2Seasonal and spatial distributions of euphotic zone and longterm variations in water transparency in a clear oligotrophic Lake Fuxian, China显示文摘To assess the seasonal and spatial variations and long-term trends in water optical properties in Lake Fuxian, investigations based on field work in four seasons and a long-term analysis of data from 1980 to 2014 were conducted. The results show that there was no significant variation in the euphotic depth(Z_(eu)) across the four seasons, and no significant correlations between Z_(eu) and potential influencing factors in seasons other than summer, suggesting that the water itself may be a major factor regulating the Z_(eu)in general. Nevertheless, significant differences in Z_(eu) between the north region(NR) and the south region(SR) were observed in all seasonal tests except spring. This finding relates to a higher abundance of chromophoric dissolved organic matter(CDOM) in the NR due to runoff, especially in the rainy seasons(summer and autumn).CDOM and its terrigenous component had an important impact on Z_(eu)in summer, with the highest precipitation, and impacts from suspended solids and non-algal particles were also found in the NR in summer. The Secchi disk depth in the lake decreased clearly over the years,with significantly negative correlations with the increasing permanganate index and air temperature, implying that organic contaminants(CDOM and/or phytoplankton) are important regulators of water transparency. We estimate that the combined effects of climate warming and changes in land use and land cover are also indirect regulating factors. 3Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. 4Perilla oil and exercise decrease expressions of tumor necrosis factor-α,plasminogen activator inhibitor-1 and highly sensitive C-reactive protein in patients with hyperlipidemia显示文摘OBJECTIVE:To verify the effects of perilla oil on the regulation of blood lipid levels in patients with hyperlipidemia.METHODS:Blood was taken from patients prior to and 8 weeks following treatment with perilla oil.Different ways to test for indexes which correlate to hyperlipidemia were performed.Some indexes,which correlate with inflammation and injury to endothelial cells,were tested using enzyme linked immunosorbent assays.RESULTS:Serum lipid levels [triglyceride(TG),total cholesterol(TC),and low-density lipoprotein-cholesterol(LDL-C)] changed significantly after 56 days of treatment.Differences were noted as early as 28 days after treatment began(P<0.05).Treatment with perilla oil showed statistically significant recovery levels of high-density lipoprotein-cholesterol(HDL-C) after 28 and 56 days of treatment.Plasma lipids levels were significantly lower after 56 days of treatment(P<0.05).Perilla oil reduced blood lipid levels in patients,and the regulation of cell signaling factor levels had no adverse effects on patients' liver or kidney function,or blood routine examinations.CONCLUSION:Perilla oil treatment is safe in clinical use,can regulate blood lipid levels and protects the function of endothelial cells.Minggang Wei Peihua Xiong Ling Zhang Mei Fei Aiping Chen Fengling Li 2013Journal of Traditional Chinese Medicine2013,33,2:10
5Combined genomic, transcriptomic, and metabolomic an alyses provide in sights into chayote (Sechium edule) evolution and fruit development显示文摘Chayote(Sechium edule)is an agricultural crop in the Cucurbitaceae family that is rich in bioactive components.To enhance genetic research on chayote,we used Nanopore third-generation sequencing combined with Hi-C data to assemble a draft chayote genome.A chromosome-level assembly anchored on 14 chromosomes(N50 contig and scaffold sizes of 8.40 and 46.56 Mb,respectively)estimated the genome size as 606.42 Mb,which is large for the Cucurbitaceae,with 65.94%(401.08 Mb)ofthe genome comprising repetitive sequences;28,237 protein-coding genes were predicted.Comparative genome analysis indicated that chayote and snake gourd diverged from sponge gourd and that a whole-genome duplication(WGD)event occurred in chayote at 25±4 Mya.Transcriptional and metabolic analysis revealed genes involved in fruit texture,pigment,fl avor,fl avonoids,antioxidants,and plant hormones during chayote fruit development.The analysis of the genome,transcriptome,and metabolome provides insights into chayote evolution and lays the groundwork for future research on fruit and tuber development and genetic improvements in chayote.Anzhen Fu Qing Wang Jianlou Mu Lili Ma Changlong Wen Xiaoyan Zhao Lipu Gao Jian Li Kai Shi Yunxiang Wang Xuechuan Zhang Xuewen Zhang Fengling Wang Donald Grierson Jinhua Zuo 2021Horticulture Research2021,8,1:6
6A flexible skin-mounted wireless acoustic device for bowel sounds monitoring and evaluation显示文摘Conventional methods of intestinal inspection play an essential role in the assessment of bowel diseases and other relevant health issues, yet fail to obtain intestinal conditions in real time because of radiation limits and operation inconvenience. Herein, a flexible, skin-mounted device is developed for longterm, real-time monitoring, and for the evaluation of bowel sounds based on the integration of a threedimensional printed elastomeric resonator with flexible electronics. The device is capable of being flexibly attached to abdominal surfaces without performance degradation during breathing. Clinical tests conduct in a normal volunteer and in patients with mechanical intestinal obstruction or paralytic ileus illustrate the usefulness of the device in capturing the characteristics of bowel sounds. 7Combined peripheral natural killer cell and circulating tumor cell enumeration enhance prognostic efficiency in patients with metastatic triple-negative breast cancer显示文摘Objective:Triple-negative breast cancer(TNBC)is a heterogeneous disease with poor prognosis.Circulating tumor cells(CTCs)are a promising predictor for breast cancer prognoses but their reliability regarding progression-free survival(PFS)is controversial.We aim to verify their predictive value in TNBC.Methods:In present prospective cohort study,we used the Pep@MNPs method to enumerate CTCs in baseline blood samples from 75 patients with TNBC(taken at inclusion in this study)and analyzed correlations between CTC numbers and outcomes and other clinical parameters.Results:Median PFS was 6.0(range:1.0–25.0)months for the entire cohort,in whom we found no correlations between baseline CTC status and initial tumor stage(P=0.167),tumor grade(P=0.783)or histological type(P=0.084).However,among those getting first-line treatment,baseline CTC status was positively correlated with ratio of peripheral natural killer(NK)cells(P=0.032),presence of lung metastasis(P=0.034)and number of visceral metastatic site(P=0.037).Baseline CTC status was predictive for PFS in first-line TNBC(P=0.033),but not for the cohort as a whole(P=0.118).This prognostic limitation of CTC could be ameliorated by combining CTC and NK cell enumeration(P=0.049).Conclusions:Baseline CTC status was predictive of lung metastasis,peripheral NK cell ratio and PFS in TNBC patients undergoing first-line treatment.We have developed a combined CTC-NK enumeration strategy that allows us to predict PFS in TNBC without any preconditions.Xiaoran Liu Ran Ran Bin Shao Hope S.Rugo Yanlian Yang Zhiyuan Hu Zewen Wei Fengling Wan Weiyao Kong Guohong Song Hanfang Jiang Xu Liang Ruyan Zhang Ying Yan Guobing Xu Huiping Li 2018Chinese Journal of Cancer Research2018,30,3:4
8Qiguiyishen decoction reduced the accumulation of extracellular matrix in the kidneys of rats with adriamycin-induced nephropathy显示文摘OBJECTIVE: To investigate the effect of Qiguiyishen decoction(QGYS) on the severity of nephropathy.METHODS: Twenty-four rats were randomly divided into four groups(Ⅰ, Ⅱ, Ⅲ, Ⅳ) according to the random number table. Group Ⅰ as control group did not establish nephropathy model. Groups Ⅱ,Ⅲ, and Ⅳ were intravenously administered Adriamycin(7.5 mg/kg) through the tail vein to establish nephropathy model. QGYS was prepared with the extracts of Huangqi(Radix Astragali Mongolici),Danggui(Radix Angelicae Sinensis),Niuxi(RadixAchyran-this Bidentatae), and Chuanxiong(Rhizoma Chuanxiong).Group Ⅳ was administered QGYS (2mL·kg-1·d-1), group Ⅲ was administered benazepril(10 mL·kg-1·d-1), and group Ⅰ, Ⅱ was administered water(2mL·kg-1·d-1)once daily for eight weeks.RESULTS: QGYS reduced the excretion of urinary protein and N-acetyl-β-D-glucosaminidase and alleviated the accumulation of extracellular matrix(ECM) in renal tissue. 9Genome-wide identification of agronomically important genes in outcrossing crops using OutcrossSeq显示文摘Many important crops(e.g.,tuber,root,and tree crops)are cross-pollinating.For these crops,no inbred lines are available for genetic study and breeding because they are self-incompatible,clonally propagated,or have a long generation time,making the identification of agronomically important genes difficult,particularly in crops with a complex autopolyploid genome.In this study,we developed a method,OutcrossSeq,for mapping agronomically important loci in outcrossing crops based on whole-genome low-coverage resequencing of a large genetic population,and designed three computation algorithms in OutcrossSeq for different types of outcrossing populations.We applied OutcrossSeq to a tuberous root crop(sweet potato,autopolyploid),a tree crop(walnut tree,highly heterozygous diploid),and hybrid crops(double-cross populations)to generate high-density genotype maps for the outcrossing populations,which enable precise identification of genomic loci underlying important agronomic traits.Candidate causative genes at these loci were detected based on functional clues.Taken together,our results indicate that OutcrossSeq is a robust and powerful method for identifying agronomically important genes in heterozygous species,including polyploids,in a cost-efficient way.The OutcrossSeq software and its instruction manual are available for downloading at www.xhhuanglab.cn/tool/OutcrossSeq.html.Mengjiao Chen Weijuan Fan Feiyang Ji Hua Hua Jie Liu Mengxiao Yan Qingguo Ma Jiongjiong Fan Qin Wang Shufeng Zhang Guiling Liu Zhe Sun Changgeng Tian Fengling Zhao Jianli Zheng Qi Zhang Jiaxin Chen Jie Qiu Xin Wei Ziru Chen Peng Zhang Dong Pei Jun Yang Xuehui Huang 2021Molecular Plant2021,14,4:4

11Ecosystem health assessment of urban rivers and lakes-Case study of “the six lakes”in Beijing,China显示文摘The assessment of the ecosystem health of urban rivers and lakes is the scientific basis for their management and ecological restoration.This study developed a three-level indicator system for its assessment.The results indicated that:Zhonghai and Nanhai are in the state of transition from unhealthy to critical state and all the other lakes are in unhealthy states.Water environmental quality,structure and function of the aquatic ecosystem,and the structure of waterfront areas were the constraints.Nanhai was ranked as poor and the others were all ranked as very poor.However,the ecological environment of Zhonghai and Nanhai were better than the others,the sums of the degree of membership to the healthy state and critical state were all close to 0.6.and the restorations of these lakes were moderate.The sums of the degree of membership to the healthy state and critical state of the other lakes were under 0.3,as it was difficult to restore these lakes.Some suggestions on scientific management and ecological restoration of the six lakes were proposed:①To control non-point pollution and to improve the water quality of six lakes and the water entering into these lakes;②To improve the hydrological conditions of six lakes;③To rehabilitate the aquatic ecosystem and waterfront areas.Fengling ZHANG Jingling LIU Zhifeng YANG Yongli LI 2008Frontiers of Environmental Science & Engineering2008,2,2:3
12Serum ficolin-2 concentrations are significantly changed in patients with hepatitis B virus infection and liver diseases显示文摘Human ficolin-2 is an important lectin complement pathway activator that is secreted from liver cells and has been implicated as an anti-infection innate immune molecule. However, the role of ficolin-2 protein and its dynamic changes over the course of and in the prognosis of chronic hepatitis B(CHB) and hepatocellular carcinoma(HCC) remain unclear. In this study, we analyzed ficolin-2 protein expression in a cohort of individuals with CHB infection, HCC and cirrhosis. A sandwich enzyme-linked immunosorbent assay(ELISA) method was used to measure serum ficolin-2 concentrations. Ficolin-2 expression in liver tissues was detected by immunohistochemical staining. Serum ficolin-2 concentrations in CHB patients were significantly higher than in healthy controls and HBV carriers. After 48 weeks of routine amelioration liver function treatment, serum ficolin-2 concentrations decreased and were positively correlated with favorable alanine aminotransferase(ALT), HBV DNA and HBe Ag-seroconversion outcomes. Interestingly, we observed much lower expression of serum and intrahepatic ficolin-2 in HCC and cirrhosis compared with healthy controls. 13Numerical simulation on optimization of structure and operating parameters of a novel lean coal decoupling burner显示文摘Due to its low volatile characteristics of lean coal,it is difficult to catch fire and burn out.Therefore,high temperature is needed to maintain combustion efficiency,while,this leads to high nitrogen oxide emission.For power plant boilers burning lean coal,stable combustion with lower nitrogen oxide emission is a challenging task.This study applied the 3D numerical simulation on the analysis of a novel de-coupling burner for low-volatile coal and its structure and operation parameters optimization.Results indicate that although it was more difficult for lean coal decoupling burner to ignite lean coal than high volatile coal,the burner formed a stepwise ignition trend,which promoted the rapid ignition of lean coal.Comparison of three central partition plate structure shows that in terms of characteristics of the flow field distribution,rich and lean separation and combustion,the structure with an inclination of 0°showed good performance,with its rich-lean air ratio being 0.85 and concentration ratio being 22.94,and there was an apparent decoupling combustion characteristic.Finally,the structure of the selected burner was optimized for its operational conditions.The optimal operating parameters was determined as the primary air velocity of 24.9 m·s^-1 and the mass flow rate of pulverized coal of 2.5 kg·s^-1,in which the pyrolysis products were utilized as reductive agent more fully.Eventually,the nitrogen oxide was efficiently reduced to nitrogen,which emission concentration was 61.88%lower than that in the design condition.Jing Wang Ruiping Zhang Fengling Yang Fangqin Cheng 2020Chinese Journal of Chemical Engineering2020,28,11:2
14Ectopic expression of antifreeze protein gene from Ammopiptanthus nanus confers chilling tolerance in maize显示文摘Improved chilling tolerance is important for maize production. Previous efforts in transgenics and marker-assisted breeding have not achieved practical results. In this study, the antifreeze protein(AnAFP) from the super-xerophyte Ammopiptanthus nanus was aligned to KnS-type dehydrins.Phosphorylation in vitro and subcellular localization showed that AnAFP was phosphorylated by maize casein kinase II and translocated from nucleus to cytoplasm under chilling stress. AnAFP also increased lactate dehydrogenase activity. A parent line of commercial maize hybrids was transformed with the AnAFP gene. Based on thermal asymmetric interlaced PCR, one hemizygous and two homozygous integration sites were identified in one T_(1) line. Ectopic expression of AnAFP in transgenic lines was confirmed by quantitative real-time PCR, RNA-seq, and Western blotting. After chilling treatment, the transgenic lines showed significantly improved phenotype, higher kernel production, survival rate and biomass, and lower relative electrolyte leakage and superoxide dismutation than the untransformed line. 15Two cases of chronic myelomonocytic leukemia combined with monoclonal gammopathy of undetermined significance and a literature review显示文摘To describe myelodysplastic syndrome(MDS)/myeloproliferative neoplasm(MPN) combined with monoclonal gammopathy of undetermined significance(MGUS) in order to investigate the potential association between these 2 diseases. Two cases of confirmed chronic myelomonocytic leukemia(CMML) combined with MGUS were reported. In addition, prior publications of cases with combined MDS or MPN with MGUS were reviewed. The first case was of a 77-year-old man whose routine blood tests showed abnormal hemogram results. The diagnosis was CMML combined with Ig M monoclonal gammopathy, and the disease course was 4 years. The CMML gradually progressed and the patient presented with anemia, thrombocytopenia, autoimmune hemolysis, and an increase in the number of immature cells in the bone marrow. Although the MGUS caused fluctuations in the concentrations of Ig M, no Ig M-associated organ damage was observed. Eventually, this patient died from a lung infection. The second case was of a 78-year-old man who sought treatment because of fever and a cough. An increase in the number of monocytes was discovered in the peripheral blood. Bone marrow smear results suggested obvious active granulocytes and an increase in the percentages of promyelocytes, myelocytes, and metamyelocytes. Unhealthy granulocytes and immature monocytes could also be observed, and the percentage of monocytes was increased. In addition, serum Ig G levels were increased, and immunofixation electrophoresis results showed Ig G-κ type M proteins. The diagnosis was CMML combined with Ig G monoclonal gammopathy. These diseases were stable and follow-up was conducted for 1 year after diagnosis. The cases in this study combined with those that were reviewed in the relevant literature indicate that the presence of these 2 diseases in the same patient might not be a coincidence. The development of the 2 diseases in case 1 was different, and we speculate that they might have had different clonal origins.
