1Review on the Research and Practice of Deep Learning and Reinforcement Learning in Smart Grids显示文摘Smart grids are the developmental trend of power systems and they have attracted much attention all over the world.Due to their complexities,and the uncertainty of the smart grid and high volume of information being collected,artificial intelligence techniques represent some of the enabling technologies for its future development and success.Owing to the decreasing cost of computing power,the profusion of data,and better algorithms,AI has entered into its new develop-mental stage and AI 2.0 is developing rapidly.Deep learning(DL),reinforcement learning(RL)and their combination-deep reinforcement learning(DRL)are representative methods and relatively mature methods in the family of AI 2.0.This article introduces the concept and status quo of the above three methods,summarizes their potential for application in smart grids,and provides an overview of the research work on their application in smart grids.Dongxia Zhang Xiaoqing Han Chunyu Deng 2018CSEE Journal of Power and Energy Systems2018,4,3:44
2A LuxR family transcriptional regulator AniF promotes the production of anisomycin and its derivatives in Streptomyces hygrospinosus var.beijingensis显示文摘The protein synthesis inhibitor anisomycin features a unique benzylpyrrolidine system and exhibits potent selective activity against pathogenic protozoa and fungi.It is one of the important effective components in Agricultural Antibiotic120,which has been widely used as naturally-originated agents for treatment of crop decay in China.The chemical synthesis of anisomycin has recently been reported,but the complex process with low productivity made the biosynthesis still to be a vital mainstay in efforts.The biosynthetic gene cluster(BGC)of anisomycin in Streptomyces hygrospinosus var.beijingensis has been identified in our previous work,while poor understanding of the regulatory mechanism limited the yield enhancement via regulation engineering of S.hygrospinosus var.beijingensis.In this study here,we characterized AniF as an indispensable LuxR family transcriptional regulator for the activation of anisomycin biosynthesis.The genetic manipulations of aniF and the real-time quantitative PCR(RT-qPCR)revealed that it positively regulated the transcription of the anisomycin BGC.Moreover,the overexpression of aniF contributed to the improvement of the production of anisomycin and its derivatives.Dissection of the mechanism underlying the function of AniF revealed that it directly activated the transcription of the genes aniR-G involved in anisomycin biosynthesis.Especially,one AniF-binding site in the promoter region of aniR was identified by DNase I footprinting assay and an inverted repeat sequence(5′-GGGC-3′)composed of two 4-nt half sites in the protected region was found.Taken together,our systematic study confirmed the positive regulatory role of AniF and might facilitate the future construction of engineering strains with high productivity of anisomycin and its derivatives.Jufang Shen Lingxin Kong Yan Li Xiaoqing Zheng Qing Wang Weinan Yang Zixin Deng Delin You 2019Synthetic and Systems Biotechnology2019,4,1:10
3Toripalimab plus chemotherapy as second-line treatment in previously EGFR-TKI treated patients with EGFR-mutant-advanced NSCLC:a multicenter phase-II trial显示文摘This multicenter phase-II trial aimed to investigate the efficacy,safety,and predictive biomarkers of toripalimab plus chemotherapy as second-line treatment in patients with EGFR-mutant-advanced NSCLC.Patients who failed from first-line EGFR-TKIs and did not harbor T790M mutation were enrolled.Toripalimab plus carboplatin and pemetrexed were administrated every three weeks for up to six cycles,followed by the maintenance of toripalimab and pemetrexed.The primary endpoint was objective-response rate(ORR).Integrated biomarker analysis of PD-L1 expression,tumor mutational burden(TMB),CD8+tumor-infiltrating lymphocyte(TIL)density,whole-exome,and transcriptome sequencing on tumor biopsies were also conducted.Forty patients were enrolled with an overall ORR of 50.0%and disease-control rate(DCR)of 87.5%.The median progression free survival(PFS)and overall survival were 7.0 and 23.5 months,respectively.The most common treatment-related adverse effects were leukopenia,neutropenia,anemia,ALT/AST elevation,and nausea.Biomarker analysis showed that none of PD-L1 expression,TMB level,and CD8+TIL density could serve as a predictive biomarker.Integrated analysis of whole-exome and transcriptome sequencing data revealed that patients with DSPP mutation had a decreased M2 macrophage infiltration and associated with longer PFS than those of wild type.Toripalimab plus chemotherapy showed a promising anti-tumor activity with acceptable safety profiles as the second-line setting in patients with EGFR-mutant NSCLC.DSPP mutation might serve as a potential biomarker for this combination.A phase-III trial to compare toripalimab versus placebo in combination with chemotherapy in this setting is ongoing(NCT03924050).Tao Jiang Pingyang Wang Jie Zhang Yanqiu Zhao Jianying Zhou Yun Fan Yongqian Shu Xiaoqing Liu Helong Zhang Jianxing He Guanghui Gao Xiaoqian Mu Zhang Bao Yanjun Xu Renhua Guo Hong Wang Lin Deng Ningqiang Ma Yalei Zhang Hui Feng Sheng Yao Jiarui Wu Luonan Chen Caicun Zhou Shengxiang Ren 2021Signal Transduction and Targeted Therapy2021,6,11:8
4Transfection of the glial cell line-derived neurotrophic factor gene promotes neuronal differentiation显示文摘Glial cell line-derived neurotrophic factor recombinant adenovirus vector-transfected bone marrow mesenchymal stem cells were induced to differentiate into neuron-like cells using inductive medium containing retinoic acid and epidermal growth factor.Cell viability,microtubule-associated protein 2-positive cell ratio,and the expression levels of glial cell line-derived neurotrophic factor,nerve growth factor and growth-associated protein-43 protein in the supernatant were significantly higher in glial cell line-derived neurotrophic factor/bone marrow mesenchymal stem cells compared with empty virus plasmid-transfected bone marrow mesenchymal stem cells.Furthermore,microtubule-associated protein 2,glial cell line-derived neurotrophic factor,nerve growth factor and growth-associated protein-43 mRNA levels in cell pellets were statistically higher in glial cell line-derived neurotrophic factor/bone marrow mesenchymal stem cells compared with empty virus plasmid-transfected bone marrow mesenchymal stem cells.These results suggest that glial cell line-derived neurotrophic factor/bone marrow mesenchymal stem cells have a higher rate of induction into neuron-like cells,and this enhanced differentiation into neuron-like cells may be associated with up-regulated expression of glial cell line-derived neurotrophic factor,nerve growth factor and growth-associated protein-43.Jie Du Xiaoqing Gao Li Deng Nengbin Chang Huailin Xiong Yu Zheng 2014Neural Regeneration Research2014,9,1:7
5LeCDJ1,a chloroplast DnaJ protein,facilitates heat tolerance in transgenic tomatoes显示文摘The roles of a tomato(Lycopersicon esculentum) chloroplast-targeted DnaJ protein(LeCDJ1) were investigated using wild-type(WT) and sense transgenic tomatoes. The LeCDJ1 expression was upregulated by 38 °C, 42 °C, 45 °C, NaCl, PEG, methyl viologen(MV) and hydrogen peroxide(H2 O2), but not by 30 °C and 35 °C. Meanwhile, LeCDJ1 was involved in the response of plants to abscisic acid(ABA). Under heat stress, the sense plants showed better growth, higher chlorophyll content, lower malondialdehyde(MDA) accumulation and relative electrical conductivity(REC), and also less PSII photoinhibition than WT. Interestingly, the sense plants treated with streptomycin(SM), an inhibitor of organellar translation, still showed higher maximum photochemistry efficiency of PSII(Fv/Fm) and D1 protein levels than the SM-untreated WT, suggesting that the protective effect of LeCDJ1 on PSII was, at least partially, independent of D1protein synthesis. Furthermore, the relatively lower superoxide radical(O2·-) and H2 O2 levels in the sense plants were considered to be due to the higher ascorbate peroxidase(APX) and superoxide dismutase(SOD) activity, which seemed unlikely dependent on their transcription level. 7Nitrogen and Sulfur Co-doped Porous Carbon Derived from ZIF-8 as Oxygen Reduction Reaction Catalyst for Microbial Fuel Cells显示文摘Nitrogen and sulfur co-doped porous nanocarbon (ZIF-C-N-S) catalyst was successfully synthesized derived from ZIF-8 and thiourea precursors.The electrochemical measurements indicate that the as-obtained ZIF-C-N-S catalyst exhibits higher electrocatalytic activity for oxygen reduction reaction (ORR) in alkaline electrolyte and superior durability-longer than commercial Pt/C catalyst.The enhancment of electrocatalytic activity mainly be come from the open pore structure,large specific surface area as well as the synergistic effect resulted from the co-doping of N and S atoms.In addition,the ZIF-C-N-S catalyst is also used as the air cathode catalyst in the microbial fuel cell (MFC) device.The maximum power density and stable output voltage of ZIF-C-N-S based MFC are 1315 mW/m2 and 0.48 V,respectively,which is better than that of Pt/C based MFC.HAN Wuli YAN Xuemin JIANG Yu PING Mei DENG Xiaoqing ZHANG Yan 2020Journal of Wuhan University of Technology(Materials Science)2020,35,2:4
8Superresolution imaging of telomeres with continuous wave stimulated emission depletion (STED) microscope显示文摘The significant role of telomeres in cells has attracted much attention since they were discovered.Fluorescence imaging is an effective method to study subcellular structures like telomeres.However,the diffraction limit of traditional optical microscope hampers further investigation on them.Recent progress on superresolution fluorescence microscopy has broken this limit.In this work,we used stimulated emission depletion(STED) microscope to observe fluorescence-labeled telomeres in interphase cell nuclei.The results showed that the size of fluorescent puncta representing telomeres under the STED microscope was much smaller than that under the confocal microscope.Two adjacent telomeres were clearly separated via STED imaging,which could hardly be discriminated by confocal microscopy due to the diffraction limit.We conclude that STED microscope is a more powerful tool that enable us to obtain detailed information about telomeres.Shaopeng Wang Suhui Deng Xiaoqing Cai Shangguo Hou Jiajun Li Zhaoshuai Gao Jiang Li Lihua Wang Chunhai Fan 2016Science China Chemistry2016,59,11:3
9Do neural precursor cells exist in a distal neurogenic region following cerebral hemorrhage?显示文摘BACKGROUND: Cerebral injury in adult mammals can induce neural precursor cells (NPCs) to proliferate and migrate towards the focal zone,but it is unclear whether endogenous NPCs can migrate towards regions distal to the hemorrhagic focus or whether NPCs differentiate in the peripheral hemorrhagic region.OBJECTIVE: To investigate the distribution of endogenous NPCs in different brain regions of rats with experimental cerebral hemorrhage,as well as NPC proliferation and differentiation with time.DESIGN,TIME AND SETTING: A randomized,controlled animal experiment was performed at the Department of Neurobiology,Luzhou Medical College,between January 2007 and October 2008.MATERIALS: Bromodeoxyuridine (BrdU) was purchased from Roche,Germany.Mouse antirat BrdU monoclonal antibody,rabbit anti-nestin polyclonal antibody,rabbit antineuron specific enolase (NSE) polyclonal antibody were purchased from Wuhan Boster,China.Rabbit antiglial fibrillary acidic protein (GFAP) polyclonal antibody was purchased from Sigma,USA.METHODS: Thirty-five adult Sprague Dawley rats were randomly divided into three groups: (1) cerebral hemorrhage group (n = 25),rats were stereotaxically administered 50 μL autologous arterial blood via the dorsal caudate putamen to induce cerebral hemorrhage;(2) sham-surgery group (n = 5),rats underwent surgery but did not receive blood injection;(3) blank control group (n = 5),rats received no surgery and blood administration.At 2 hours after surgery,all rats were intraperitoneally administered BrdU.MAIN OUTCOME MEASURES: Distribution and proliferation of BrdU-positive cells were observed by immunohistochemical staining.BrdU-positive cell differentiation into neurons and glial cells in the peripheral hemorrhagic region was detected by double-label immunofluorescence.RESULTS: Immunohistochemistry results revealed that BrdU-positive cells existed not only in the peripheral hemorrhagic region,such as the subependymal layer and hippocampal dentate gyrus,but also in the lateral septal nucleus,diagonal band,habenular nucleus,and cerebral cortex.Following cerebral hemorrhage,BrdU-positive cells in the peripheral hemorrhagic region gradually in-creased (P < 0.05),and peaked at 7-14 days.Double-label immunofluorescence showed that with time after cerebral hemorrhage,BrdU/nestin-positive cells decreased,but BrdU/GFAPand BrdU/NSE-positive cells increased in the peripheral cerebral hemorrhagic region (P< 0.05).CONCLUSION: Cerebral hemorrhage can induce the proliferation of endogenous NPCs,which peaks at 1-2 weeks after hemorrhage.NPCs can also migrate towards the regions distal to the hemorrhagic focus,such as a diagonal band or lateral septal nucleus.NPCs can gradually differentiate with increasing time after hemorrhage.Chaoxian Yang Ling Zhou Li Deng Xiaoqing Gao 2010Neural Regeneration Research2010,5,5:2
10China's battery electric vehicles lead the world:achievements in technology system architecture and technological breakthroughs显示文摘Developing new energy vehicles has been a worldwide consensus,and developing new energy vehicles characterized by pure electric drive has been China's national strategy.After more than 20 years of high-quality development of China's electric vehicles(EVs),a technological R&D layout of“Three Verticals and Three Horizontals”has been created,and technological advantages have been accumulated.As a result,China's new energy vehicle market has ranked first in the world since 2015.To systematically solve the key problems of battery electric vehicles(BEVs)such as“driving range anxiety,long battery charging time,and driving safety hazards”,China took the lead in putting forward a“system engineering-based technology system architecture for BEVs”and clarifying its connotation.This paper analyzes the research status and progress of the three core components of this architecture,namely,“BEV platform,charging/swapping station,and real-time operation monitoring platform”,and their key technological points.The three major demonstration projects of the 2008 Beijing Olympic Games,the 2022 Beijing Winter Olympics,and the intelligent and connected autonomous battery electric bus project are discussed to specify the applications of BEVs in China.The key research directions for upgrading BEV technologies remain to be further improving the vehicle-level all-climate environmental adaptability and all-day safety of BEVs,systematically solving the charging problem of BEVs and improving their application convenience,and safeguarding safety with early warning and implementing active/passive safety protection for the whole life cycle of power batteries on the basis of BEVs'operation big data.BEVs have acquired new technological features such as intelligent and networked technology empowerment,extensive integration of control-by-wire systems,a platform of chassis hardware,and modularization of functional software.Hongwen He Fengchun Sun Zhenpo Wang Cheng Lin Chengning Zhang Rui Xiong Junjun Deng Xiaoqing Zhu Peng Xie Shuo Zhang Zhongbao Wei Wanke Cao Li Zhai 2022Green Energy and Intelligent Transportation2022,1,1:2

12F0X01 and F0X03a sensitize non-small-cell lung cancer cells to cisplatin-induced apoptosis independent of Bim显示文摘Low sensitivity to chem otherapy has been a m ajor challenge in the treatm ent of non-small-cell lung cancer(NSCLC).It is of great clinical significance to discover its mechanisms to improve cell sensitivity to chem otherapeutic drugs.The forkhead box subfam ily O(FOXO)transcriptional factors are downstream factors of the phosphatidylinositol 3-kinase/protein kinase B(PI3K/AKT)pathway and are reported to play pro-apoptotic roles in a variety of cells including NSCLC cells.But th e ir roles and mechanisms in m ediating cell response to chem otherapy remain to be discovered.We proposed that FOX01 and FOX03a may increase the sensitivity o f NSCLC cells to cisplatin.Moreover,we presumed that LY294002,an inh ib ito r of the PI3K/AKT pathway,may enhance the cytotoxic effects of cisplatin through upregulating FOX01 and FOX03a.In the present study,we found that cisplatin initially increased the expressions and nuclear accum ulation o f FOX01 and FOX03a in NSCLC.Knockdown of FOX01 and FOX03a significantly decreased the cell sensitivity to cisplatin in vitro and in vivo.Moreover,inhibition of F0X01 and FOX03a attenuated cisplatininduced cell apoptosis independent of Bim,a pro-apoptotic protein downstream of the FOXOs.Moreover,LY294002 synergistically increased the cytotoxic effects of cisplatin.Mechanistically,LY294002 increased the expressions and nuclear accumulation of FOX01 and FOX03a.Knockdown of FOXCH and FOX03a abrogated the enhancing effect of LY294002 on cisplatin.Taken together,our results suggested that FOX01 and FOX03a sensitize NSCLC cells to cisplatin and mediate the enhancing effects of LY294002 on cisplatin.Tianyu Sun Jingge Zhang Bo Deng Xiaoqing Fan Tan Long Hua Jin Shaolin Tao Poming Kang Qunyou Tan 2020Acta Biochimica et Biophysica Sinica2020,52,12:2
