| 1 | Multigene editing via CRISPR/Cas9 guided by a single-sgRNA seed in Arabidopsis显示文摘We report that a solo single-guide RNA(sg RNA) seed is capable of guiding Clustered Regularly Interspaced Short Palindromic Repeats(CRISPR)/CRISPRàassociated 9(CRISRP/Cas_9) to simultaneously edit multiple genes At RPL_(10)A, At RPL_(10)B and At RPL_(10)C in Arabidopsis. Our results also demonstrate that it is possible to use CRISPR/Cas_9 technology to create At RPL_(10) triple mutants which otherwise cannot be generated by conventional genetic crossing. Compared to other conventional multiplex CRISPR/Cas systems, a single sg RNA seed has the advantage of reducing off-target gene-editing. | 4 | Safety of SARS-CoV-2 vaccines:a systematic review and meta-analysis of randomized controlled trials显示文摘Background Various modalities of vaccines against coronavirus disease 2019(COVID-19),based on different platforms and immunization procedures,have been successively approved for marketing worldwide.A comprehensive review for clinical trials assessing the safety of COVID-19 vaccines is urgently needed to make an accurate judgment for mass vaccination.Main text A systematic review and meta-analysis was conducted to determine the safety of COVID-19 vaccine candidates in randomized controlled trials(RCTs).Data search was performed in PubMed,Embase,Cochrane library,Scopus,Web of Science,and MedRxiv.Included articles were limited to RCTs on COVID-19 vaccines.A total of 73,633 subjects from 14 articles were included to compare the risks of adverse events following immunization(AEFI)after vaccinating different COVID-19 vaccines.Pooled risk ratios(RR)of total AEFI for inactivated vaccine,viral-vectored vaccine,and mRNA vaccine were 1.34[95%confidence interval(CI)1.11–1.61,P<0.001],1.65(95%CI 1.31–2.07,P<0.001),and 2.01(95%CI 1.78–2.26,P<0.001),respectively.No significant differences on local and systemic AEFI were found between the first dose and second dose.In addition,people aged≤55 years were at significantly higher risk of AEFI than people aged≥56 years,with a pooled RR of 1.25(95%CI 1.15–1.35,P<0.001).Conclusions The safety and tolerance of current COVID-19 vaccine candidates are acceptable for mass vaccination,with inactivated COVID-19 vaccines candidates having the lowest reported AEFI.Long-term surveillance of vaccine safety is required,especially among elderly people with underlying medical conditions. | Musha Chen Yue Yuan Yiguo Zhou Zhaomin Deng Jin Zhao Fengling Feng Huachun Zou Caijun Sun | 2021 | Infectious Diseases of Poverty2021,10,4: | 2 |

| 7 | Bioactive glass promotes the barrier functional behaviors of keratinocytes and improves the Re-epithelialization in wound healing in diabetic rats显示文摘Upon skin injury,re-epithelialization must be triggered promptly to restore the integrity and barrier function of the epidermis.However,this process is often delayed or interrupted in chronic wounds like diabetic foot ulcers.Considering that BG particles can activate multiple genes in various cells,herein,we hypothesized that bioactive glass(BG)might be able to modulate the barrier functional behaviors of keratinocytes.By measuring the transepithelial electrical resistance(TEER)and the paracellular tracer flux,we found the 58S-BG extracts substantially enhanced the barrier function of keratinocyte monolayers.The BG extracts might exert such effects by promoting the keratinocyte differentiation and the formation of tight junctions,as evidenced by the increased expression of critical differentiation markers(K10 and involucrin)and TJ protein claudin-1,as well as the altered subcellular location of four major TJ proteins(claudin-1,occludin,JAM-A,and ZO-1).Besides,the cell scratch assay showed that BG extracts induced the collective migration of keratinocytes,though they did not accelerate the migration rate compared to the control.The in vivo study using a diabetic rat wound model demonstrated that the BG extracts accelerated the process of re-epithelialization,stimulated keratinocyte differentiation,and promoted the formation of tight junctions in the newly regenerated epidermis.Our findings revealed the crucial effects of BGs on keratinocytes and highlighted its potential application for chronic wound healing by restoring the barrier function of the wounded skin effectively. | Fengling Tang Junliang Li Weihan Xie Yunfei Mo Lu Ouyang Fujian Zhao Xiaoling Fu Xiaofeng Chen | 2021 | Bioactive Materials2021,6,10: | 1 |
| 8 | Low-coordinated surface sites make truncated Pd tetrahedrons as robust ORR electrocatalysts outperforming Pt for DMFC devices显示文摘Developing highly stable and active non-Pt oxygen reduction reaction(ORR)electrocatalysts for power generation device raises great concerns and remains a challenge.Here,we report novel truncated Pd tetrahedrons(T-Pd-Ths)enclosed by{111}facets with excellent uniformity,which have both low-coordinated surface sites and distinct lattice distortions that would induce“local strain”.In alkaline electrolyte,the T-Pd-Ths/C achieves remarkable ORR specific/mass activity(SA/MA)of 2.46 mA·cm^(−2)/1.69 A·mgPd^(−1),which is 12.3/16.9 and 10.7/14.1 times higher than commercial Pd/C and Pt/C,respectively.The T-Pd-Ths/C also exhibits high in-situ carbon monoxide(CO)tolerance and 50,000 cycles durability with an activity loss of 7.69%and morphological stability.The rotating ring-disk electrode(RRDE)measurements show that a 4-electron process occurs on T-PdThs/C.Theoretical calculations demonstrate that the low-coordinated surface sites contribute largely to the enhancement of ORR activity.In actual direct methanol fuel cell(DMFC)device,the T-Pd-Ths/C delivers superior open-circuit voltage(OCV)and peak power density(PPD)to commercial Pt/C from 25 to 80℃,and the maximum PPD can reach up to 163.7 mW·cm−2.This study demonstrates that the T-Pd-Ths/C holds promise as alternatives to Pt for ORR in DMFC device. | Xiaoling Wang Jingwei Li Xiaotong Yang Fengling Zhao Yongfei Li Daliang Zhang Li-Yong Gan Ke Xin Yao Qiang Yuan | 2022 | Nano Research2022,15,9: | 1 |
| 9 | Bioceramic scaffolds with triply periodic minimal surface architectures guide early-stage bone regeneration显示文摘The pore architecture of porous scaffolds is a critical factor in osteogenesis,but it is a challenge to precisely configure strut-based scaffolds because of the inevitable filament corner and pore geometry deformation.This study provides a pore architecture tailoring strategy in which a series of Mg-doped wollastonite scaffolds with fully interconnected pore networks and curved pore architectures called triply periodic minimal surfaces(TPMS),which are similar to cancellous bone,are fabricated by a digital light processing technique.The sheet-TPMS pore geometries(s-Diamond,s-Gyroid)contribute to a 3‒4-fold higher initial compressive strength and 20%-40%faster Mg-ion-release rate compared to the other-TPMS scaffolds,including Diamond,Gyroid,and the Schoen’s I-graph-Wrapped Package(IWP)in vitro.However,we found that Gyroid and Diamond pore scaffolds can significantly induce osteogenic differentiation of bone marrow mesenchymal stem cells(BMSCs).Analyses of rabbit experiments in vivo show that the regeneration of bone tissue in the sheet-TPMS pore geometry is delayed;on the other hand,Diamond and Gyroid pore scaffolds show notable neo-bone tissue in the center pore regions during the early stages(3-5 weeks)and the bone tissue uniformly fills the whole porous network after 7 weeks.Collectively,the design methods in this study provide an important perspective for optimizing the pore architecture design of bioceramic scaffolds to accelerate the rate of osteogenesis and promote the clinical translation of bioceramic scaffolds in the repair of bone defects. | Miaoda Shen Yifan Li Fengling Lu Yahui Gou Cheng Zhong Shukun He Chenchen Zhao Guojing Yang Lei Zhang Xianyan Yang Zhongru Gou Sanzhong Xu | 2023 | Bioactive Materials2023,,7: | 1 |
| 10 | Ficolin-2 binds to HIV-1 gp120 and blocks viral infection显示文摘Ficolin-2 is a lectin complement pathway activator present in normal human plasma and usually associated with infectious diseases, but little is known about the role of ficolin-2 in human immunodeficiency virus(HIV) infection. Here, we describe our novel findings that serum ficolin-2concentrations of 103 HIV-1 patients were much higher compared to those of 57 healthy donors. In vitro analysis showed that HIV-1 infection could enhance ficolin-2 expression. We further demonstrated that recombinant ficolin-2 protein could bind with HIV-1 envelope glycoprotein gp120, and subsequently induce complement dependent cytotoxicity. Moreover, ficolin-2 could block the entry of HIV-1 into target cells(TZM-b1 and MT-2 cells) and infection in a ficolin-2 dosedependent manner. | 12 | Interface-rich Au-doped PdBi alloy nanochains as multifunctional oxygen reduction catalysts boost the power density and durability of a direct methanol fuel cell device显示文摘The development of cathode oxygen reduction reaction(ORR)catalysts with high characteristics for practical,direct methanol fuel cells(DMFCs)has continuously increased the attention of researchers.In this work,interface-rich Au-doped PdBi(PdBiAu)branched one-dimensional(1D)alloyed nanochains assembled by sub-6.5 nm particles have been prepared,exhibiting an ORR mass activity(MA)of 6.40 A·mgPd^(−1) and long-term durability of 5,000 cycles in an alkaline medium.The MA of PdBiAu nanochains is 46 times and 80 times higher than that of commercial Pt/C(0.14 A·mgPt^(−1))and Pd/C(0.08 A·mgPd^(−1)).The MA of binary PdBi nanochains also reaches 5.71 A·mgPd^(−1).Notably,the PdBiAu nanochains exhibit high in-situ carbon monoxide poisoning resistance and high methanol tolerance.In actual DMFC device tests,the PdBiAu nanochains enhance power density of 140.1 mW·cm^(−2)(in O_(2))/112.4 mW·cm^(−2)(in air)and durability compared with PdBi nanochains and Pt/C.The analysis of the structure–function relationship indicates that the enhanced performance of PdBiAu nanochains is attributed to integrated functions of surficial defect-rich 1D chain structure,improved charge transfer capability,downshift of the d-band center of Pd,as well as the synergistic effect derived from“Pd-Bi”and/or“Pd-Au”dual active sites. | Xin Li Ke Xin Yao Fengling Zhao Xiaotong Yang Jingwei Li Yongfei Li Qiang Yuan | 2022 | Nano Research2022,15,7: | 0 |
| 13 | GRK2 inhibits Flt-1^(+)macrophage infiltration and its proangiogenic properties in rheumatoid arthritis显示文摘Rheumatoid arthritis(RA)is an autoimmune disease with a complex etiology.Monocyte-derived macrophages(MDMs)infiltration are associated with RA severity.We have reported the deletion of G-protein-coupled receptor kinase 2(GRK2)reprograms macrophages toward an anti-inflammatory phenotype by recovering G-protein-coupled receptor signaling.However,as more GRK2-interacting proteins were discovered,the GRK2 interactome mechanisms in RA have been understudied.Thus,in the collagen-induced arthritis mouse model,we performed genetic GRK2 deletion using GRK2^(f/f)Lyz2-Cre^(+/−)mice.Synovial inflammation and M1 polarization were improved in GRK2^(f/f)Lyz2-Cre^(+/−)mice.Supporting experiments with RNA-seq and dual-luciferase reporter assays identified peroxisome proliferator-activated receptorγ(PPARγ)as a new GRK2-interacting protein.We further confirmed that fms-related tyrosine kinase 1(Flt-1),which promoted macrophage migration to induce angiogenesis,was inhibited by GRK2-PPARγsignaling.Mechanistically,excess GRK2 membrane recruitment in CIA MDMs reduced the activation of PPARγligand-binding domain and enhanced Flt-1 transcription.Furthermore,the treatment of mice with GRK2 activity inhibitor resulted in significantly diminished CIA pathology,Flt-1^(+)macrophages induced-synovial inflammation,and angiogenesis.Altogether,we anticipate to facilitate the elucidation of previously unappreciated details of GRK2-specific intracellular signaling.Targeting GRK2 activity is a viable strategy to inhibit MDMs infiltration,affording a distinct way to control joint inflammation and angiogenesis of RA. | Xuezhi Yang Yingjie Zhao Qi Wei Xuemin Zhu Luping Wang Wankang Zhang Xiaoyi Liu Jiajie Kuai Fengling Wang Wei Wei | 2024 | Acta Pharmaceutica Sinica B2024,14,1: | 0 |
| 14 | PtCu_(3) nanoalloy@porous PWO_(x) composites with oxygen container function as efficient ORR electrocatalysts advance the power density of room-temperature hydrogen-air fuel cells显示文摘It is challenging and desirable to construct Pt-based nanocomposites with oxygen storage function as efficient oxygen reduction reaction(ORR)catalysts for practical proton exchange membrane fuel cells(PEMFCs).Herein,we achieve novel porous nanocomposites of PtCu_(3) nanoalloys-embedded in the PWO_(x) matrix(PtCu_(3)@PWO_(x)),which has an oxygen container feature.The PtCu_(3)@PWO_(x)/C exhibits an ultrahigh mass activity(MA)of 3.94 A·mgPt−1 for ORR,which is 26.3 times as high as the commercial Pt/C and the highest value ever reported for PtCu-based binary system.Theoretical calculations reveal that the compressive strain and d-band center downshift of Pt intrinsically contribute to the excellent ORR performance.In H_(2)-air PEMFCs at room temperature,furthermore,the PtCu_(3)@PWO_(x)/C delivers a high power density(218.6 mW·cm^(−2)),much superior to commercial Pt/C(131.6 mW·cm^(−2)).In H_(2)-O_(2) PEMFCs,PtCu_(3)@PWO_(x)/C outputs a maximum power density of 420.1 mW·cm^(−2).This work provides an effective idea for designing oxygen-storing ORR catalysts used for practical room-temperature H_(2)-air fuel cells. | Rui Chen Tie Shu Fengling Zhao Yongfei Li Xiaotong Yang Jingwei Li Daliang Zhang Li-Yong Gan Ke Xin Yao Qiang Yuan | 2022 | Nano Research2022,15,10: | 0 |
| 15 | Follow-up of two victims of a 60Co-source radiation accident in 1986, Kaifeng, Henan Province, China显示文摘Objective:To explore the late effects of acute radiation exposure through the follow-up of two victims of a 60Cosource radiation accident,who suffered moderate acute radiation sickness of bone marrow type.Methods:On May 26,1986,a power failure occurred suddenly at an irradiation plant in Kaifeng,Henan Province,China,during which two workers at the site,named Liang and Yan,accidentally entered the 60Co-source irradiation cell in service.Long-term medical observations were carried out on them following the accident.Results:The late effects of the accident on Liang and Yan were observed.The main manifestations of Liang were hypomnesia and visual impairment.A typical radiation-induced cataract developed 2 years after exposure.Local cutaneous radiation injury and resultant five operations were performed within a period of five years after exposure.The decrease in the number and low viability of his sperms was found 1 year after exposure.The decrease in his leukocytes and platelets occurred during 1–5 years after exposure.He died from intrahepatic cholangiocarcinoma 21 years after exposure.For Yan,the main manifestations included headache,dizziness,fatigue,insomnia,and susceptibility to cold just after exposure.She had intermittent menolipsis at age 41.The leukocytes was fluctuating in the range of(3.1–3.9)109/L 14–24 years after exposure.A decrease in estradiol(reproductive hormone)and an increase in follicle-stimulating hormone were observed.Thyroid nodules were detected by thyroid ultrasound 21 years after exposure.She died from acute myeloid leukemia(AML)32 years after exposure.Conclusions:Follow-up of victims exposed to a high activity radiation source should focus on the organs sensitive to radiation and on the radiation-induced carcinogenesis. | Fengling Zhao Jifeng Yang Yumin Lyu Quanfu Sun Baohua Fu | 2020 | Radiation Medicine and Protection2020,1,4: | 0 |

| 17 | Investigation on step overcharge to self-heating behavior and mechanism analysis of lithium ion batteries显示文摘To obtain intrinsic overcharge boundary and investigate overcharge mechanism,here we propose an innovative method,the step overcharge test,to reduce the thermal crossover and distinguish the overcharge thermal behavior,including 5%state of charge(SOC)with small current overcharge and resting until the temperature equilibrium under adiabatic conditions.The intrinsic thermal response and the self-excitation behaviour are analysed through temperature and voltage changes during the step overcharge period.Experimental results show that the deintercalated state of the cathode is highly correlated to self-heating parasitic reactions.Before reaching the upper limit of Negative/Positive(N/P)ratio,the temperature changes little,the heat generation is significantly induced by the reversible heat(endothermic)and ohmic heat,which could balance each other.Following that the lithium metal is gradually deposited on the surface of the anode and reacts with electrolyte upon overcharge,inducing selfheating side reaction.However,this spontaneous thermal reaction could be“self-extinguished”.When the lithium in cathode is completely deintercalated,the boundary point of overcharge is about 4.7 V(~148%SOC,>40℃),and from this point,the self-heating behaviour could be continuously triggered until thermal runaway(TR)without additional overcharge.The whole static and spontaneous process lasts for 115 h and the side reaction heat is beyond 320,000 J.The continuous self-excitation behavior inside the battery is attributed to the interaction between the highly oxidized cathode and the solvent,which leads to the dissolution of metal ions.The dissolved metal ions destroy the SEI(solid electrolyte interphase)film on the surface of the deposited Li of anode,which induces the thermal reaction between lithium metal and the solvent.The interaction between cathode,the deposited Li of anode,and solvent promotes the temperature of the battery to rise slowly.When the temperature of the battery reaches more than 60℃,the reaction between lithium metal and solvent is accelerated.After the temperature rises rapidly to the melting point of the separator,it triggers the thermal runaway of the battery due to the short circuit of the battery. | Fengling Yun Shiyang Liu Min Gao Xuanxuan Bi Weijia Zhao Zenghua Chang Minjuan Yuan Jingjing Li Xueling Shen Xiaopeng Qi Ling Tang Yi Cui Yanyan Fang Lihao Guo Shangqian Zhao Xiangjun Zhang Shigang Lu | 2023 | Journal of Energy Chemistry2023,,4: | 0 |
| 18 | Single-cell landscape analysis reveals distinct regression trajectories and novel prognostic biomarkers in primary neuroblastoma显示文摘Neuroblastoma(NB),which is the most common pediatric extracranial solid tumor,varies widely in its clinical presentation and outcome.NB has a unique ability to spontaneously differentiate and regress,suggesting a potential direction for therapeutic intervention.However,the underlying mechanisms of regression remain largely unknown,and more reliable prognostic biomarkers are needed for predicting trajectories for NB.We performed scRNAseq analysis on 17 NB clinical samples and three peritumoral adrenal tissues.Primary NB displayed varied cell constitution,even among tumors of the same pathological subtype.Copy number variation patterns suggested that neuroendocrine cells represent the malignant cell type.Based on the differential expression of sets of related marker genes,a subgroup of neuroendocrine cells was identified and projected to differentiate into a subcluster of benign fibroblasts with highly expressed CCL2 and ZFP36,supporting a progressive pathway of spontaneous NB regression.We also identified prognostic markers(STMN2,TUBA1A,PAGE5,and ETV1)by evaluating intra-tumoral heterogeneity.Lastly,we determined that ITGB1 in M2-like macrophages was associated with favorable prognosis and may serve as a potential diagnostic marker and therapeutic target.In conclusion,our findings reveal novel mechanisms underlying regression and potential prognostic markers and therapeutic targets of NB. | Qingqing Liu Zhenni Wang Yan Jiang Fengling Shao Yue Ma Mingzhao Zhu Qing Luo Yang Bi Lijian Cao Liang Peng Jianwu Zhou Zhenzhen Zhao Xiaobin Deng Tong-Chuan He Shan Wang | 2022 | Genes & Diseases2022,9,6: | 0 |

| 20 | PtCu subnanoclusters epitaxial on octahedral PtCu/Pt skin matrix as ultrahigh stable cathode electrocatalysts for room-temperature hydrogen fuel cells显示文摘Achieving stable surface structures of metal catalysts is an extreme challenge for obtaining long-term durability and meeting industrial application requirements.We report a new class of metal catalyst,Pt-rich PtCu heteroatom subnanoclusters epitaxially grown on an octahedral PtCu alloy/Pt skin matrix(PtCu1.60),for the oxygen reduction reaction(ORR)in an acid electrolyte.The PtCu1.60/C exhibits an 8.9-fold enhanced mass activity(1.42 A·mgPt^(−1))over that of commercial Pt/C(0.16 A·mgPt^(−1)).The PtCu1.60/C exhibits 140,000 cycles durability without activity decline and surface PtCu cluster stability owing to unique structure derived from the matrix and epitaxial growth pattern,which effectively prevents the agglomeration of clusters and loss of near-surface active sites.Structure characterization and theoretical calculations confirm that Pt-rich PtCu clusters favor ORR activity and thermodynamic stability.In room-temperature polymer electrolyte membrane fuel cells,the PtCu1.60/C shows enhanced performance and delivers a power density of 154.1/318.8 mW·cm^(−2)and 100 h/50 h durability without current density decay in an air/O_(2)feedstock. | Fengling Zhao Lirong Zheng Qiang Yuan Qinghua Zhang Tian Sheng Xiaotong Yang Lin Gu Xun Wang | 2023 | Nano Research2023,16,2: | 0 |
