| 1 | Polysaccharides from Angelica sinensis alleviate neuronal cell injury caused by oxidative stress显示文摘Angelica sinensis has antioxidative and neuroprotective effects. In the present study, we aimed to determine the neuroprotective effect of polysaccharides isolated from Angelica sinensis. In a preliminary experiment, Angelica sinensis polysaccharides not only protected PC12 neuronal cells from H 2 O 2-induced cytotoxicity, but also reduced apoptosis and intracellular reactive oxygen species levels, and increased the mitochondrial membrane potential induced by H 2 O 2 treatment. In a rat model of local cerebral ischemia, we further demonstrated that Angelica sinensis polysaccharides enhanced the antioxidant activity in cerebral cortical neurons, increased the number of microvessels, and improved blood flow after ischemia. | 5 | Influence of synthesis temperatures on the crystalline grain growth and morphology of lanthanum magnesium hexaaluminate显示文摘Lanthanum magnesium hexaaluminate(LaMgAl_(11)O_(19), LMA) was prepared at different temperatures by solid-state reaction. Phase compositions and crystal morphologies of specimens synthesized at different temperatures were investigated using X-ray diffraction(XRD), scanning electron microscopy(SEM). It was observed that the crystalline grain size of LMA was not only dependent on the preparation temperature but also on its powder morphology. In the temperature range of 1300 e1550℃, LMA showed platelet grain and the average crystalline grain size increases with the increase in temperature. At1600℃, if the powder was sintered for two times, the equiaxed grain could be found with the decrease in grain space, resulting in the reduction of the crystalline grain size. Styles of specimens(powder or disk) might have no obvious influence on morphologies and sizes of LMA crystalline grains which were synthesized with the well-dispersed raw material mixtures. | 8 | Comprehensive identification of lysine 2-hydroxyisobutyrylated proteins in Ustilaginoidea virens reveals the involvement of lysine 2-hydroxyisobutyrylation in fungal virulence显示文摘Lysine 2-hydroxyisobutyrylation(Khib) is a newly identified post-translational modification(PTM) that plays important roles in transcription and cell proliferation in eukaryotes. However, its function remains unknown in phytopathogenic fungi. Here,we performed a comprehensive assessment of Khibin the rice false smut fungus Ustilaginoidea virens, using Tandem Mass Tag(TMT)-based quantitative proteomics approach. A total of 3 426 Khibsites were identified in 977 proteins, sugg esting that Khibis a common and complex PTM in U. virens. Our data demonstrated that the2-hydroxyisobutyrylated proteins are involved in diverse biological processes. Network analysis of the modified proteins revealed a highly interconnected protein network that included many well-studied virulence factors. We confirmed that the Zn-binding reduced potassium dependency3-type histone deacetylase(UvRpd3) is a major enzyme that removes 2-hydroxyisobutyrylation and acetylation in U. virens. Notably, mutations of Khib sites in the mitogen-activated protein kinase(MAPK)UvSlt2 significantly reduced fungal virulence and decreased the enzymatic activity of UvSlt2. Molecular dynamics simulations demonstrated that 2-hydroxyisobutyrylation in UvSlt2 increased the hydrophobic solvent-accessible surface area and thereby affected binding between the UvSlt2 enzyme and its substrates. | 20 | A lipid droplet-associated protein Nem1 regulates appressorium function for infection of Magnaporthe oryzae显示文摘Lipid droplets are important storages in fungal conidia and can be used by plant pathogenic fungi for infection.However,the regulatory mechanism of lipid droplets formation and the utilization during fungal development and infection are largely unknown.Here,in Magnaporthe oryzae,we identified a lipid droplet-associated protein Nem1 that played a key role in lipid droplets biogenesis and utilization.Nem1 was highly expressed in conidia,but lowly expressed in appressoria,and its encoded protein was localized to lipid droplets.Deletion of NEM1 resulted in reduced numbers of lipid droplets and decreased content of diacylglycerol(DAG)or triacylglycerol(TAG).NEM1 was required for asexual development especially conidia production.TheΔnem1 mutant was nearly loss of virulence to host plants due to defects in appressorial penetration and invasive growth.Remarkably,Nem1 was regulated by the TOR signaling pathway and involved in the autophagy process.The Ser303 residue of Nem1 could be phosphorylated by the cAMP-PKA signaling pathway and was important for biological function of Nem1.Together,our study revealed a regulatory mechanism of lipid biogenesis and metabolism during the conidium and appressorium formation of the rice blast fungus. | Deng Chen Xuan Cai Junjie Xing Shen Chen Juan Zhao Zhiguang Qu Guotian Li Hao Liu Lu Zheng Junbin Huang Xiao-Lin Chen | 2023 | aBIOTECH2023,4,2: | 0 |
