Protective effect of periplaneta americana extract on intestinal mucosal barrier function in patients with sepsis显示文摘OBJECTIVE: To investigate the effect of the periplaneta americana extract on the intestinal mucosal barrier and prognostic implications in patients with sepsis. METHODS: Sixty and six patients with sepsis were assigned randomly to treatment group (32 cases) and control group (32 cases). The extractfrom periplaneta americana plus conventional medication for sepsis was administered to the treatment group, while the control group only received conventional treatment. The gastrointestinal function scores and acute physiology and chronic health evaluation II (APACHEⅡ) scores of all subjects were documented at baseline, at days 1, 3 and 7 after treatment respectively and their blood endotoxin was tested at the same time points as well. The incidence of death was recorded for both groups throughout the trial. RESULTS: At days 3 and 7 after treatment, gastroin-testinal function score, APACHE II, and endotoxin level in treatment group wasbetter than that in control group and the difference between them was significant (both P<0.05). Although the incidence of death in treatment group was less than that in control group, the difference between the two groups was not significant (P> 0.05). CONCLUSION: The extract of periplaneta americana had protective effect on intestinal mucosal barrier and could improve the condition and prognosis in patients with sepsis.Hongwei Zhang Liyou Wei Zhenyu Zhang Shuzheng Liu Gang Zhao Jing Zhang Yanling Hu 2013Journal of Traditional Chinese Medicine2013,33,1:21
TD-AltBOC:A new COMPASS B2 modulation显示文摘AltBOC(15,10) is the baseline of COMPASS B2 signal modulation. It is a BOC-like signal having different PN codes in the lower and the upper main split lobes, which allow one signal service per lobe. The two lobes can be received and processed separately like two BPSK(10) signals,or coherently processed to achieve better performance.Interoperability among COMPASS B2,Galileo E5 and GPS L5 is also achievable using AltBOC modulation. However,Galileo's 4-code AltBOC has drawbacks such as low efficiency and great receiver complexity. This paper presents a new modulation type named TD-AltBOC (Time Division AltBOC).The signal generation scheme and receiving method are presented, and are compared with AltBOC in the areas of power spectrum, ranging accuracy,anti-multipath performance,anti-interference performance, processing flexibility and complexity.Analysis results show TD-AltBOC has similar spectrum characteristics, interoperability, flexibility and anti-interference performance with AltBOC.When the frontend bandwidth is more than 50 MHz,TD-AltBOC can achieve better ranging accuracy and anti-multipath performance. It also has such advantages as high efficiency and low receiver complexity.TD-AltBOC could be a good solution to COMPASS B2 navigation signal.TANG ZuPing ZHOU HongWei WEI JiaoLong YAN Tao LIU YuQi RAN YiHang ZHOU YanLing 2011Science China(Physics,Mechanics & Astronomy)2011,54,6:20
Analysis and Numerical Simulation of Rolling Contact between Sphere and Cone显示文摘In non-conforming rolling contact, the contact stress is highly concentrated in the contact area. However, there are some limitations of the special contact model and stress model used for the theoretical study of the phenomenon, and this has prevented in-depth analysis of the associated friction, wear, and failure. This paper is particularly aimed at investigating the area of rolling contact between a sphere and a cone, for which purpose the boundary is determined by the Hertz theory and the geometries of the non-conforming surfaces. The phenomenon of stick-slip contact is observed to occur in the contact area under the condition of no-full-slip(Q < μ·P). Using the two-dimensional rolling contact theory developed by CARTER, the relative positions of the stick and slip regions and the distribution of the tangential force over the contact area are analyzed. Furthermore, each stress component is calculated based on the Mc Ewen theory and the idea of narrow band. The stress equations for the three-dimensional rolling contact between the sphere and the cone are obtained by the principle of superposition, and are used to perform some numerical simulations. The results show that the stress components have a large gradient along the boundary between the stick and slip regions, and that the maximum stress is inversely proportional to the contact coefficient and proportional to the friction coefficient. A new method for investigating the stress during non-classical three-dimensional rolling contact is proposed as a theoretical foundation for the analysis of the associated friction, wear, and failure.ZHAO Yanling XIA Chengtao WANG Hongbo XUAN Jiaping XIANG Jingzhong LIU Xianli SU Xiangguo 2015Chinese Journal of Mechanical Engineering2015,28,3:18
Hybrid Laser Ablation and Chemical Modification for Fast Fabrication of Bio-inspired Super-hydrophobic Surface with Excellent Self-cleaning,Stability and Corrosion Resistance显示文摘Although laser ablation is considered as a facile technique to fabricate bio-inspired super-hydrophobic surfaces, the issue is that the initial laser treated metallic surfaces show super-hydrophilic property. It will take a long period to reach super-hydrophobic state under ambient air.It is reported that these super-hydrophobic surfaces could be easily damaged by thermal heating effect or interaction with other liquids, causing uncontrolled loss of super-hydrophobicity.In this study,a stable super-hydrophobic aluminum surface was rapidly fab-ricated via the hybrid laser ablation and surface chemical modification of(heptadecafluoro-1,1,2,2-tetradecyl)triethoxysilane(AC-FAS).Surface morphology and chemistry were systematically investigated to explore the generation mechanism of super-hydrophobicity.The water contact angle of the treated surfaces can reach up to 160.6°±1.5°with rolling angle of 3.0°±1.0°,exhibiting perfect self-cleaning capability,long-term stability, and excellent chemical stability in acidic as well as alkaline solutions. The potentiodynamic polarization tests implied that the super-hydrophobic surfaces showed better anti-corrosion performance. This hybrid laser ablation and surface chemical modification are very time-saving and low-cost,which offers a rapid way for quantity production of super-hydrophobic surface on aluminum material.Zhen Yang Xianping Liu Yanling Tian 2019Journal of Bionic Engineering2019,16,1:15
Effect of stimulating acupoint Guanyuan(CV 4) on lower back pain by burning moxa heat for different time lengths: a randomized controlled clinical trial显示文摘OBJECTIVE: To investigate the effect of different heat-stimulating time lengths on lower back pain.METHODS: Forty participants were randomly assigned to four groups of various heating time lengths. The short heating time length group(SL),moderate heating time length group(ML), and long heating time length group(LL) respectively received 15, 30, and 60 min of moxibustion therapy stimulating the acupoint Guanyuan(CV 4). The conventional acupuncture group(CA) received needle acupuncture treatment as a control group. The participants were treated continuously over a 2-week treatment period for a total of 10 sessions, with five sessions given per week. Participants were assessed weekly by blinded assessors, using the visual analogue scale(VAS) and Roland Morris Questionnaire(RMQ).RESULTS: The VAS and RMQ scores reduced in all four groups during treatment. There were significant differences in VAS scores(P < 0.01) and RMQ scores(P < 0.01) between before treatment and after 2 weeks of treatment in the LL group. After treatment, the LL group reported significantly lower VAS scores compared with the CA group, ML group, and SL group(P < 0.05).CONCLUSION: The long and moderate lengths of heat-stimulating time of 30 and 60 min may be more effective for relieving lower back pain than that of short stimulating time lengths.Xu Jianfeng Lin Ruizhu Wu Yongli Wang Yingxu Liu Jian Zhang Yanling Zhang Yuequan Xi Chaolei Wu Qiang Li Xinjian 2015Journal of Traditional Chinese Medicine2015,35,1:13
Roux-en-Y gastric bypass for Chinese type 2 diabetes mellitus patients with a BMI,28kg/m^2:a multi-institutional study显示文摘Roux-en-Y gastric bypass surgery(RYGB) has been demonstrated to be successful for treating type-II diabetes2mellitus(T2DM) patients with a body mass index(BMI),30 kg/m,but reports of RYGB for T2 DM patients with22 a BMI,28 kg/mare lacking.T2 DM patients with a BMI,28 kg/mwere prospectively recruited to participate in this study in four hospitals. The endpoint was T2 DM remission(defined by fasting blood glucose(FBG) level,110 mg/d L and hemoglobin(Hb)A1c level,6.0% at 12 months postoperatively).Predictors of remission were investigated by univariate and multivariate analyses.Eighty-six patients were assessed.Eighty-five patients underwent RYGB,with one conversion to open surgery. We compared the values of various variables before and after2 surgery. The mean BMI decreased from 24.68±2.12 to 21.72±2.43 kg/m(P,0.001).Fifty-eight(67.4%) patients were not treated by drugs or insulin after surgery, and 20 patients(23.3%) had complete remission of T2 DM at12 months after surgery with an acceptable number of complications. The mean Hb A1 c level in the remission group was significantly lower than that in the non-remission group.Patients with a higher weight, lower Hb A1 c level, higher C-peptide level, and higher FBG level were more likely to have T2 DM remission in multivariate2 analyses. In conclusion,RYGB was effective and safe for treating T2 DM patients with a BMI,28 kg/m.Complete remission can be predicted by cases having a higher weight, lower Hb A1 c level, higher C-peptide level, and higher FBG level.Hui Liang Wei Guan Yanling Yang Zhongqi Mao Yijun Mei Huan Liu Yi Miao 2015The Journal of Biomedical Research2015,29,2:13
mTOR/miR-145-regulated exosomal GOLM1 promotes hepatocellular carcinoma through augmented GSK-3b/MMPs显示文摘Golgi membrane protein 1(GOLM1/GP73)is a serum marker of hepatocellular carcinoma(HCC).We have previously shown that mTOR promoted tumorigenesis of HCC through stimulating GOLM1 expression. In this study,we demonstrated that the mammalian target of rapamycin(mTOR)was a negative regulator of microRNA-145(miR-145)expression.miR-145 inhibited GOLM1 expression by targeting a coding sequence of GOLM1 gene.GOLM1 and miR-145 were inversely correlated in human HCC tissues.GOLM1-enriched exosomes activated the glycogen synthase kinase-3β/matrix metalloproteinases(GSK-3β/MMPs)signaling axis of recipient cells and accelerated cell proliferation and migration. In contrast,miR-145 suppressed tumorigenesis and metastasis. We suggest that mTOR/miR-145/GOLM1 signaling pathway should be targeted for HCC treatment.Xiaochen Gai Bufu Tang Fangming Liu Yuting Wu Fang Wang Yanling Jing Fuqiang Huang Di Jin Ling Wang Hongbing Zhang 2019Journal of Genetics and Genomics2019,46,5:13
Coordinated Transcriptional Regulation by the UVB Photoreceptor and Multiple Transcription Factors for Plant UV-B Responses显示文摘Non-damaging ultraviolet B(UV-B)light promotes photomorphogenic development and stress acclimation through UV-B-specific signal transduction in Arabidopsis.UV-B irradiation induces monomerization and nuclear translocation of the UV-B photoreceptor UV RESISTANCE LOCUS 8(UVR8).However,it is not clear how the nuclear localization of UVR8 leads to changes in global gene expression. Here,we reveal that nuclear UVR8 governs UV-B-responsive transcriptional networks in concert with several previously known transcription factors, including ELONGATED HYPOCOTYL 5(HY5)and PHYTOCHROME INTERACTING FACTOR 4(PIF4).Based on the transcriptomic analysis,we identify MYB13 as a novel positive regulator in UV-B-induced cotyledon expansion and stress acclimation.MYB13 is UV-B inducible and is predominantly expressed in the cotyledons. Our results demonstrate that MYB13 protein functions as a transcription factor to regulate the expression of genes involved in auxin response and flavonoid biosynthesis through direct binding with their promoters. In addition, photoactivated UVR8 interacts with MYB13 in a UV-B-dependent manner and differentially modulates the affinity of MYB13 with its targets.Taken together, our results elucidate the cooperative function of the UV-B photoreceptor UVR8 with various transcription factors in the nucleus to orchestrate the expression of specific sets of downstream genes and, ultimately, mediate plant responses to UV-B light.Chongzhen Qian Zhiren Chen Qing Liu Weiwei Mao Yanling Chen Wei Tian Yan Liu Jiupan Han Xinhao Ouyang Xi Huang 2020Molecular Plant2020,13,5:13
Generation of a Hutchinson-Gilford progeria syndrome monkey model by base editing显示文摘Many human genetic diseases, including Hutchinson-Gilford progeria syndrome(HGPS),are caused by single point mutations.HGPS is a rare disorder that causes premature aging and is usually caused by a de novo point mutation in the LMNA gene.Base editors(BEs)composed of a cytidine deaminase fused to CRISPR/Cas9 nickase are highly efficient at inducing C to T base conversions in a programmable manner and can be used to generate animal disease models with single amino-acid substitutions. Here,we generated the first HGPS monkey model by delivering a BE mRNA and guide RNA(gRNA)targeting the LMNA gene via microinjection into monkey zygotes.Five out of six newborn monkeys carried the mutation specifically at the target site.HGPS monkeys expressed the toxic form of lamin A,progerin, and recapitulated the typical HGPS phenotypes including growth retardation, bone alterations, and vascular abnormalities. Thus, this monkey model genetically and clinically mimics HGPS in humans, demonstrating that the BE system can efficiently and accurately generate patient-specific disease models in non-human primates.Fang Wang Weiqi Zhang Qiaoyan Yang Yu Kang Yanling Fan Jingkuan Wei Zunpeng Liu Shaoxing Dai Hao Li Zifan Li Lizhu Xu Chu Chu Jing Qu Chenyang Si Weizhi Ji Guang-Hui Liu Chengzu Long Yuyu Niu 2020Protein & Cell2020,11,11:11
Single-cell transcriptomic atlas of primate cardiopulmonary aging显示文摘Aging is a major risk factor for many diseases, especially in highly prevalent cardiopulmonary comorbidities and infectious diseases including Coronavirus Disease 2019(COVID-19).Resolving cellular and molecular mechanisms associated with aging in higher mammals is therefore urgently needed. Here,we created young and old non-human primate single-nucleus/cell transcriptomic atlases of lung, heart and artery, the top tissues targeted by SARS-CoV-2.Analysis of cell type-specific aging-associated transcriptional changes revealed increased systemic inflammation and compromised virus defense as a hallmark of cardiopulmonary aging. With age, expression of the SARS-CoV-2 receptor angiotensin-converting enzyme 2(ACE2)was increased in the pulmonary alveolar epithelial barrier, cardiomyocytes, and vascular endothelial cells. We found that interleukin 7(IL7)accumulated in aged cardiopulmonary tissues and induced ACE2 expression in human vascular endothelial cells in an NF-κB-dependent manner.Furthermore, treatment with vitamin C blocked IL7-induced ACE2 expression.Altogether, our findings depict the first transcriptomic atlas of the aged primate cardiopulmonary system and provide vital insights into age-linked susceptibility to SARS-CoV-2,suggesting that geroprotective strategies may reduce COVID-19 severity in the elderly.Shuai Ma Shuhui Sun Jiaming Li Yanling Fan Jing Qu Liang Sun Si Wang Yiyuan Zhang Shanshan Yang Zunpeng Liu Zeming Wu Sheng Zhang Qiaoran Wang Aihua Zheng Shuguang Duo Yang Yu Juan Carlos Izpisua Belmonte Piu Chan Qi Zhou Moshi Song Weiqi Zhang Guang-Hui Liu 2021Cell Research2021,31,4:10
Comprehensive analysis of NAC transcription factors and their expression during fruit spine development in cucumber (Cucumis sativus L.)显示文摘The cucumber(Cucumis sativus L.)is an important vegetable crop worldwide, and fruit trichomes or spines are an important trait for external fruit quality. The mechanisms underlying spine formation are not well understood, but the plant-specific NAC family of transcription factors may play important roles in fruit spine initiation and development. In this study,we conducted a genome-wide survey and identified 91 NAC gene homologs in the cucumber genome.Clustering analysis classified these genes into six subfamilies;each contained a varying number of NAC family members with a similar intron–exon structure and conserved motifs.Quantitative real-time PCR analysis revealed tissue-specific expression patterns of these genes, including 10 and 12 that exhibited preferential expression in the stem and fruit, respectively.Thirteen of the 91 NAC genes showed higher expression in the wild-type plant than in its near-isogenic trichome mutant, suggesting their important roles in fruit spine development.Exogenous application of four plant hormones promoted spine formation and increased spine density on the cucumber fruits;several NAC genes showed differential expression over time in response to phytohormone treatments on cucumber fruit, implying their essential roles in fruit-trichome development.Among the NAC genes identified,12 were found to be targets of 13 known cucumber micro-RNAs.Collectively,these findings provide a useful resource for further analysis of the interactions between NAC genes and genes underlying trichome organogenesis and development during fruit spine development in cucumber.Xingwang Liu Ting wang Ezra Bartholomew Kezia Black Mingming Dong Yaqi Zhang Sen Yang Yanling Cai Shudan Xue Yiqun Weng Huazhong Ren 2018Horticulture Research2018,5,1:10
Effect of ferric and bromide ions on the formation and speciation of disinfection byproducts during chlorination显示文摘The effects of ferric ion, pH, and bromide on the formation and distribution of disinfection byproducts (DBPs) during chlorination were studied. Two raw water samples from Huangpu River and Yangtze River, two typical drinking water sources of Shanghai, were used for the investigation. Compared with the samples from Huangpu River, the raw water samples from Yangtze River had lower content of total organic carbon (TOC) and ferric ions, but higher bromide concentrations. Under controlled chlorination conditions, four trihalomethanes (THMs), nine haloacetic acids (HAAs), total organic halogen (TOX) and its halogen species fractions, including total organic chlorine (TOCl) and total organic bromide (TOBr), were determined. The results showed that co-existent ferric and bromide ions significantly promoted the formation of total THMs and HAAs for both raw water samples. Higher concentration of bromide ions significantly changed the speciation of the formed THMs and HAAs. There was an obvious shift to brominated species, which might result in a more adverse influence on the safety of drinking water. The results also indicated that high levels of bromide ions in raw water samples produced higher percentages of unknown TOBr.Shaogang Liu Zhiliang Zhu Yanling Qiu Jianfu Zhao 2011Journal of Environmental Sciences2011,23,5:9

Investigation of benzylisoquinoline alkaloid biosynthetic pathway and its transcriptional regulation in lotus显示文摘Lotus predominantly accumulates benzylisoquinoline alkaloids(BIAs),but their biosynthesis and regulation remain unclear. Here,we investigated structural and regulatory genes involved in BIA accumulation in lotus. Two clustered CYP80 genes were identified to be responsible for the biosynthesis of bis-BIAs and aporphine-type BIAs,respectively, and their tissue-specific expression causes divergence in alkaloid component between leaf and embryo. In contrast with the common(S)-reticuline precursor for most BIAs,aporphine alkaloids in lotus leaf may result from the(S)-N-methylcoclaurine precursor.Structural diversity of BIA alkaloids in the leaf is attributed to enzymatic modifications, including intramolecular C–C phenol coupling on ring A and methylation and demethylation at certain positions.Additionally, most BIA biosynthetic pathway genes show higher levels of expression in the leaf of high-BIA cultivar compared with low-BIA cultivar, suggesting transcriptional regulation of BIA accumulation in lotus.Five transcription factors, including three MYBs,one ethylene-responsive factor, and one basic helix–loop–helix(bHLH),were identified to be candidate regulators of BIA biosynthesis in lotus. Our study reveals a BIA biosynthetic pathway and its transcriptional regulation in lotus, which will enable a deeper understanding of BIA biosynthesis in plants.Xianbao Deng Li Zhao Ting Fang Yaqian Xiong Collins Ogutu Dong Yang Sornkanok Vimolmangkang Yanling Liu Yuepeng Han 2018Horticulture Research2018,5,1:8
RBD-Fc-based COVID-19 vaccine candidate induces highly potent SARS-CoV-2 neutralizing antibody response显示文摘The pandemic of coronavirus disease 2019(COVID-19)caused by severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)has posed serious threats to global health and economy, thus calling for the development of safe and effective vaccines. The receptorbinding domain(RBD)in the spike protein of SARS-CoV-2 is responsible for its binding to angiotensin-converting enzyme 2(ACE2)receptor.It contains multiple dominant neutralizing epitopes and serves as an important antigen for the development of COVID-19 vaccines. Here,we showed that immunization of mice with a candidate subunit vaccine consisting of SARS-CoV-2 RBD and Fc fragment of human IgG,as an immunopotentiator, elicited high titer of RBD-specific antibodies with robust neutralizing activity against both pseudotyped and live SARS-CoV-2 infections. The mouse antisera could also effectively neutralize infection by pseudotyped SARS-CoV-2 with several natural mutations in RBD and the IgG extracted from the mouse antisera could also show neutralization against pseudotyped SARS-CoV and SARS-related coronavirus(SARSr-CoV).Vaccination of human ACE2 transgenic mice with RBD-Fc could effectively protect mice from the SARS-CoV-2 challenge. These results suggest that SARS-CoV-2 RBD-Fc has good potential to be further developed as an effective and broad-spectrum vaccine to prevent infection of the current SARS-CoV-2 and its mutants,as well as future emerging SARSr-CoVs and re-emerging SARS-CoV.Zezhong Liu Wei Xu Shuai Xia Chenjian Gu Xinling Wang Qian Wang Jie Zhou Yanling Wu Xia Cai Di Qu Tianlei Ying Youhua Xie Lu Lu Zhenghong Yuan Shibo Jiang 2020Signal Transduction and Targeted Therapy2020,5,1:7
One century sedimentary record of lead and zinc pollution in Yangzong Lake, a highland lake in southwestern China显示文摘Reconstruction of trace metal pollution histories and sources may help us to regulate current pollutant discharge. This is especially important for the highland lakes in southwestern China, which are facing trace metals pollution. We present sedimentary records of 11 metals accumulated in Yangzong Lake since the 1870's, a highland lake in southwestern China. Pollution of lead and zinc (Pb and Zn) was differentiated based on principal component analysis, geochemical normalization, and lead isotope ratios. Nearly all the metals as well as grain size composition show generally constant values before the mid-1980's, denoting stable detrital input in the catchment. Fluctuations in the concentrations of the metals as well as grain size composition since the mid-1980's indicate an increase in soil erosion with strengthened human disturbance in the catchment. After geochemical normalization, Pb and Zn showed constant values before 1990 AD and then a gradual increase in parallel with the variations in 208Pb/206Pb and 207Pb/206Pb ratios, indicating that Pb and Zn pollution occurred. Combining the data of 208Pb/206Pb and 207Pb/206Pb ratios in the sediments of Yangzong Lake, leaded gasoline, Pb-Zn ore and coal, and consumption or production historical trends, we deduced that the enhanced Pb and Zn pollution in Yangzong Lake is caused primarily by ore mining and refining.Enlou Zhang Enfeng Liu Ji Shen Yanmin Cao Yanling Li 2012Journal of Environmental Sciences2012,24,7:7
Jiaweisinisan facilitates neurogenesis in the hippocampus after stress damage显示文摘The traditional Chinese medicine Jiaweisinisan has antidepressant effects, and can inhibit hypothalamus-pituitary-adrenal gland axis hyperactivity in stress-induced depression. In this study, rat hippocampal neural precursor cells were cultured in serum-free medium in vitro and a stress damage model was established with 120 μM corticosterone. Cells were treated with 10% (v/v) Jiaweisinisan drug-containing serum and the corticosterone antagonist RU38486. Results of the 3-(4,5-dimethylthiazol-2-yl)-3,5-di-phenytetrazoliumromide assay showed that both Jiaweisinisan drug-containing serum and RU38486 promoted the proliferation of neural precursor cells after corticosterone exposure. Immunofluorescence detection showed that after Jiaweisinisan drug-containing serum and RU38486 treatment, the 5-bromo-2-deoxyuridine/terminal deoxynucleotidyl transferase dUTP nick end labeling ratio in hippocampal neural precursor cells significantly increased, and the apoptotic rates of glial cells reduced, and neuron-like cell differentiation from neural precursor cells significantly increased. Our experimental findings indicate that Jiaweisinisan promotes hippocampal neurogenesis after stress damage.Lili Wu Chuanlian Ran Shukao Liu Lizhen Liao Yanling Chen Hualei Guo Weikang Wu Can Yan 2013Neural Regeneration Research2013,8,12:7
