| 1 | Structure and deformation around the Gyirong basin, north Himalaya, and onset of the south Tibetan detachment system显示文摘Gyirong basin and its adjacent area are located at a special position in the Himalayan orogen, where the south Tibetan detachment system (STDS) and N-S trending rift converged. The north Himalayan orogen here can be divided into five petrologic-tectonic units successively from south to north: 1) the Greater Himalayan crystalline complex (GHC); 2) the STDS shear zone; 3) the Tethyan Himalayan sedimentary sequence (THS); 4) the late Cenozoic sedimentary basins, such as Gyirong and Oma basins; and 5) the Malashan gneiss dome. Structural studies show that this area experienced four stages of deformation: 1) the earlier south-directed thrusting, preserved both in the GHC and THS; 2) top-down-to-north slip along the STDS, normal faults related to this slip formed the early controlling structures of the Cenozoic basins,and the tilted pattern of the blocks between the basins indicated a north-directed slip; 3) east-west extension, the resultant N-S trending normal fault formed the eastern boundary of the basins; and 4) late gravitational collapse. | 2 | In vitro Anthocyanin Induction and Metabolite Analysis in Malus spectabilis Leaves Under Low Nitrogen Conditions显示文摘Anthocyanins are the most widely produced secondary metabolites in plants,and they play an important role in plant growth and reproduction.The nitrogen source is an important factor affecting anthocyanin production,but the nitrogen concentrations on metabolism and the underlying genetic basis remain unclear.In this study,in vitro anthocyanin induction was conducted on Malus spectabilis.The leaf explants were cultivated in media containing different nitrogen concentrations.The results suggested that when the nitrogen contents decreased in limit,the color of leaf explants turned from green to red,and increased anthocyanin accumulation led to a change in phenotype.Furthermore,the content of other flavonoids,such as dihydroquercetin,epicatechin,and catechin,increased under low nitrogen conditions.The transcription levels of the general flavonoid pathway genes,from phenylalanine ammonia lyase(PAL)to anthocyanidin synthase(ANS),were associated with the concentration of corresponding flavonoid compounds and phenotype changes.In particular,the expression level of ANS increased substantially under a low nitrogen treatment,which was significantly and positively correlated with the anthocyanin levels(R2=0.72,P<0.05).The increased expression patterns of anthocyanin pathway genes were similar to that of the transcription factor MYB10.We further verified MYB10 played an important role in the anthocyanin pathway in leaves of Malus spectabilis.These results suggested that we can improve the desirable ornamental plant phenotypes by controlling nitrogen content.This process may offer clues to further development of new agricultural practices. | Jiaxin Meng Yan Gao Meiling Han Pengyuan Liu Chen Yang Ting Shen Houhua Li | 2020 | Horticultural Plant Journal2020,6,5: | 13 |
| 3 | The hepatitis B virus-associated tumor microenvironment in hepatocellular carcinoma显示文摘In contrast to a majority of cancer types, the initiation of hepatocellular carcinoma(HCC) is intimately associated with a chronically diseased liver tissue, with one of the most prevalent etiological factors being hepatitis B virus(HBV). Transformation of the liver in HBV-associated HCC oten follows from or accompanies long-term symptoms of chronic hepatitis, inlammation and cirrhosis, and viral load is a strong predictor for both incidence and progression of HCC. | 5 | Comparison of alternative extraction methods for secretome profiling in human hepatocellular carcinoma cells显示文摘Secreted proteins are important sources for early detection and diagnosis of disease, and as such have received considerable attention. The extraction of low concentration proteins from large volumes of culture media, which are rich in salts and other compounds that interfere with most proteomics techniques, presents a problem for secretome studies. Ultrafiltration, precipitation, and dialysis are three major extraction methods that can be used to overcome this problem. The present study for the first time, compared the merits and shortcomings of these three methods, without bias. Centrifugal ultrafiltration provided the best extraction efficiency, and precipitation provided the highest number of identifiable proteins. The three methods yielded closely related, but different, information on the secretome; thus, they should be considered complementary or, at least, supplementary methods. Three hundred and sixty unique proteins were identified, including 211 potential secreted proteins. Compared with previous studies, this study also identified 42 new secreted proteins. | 14 | TiO_2-functionalized mesoporous materials for sensitive analysis of multi-phosphopeptides显示文摘Protein phosphorylation as one of the most important post-translational modifications in mammalian cells regulates numerous biological processes. Here we propose a novel strategy for the selective isolation and sensitive analysis of multi-phosphopeptides based on TiO2-gratfed mesoporous materials, in which MCM-41 and SBA-15 were chosen as the hard templates. The commercialized IMAC and TiO2 nanopartices were further investigated in the phosphopeptide analysis for comparison. The enrichment efficiency was evaluated and measured by MALDI-TOF mass spectrometry. The results indicated that both TiO2-SBA-15 and TiO2-MCM-41 exhibited the preferential affinity to multi-phosphopeptides compared with the other two widely used strategies.
