1Mogroside V inhibits LPS-induced COX-2 expression/ROS production and overexpression of HO-1 by blocking phosphorylation of AKT1 in RAW264.7 cells显示文摘Momordica grosvenori is a valuable edible plant with medicinal purposes, and it is widely used in medicated diets and traditional Chinese medicine in Asia. Mogroside V (MV), the main bioactive component from M. grosvenori, is comm only used as a natural sweetener. M. grosvenori extracts have been reported to exert potent anti-inflammatory property, however the underlying molecular mechanism still remains unknown. In the present study, the biological effect of MV in inflammation was investigated in lipopolysaccharide (LPS)-stimulated RAW264.7 cells. The ELISA and western blot analysis results showed that MV significantly inhibited LPS-induced prostaglandin E2 (PGE2) production and cyclooxygenase-2 (COX-2) expression. MV markedly decreased the phosphorylation of IxB-a, increased IkB-oc, and reduced nuclear p-65 and C/EBP6. Furthermore, MV attenuated LPS-induced phosphorylation of MAPKs and AKT1, and only the phosphorylation status of AKT1 was found to be consistent with the expression trend of COX-2. Moreover, MV reduced ROS level and restored overexpressed HO-1 and AP-1 to basal level, which can be markedly reversed by AKT1 inhibitor LY294002. These results revealed that AKT1 plays a key role in LPS-induced COX-2 expression, and acts as a mediator to keep the redox balance in LPS-stimulated RAW264.7 cells. MV exerts anti-inflammatory property by blocking AKT1-mediated NF-kB and C/EBP6 activation, ROS generation and AP-1/ HO-1 expression. 4Quantitative trait locus mapping with background control in genetic populations of clonal F_1 and double cross显示文摘In this study, we considered five categories of molecular markers in clonal F_1 and double cross populations,based on the number of distinguishable alleles and the number of distinguishable genotypes at the marker locus. Using the completed linkage maps, incomplete and missing markers were imputed as fully informative markers in order to simplify the linkage mapping approaches of quantitative trait genes. Under the condition of fully informative markers, we demonstrated that dominance effect between the female and male parents in clonal F_1 and double cross populations can cause the interactions between markers. We then developed an inclusive linear model that includes marker variables and marker interactions so as to completely control additive effects of the female and male parents, as well as the dominance effect between the female and male parents. The linear model was finally used for background control in inclusive composite interval mapping(ICIM) of quantitative trait locus(QTL). The ef ficiency of ICIM was demonstrated by extensive simulations and by comparisons with simple interval mapping, multiple-QTL models and composite interval mapping. 5Degradable Hybrid CuS Nanoparticles for Imaging-Guided Synergistic Cancer Therapy via Low-Power NIR-II Light Excitation显示文摘Near-infrared(NIR)-II light-excitable photonic agents capable of generating tumor hyperthermia and cytotoxic free radicals are promising for synergistic phototherapy of tumors.However,the lack of NIR-II excitable agents makes it challenging to achieve combinational tumor phototherapy.Here,the authors have reported on a tumor-targeting and degradable hybrid copper sulfide(CuS)nanoparticle(AIBA@CuS-FA)via loading a hydrophilic Azo initiator(AIBA)into an amphiphilic lipid-encapsulating CuS nanoparticle.AIBA@CuS-FA shows high photothermal conversion efficiency(PCE≈47.5%)at 1064 nm,enabling heat production to trigger tumor hyperthermia and thermal decomposition of AIBA into cytotoxic free alkyl radicals upon irradiation with a 1064-nm laser under low-power density(0.5 W/cm2).Moreover,alkyl radicals can drive degradation of AIBA@CuS-FA and embedded CuS nanodisks,releasing Cu^(2+)ions that can catalyze a Fenton-like reaction for hydroxyl radical(•OH)production to promote tumor therapy.Findings demonstrate promise for combinational photothermal therapy(PTT),oxygen-independent alkyl radical therapy,and chemodynamic therapy(CDT)of tumors.Yidan Sun Hua Shi Xiaoyang Cheng Luyan Wu Yuqi Wang Zhengyang Zhou Jian He Hong-Yuan Chen Deju Ye 2021CCS Chemistry2021,3,5:2

8An activatable ratiometric near-infrared fluorescent probe for hydrogen sulfide imaging in vivo显示文摘Ratiometric fluorescent probes hold great promise for in vivo imaging;however,stimuli-activatable ratiometric probes with fluorescence emissions in near-infrared(NIR)region are still very few.Herein,we report a hydrogen sulfide(H_2S)-activatable ratiometric NIR fluorescent probe(1-SPN)by integrating a H_2S-responsive NIR fluorescent probe 1 into a H_2S-inert poly[2,6-(4,4-bis-(2-ethylhexyl)-4 H-cyclopenta[2,1-b;3,4-b′]dithiophene)-alt-4,7(2,1,3-benzothiadiazole)](PCPDTBT)-based NIR semiconducting polymer nanoparticle(SPN).1-SPN shows'always on'PCPDTBT fluorescence at 830 nm and weak probe 1 fluorescence at 725 nm under excitation at 680 nm.The ratio of NIR fluorescence intensities between 725 and 830 nm(I_(725)/I_(830))is small.Upon interaction with H_2S,the fluorescence at 725 nm is rapidly switched on,resulting in a large enhancement of I_(725)/I_(830),which is allowed for sensitive visualization and quantification of H_2S concentrations in living cells.Taking advantage of enhanced tissue penetration depth of NIR fluorescence,1-SPN is also applied for real-time ratiometric fluorescence imaging of hepatic and tumor H_2S in living mice.This study demonstrates that activatable ratiometric NIR fluorescent probes hold great potential for in vivo imaging.Luyan Wu Wenhui Zeng Liandong Feng Yuxuan Hu Yidan Sun Yingxiao Yan Hong-Yuan Chen Deju Ye 2020Science China Chemistry2020,63,5:0
9External control of qubit-photon interaction and multi-qubit reset in a dissipative quantum network显示文摘A quantum network is a promising quantum many-body system because of its tailored geometry and controllable interaction. Here,we propose an external control scheme for the qubit-photon interaction and multiqubit reset in a dissipative quantum network,which comprises superconducting circuit chains with microwave drives and filter-filter couplings. The traditional multiqubit reset of the quantum network requires physically disconnected qubits to prevent their entanglement. However, we use an original effect of dissipation, i.e., consuming the entanglement generated by qubits’ interaction, to achieve an external control of the multiqubit reset in an always-connected superconducting circuit. The reset time is independent of the number of qubits in the quantum network. 10Research on error accumulation control of three-dimensional adjustment with offset constraint显示文摘Background Currently,laser tracker is the primary instrument used to carry out three-dimensional position measurement in accelerator alignment.Theoretically,three-dimensional measuring data processed by three-dimensional adjustment are more rigorous,however,error accumulation is found in practice.Purpose In order to control error accumulation and further improve the measurement accuracy of accelerator alignment,this research introduces the laser alignment system into the activity of measurement and data processing.Methods A measurement scheme combining laser tracker and laser alignment system is proposed.To construct the constraint condition,the offset values from the measuring points to the laser straight-line datum were used.To carry out the three-dimensional adjustment with offset constraint,the laser tracker observations were used.Results A three-dimensional adjustment function model of laser tracker observations is given.The construction method of the constraint equation is researched,and the calculation formulas of the three-dimensional adjustment with offset constraint are derived.A 200 m linac tunnel control network is designed,using simulation measurement method,the measuring data of laser tracker and the offset values from the measuring points to the laser straight-line datum were generated.The simulated data are calculated by the method given in this paper and the result is analyzed.Conclusion Simulation result shows introducing the laser alignment system into laser tracker measurement and applying the three-dimensional adjustment with offset constraint can effectively suppress the error accumulation caused by long distance move station measurement.Wang Xiaolong Kang Ling Dong Lan Li Bo Men Lingling Luo Tao Wang Tong Liang Jing He Zhenqiang Ke Zhiyong Ma Na Lu Shang Han Yuanying Yan Luping Zhang Luyan Liu Xiaoyang Yan Haoyue Li Chunhua Wu Lei 2022Radiation Detection Technology and Methods2022,6,4:0
