1The herbal compound geniposide rescues formaldehyde-induced apoptosis in N2a neuroblastoma cells显示文摘The herbal medicine Tong Luo Jiu Nao(TLJN)contains geniposide(GP)and ginsenoside Rg1 at a molar ratio of 10:1.Rg1 is the major component of another herbal medicine,panax notoginseng saponin(PNS).TLJN has been shown to strengthen brain function in humans,and in animals it improves learning and memory.We have previously shown that TLJN reduces amyloidogenic processing in Alzheimer’s disease(AD)mouse models.Together this suggests TLJN may be a potential treatment for patients with dementia.Because chronic damage of the central nervous system by formaldehyde(FA)has been presented as a risk factor for age-associated cognitive dysfunction,in the present study we investigated the protective effect of both TLJN and GP in neuron-like cells exposed to FA.FA-exposed murine N2a neuroblastoma cells were incubated with TLJN,its main ingredient GP,as well as PNS,to measure cell viability and morphology,the rate of apoptosis and expression of genes encoding Akt,FOXO3,Bcl2 and p53.The CCK-8 assay,cytoskeletal staining and flow cytometry were used to test cell viability,morphology and apoptosis,respectively.Fluorescent quantitative real-time PCR(qRT-PCR)was used to monitor changes in gene expression,and HPLC to determine the rate of FA clearance.Treatment of N2a cells with 0.09 mmol L?1 FA for 24 h significantly reduced cell viability,changed cell morphology and promoted apoptosis.Both TLJN and GP conferred neuroprotection to FA-treated N2a cells,whereas PNS,which had to be used at lower concentrations because of its toxicity,did not.Our data demonstrate that TLJN can rescue neuronal damage caused by FA and that its main ingredient,GP,has a major role in this efficacy.This presents purified GP as a drug or lead compound for the treatment of AD.CHEN JinYan SUN MengRu WANG XingHua LU Jing WEI Yan TAN Yan LIU Ying GTZ Jürgen HE RongQiao HUA Qian 2014Science China(Life Sciences)2014,57,4:15
2Genome Characteristics and Evolution of Pseudorabies Virus Strains in Eastern China from 2017 to 2019显示文摘Since late 2011,outbreaks of pseudorabies virus(PRV)have occurred in southern China causing major economic losses to the pig industry.We previously reported that variant PRV forms and recombination in China could be the source of continued epidemics.Here,we analyzed samples from intensive pig farms in eastern China between 2017 and 2019,and sequenced the main glycoproteins(gB,gC,gD,and gE)to study the evolution characteristics of PRV.Based on the gC gene,we found that PRV variants belong to clade 2 and detected a founder effect during by the PRV epidemic.In addition,we detected inter-and intra-clade recombination;in particular,inter-clade recombination in the gB genes of strains FJ-ZXF and FJ-W2,which were recombinant with clade 1 strains.We also found specific amino-acid changes and positively selected sites,possibly associated with functional changes.This analysis of the emergence of PRV in China illustrates the need for continuous monitoring and the development of vaccines against specific variants of PRV.Xiaofeng Zhai Wen Zhao Kemang Li Cheng Zhang Congcong Wang Shuo Su Jiyong Zhou Jing Lei Gang Xing Haifeng Sun Zhiyu Shi Jinyan Gu 2019Virologica Sinica2019,34,6:12

4Production of Two Elite Glutinous Rice Varieties by Editing Wx Gene显示文摘The waxy gene(Wx) in rice, which encodes the granule bound starch synthase enzyme, is responsible for amylose synthesis. Glutinous(sticky) rice has little or no amylose that can be used in various applications, such as brewing. In this study, knockout of the Wx gene with CRISPR/Cas9 technology was conducted in two elite japonica rice lines, Huaidao 5(HD5) and Suken 118(SK118), aiming to develop elite sticky rice varieties. We achieved six homozygous T_0 plants with more than 200 bp deletion in the Wx gene, as well as 36 wx-HD5 and 18 wx-SK118 homozygous transgene-free plants in the T_1 generation. The seeds of all the mutants were white and opaque, similar to those of sticky rice, and contained only 2.6%–3.2% amylose. Results of scanning electron microscopy showed that the quality of rice did not change. 5Hepatitis B virus X protein enhances hepatocarcinogenesis by depressing the targeting of NUSAP1 mRNA by miR-18b显示文摘Objective: The aim of this study was to investigate the underlying mechanism whereby HBx modulates the targeting of NUSAP1 by miR-18b to enhance hepatocarcinogenesis.Methods: We employed an integrated approach of bioinformatics analysis and molecular experiments in hepatoma cells, HBV transgenic mice, and clinical liver cancer tissues to investigate the role of HBx-regulated miR-18b in the development of liver cancer.Results: In this study, we report that the HBx-mediated tumor suppressor miR-18b modulates hepatocarcinogenesis during the host-HBV interaction. The expression levels of miR-18b were lower in clinical HBV-positive liver cancer tissues and liver tissues of HBV-transgenic mice. Interestingly, HBx inhibited miR-18b expression by inducing the methylation of CpG islands in its promoter. Accordingly, we tested the hypothesis that HBx enhanced hepatocarcinogenesis by increasing the expression of target genes of miR-18b. Moreover, we identified nucleolar spindle-associated protein 1(NUSAP1) as one of the target genes of miR-18b.NUSAP1 was expressed at high levels in liver cancer tissues. Interestingly, HBx up-regulated NUSAP1 by suppressing miR-18b.Functionally, miR-18b significantly inhibited the proliferation of hepatoma cells by depressing NUSAP1 levels in vivo and in vitro.Conclusions: Thus, we conclude that the targeting of NUSAP1 mRNA by the tumor suppressor miR-18b is controlled by HBxmodulated promoter methylation during the host-virus interaction, leading to hepatocarcinogenesis. 6Two-dimensional MOS2 for hydrogen evolution reaction catalysis: The electronic structure regulation显示文摘Molybdenum disulfide (MoS2) has been recognized as one of the most promising candidates to replace precious Pt for hydrogen evolution reaction (HER) catalysis, due to the natural abundance, low cost, tunable electronic properties, and excellent chemical stability. Although notable processes have been achieved in the past decades, their performance is still far less than that of Pt. Searching effective strategies to boosting their HER performance is still the primary goal. In this review, the recent process of the electronic regulation of MoS2 for HER is summarized, including band structure engineering, electronic state modulation, orbital orientation regulation, interface engineering. 7Novel constant-cladding tapered-core ytterbium-doped fiber for high-power fiber laser oscillator显示文摘Power scaling based on traditional ytterbium-doped fibers(YDFs)is limited by optical nonlinear effects and transverse mode instability(TMI)in high-power fiber lasers.Here,we propose a novel long tapered fiber with a constant cladding and tapered core(CCTC)along its axis direction.The tapered-core region of the fiber is designed to enhance the stimulated Raman scattering(SRS)threshold and suppress higher-order mode resonance in the laser cavity.The CCTC YDF was fabricated successfully with a modified chemical vapor deposition(MCVD)method combined with solution doping technology,which has a cladding diameter of 400µm and a varying core with a diameter of~24μm at both ends and~31μm in the middle.To test the performance of the CCTC fiber during high-power operation,an all-fiber laser oscillator based on a CCTC YDF was investigated experimentally.As a result,a maximum output power of 3.42 kW was achieved with an optical-to-optical efficiency of 55.2%,although the TMI effect was observed at an output power of~3.12 kW.The measured beam quality(M^(2)factor)was~1.7,and no sign of the Raman component was observed in the spectrum.We believe that CCTC YDF has great potential to simultaneously mitigate the SRS and TMI effects,and further power scaling is promising by optimizing the structure of the YDF.Yun Ye Xianfeng Lin Xiaoming Xi Chen Shi Baolai Yang Hanwei Zhang Xiaolin Wang Jinyan Li Xiaojun Xu 2021High Power Laser Science and Engineering2021,9,2:5

9Geniposide, the component of the Chinese herbal formula Tongluojiunao, protects amyloid-β peptide(1–42)-mediated death of hippocampal neurons via the non-classical estrogen signaling pathway显示文摘Tongluojiunao(TLJN) is an herbal medicine consisting of two main components, geniposide and ginsenoside Rg1. TLJN has been shown to protect primary cultured hippocampal neurons. However, its mechanism of action remains unclear. In the present study, primary cultured hippocampal neurons treated with Aβ1–42(10 μmol/L) significantly increased the release of lactate dehydrogenase, which was markedly reduced by TLJN(2 μL/mL), specifically by the component geniposide(26 μmol/L), but not ginsenoside Rg1(2.5 μmol/L). The estrogen receptor inhibitor, ICI182780(1 μmol/L), did not block TLJN- or geniposide-mediated decrease of lactate dehydrogenase under Aβ1–42-exposed conditions. However, the phosphatidyl inositol 3-kinase or mitogen-activated protein kinase pathway inhibitor, LY294002(50 μmol/L) or U0126(10 μmol/L), respectively blo cked the decrease of lactate dehydrogenase mediated by TLJN or geniposide. 12Theories and calculation methods for regional objective ET显示文摘The regional objective ET (Evapotranspiration) is a new concept in water resources research,which refers to the total amount of water that could be exhausted from a region in the form of vapor per year. The objective-ET based water resources management allocates water to different regions in terms of ET. It controls the water exhausted from a region to meet the objective ET. The regional objective ET must be adapted to fit the region’s local available water resources. By improving the water utilization effi-ciency and reducing the unrecoverable water in the social water circle,it is saved so that water related production is maintained or even increased under the same water consumption conditions. Regional water balance is realized by rationally deploying the available water among different industries,adjusting industrial structures,and adopting new water-saving technologies,therefore to meeting the requirements for groundwater conservation,agricultural income stability,and avoiding environmental damages. Furthermore,water competition among various departments and industries (including envi-ronmental and ecological water use) may be avoided. This paper proposes an innovative definition of objective ET,and its principles,sub-index systems. 13Theories and calculation methods for regional objective ET(evapotranspiration):Applications显示文摘The regional objective ET(evapotranspiration) is defined as the quantity of water that could be con-sumed in a particular region.It varies with the water conditions and economic development stages in the region.It is also constrained by the requirement of benign environment cycle.At the same time,it must meet the demands of sustainable economic growth and the construction of harmony society.Objective ET based water resources distribution will replace the conventional method,which empha-sizes the balance between the water demand and the water supply.It puts focus on the reasonable water consumption instead of the forecasted water demand,which is usually greater than the actual one.In this paper,we calculated the objective ET of 2010 year level in Tianjin by an analysis-integra-tion-assessment method.Objective ET can be classified into two parts:controllable ET and uncontrol-lable ET.Controllable ET includes the ET from irrigation land and the ET from resident land,among which the former can be calculated with soil moisture model and evapotranspiration model,while the latter can be calculated by water use ration and water consumption rate.The uncontrollable ET can be calculated with the distributed hydrological model and the remote sensing monitoring model.The two models can be mutually calibrated.In this paper,eight schemes are put forward based on different portfolios of water resources.The objective ET of each scheme was calculated and the results were assessed and analyzed.Finally,an optimal scheme was recommended.LIU JiaHong QIN DaYong WANG MingNa LüJinYan SANG XueFeng ZHANG RuiMei 2009Science China(Technological Sciences)2009,52,5:2
14Temporal variations in aerosol composition at Lanzhou City, Northwestern China显示文摘Although Lanzhou is one of the most heavily polluted cities in China, the composition of its air pollutants have not yet been stud- ied in detail. The data of four months on the concentrations and compositions of ions from daily air-filter samples in the winter and spring during 2007-2008 were analyzed to investigate temporal trends and their possible causes. The results indicate that mean concentrations of almost all the aerosol components are greater in winter than in spring due to stronger emissions in winter and weaker localized dispersion. Spring dust storms are the major cause of the highest peak PM10 concentrations recorded during the study period; however, these elevated levels were transient. Ion concentrations related to coal combustion show the greatest reduc- tion from winter to spring, while the concentrations of strong crustal components show a less-pronounced reduction. Strong cycles in the levels of particulate matter (PM) and all ions are also observed during the winter months due to meteorological conditions. Depending on the season, nine different ions accounted for 20%-50% of the PM concentration. The particle size of polluting ions was constant at times of increasing PM concentrations (particularly during winter), whereas the particle size of crustal ions in- creased dramatically during spring dust storms. Local meteorological conditions (especially wind speed) have a strong influence on the levels of pollutants. Four dust storms were noted, including one during winter. In summary, the regional transport of desert dust from the Gobi can significantly affect air quality and the chemical composition of aerosols in Lanzhou. The dust storms can strongly increase concentration of crustal ions, which are characteristics of deserts in northwestern China. This observation is in agreement with back-trajectories, which show reduced levels of pollutant ions during dust storms. 16Tumor Microenvironment Cascade‑Responsive Nanodrug with Self‑Targeting Activation and ROS Regeneration for Synergistic Oxidation‑Chemotherapy显示文摘Carrier-free nanodrug with exceptionally high drug payload has attracted increasing attentions.Herein,we construct a pH/ROS cascade-responsive nanodrug which could achieve tumor acidity-triggered targeting activation followed by circularly amplified ROS-triggered drug release via positive-feedback loop.The di-selenide-bridged prodrug synthesized from vitamin E succinate and methotrexate(MTX)self-assembles into nanoparticles(VSeM);decorating acidity-cleavable PEG onto VSeM surface temporarily shields the targeting ability of MTX to evade immune clearance and consequently elongate circulation time.Upon reaching tumor sites,acidity-triggered detachment of PEG results in targeting recovery to enhance tumor cell uptake.Afterward,the VSeM could be dissociated in response to intracellular ROS to trigger VES/MTX release;then the released VES could produce extra ROS to accelerate the collapse of VSeM.Finally,the excessive ROS produced from VES could synergize with the released MTX to efficiently suppress tumor growth via orchestrated oxidation-chemotherapy.Our study provides a novel strategy to engineer cascade-responsive nanodrug for synergistic cancer treatment.Yang Li Jinyan Lin Peiyuan Wang Qiang Luo Fukai Zhu Yun Zhang Zhenqing Hou Xiaolong Liu Jingfeng Liu 2020Nano-Micro Letters2020,12,12:2
17New Progress in Paleoearthquake Studies of the East Sertengshan Piedmont Fault, Inner Mongolia, China显示文摘The two eastern segments of the Sertengshan piedmont fault have moved considerably since the Holocene. Several paleoseismic events have occurred along the fault since 30 ka BP. Paleoearthquake studies have been advanced by digging new trenches and combining the results with the findings of previous studies. Comprehensive analyses of the trenches revealed that 6 paleoseismic events have occurred on the Kuoluebulong segment since approximately 30 ka BP within the following successive time periods: 19.01–37.56, 18.73, 15.03–15.86, 10.96, 5.77–6.48, and 2.32 ka BP. The analyses also revealed that 6 paleoseismic events have occurred on the Dashetai segment since approximately 30 ka BP, and the successive occurrence times are 29.07, 19.12–28.23, 13.92–15.22, 9.38–9.83, 6.08–8.36, and 3.59 ka BP. The results indicate that quasi-periodic recurrences occurred along the two segments with an approximate 4 000 a mean recurrence interval. The consistent timing of the 6 events between the two segments indicates that the segments might conform to the cascade rupturing model between the two segments. The gut microbiota change in A. japonicus was then analyzed through high-throughput sequencing during the immersion bathing in B. velezensis DY-6 at the optimum concentration for 49 d. The results illustrate that the body weight growth rate of all bathing groups was higher than that of the control. The highest growth rate (25.3%) was achieved when the bacterial concentration was 1×10^3 CFU/mL. The activities of non-specific immune enzymes (ACP, AKP, SOD and LZM) of all bathing groups increased, and the activities of the enzymes of groups bathed with the bacterium at 1×10^3 and 1×10^4 CFU/mL reached the highest on day 21 and day 28. Taking the growth rate and economic cost into consideration, the optimum concentration of B. velezensis DY-6 was 1×10^3 CFU/mL. The influences of immersion bathing in B. velezensis DY-6 at 1×10^3 CFU/mL on the gut microbiota of A. japonicus were then evaluated through 16S rDNA sequencing analysis. Results showed that the gut microbiota changed with the addition of B. velezensis DY-6, and the richness and diversity of the gut microbiota peaked twice on day 14 and day 21, respectively. In association with the non-specific immune enzyme activities and if day 28 was selected as the dividing point, the community structure of the gut microbiota could be obviously divided into two types. The correlation analysis revealed that the non-specific immune enzyme activities were correlated significantly to some gut bacteria (in the phyla Firmicutes, Proteobacteria, and Bacteroidetes) after immersion bathing in B. velezensis DY-6. 20Building highly active hybrid double-atom sites in C_(2)N for enhanced electrocatalytic hydrogen peroxide synthesis显示文摘Two-electron(2 e^(-))oxygen reduction reaction(ORR)shows great promise for on-site electrochemical synthesis of hydrogen peroxide(H_(2)O_(2)).However,it is still a great challenge to design efficient electrocatalysts for H_(2)O_(2)synthesis.To address this issue,the logical design of the active site by controlling the geometric and electronic structures is urgently desired.Therefore,using density functional theory(DFT)computations,two kinds of hybrid double-atom supported on C_(2)N nanosheet(RuCu@C_(2)N and PdCu@C_(2)N)are screened out and their H_(2)O_(2)performances are predicted.PdCu@C_(2)N exhibits higher activity for H_(2)O_(2)synthesis with a lower overpotential of 0.12 V than RuCu@C_(2)N(0.59 V),Ru_(3)Cu(110)facet(0.60 V),and PdCu(110)facet(0.54 V).In aqueous phase,the adsorbed O_(2)is further stabilized with bulk H_(2)0 and the thermodynamic rate-determining step of 2 e^(-) ORR change.The activation barrier on PdCu@C_(2)N is 0.43 eV lower than the one on RuCu@C_(2)N with 0.68 eV.PdCu@C_(2)N is near the top of 2 e^(-) ORR volcano plot,and exhibits high selectivity of H_(2)O_(2.)This work provides guidelines for designing highly effective hybrid double-atom electrocatalysts(HDACs)for H_(2)O_(2)synthesis.Yongyong Cao JinYan Zhao Xing Zhong Guilin Zhuang Shengwei Deng Zhongzhe Wei Zihao Yao Jianguo Wang 2021Green Energy & Environment2021,6,6:2
