neurotoxin to cause neurodegeneration,especially in hippocampus.This study aimed to identify dietary components that can effectively attenuate TMT-induced neurodegeneration in humans.The predominant anthocyanin in human diets,cyanidin-3-O-glucoside(C3G,5 or 50 mg/kg),was given to mice for 16 days,and TMT(2.7 mg/kg)was injected intraperitoneally once on the eighth day.C3G(50 mg/kg)significantly alleviated TMT-induced seizures and subsequent cognitive impairment by ameliorating hippocampal neurodegeneration and synaptic dysfunction.Furthermore,C3G treatment restored glutamate homeostasis in brain and reversed glutamine synthetase(GS)inhibition in reactive astrogliosis and neuroinflammation,which are critical for C3G's neuroprotective effects.Notably,C3G decreased the lipopolysaccharide,tumor necrosis factor-α,interleukin-6,and interleukin-1βlevels in the mice,which potentially by modulating the relative abundance of Atopobiaceae and Lachnospiraceae in the gut.C3G may be a promising and practical dietary component for reducing TMT-induced neurodegeneration.Yu Xi Wenhui Li Junru Wang Meihong Yu Xiangquan Zeng He Li Jian Li 2024Food Science and Human Wellness2024,13,2:0
