12018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents显示文摘Syncope belongs to the transient loss of consciousness(TLOC), characterized by a rapid onset, short duration, and spontaneous complete recovery. It is common in children and adolescents, accounting for 1% to 2% of emergency department visits.Recurrent syncope can seriously affect children's physical and mental health, learning ability and quality of life and sometimes cardiac syncope even poses a risk of sudden death. The present guideline for the diagnosis and treatment of syncope in children and adolescents was developed for guiding a better clinical management of pediatric syncope. Based on the globally recent development and the evidence-based data in China, 2018 Chinese Pediatric Cardiology Society(CPCS) guideline for diagnosis and treatment of syncope in children and adolescents was jointly prepared by the Pediatric Cardiology Society, Chinese Pediatric Society, Chinese Medical Association(CMA)/Committee on Pediatric Syncope, Pediatricians Branch, Chinese Medical Doctor Association(CMDA)/Committee on Pediatric Cardiology, Chinese College of Cardiovascular Physicians, Chinese Medical Doctor Association(CMDA)/Pediatric Cardiology Society, Beijing Pediatric Society, Beijing Medical Association(BMA). 2Genetic basis and biotechnological manipulation of sexual dimorphism and sex determination in fish显示文摘Aquaculture has made an enormous contribution to the world food production,especially to the sustainable supply of animal proteins.The utility of diverse reproduction strategies in fish,such as the exploiting use of unisexual gynogenesis,has created a typical case of fish genetic breeding.A number of fish species show substantial sexual dimorphism that is closely linked to multiple economic traits including growth rate and body size,and the efficient development of sex-linked genetic markers and sex control biotechnologies has provided significant approaches to increase the production and value for commercial purposes.Along with the rapid development of genomics and molecular genetic techniques,the genetic basis of sexual dimorphism has been gradually deciphered,and great progress has been made in the mechanisms of fish sex determination and identification of sex-determining genes.This review summarizes the progress to provide some directive and objective thinking for further research in this field.MEI Jie GUI Jian-Fang 2015Science China(Life Sciences)2015,58,2:45
3Identification of long-range transport pathways and potential sources of PM2.5 and PM10 in Beijing from 2014 to 2015显示文摘Trajectory clustering, potential source contribution function(PSCF) and concentration-weighted trajectory(CWT) methods were applied to investigate the transport pathways and identify potential sources of PM_(2.5)and PM_(10) in different seasons from June 2014 to May 2015 in Beijing.The cluster analyses showed that Beijing was affected by trajectories from the south and southeast in summer and autumn. In winter and spring, Beijing was not only affected by the trajectories from the south and southeast, but was also affected by trajectories from the north and northwest. In addition, the analyses of the pressure profile of backward trajectories showed that backward trajectories, which have important influence on Beijing, were mainly distributed above 970 h Pa in summer and autumn and below 950 h Pa in spring and winter. This indicates that PM_(2.5)and PM_(10) were strongly affected by the near surface air masses in summer and autumn and by high altitude air masses in winter and spring. Results of PSCF and CWT analyses showed that the largest potential source areas were identified in spring, followed by winter and autumn,then summer. In addition, potential source regions of PM_(10) were similar to those of PM_(2.5). There were a clear seasonal and spatial variation of the potential source areas of Beijing and the airflow in the horizontal and vertical directions. 5Optimal time for mesenchymal stem cell transplantation in rats with myocardial infarction显示文摘Background:Bone marrow mesenchymal stem cell (MSC) transplantation is a promising strategy in the treatment of myocardial infarction (MI). However, the time for transplanting cells remains controversial. The aim of this study was to find an optimal time point for cell transplantation. Methods: MSCs were isolated and cultured from Sprague-Dawley (SD) rats. MI model was set up in SD rats by permanent ligation of left anterior descending coronary artery. MSCs were directly injected into the infarct border zone at 1 h, 1 week and 2 weeks after MI, respectively. Sham-operated and MI control groups received equal volume of phosphate buffered saline (PBS). At 4 weeks after MI, cardiac function was assessed by echocardiography; vessel density was analyzed on hematoxylin-eosin stained slides by light microscopy; the apoptosis of cardiomyocytes was evaluated by terminal deoxynucleotidyl transferase-mediated dUTP nick end-labeling (TUNEL) assay; the expressions of proteins were analyzed by Western blot. Results: MSC transplantation improved cardiac function, reduced the apoptosis of cardiomyocytes and increased vessel density. These benefits were more obvious in 1-week group than in 1-h and 2-week groups. There are more obvious in-creases in the ratio of bcl-2/bax and the expression of vascular endothelial growth factor (VEGF) and more obvious decreases in the expression of cleaved-caspase-3 in 1-week group than those in other two groups. Conclusion: MSC transplantation was beneficial for the recovery of cardiac function. 7Identification of a Newly Isolated Getah Virus in the China-Laos Border,China显示文摘In this study, we isolated a virus strain(YN12031) from specimens of Armigeres subalbatus collected in the China-Laos border. BHK-21 cells infected with YN12031 exhibited an evident cytopathic effect(CPE) 32 h post-infection. The virus particles were spherical, 70 nm in diameter, and enveloped; they also featured surface fibers. Molecular genetic analysis revealed that YN12031 was closely related to alpha viruses such as Chikungunya virus and Sindbis virus, and located in the same clade as MM2021, the prototype of Getahvirus(GETV) isolated in Malaysia in 1955. Phylogenetic analysis of the E2 and capsid genes further revealed that YN12031 was located in the same clade as the Russian isolate LEIV/16275/Mag. Analysis of the homology of nucleotides and amino acids in the coding area and E2 gene demonstrated that the YN12031 isolated from the China-Laos border(tropical region) was related closest to the LEIV/16275/Mag isolate obtained in Russia(North frigid zone area) among other isolates studied. These results suggest that GETV can adapt to different geographical environments to propagate and evolve. 8Detrimental Effect of Electromagnetic Pulse Exposure on Permeability of In Vitro Blood-brain-barrier Model显示文摘Objective To study the effect of electromagnetic pulse (EMP) exposure on permeability of in vitro blood-brain-barrier (BBB) model. Methods An in vitro BBB model, established by co-culturing brain microvascular endothelial cells (BMVEC) and astroglial cells (AC) isolated from rat brain, was exposed to EMP at 100 kV/m and 400 kV/m, respectively. Permeability of the model was assayed by measuring the transendothelial electrical resistance (TEER) and the horseradish peroxidase (HRP) transmission at different time points. Levels of BBB tight junction-related proteins were measured at 0, 1, 2, 4, 8, 12, 16, 20, 24 h after EMP exposure by Western blotting. Results The TEER level was lower in BBB model group than in control group at 12 h after EMP, exposure which returned to its normal level at 24 h. The 24 h recovery process was triphasic and biphasic respectively after EMP exposure at 100 kV/m and 400 kV/m. Following exposure to 400 kV/m EMP, the HRP permeability increased at 1-12 h and returned to its normal level at 24 h. Western blotting showed that the claudin-5 and ZO-1 protein levels were changed after EMP exposure. 9Development of a static test apparatus for evaluating the performance of three PM2.5 separators commonly used in China显示文摘PM2.5 separator directly affects the accuracy of PM2.5 sampling.The specification testing and evaluation for PM2.5 separator is particularly important,especially under China’s wide variation of terrain and climate.In this study,first a static test apparatus based on polydisperse aerosol was established and calibrated to evaluate the performance of the PM2.5 separators.A uniform mixing chamber was developed to make particles mix completely.The aerosol concentration relative standard deviations of three test points at the same horizontal chamber position were less than 0.57%,and the particle size distribution obeyed logarithmic normal distribution with an R2 of 0.996.The flow rate deviation between the measurement and the set point flow rate agreed to within±1.0%in the range of-40 to 50℃.Secondly,the separation,flow and loading characteristics of three cyclone separators(VSCC-A,SCC-A and SCC112)were evaluated using this system.The results showed that the 50%cutoff sizes(D50)of the three cyclones were 2.48,2.47 and 2.44μm when worked at the manufacturer’s recommended flow rates,respectively.The geometric standard deviation(GSD)of the capture efficiency of VSCCA was 1.23,showed a slightly sharper than SCC-A(GSD=1.27),while the SCC112 did not meet the relevant indicator(GSD=1.2±0.1)with a GSD=1.44.The flow rate and loading test had a great effect on D50,while the GSD remained almost the same as before.In addition,the maintenance frequency under different air pollution conditions of the cyclones was summarized according to the loading test.Peng Du Jianguo Liu Huaqiao Gui Jiaoshi Zhang Tongzhu Yu Jie Wang Yin Cheng Yihuai Lu Yawei Yao Qiang Fu ChihChieh Chen 2020Journal of Environmental Sciences2020,32,1:9

12Loss of stat3 function leads to spine malformation and immune disorder in zebrafish显示文摘STAT(Signal Transducers and Activators of Transcription) gene family members have been revealed to be involved in cell growth and differentiation in vertebrates. Despite their physiological importance,their functions are poorly studied at organ and systemic levels. In this study, we performed a genome-wide analysis using data from invertebrates to vertebrates to identify STAT genes and analyze their evolutionary history. Interestingly, the STAT gene family undergoes genome duplications during the evolutionary history with STAT3 homologues firstly appearing in the basal extant vertebrate, sea lamprey, suggesting its possible roles in spine formation. To investigate the functions of stat3 in fish species, TALEN technology was performed to generate mutant zebrafish lines. Stat3 mutant zebrafish showed no obvious defects at early developmental stage but displayed severe lateral and vertical curvature of the spine(scoliosis), spine fracture and the incomplete bone joints with narrower junction between vertebrae at early juvenile stage, as indicated by Alizarin red and Alcian blue staining,radiography and micro-computed tomography(Micro CT) analysis. Transcriptome analysis reveals dramatic alterations in a number of genes involved in immune and infection response, skeletal development and somatic growth, especially downregulated expression of collagen gene family, in the juvenile stat3 mutant zebrafish. Moreover, most of the collagen genes were detected to have abnormal expression pattern during the formation of spine deformities in stat3 mutants. 14Bioinformatic identification of genes encoding Clq-domaincontaining proteins in zebrafish显示文摘C1q is the first subcomponent of classical pathway in the complement system and a major link between innate and acquired immuni- ties. The globular (gC1q) domain similar with C1q was also found in many non-complement C1q-domain-containing (C1qDC) proteins which have similar crystal structure to that of the multifunctional tumor necrosis factor (TNF) ligand family, and also have diverse func- tions. In this study, we identified a total of 52 independent gene sequences encoding C1q-domain-containing proteins through compre- hensive searches of zebrafish genome, cDNA and EST databases. In comparison to 31 orthologous genes in human and different numbers in other species, a significant selective pressure was suggested during vertebrate evolution. Domain organization of C1q-domain-con- taining (C1qDC) proteins mainly includes a leading signal peptide, a collagen-like region of variable length, and a C-terminal C1q do- main. There are 11 highly conserved residues within the C1q domain, among which 2 are invariant within the zebrafish gene set. A more extensive database searches also revealed homologous C1qDC proteins in other vertebrates, invertebrates and even bacterium, but no homologous sequences for encoding C1qDC proteins were found in many species that have a more recent evolutionary history with zebrafish. 15A WRKY transcription factor confers aluminum tolerance via regulation of cell wall modifying genes显示文摘Modification of cell wall properties has been considered as one of the determinants that confer aluminum(Al)tolerance in plants,while how cell wall modifying processes are regulated remains elusive.Here,we present a WRKY transcription factor WRKY47 involved in Al tolerance and root growth.Lack of WRKY47 significantly reduces,while overexpression of it increases Al tolerance.We show that lack of WRKY47 substantially affects subcellular Al distribution in the root,with Al content decreased in apoplast and increased in symplast,which is attributed to the reduced cell wall Al-binding capacity conferred by the decreased content of hemicellulose I in the wrky47-1 mutant.Based on microarray,real time-quantitative polymerase chain reaction and chromatin immunoprecipitation assays,we further show that WRKY47 directly regulates the expression of EXTENSIN-LIKE PROTEIN(ELP)and XYLOGLUCAN ENDOTRANSGLUCOSYLASE-HYDROLASES17(XTH17)responsible for cell wall modification.Increasing the expression of ELP and XTH17 rescued Al tolerance as well as root growth in wrky47-1 mutant.In summary,our results demonstrate that WRKY47 is required for root growth under both normal and Al stress conditions via direct regulation of cell wall modification genes,and that the balance of Al distribution between root apoplast and symplast conferred by WRKY47 is important for Al tolerance.Chun Xiao Li Jing Ying Yan Jiang Yuan Ren Li Sun Chen Xu Gui Xin Li Zhong Jie Ding Shao Jian Zheng 2020Journal of Integrative Plant Biology2020,62,8:7
16LBE-DEM coupled simulation of gas-solid two-phase cross jets显示文摘This paper establishes a lattice Boltzmann equation-discrete element method (LBE-DEM) coupled simulation method under the Eulerian-Lagrangian framework at first, and applies it to simulating a two-dimensional gas-solid two-phase cross jet. The gas phase is simulated by the lattice-Boltzmann method via the TD2G9 model; the solid phase is traced by the Lagrangian method and the inter-particle collision is calculated by the DEM method. Three values of the Stokes number St=10, 25, and 50 are simulated under the same mass loading. This paper focuses on the characteristics of vortex structure, particle distribution, and the reverse-flow/rebounding rate in cross jets. We analyze the characteristics of fluid vortex motion, particle cluster distribution, rebounding rate of particles and the influencing factors for them. The results show the existence of joint distribution of discrete clusters and discrete particles in cross jets. Meanwhile, it shows that a larger concentration of particles in the early stage of jet evolution or a smaller Stokes number under the same mass loading can produce a larger rebounding rate. 17Grain refinement of commercial pure Al treated by Pulsed Magneto-Oscillation on the top surface of melt显示文摘Commercial pure Al can be refined by Pulsed Magneto-Oscillation(PMO) treatment applied via a plate induction coil above the top surface of the melt. The proportion of the equiaxed zone area increases with decreasing Height to Diameter(H/D) ratios from 3.5 to1.8 and further to 1.0. Meanwhile, it increases and then decreases with increasing peak current for the three kinds of ingots with H/D ratios of 3.5, 1.8 and 1.0, respectively. However, when the H/D ratio decreases to 0.44, the area proportion of equiaxed zone can reach the maximum value with a lower peak current. FEA software simulation indicates that smaller H/D ratio results in larger current density, electromagnetic force and convection on the top surface of the melt, favoring nucleation and subsequent grain formation. Through evaluating Joule heating effect by PMO, it was found that the proper amount of Joule heating benefits grain refinement.
