1Peripheral T-lymphocyte subpopulations in different clinical stages of chronic HBV infection correlate with HBV load显示文摘AIM: To characterize the peripheral T-cell subpopulation profiles and their correlation with hepatitis B virus (HBV) replication in different clinical stages of chronic HBV infection.METHODS: A total of 422 patients with chronic HBV infection were enrolled in this study. The patients were divided into three stages: immune-tolerant stage, immune active stage, and immune-inactive carrier stage. Composition of peripheral T-cell subpopulations was determined by flow cytometry. HBV markers were detected by enzyme-linked immunosorbent assay. SerumHBV DNA load was assessed by quantitative real-time polymerase chain reaction.RESULTS: CD8+ T-cells were significantly higher in patients at the immune-tolerant stage than in patients at the immune-active and -inactive carrier stages (36.87 ± 7.58 vs 34.37 ± 9.07, 36.87 ± 7.58 vs 28.09 ± 5.64, P < 0.001). The peripheral blood in patients at the immune-tolerant and immune active stages contained more CD8+ T-cells than CD4+ T-cells (36.87 ± 7.58 vs 30.23 ± 6.35, 34.37 ± 9.07 vs 30.92 ± 7.40, P < 0.01), whereas the peripheral blood in patients at the immune-inactive carrier stage and in normal controls contained less CD8+ T-cells than CD4+ T-cells (28.09 ± 5.64 vs 36.85 ± 6.06, 24.02 ± 4.35 vs 38.94 ± 3.39, P < 0.01). ANOVA linear trend test showed that CD8+ T-cells were signif icantly increased in patients with a high viral load (39.41 ± 7.36, 33.83 ± 7.50, 31.81 ± 5.95 and 26.89 ± 5.71, P < 0.001), while CD4+ T-cells were signif icantly increased in patients with a low HBV DNA load (37.45 ± 6.14, 33.33 ± 5.61, 31.58 ± 6.99 and 27.56 ± 5.49, P < 0.001). Multiple regression analysis displayed that log copies of HBV DNA still maintained its highly signif icant coefficients for T-cell subpopulations, and was the strongest predictors for variations in CD3+, CD4+ and CD8+ cells and CD4+/CD8+ ratio after adjustment for age at HBV-infection, maternal HBV-infection status, presence of hepatitis B e antigen and HBV mutation.CONCLUSION: Differences in peripheral T-cell subpopulation profi les can be found in different clinical stages of chronic HBV infection. 2Effect of viral load on T-lymphocyte failure in patients with chronic hepatitis B显示文摘AIM: To investigate peripheral T-lymphocyte sub-population profile and its correlation with hepatitis B virus (HBV) replication in patients with chronic hepatitis B (CHB).METHODS: Distribution of T-lymphocyte subpopulations in peripheral blood was measured by flow cytometry in 206 CHB patients. HBV markers were detected with ELISA. Serum HBV DNA load was assessed with quantitative real-time polymerase chain reaction (PCR). The relationship between HBV replication and variation in peripheral T-cell subsets was analyzed.RESULTS: CHB patients had significantly decreased CD3+ and CD4+ cells and CD4+/CD8+ ratio, and increased CD8+ cells compared with uninfected controls (55.44 ± 12.39 vs 71.07 ± 4.76, 30.92 ± 7.48 vs 38.94 ± 3.39, 1.01 ± 0.49 vs 1.67 ± 0.33, and 34.39 ± 9.22 vs 24.02 ± 4.35; P < 0.001, respectively). Univariate analysis showed a similar pattern of these parameters was significantly associated with high viral load, presence of serum hepatitis B e antigen (HBeAg) expression, liver disease severity, history of maternal HBV infection, and young age at HBV infection, all with P < 0.01. There was a significant linear relationship between viral load and these parameters of T-lymphocyte subpopulations (linear trend test P < 0.001). There was a negative correlation between the levels of CD3+ and CD4+ cells and CD4+/CD8+ ratio and serum level of viral load in CHB patients (r = -0.68, -0.65 and -0.75, all P < 0.0001), and a positive correlation between CD8+ cells and viral load (r = 0.70, P < 0.0001). There was a significant decreasing trend in CD3+ and CD4+ cells and CD4+/CD8+ ratio with increasing severity of hepatocyte damage and decreasing age at HBV infection (linear trend test P < 0.01). In multiple regression (after adjustment for age at HBV infection, maternal HBV infection status and hepatocyte damage severity) log copies of HBV DNA maintained a highly significant predictive coefficient on T-lymphocyte subpopulations, and was the strongest predictor of variation in CD3+, CD4+, CD8+ cells and CD4+/CD8+ ratio. However, the effect of HBeAg was not significant.CONCLUSION: T-lymphocyte failure was signifi-cantly associated with viral replication level. 3Dynamic Microwave-assisted Extraction of Flavonoids from Radix Scutellariae显示文摘Dynamic microwave-assisted extraction(DMAE) of flavonoids from Radix Scutellarie was described in this article. A TM010 microwave resonant cavity was used as the energy coupler. Several parameters of DMAE, including concentration, flow rate, volume of the extraction solvent, and irradiation power, were optimized. The extraction was performed under the optimum conditions for the extraction of total flavonoids. The total flavonoids were determined by UV-Vis spectrophotometry. The main bioactive components, including baicalin, baicalein, and wogonin, were determined by HPLC. 4Efficacy of thymosin alpha-1 and interferon alpha in treatment of chronic viral hepatitis B:A randomized controlled study显示文摘AIM: To observe the efficiency and safety of thymosin-α1 treatment in patients with hepatitis B e antigen (HBeAg) and HBV DNA positive chronic hepatitis. METHODS: Sixty-two patients were randomly divided into groups A and B. The patients in group A received subcutaneous injection of 1.6 mg thymosin-α1, twice a week (T-α1 group) for six months, and the patients in group B received 5 MU interferon alpha (IFN-α) each day for fifteen days, then three times weekly (IFN-α group) for six months. The results between two groups treated with and the group untreated with IFN-α which was followed up for 12 mo (historical control group consisting of 30 patients) were compared, and three groups were comparable between each other (P > 0.05) at baseline (age, sex, clinical history, biochemical, and serological parameters). RESULTS: At the end of treatment, complete response, which was defined as alanine aminotransferase (ALT) normalization and HBV DNA and HBeAg loss, occurred in 9 of 29 (31.0%) patients in the T-α1 group and in 15 of 33 (45.5%) patients in the IFN-α group (c2 = 1.36, P >0.05). After a follow-up period of six months, a complete response was observed in 14 of 29 (48.3%) patients in the T-α1 group and in 9 of 33 (27.3%) patients in the IFN-α group (c2 = 2.93, P > 0.05). Compared with the results observed in the historical control (HC) group untreated with IFN-α which was followed up for 12 mo, the rate of complete response was significantly higher in IFN-α group at the end of therapy (1 of 30 vs 15 of 33, c2 = 14.72, P < 0.001) and in the T-α1 group at the end of follow-up (1 of 30 vs 14 of 29, c2 = 15.71, P < 0.001). In T-α1 and IFN-α treatment groups, the area under (the plasma concentration time) curve (AUC) of negative HBV DNA and HBeAg was 34%, 17%, 31% and 19% smaller than that in the HC group. By the end of the follow- up period, the proportions of ALT normalization and negative HBV DNA in the T-α1 group were significantly higher than those in the IFN-α and HC groups. The odds of ALT normalization and negative HBV DNA at the end of the follow-up was three-fold higher in the T-α1 group than in the IFN-α group. Unlike IFN-α, T-α1 was well tolerated by all patients, and no side effects appeared in T-α1 group.CONCLUSION: The results suggest that a 6-mo course of T-α1 therapy is effective and safe in patients with chronic hepatitis B. T-α1 is able to reduce HBV replication in patients with chronic hepatitis B. Furthermore, T-α1 is better tolerated than IFN-α and can gradually induce more sustained ALT normalization and HBV DNA and HBeAg loss. However, a response rate of 48.3% is still less ideal. 5A new perylene diimide-based colorimetric and fluorescent sensor for selective detection of Cu^(2+) cation显示文摘A new perylene diimide (PDI) ligand (1) functionalized with a dipicolylethylenediamine (DPEN) moiety was synthesized and first used as a colorimetric and fluorometric dual-channel sensor to specifically detect the presence of Cu2+ over a wide range of other cations. The solution of 1 (10 μmol/L) upon ad-dition of Cu2+ displayed distinguishing pink color compared with other cations including K+,Ni2+,Ca2+,Mn2+,Na+,Sr2+,Zn2+,Co2+,Cd2+,Mg2+,Cr3+,Ag+,and Ba2+,indicating the sensitivity and selectivity of 1 to Cu2+. Thus,the advantage of this assay is that naked-eye detection of Cu2+ becomes possible. More-over,among these metal ions investigated,only Cu2+ quenched more than half fluorescent intensity of 1. 9Efficacy of water fraction from Dioscorea cirrhosa on oxidative stress and apoptosis in H9c2 cardiomyocytes induced by H2O2显示文摘OBJECTIVE:To investigate the efficacy of water fraction from Dioscorea cirrhosa(WF)on oxidative damage and apoptosis of cardiomyocytes induced by H2O2,and to study its mechanism.METHODS:Cell viability was measured by the MST assay kit.The content of malondialdehyde(MDA),release of lactate dehydrogenase(LDH)and activity of catalase(CAT)and superoxide dismutase(SOD)were detected by biochemical kit.The content of reactive oxygen species(ROS)was assessed by nonfluorescent probe 2′,7′-dichlorofluorescin diacetate(DCFH-DA).JC-1 was used to analyze the mitochondrial membrane potential(mtΔΨ)and Annexin-V-FITC/PI staining was applied to assess apoptosis of H9c2 by flow cytometry.Moreover,the expression of B-cell lymphoma-2(Bcl-2),Bcl-2-associated X(Bax),caspase-3,caspase-9,cleaved-caspase-3 and cleaved-caspase-9 proteins was determined by western blot analysis.RESULTS:WF increased cell viability and decreased LDH leakage in H9c2 cells exposed to H2O2.WF treatment decreased ROS and MDA level,enhanced SOD and CAT activities,improved mtΔΨand inhibited apoptosis.Western blot analysis demonstrated that the ratio of Bcl-2/Bax was increased and the expression cleaved-caspase-3,caspase-3,cleaved-caspase-9 and caspase-9 were decreased in group treated with WF.CONCLUSION:WF protects H9c2 myocardial cells on H2O2-induced oxidative stress and apoptosis by scavenging ROS,improving antioxidant capacity,protecting mitochondrial and regulating the proteins expression related to apoptosis.LIU Chunhua PAN Jie WANG Aimin LAN Yanyu LI Yongjun LU Yuan SUN Jia WANG Yonglin LIU Ting LU Dingyan YOU Jingrui 2021Journal of Traditional Chinese Medicine2021,41,1:4

12Activation of Piezo1 contributes to matrix stiffness-induced angiogenesis in hepatocellular carcinoma显示文摘Background:Despite integrin being highlighted as a stiffness-sensor molecule in matrix stiffness-driven angiogenesis,other stiffness-sensor molecules and their mechanosensory pathways related to angiogenesis in hepatocellular carcinoma(HCC)remain obscure.Here,we explored the interplay between Piezo1 and integrinβ1 in the mechanosensory pathway and their effects on HCC angiogenesis to better understand matrix stiffness-induced angiogenesis.Methods:The role of Piezo1 in matrix stiffness-induced angiogenesis was investigated using orthotopic liver cancer SD rat models with high liver stiffness background,and its clinical significance was evaluated in human HCC tissues.Matrix stiffness-mediated Piezo1 upregulation and activation were assayed using an in vitro fibronectin(FN)-coated cell culture system with different stiffness,Western blotting and Ca^(2+)probe.The effects of shPiezo1-conditioned medium(CM)on angiogenesis were examined by tube formation assay,wound healing assay and angiogenesis array.The underlying mechanism by which Piezo1 participated in matrix stiffness-induced angiogenesis was analyzed by microRNA quantitative real-time polymerase chain reaction(qRT-PCR),matrix stiffness measurement,dual-luciferase reporter assay,ubiquitination assay and co-immunoprecipitation.Results:Increased matrix stiffness significantly upregulated Piezo1 expression at both cellular and tissue levels,and high expression of Piezo1 indicated an unfavorable prognosis.High matrix stiffness also noticeably enhanced the activation level of Piezo1,similar to its expression level.Piezo1 knockdown significantly suppressed tumor growth,angiogenesis,and lung metastasis of HCC rat models with high liver stiffness background.shPiezo1-CM from HCC cells attenuated tube formation and migration abilities of vascular endothelial cells remarkably,and analysis of differentially expressed pro-angiogenic factors revealed that Piezo1 promoted the expression and secretion of vascular endothelial growth factor(VEGF),CXC chemokine ligand 16(CXCL16)and insulin-like growth factor binding protein 2(IGFBP2).Matrix stiffness-caused Piezo1 upregulation/activation restrained hypoxia inducible factor-1α(HIF-1α)ubiquitination,subsequently enhanced the expression of downstream pro-angiogenic factors to accelerate HCC angiogenesis.Besides,collagen 1(COL1)-reinforced tissue stiffening resulted in more expression of Piezo1 via miR-625-5p.Conclusions:This study unravels a new mechanism by which the inte-grinβ1/Piezo1 activation/Ca2+influx/HIF-1αubiquitination/VEGF,CXCL16 and IGFBP2 pathway participates in matrix stiffness-driven HCC angiogenesis.Simultaneously,a positive feedback regulation loop as stiff matrix/integrinβ1/miR-625-5p/Piezo1 and COL1/stiffer matrix mediates matrix stiffness-caused Piezo1 upregulation.Miao Li Xi Zhang Mimi Wang Yaohui Wang Jiali Qian Xiaoxia Xing Zhiming Wang Yang You Kun Guo Jie Chen Dongmei Gao Yan Zhao Lan Zhang Rongxin Chen Jiefeng Cui Zhenggang Ren 2022Cancer Communications2022,42,11:3

17Gold nanoparticle-directed autophagy intervention for antitumor immunotherapy via inhibiting tumor-associated macrophage M2 polarization显示文摘Tumor-associated macrophages(TAMs),one of the dominating constituents of tumor microenvironment,are important contributors to cancer progression and treatment resistance.Therefore,regulation of TAMs polarization from M2 phenotype towards M1 phenotype has emerged as a new strategy for tumor immunotherapy.Herein,we successfully initiated antitumor immunotherapy by inhibiting TAMs M2 polarization via autophagy intervention with polyethylene glycol-conjugated gold nanoparticles(PEG-Au NPs).PEG-Au NPs suppressed TAMs M2 polarization in both in vitro and in vivo models,elicited antitumor immunotherapy and inhibited subcutaneous tumor growth in mice.As demonstrated by the m RFP-GFP-LC3 assay and analyzing the autophagy-related proteins(LC3,beclin1 and P62),PEGAu NPs induced autophagic flux inhibition in TAMs,which is attributed to the PEG-Au NPs induced lysosome alkalization and membrane permeabilization.Besides,TAMs were prone to polarize towards M2phenotype following autophagy activation,whereas inhibition of autophagic flux could reduce the M2polarization of TAMs.Our results revealed a mechanism underlying PEG-Au NPs induced antitumor immunotherapy,where PEG-Au NPs reduce TAMs M2 polarization via induction of lysosome dysfunction and autophagic flux inhibition.This study elucidated the biological effects of nanomaterials on TAMs polarization and provided insight into harnessing the intrinsic immunomodulation capacity of nanomaterials for effective cancer treatment.Siyue Zhang Fangyuan Xie Kaichun Li He Zhang You Yin Yuan Yu Guangzhao Lu Shihao Zhang Yan Wei Ke Xu Yan Wu Hong Jin Lan Xiao Leilei Bao Can Xu Yulin Li Ying Lu Jie Gao 2022Acta Pharmaceutica Sinica B2022,12,7:2

19Tandem Biocatalysis by CotA-TJ102@UIO-66-NH2 and Novozym 435 for Highly Selective Transformation of HMF into FDCA显示文摘2,5-Furandicarboxylic acid (FDCA) is a potential biorenewable chemical for applications including plastics, polyamides, drugs, etc. The selective biosynthesis of FDCA from 5-hydroxymethylfurfural (HMF) by a speci c enzyme poses a great challenge. In this study, we reported an e cient strategy to produce FDCA from HMF by the tandem biocatalysis of laccase (CotA-TJ102@UIO-66-NH 2 ) and Novozym 435. For the rst step, a nanoparticle metal organic framework was synthesized as a carrier to immobilize CotA-TJ102@UIO-66-NH 2 , which was assigned for the production of 5-formyl-2-furancarboxylic acid (FFCA) and featured an enzyme loading of 255.54 mg/g, speci c activity of 135.90 U/mg, and solid loading ratio of 99.65%. Under optimal conditions, an ideal FFCA yield of 98.5% was achieved, and the CotA-TJ102@UIO-66-NH2 pre- sented a high recycling capacity after 10 cycles. For the second step, Novozym 435 was applied for the further conversion of FFCA into FDCA, presenting a high FDCA yield of 95.5% under the optimized conditions. Novozym 435 also exhibited a high recyclability after eight cycles.
