1Sintilimab versus docetaxel as second-line treatment in advanced or metastatic squamous non-small-cell lung cancer:an open-label,randomized controlled phase 3 trial(ORIENT-3)显示文摘Background:Treatment options for Chinese patients with locally advanced or metastatic squamous-cell non-small-cell lung cancer(sqNSCLC)after failure of first-line chemotherapy are limited.This study(ORIENT-3)aimed to evaluate the efficacy and safety of sintilimab versus docetaxel as second-line treatment in patients with locally advanced or metastatic sqNSCLC.Methods:ORIENT-3 was an open-label,multicenter,randomized controlled phase 3 trial that recruited patients with stage IIIB/IIIC/IV sqNSCLC after failure with first-line platinum-based chemotherapy.Patients were randomized in a 1:1 ratio to receive either 200 mg of sintilimab or 75 mg/m^(2) of docetaxel intravenously every 3 weeks,stratified by the Eastern Cooperative Oncology Group performance status.The primary endpoint was overall survival(OS)in the full analysis set(FAS).Secondary endpoints included progression-free survival(PFS),objective response rate(ORR),disease control rate(DCR),duration of response(DoR)and safety.Results:Between August 25,2017,and November 7,2018,290 patients were randomized.For FAS,10 patients fromthe docetaxel armwere excluded.Themedian OS was 11.79(n=145;95%confidence interval[CI],10.28-15.57)months with sintilimab versus 8.25(n=135;95%CI,6.47-9.82)months with docetaxel(hazard ratio[HR]:0.74;95%CI,0.56-0.96;P=0.025).Sintilimab treatment significantly prolonged PFS(median 4.30 vs.2.79 months;HR:0.52;95%CI,0.39-0.68;P<0.001)and showed higher ORR(25.50%vs.2.20%,P<0.001)and DCR(65.50%vs.37.80%,P<0.001)than the docetaxel arm.The median DoRwas 12.45(95%CI,4.86-25.33)months in the sintilimab arm and 4.14(95%CI,1.41-7.23)months in the docetaxel arm(P=0.045).Treatment-related adverse events of grade≥3were reported in 26(18.1%)patients in the sintilimab arm and 47(36.2%)patients in the docetaxel arm.Exploratory biomarker analysis showed potential predictive values of expression levels of two transcription factors,including OVOL2(HR:0.35;P<0.001)and CTCF(HR:3.50;P<0.001),for sintilimab treatment.Conclusions:Compared with docetaxel,sintilimab significantly improved the OS,PFS,and ORR of Chinese patients with previously treated locally advanced or metastatic sqNSCLC.Yuankai Shi Lin Wu Xinmin Yu Puyuan Xing Yan Wang Jianying Zhou Airong Wang Jianhua Shi Yi Hu Ziping Wang Guangyu An Yong Fang Sanyuan Sun Caicun Zhou Changli Wang Feng Ye Xingya Li Junye Wang Mengzhao Wang Yunpeng Liu Yanqiu Zhao Ying Yuan Jifeng Feng Zhendong Chen Jindong Shi Tao Sun Gang Wu Yongqian Shu Qisen Guo Yi Zhang Yong Song Shucai Zhang Yuan Chen Wei Li Hongrui Niu Wenwei Hu Lijun Wang Jianan Huang Yang Zhang Ying Cheng Zhengdong Wu Bo Peng Jiya Sun Christoph Mancao Yanqi Wang Luyao Sun 2022Cancer Communications2022,42,12:8

3Probabilistic stability analysis of Bazimen landslide with monitored rainfall data and water level fluctuations in Three Gorges Reservoir, China显示文摘Landslide is a common geological hazard in reservoir areas and may cause great damage to local residents’life and property.It is widely accepted that rainfall and periodic variation of water level are the two main factors triggering reservoir landslides.In this study,the Bazimen landslide located in the Three Gorges Reservoir(TGR)was back-analyzed as a case study.Based on the statistical features of the last 3-year monitored data and field instrumentations,the landslide susceptibility in an annual cycle and four representative periods was investigated via the deterministic and probabilistic analysis,respectively.The results indicate that the fluctuation of the reservoir water level plays a pivotal role in inducing slope failures,for the minimum stability coefficient occurs at the rapid decline period of water level.The probabilistic analysis results reveal that the initial sliding surface is the most important area influencing the occurrence of landslide,compared with other parts in the landslide.The seepage calculations from probabilistic analysis imply that rainfall is a relatively inferior factor affecting slope stability.This study aims to provide preliminary guidance on risk management and early warning in the TGR area.Wengang ZHANG Libin TANG Hongrui LI Lin WANG Longfei CHENG Tingqiang ZHOU Xiang CHEN 2020Frontiers of Structural and Civil Engineering2020,14,5:1

5A novel TNKS/USP25 inhibitor blocks the Wnt pathway to overcome multi-drug resistance in TNKS-overexpressing colorectal cancer显示文摘Modulating Tankyrases(TNKS),interactions with USP25 to promote TNKS degradation,rather than inhibiting their enzymatic activities,is emerging as an alternative/specific approach to inhibit the Wnt/β-catenin pathway.Here,we identified UAT-B,a novel neoantimycin analog isolated from Streptomyces conglobatus,as a small-molecule inhibitor of TNKS-USP25 protein-protein interaction(PPI)to overcome multi-drug resistance in colorectal cancer(CRC).The disruption of TNKS-USP25 complex formation by UAT-B led to a significant decrease in TNKS levels,triggering cell apoptosis through modulation of the Wnt/β-catenin pathway.Importantly,UAT-B successfully inhibited the CRC cells growth that harbored high TNKS levels,as demonstrated in various in vitro and in vivo studies utilizing cell line-based and patient-derived xenografts,as well as APC^(min/+)spontaneous CRC models.Collectively,these findings suggest that targeting the TNKS-USP25 PPI using a small-molecule inhibitor represents a compelling therapeutic strategy for CRC treatment,and UAT-B emerges as a promising candidate for further preclinical and clinical investigations.Hongrui Zhu Yamin Gao Liyun Liu Mengyu Tao Xiao Lin Yijia Cheng Yaoyao Shen Haitao Xue Li Guan Huimin Zhao Li Liu Shuping Wang Fan Yang Yongjun Zhou Hongze Liao Fan Sun Houwen Lin 2024Acta Pharmaceutica Sinica B2024,14,1:0

8Homojunction and ohmic contact coexisting carbon nitride for efficient photocatalytic hydrogen evolution显示文摘Carbon nitride(CN)has attracted intensive attention as a visible light photocatalyst,but the rapid recombination of photogenerated charge carriers limits its photocatalytic activity.Herein,we develop a new strategy to construct both homojunction and ohmic junction into CN via selectively introducing metallized CN(MCN),which leads to rapid separation and transfer of photogenerated charge carriers.The polymerization of urea in the presence of KOH creates CN homojunction with amino and cyano groups.The subsequent molten salt treatment induces a new type of cyano-terminated CN that can be converted to MCN through photodoping,forming homojunction and ohmic contact coexisting CN(HOCN).The formed HOCN photocatalyst exhibits a high photocatalytic H_(2)evolution rate of 18.5 mmol·g^(-1)·h^(-1)under visible light irradiation,45-fold higher than that of bulk CN.This strategy provides a new idea for designing ohmic contact between semiconductor and metal,and realizing efficient photocatalysis by improving charge separation and transfer.Xiao Fang Lu Chen Hongrui Cheng Xiaoqiong Bian Wenhao Sun Kaining Ding Xinghe Xia Xin Chen Jiefang Zhu Yuanhui Zheng 2023Nano Research2023,16,7:0
9Quantitative Inversion of REEs in Ion-Adsorbed Rare Earth Ores from the Liutang Area(South China),Based on Measured Hyperspectral Data显示文摘Rare earth minerals are important strategic resources to economic development all over the world.In this study,multiple linear regression and back propagation(BP) neural network methods are used to invert the contents of ion adsorbed rare earth elements(REEs) and exploring the feasibility of quantitative inversion of REEs through measured hyperspectral data in Liutang rare earth mines,South China.The result shows that the spectral curve of the rare earth ore samples has obvious absorption characteristics around 390,930,1 400,1 900 and 2 200 nm,and continuum removal and the 1st derivative treatment can highlight the absorption characteristics.The modeling accuracies of BP neural network are higher than that of multiple linear regression model.The BP neural network model of the 1st derivative data in 400–1 000 nm bands has the best inversion result of the total content of REEs,R2 reaches 0.98,the ratio of the performance to deviation(RPD) is larger than 3.0.The quantitative inversion model of each REE(except for Ce) has high precision,R2 is greater than 0.90 and RPD is greater than 3.0.The results indicate that quantitative inversion of REEs using measured spectra not only has great potential and feasibility in the exploration of rare earth minerals,but also provides a rapid test method for the content of ion-adsorbed rare earth elements.Gong Cheng Hongrui Zhang Huan Li Xiaoqing Deng Safiyanu Muhammad Elatikpo Jiaxuan Li Zhenguang Hu Guangqiang Li 2023Journal of Earth Science2023,34,4:0
