1Prognostic nutritional index predicts postoperative complications and long-term outcomes of gastric cancer显示文摘AIM:To investigate the impact of prognostic nutritional index(PNI)on the postoperative complications and long-term outcomes in gastric cancer patients undergoing total gastrectomy.METHODS:The data for 386 patients with gastric cancer were extracted and analyzed between January2003 and December 2008 in our center.The patients were divided into two groups according to the cutoff value of the PNI:those with a PNI≥46 and those with a PNI<46.Clinicopathological features were compared between the two groups and potential prognostic factors were analyzed.The relationship between postoperative complications and PNI was analyzed by logistic regression.The univariate and multivariate hazard ratios were calculated using the Cox proportional hazard model.RESULTS:The optimal cutoff value of the PNI was set at 46,and patients with a PNI≥46 and those with a PNI<46 were classified into PNI-high and PNI-low groups,respectively.Patients in the PNI-low group were more likely to have advanced tumor(T),node(N),and TNM stages than patients in the PNI-high group.The low PNI is an independent risk factor for the incidence of postoperative complications(OR=2.223).The 5-year overall survival(OS)rates were 54.1%and21.1%for patients with a PNI≥46 and those with a PNI<46,respectively.The OS rates were significantly lower in the PNI-low group than in the PNI-high group among patients with stagesⅡ(P=0.001)andⅢ(P<0.001)disease.CONCLUSION:The PNI is a simple and useful marker not only to identify patients at increased risk for postoperative complications,but also to predict long-term survival after total gastrectomy.The PNI should be included in the routine assessment of advanced gastric cancer patients.Nan Jiang Jing-Yu Deng Xue-Wei Ding Bin Ke Ning Liu Ru-Peng Zhang Han Liang 2014World Journal of Gastroenterology2014,20,30:41
2Positive impact of adding No.14v lymph node to D2 dissection on survival for distal gastric cancer patients after surgery with curative intent显示文摘Background:D2 lymphadenectomy has been increasingly regarded as standard surgical procedure for advanced gastric cancer(GC),while the necessity of No.14 v lymph node(14v) dissection for distal GC is still controversial.Methods:A total of 920 distal GC patients receiving at least a D2 lymph node dissection in Department of Gastric Cancer,Tianjin Medical University Cancer Institute and Hospital were enrolled in this study,of whom,243 patients also had the 14 v dissected.Other 677 patients without 14 v dissection were used for comparison.Results:Forty-five(18.5%) patients had 14 v metastasis.There was no significant difference in 3-year overall survival(OS) rate between patients with and without 14 v dissection.Following stratified analysis,in TNM stages I,II,IIIa and IV,14 v dissection did not affect 3-year OS;in contrast,patients with 14 v dissection had a significant higher 3-year OS than those without in TNM stages IIIb and IIIc.In multivariate analysis,14 v dissection was found to be an independent prognostic factor for GC patients with TNM stage IIIb/IIIc disease [hazard ratio(HR),1.568;95% confidence interval(CI):1.186-2.072;P=0.002].GC patients with 14 v dissection had a significant lower locoregional,especially lymph node,recurrence rate than those without 14 v dissection(11.7 % vs.21.1%,P=0.035).Conclusions:Adding 14 v to D2 lymphadenectomy may be associated with improved 3-year OS for distal GC staged TNM Ⅲb/Ⅲc.Yuexiang Liang Liangliang Wu Xiaona Wang Xuewei Ding Hongmin Liu Bin Li Baogui Wang Yuan Pan Rupeng Zhang Ning Liu Han Liang 2015Chinese Journal of Cancer Research2015,27,6:22

5BMAL1 regulates mitochondrial fission and mitophagy through mitochondrial protein BNIP3 and is critical in the development of dilated cardiomyopathy显示文摘Dysregulation of circadian rhythms associates with cardiovascular disorders.It is known that deletion of the core circadian gene Bma/1 in mice causes dilated car-diomyopathy.However,the biological rhythm regulation system in mouse is very different from that of humans.Whether BMAL1 plays a role in regulating human heart function remains unclear.Here we generated a BMAL1 knockout human embryonic stem cell(hESC)model and further derived human BMAL1 deficient cardiomy-ocytes.We show that BMAL1 deficient hESC-derived cardiomyocytes exhibited typical phenotypes of dilated cardiomyopathy including attenuated contractility,cal-cium dysregulation,and disorganized myofilaments.In addition,mitochondrial fission and mitophagy were suppressed in BMAL1 deficient hESC-cardiomyocytes,which resulted in significantly attenuated mitochondrial oxidative phosphorylation and compromised cardiomy-ocyte function.We also found that BMAL1 binds to the E-box element in the promoter region of BNIP3 gene and specifically controls BNIP3 protein expression.BMAL1 knockout directly reduced BNIP3 protein level,causing compromised mitophagy and mitochondria dysfunction and thereby leading to compromised cardiomyocyte function.Our data indicated that the core circadian gene S/VMLf is critical for normal mitochondria activities and cardiac function.Circadian rhythm disruption may directly link to compromised heart function and dilated cardiomyopathy in humans.Ermin Li Xiuya Li Jie Huang Chen Xu Qianqian Liang Kehan Ren Aobing Bai Chao Lu Ruizhe Qian Ning Sun 2020Protein & Cell2020,11,9:17

8High levels of serum interleukin-6 increase mortality of hepatitis B virus-associated acute-on-chronic liver failure显示文摘BACKGROUND Patients with hepatitis B virus-associated acute-on-chronic liver failure(HBVACLF)present a complex and poor prognosis.Systemic inflammation plays an important role in its pathogenesis,and interleukin-6(IL-6)as a pro-inflammatory cytokine is related with severe liver impairment and also plays a role in promoting liver regeneration.Whether serum IL-6 influences HBV-ACLF prognosis has not been studied.AIM To determine the impact of serum IL-6 on outcome of patients with HBV-ACLF.METHODS We performed a retrospective study of 412 HBV-ACLF patients.The findings were analyzed with regard to mortality and the serum IL-6 level at baseline,as well as dynamic changes of serum IL-6 within 4 wk.RESULTS The serum IL-6 level was associated with mortality.Within 4 wk,deceased patients had significantly higher levels of IL-6 at baseline than surviving patients[17.9(7.3-57.6)vs 10.4(4.7-22.3),P=0.011].Patients with high IL-6 levels(>11.8 pg/mL)had a higher mortality within 4 wk than those with low IL-6 levels(≤11.8 pg/mL)(24.2%vs 13.2%,P=0.004).The odds ratios calculated using univariate and multivariate logistic regression were 2.10(95%confidence interval[CI]:1.26-3.51,P=0.005)and 2.11(95%CI:1.15-3.90,P=0.017),respectively.The mortality between weeks 5 and 8 in patients with high IL-6 levels at 4 wk was 15.0%,which was significantly higher than the 6.6%mortality rate in patients with low IL-6 levels at 4 wk(hazard ratio=2.39,95%CI:1.05-5.41,P=0.037).The mortality was 5.0%in patients with high IL-6 levels at baseline and low IL-6 levels at 4 wk,7.5%in patients with low IL-6 levels both at baseline and at 4 wk,11.5%in patients with low IL-6 levels at baseline and high IL-6 levels at 4 wk,and 16.7%in patients with high IL-6 levels both at baseline and at 4 wk.The increasing trend of the mortality rate with the dynamic changes of IL-6 was significant(P for trend=0.023).CONCLUSION A high level of serum IL-6 is an independent risk factor for mortality in patients with HBV-ACLF.Furthermore,a sustained high level or dynamic elevated level of serum IL-6 indicates a higher mortality.Chao Zhou Ning Zhang Ting-Ting He Yao Wang Li-Fu Wang Yong-Qiang Sun Jing Jing Jing-Jing Zhang Shuang-Nan Fu Xuan Wang Xiao-Xiao Liang Xin Li Man Gong Jun Li 2020World Journal of Gastroenterology2020,26,30:15
9The Intervention Effect of Rosiglitozone in Ovarian Fibrosis of PCOS Rats显示文摘Objective To explore the Intervention effect of Rosiglitozone in ovarian fibrosis of PCOS rats.Methods 60 female SD rats were randomly divided into 3 groups:control group,model group and treatment group.The model and treatment groups were established by subcutaneous injection of DHEA,while the treatment group was given RGZ.The serum hormone values,pathohistology of ovarian structure of rats,ovarian ultrastructure and the expressions of TGF-β 1 and CTGF were detected.Results The PCOS model was established successfully.The expression intensity of TGF-β 1 and CTGF in Oocytes of the PCOS groups was 9.545±2.954 and 9.665±2.400,respectively and was significantly higher than that of the control group 6.636±2.264 and 7.036±2.133;after treatment with rosiglitazone,the expression was significantly decreased 6.980±2.421 and 6.642±2.721 as compared with that of the model group (P<0.05,P<0.001).The values in serum of the PCOS groups were 3.749±2.054 and 0.265±0.129,and 1.914±1.801 and 0.096±0.088 in the control group which had statistically significant difference (P<0.05,P<0.001).After treatment with rosiglitazone,the values were 2.3100±1.825 and 0.112±0.187 and were significantly different with those of the model group (P<0.05,P<0.001).Conclusion TGF-β 1 and CTGF play an important role in the development of ovary fibrosis in PCOS.However,RGZ may postpone the development of fibrosis by decreasing the levels of TGF-β 1 and CTGF.MIAO Zhu Lin GUO Liang WANG Yong Xia CUI Rong YANG Ning HUANG Mi Qiong QIN Wei Bing CHEN Jin LI Hong Mei WANG Zi Neng WEI Xiang Cai 2012Biomedical and Environmental Sciences2012,25,1:14
10Application of next-generation sequencing technology to precision medicine in cancer: joint consensus of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology显示文摘Next-generation sequencing(NGS) technology is capable of sequencing millions or billions of DNA molecules simultaneously.Therefore, it represents a promising tool for the analysis of molecular targets for the initial diagnosis of disease, monitoring of disease progression, and identifying the mechanism of drug resistance. On behalf of the Tumor Biomarker Committee of the Chinese Society of Clinical Oncology(CSCO) and the China Actionable Genome Consortium(CAGC), the present expert group hereby proposes advisory guidelines on clinical applications of NGS technology for the analysis of cancer driver genes for precision cancer therapy. This group comprises an assembly of laboratory cancer geneticists, clinical oncologists, bioinformaticians,pathologists, and other professionals. After multiple rounds of discussions and revisions, the expert group has reached a preliminary consensus on the need of NGS in clinical diagnosis, its regulation, and compliance standards in clinical sample collection. 11Hypoxia-inducible factor-1α induces the epithelial-mesenchymal transition of human prostatecancer cells显示文摘Background Hypoxia-inducible factor-1α (HIF-1α) is a transcriptional factor that could improve the stimulation of angiogenesis and the metabolic adaptation of tumor cells to hypoxia. A recent study showed that HIF-1α could induce colon cancer cells epithelial-mesenchymal transition (EMT). However, no evidence indicates a similar correlation in human prostate cancer cells. This study was designed to evaluate the effect of HIF-1α over-expression on the EMT in human prostate cancer cells. Methods We selected the appropriate cell line for HIF-1α induction from those EMT negative prostate cell lines through vimentin gene detection by RT-PCR. As the result, LNCaP cell line is the best one for further experiment. LNCaP cells were transfected with recombinant plasmid pcDNA3.1(-)/HIF-1α and pcDNA3.1(-) control vector by Lipofectamine 2000 system. The positive cell colonies were confirmed by indirect immunofluorescence labeling. Then Transwell polycarbonate filter was used to analyze the invasive potency. The expression of EMT associated proteins, E-cadherin and vimentin, was detected by Western blotting. Results Among four of the EMT negative cell lines, LNCaP was the only one expressed the vimentin gene but not the associated protein. The expression level of HIF-1α in LNCaP/HIF-1α was distinctly higher than that in LNCaP/pcDNA3.1 and LNCaP. The cell numbers of LNCaP/HIF-1α that penetrated through the Transwell filter were higher than that of LNCaP/pcDNA3.1 and LNCaP. Compared with the LNCaP/pcDNA3.1 and LNCaP cells, the expression of vimentin was up-regulated in LNCaP/HIF-1α, whereas the expression of E-cadherin was down-regulated. Conclusions Over-expression of HIF-1α stimulates the invasion potency of human prostate carcinoma cells through EMT pathway. 15Trabecular-like Ti-6Al-4V scaffolds for orthopedic: fabrication by selective laser melting and in vitro biocompatibility显示文摘Porous metal scaffolds play an important role in the orthopedic field, due to their wide applications in prostheses implantation. Some previous studies showed that the scaffolds with trabecular bone structure reconstructed via computed tomography had satisfactory biocompatibility. However, the reverse modeling scaffolds were inflexible for customized design. Therefore, a top-down designing biomimetic bone scaffold with favorable mechanical performances and cytocompatibility is urgently demanded for orthopedic implants. An emerging additive manufacturing technique, selective laser melting, was employed to fabricate the trabecular-like porous Ti-6Al-4 V scaffolds with varying irregularities(0.05-0.5) and porosities(48.83%–74.28%) designed through a novel Voronoi-Tessellation based method. Micro-computed tomography and scanning electron microscopy were used to characterize the scaffolds’ morphology.Quasi-static compression tests were performed to evaluate the scaffolds’ mechanical properties. The MG63 cells culture in vitro experiments, including adhesion, proliferation, and differentiation, were conducted to study the cytocompatibility of scaffolds. Compressive tests of scaffolds revealed an apparent elastic modulus range of 1.93–5.24 GPa and an ultimate strength ranging within 44.9–237.5 MPa, which were influenced by irregularity and porosity, and improved by heat treatment. Furthermore, the in vitro assay suggested that the original surface of the SLM-fabricated scaffolds was favorable for osteoblasts adhesion and migration because of micro scale pores and ravines. The trabecular-like porous scaffolds with full irregularity and higher porosity exhibited enhanced cells proliferation and osteoblast differentiation at earlier time, due to their preferable combination of small and large pores with various shapes. 18Identification of a Newly Isolated Getah Virus in the China-Laos Border,China显示文摘In this study, we isolated a virus strain(YN12031) from specimens of Armigeres subalbatus collected in the China-Laos border. BHK-21 cells infected with YN12031 exhibited an evident cytopathic effect(CPE) 32 h post-infection. The virus particles were spherical, 70 nm in diameter, and enveloped; they also featured surface fibers. Molecular genetic analysis revealed that YN12031 was closely related to alpha viruses such as Chikungunya virus and Sindbis virus, and located in the same clade as MM2021, the prototype of Getahvirus(GETV) isolated in Malaysia in 1955. Phylogenetic analysis of the E2 and capsid genes further revealed that YN12031 was located in the same clade as the Russian isolate LEIV/16275/Mag. Analysis of the homology of nucleotides and amino acids in the coding area and E2 gene demonstrated that the YN12031 isolated from the China-Laos border(tropical region) was related closest to the LEIV/16275/Mag isolate obtained in Russia(North frigid zone area) among other isolates studied. These results suggest that GETV can adapt to different geographical environments to propagate and evolve. 19Mitosin/CENP-F is a conserved kinetochore protein subjected to cyto plasmic dynein-mediated poleward transport显示文摘Mitosin/CENP-F is a human nuclear protein transiently associated with the outer kinetochore plate in M phase and is involved in M phase progression. LEK1 and CMF1, which are its murine and chicken orthologs, however, are implicated in muscle differentiation and reportedly not distributed at kinetochores.We therefore conducted several assays to clarify this issue. The typical centromere staining patterns were observed in mitotic cells from both human primary culture and murine, canine, and mink cell lines. A C-terminal portion of LEK1 also conferred centromere localization. Our analysis further suggests conserved kinetochore localization of mammalian mitosin orthologs. Moreover, mitosin was associated preferentially with kinetochores of unaligned chromosomes. It was also constantly transported from kinetochores to spindle poles by cytoplasmic dynein. These properties resemble those of other kinetochore proteins important for the spindle checkpoint, thus implying a role of mitosin in this checkpoint.
