1The lncRNA APOLO interacts with the transcription factor WRKY42 to trigger root hair cell expansion in response to cold显示文摘Plant long noncoding RNAs(lncRNAs)have emerged as important regulators of chromatin dynamics,impacting on transcriptional programs leading to different developmental outputs.The lncRNA AUXIN-REGULATED PROMOTER LOOP(APOLO)directly recognizes multiple independent loci across the Arabidopsis genome and modulates their three-dimensional chromatin conformation,leading to transcriptional shifts.Here,we show that APOLO recognizes the locus encoding the root hair(RH)master regulator ROOT HAIR DEFECTIVE 6(RHD6)and controls RHD6 transcriptional activity,leading to cold-enhanced RH elongation through the consequent activation of the transcription factor gene RHD6-like RSL4.Furthermore,we demonstrate that APOLO interacts with the transcription factor WRKY42 and modulates its binding to the RHD6 promoter.WRKY42 is required for the activation of RHD6 by low temperatures and WRKY42 deregulation impairs cold-induced RH expansion.Collectively,our results indicate that a novel ribonucleoprotein complex with APOLO and WRKY42 forms a regulatory hub to activate RHD6 by shaping its epigenetic environment and integrate signals governing RH growth and development.Michael Moison Javier Martinez Pacheco Leandro Lucero Camille Fonouni-Farde Johan Rodriguez-Melo Natanael Mansilla Aurelie Christ Jeremie Bazin Moussa Benhamed Fernando Ibanez Martin Crespi Jose M.Estevez Federico Ariel 2021Molecular Plant2021,14,6:8
2A comparison of survival and pathologic features of non-alcoholic steatohepatitis and hepatitis C virus patients with hepatocellular carcinoma显示文摘AIM:To compare the clinical outcome and pathologic features of non-alcoholic steatohepatitis(NASH) patients with hepatocellular carcinoma(HCC) and hepatitic C virus(HCV) patients with HCC(another group in which HCC is commonly seen) undergoing liver transplantation.METHODS:Patients transplanted for HCV and NASH at our institution from January 2000 to April 2011 were analyzed.All explanted liver histology and pre-transplant liver biopsies were examined by two specialist liver histopathologists.Patient demographics,disease free survival,explant liver characteristics and HCC features(tumour number,cumulative tumour size,vascular invasion and differentiation) were compared between HCV and NASH liver transplant recipients.RESULTS:A total of 102 patients with NASH and 283 patients with HCV were transplanted.The incidence of HCC in NASH transplant recipients was 16.7%(17/102).The incidence of HCC in HCV transplant recipients was 22.6%(64/283).Patients with NASH-HCC were statistically older than HCV-HCC patients(P < 0.001).A significantly higher proportion of HCV-HCC patients had vascular invasion(23.4% vs 6.4%,P = 0.002) and poorly differentiated HCC(4.7% vs 0%,P < 0.001) compared to the NASH-HCC group.A trend of poorer recurrence free survival at 5 years was seen in HCV-HCC patients compared to NASH-HCC who underwent a Liver transplantation(P = 0.11).CONCLUSION:Patients transplanted for NASH-HCC appear to have less aggressive tumour features compared to those with HCV-HCC,which likely in part accounts for their improved recurrence free survival.Roberto Hernandez-Alejandro Kris P Croome Martin Drage Nathalie Sela Jeremy Parfitt Natasha Chandok Paul Marotta Cheryl Dale William Wall Douglas Quan 2012World Journal of Gastroenterology2012,18,31:5
3Hybrid multi-chip assembly of optical communication engines by in situ 3D nanolithography显示文摘Three-dimensional(3D)nano-printing of freeform optical waveguides,also referred to as photonic wire bonding,allows for efficient coupling between photonic chips and can greatly simplify optical system assembly.As a key advantage,the shape and the trajectory of photonic wire bonds can be adapted to the mode-field profiles and the positions of the chips,thereby offering an attractive alternative to conventional optical assembly techniques that rely on technically complex and costly high-precision alignment.However,while the fundamental advantages of the photonic wire bonding concept have been shown in proof-of-concept experiments,it has so far been unclear whether the technique can also be leveraged for practically relevant use cases with stringent reproducibility and reliability requirements.In this paper,we demonstrate optical communication engines that rely on photonic wire bonding for connecting arrays of silicon photonic modulators to InP lasers and single-mode fibres.In a first experiment,we show an eight-channel transmitter offering an aggregate line rate of 448 Gbit/s by low-complexity intensity modulation.A second experiment is dedicated to a four-channel coherent transmitter,operating at a net data rate of 732.7 Gbit/s-a record for coherent silicon photonic transmitters with co-packaged lasers.Using dedicated test chips,we further demonstrate automated mass production of photonic wire bonds with insertion losses of(0.7±0.15)dB,and we show their resilience in environmental-stability tests and at high optical power.These results might form the basis for simplified assembly of advanced photonic multi-chip systems that combine the distinct advantages of different integration platforms.Matthias Blaicher Muhammad Rodlin Billah Juned Kemal Tobias Hoose Pablo Marin-Palomo Andreas Hofmann Yasar Kutuvantavida Clemens Kieninger Philipp-Immanuel Dietrich Matthias Lauermann Stefan Wolf Ute Troppenz Martin Moehrle Florian Merget Sebastian Skacel Jeremy Witzens Sebastian Randel Wolfgang Freude Christian Koos 2020Light(Science & Applications)2020,9,1:4

17The PIWI protein Aubergine recruits eIF3 to activate translation in the germ plasm显示文摘Piwi-interacting RNAs(piRNAs)and PIWI proteins are essential in germ cells to repress transposons and regulate mRNAs.In Drosophila,piRNAs bound to the PIWI protein Aubergine(Aub)are transferred maternally to the embryo and regulate maternal mRNA stability through two opposite roles.They target mRNAs by incomplete base pairing,leading to their destabilization in the soma and stabilization in the germ plasm.Here,we report a function of Aub in translation.Aub is required for translational activation of nanos mRNA,a key determinant of the germ plasm.Aub physically interacts with the poly(A)-binding protein(PABP)and the translation initiation factor elF3.Polysome gradient profiling reveals the role of Aub at the initiation step of translation.In the germ plasm,PABP and elF3d assemble in foci that surround Aub-containing germ granules,and Aub acts with elF3d to promote nanos translation.These results identify translational activation as a new mode of mRNA regulation by Aub,highlighting the versatility of PIWI proteins in mRNA regulation.Anne Ramat Maria-Rosa Garcia-Silva Camille Jahan Rima Nait-Saidi Jeremy Dufourt Celine Garret Aymeric Chartier Julie Cremaschi Vipul Patel Mathilde Decourcelle Amandine Bastide Francois Juge Martine Simonelig 2020Cell Research2020,30,5:1
