1Biosynthesis of Biocompatible Cadmium Telluride Quantum Dots Using Yeast Cells显示文摘We demonstrate a simple and efficient biosynthesis method to prepare easily harvested biocompatible cadmium telluride(CdTe)quantum dots(QDs)with tunable fluorescence emission using yeast cells.Ultraviolet-visible(UV-vis)spectroscopy,photoluminescence(PL)spectroscopy,X-ray diffraction(XRD),and transmission electron microscopy(TEM)confirm that the CdTe QDs are formed via an extracellular growth and subsequent endocytosis pathway and have size-tunable optical properties with fluorescence emission from 490 to 560 nm and a cubic zinc blende structure with good crystallinity.In particular,the CdTe QDs with uniform size(2-3.6 nm)are protein-capped,which makes them highly soluble in water,and in situ bio-imaging in yeast cells indicates that the biosynthesized QDs have good biocompatibility.This work provides an economic and environmentally friendly approach to synthesize highly fluorescent biocompatible CdTe QDs for bio-imaging and bio-labeling applications.Haifeng Bao Na Hao Yunxia Yang Dongyuan Zhao 2010Nano Research2010,3,7:11
2Rapid genomic changes in polyploid wheat and related species:implications for genome evolution and genetic improvement显示文摘A polyploid organism by possessing more than two sets of chromosomes from one species (autopolyploidy) or two or more species (allopolyploidy) is known to have evolutionary advantages. However, by what means a polyploid accommodates increased genetic dosage or divergent genomes (allopolyploidy) in one cell nucleus and cytoplasm constitutes an enormous challenge. Recent years have witnessed efforts and progress in exploring the possible mechanisms by which these seemingly intangible hurdles of polyploidy may be ameliorated or eventually overcome. In particular, the documentation of rapid and extensive non-Mendelian genetic and epigenetic changes that often accompany nascent polyploidy is revealing: the resulting non-additive and novel gene expression at global, regional and local levels, and timely restoration of meiotic chromosomal behavior towards bivalent pairing and disomic inheritance may ensure rapid establishment and stabilization as well as its long-term evolutionary success. Further elucidation on these novel mechanisms underpinning polyploidy will promote our understanding on fundamental issues in evolutionary biology and in our manipulation capacities in future genetic improvement of important crops that are currently polyploids in genomic constitution. 4Strengths, weaknesses, opportunities and threats (SWOT) analysis of reinitiation into methamphetamine abusers: qualitative findings from an exploration of methamphetamine abusers in Shanghai, China显示文摘Background Despite high relapse rate among methamphetamine (MA) abusers, there still have been little empirical data to date detecting the risk factors related to craving and relapse from the perspective of MA abusers. Therefore, the aim of this study was to use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors that could facilitate or impede individuals' abstinence in a real-life context from the perspectives of MA abusers. Aim To use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors relating to individuals' abstinence in a real-life context. Method Semistructured interviews were conducted with 32 MA abusers recruited in Shanghai guided by open-ended questions on narrating the real-life catalysts and inhibitors related to craving, initiation, relapse and abstinence. All data were transcribed verbatim and analysed using a strengths, weaknesses, opportunities and threats (SWOT) analysis. Results The results of the SWOT analysis revealed that contextual factors including peer influence, prevalence and availability of MA, familiar venue of MA use, discrimination, sexual behaviours, alcohol, emotional states and their attitudes towards smoking MA were important factors that contribute to reinitiation and relapse. Surveillance systems, antidrug social workers, vocational skills trainings, moving to another city and family responsibility might serve as counter measures targeti叩 those mentioned weaknesses and threats above. Conclusion This SWOT analysis highlights the complex nature of relapse. 5A tree-ring-based reconstruction of the Yimin River annual runoff in the Hulun Buir region,Inner Mongolia,for the past 135 years显示文摘Based on the relationships between the regional tree-ring chronology(RC) of moisture-sensitive Pinus sylvestris var.mongolica and the monthly mean maximum temperature,annual precipitation and annual runoff,a reconstruction of the runoff of the Yimin River was performed for the period 1868-2002.The model was stable and could explain 52.2% of the variance for the calibration period of 1956-2002.During the past 135 years,21 extremely dry years and 19 extremely wet years occurred.These years represented 15.6% and 14.1% of the total study period,respectively.Six severe drought events lasting two years or more occurred in 1950-1951,1986-1987,1905-1909,1926-1928,1968-1969 and 1919-1920.Four wetter events occurred during 1954-1959,1932-1934,1939-1940 and 1990-1991.Comparisons with other tree-ring-based streamflow reconstructions or chronologies for surrounding areas supplied a high degree of confidence in our reconstruction.Power spectrum and wavelet analyses suggested that the reconstructed annual runoff variation in the Hulun Buir region and surrounding area could be associated with large-scale atmospheric-oceanic variability,such as the Pacific Decadal Oscillation(PDO) and El Ni o-Southern Oscillation(ENSO),and sunspot activity.BAO Guang LIU Yu LIU Na 2012Chinese Science Bulletin2012,57,36:9

7A fundamental theorem for eco-environmental surface modelling and its applications显示文摘We propose a fundamental theorem for eco-environmental surface modelling(FTEEM) in order to apply it into the fields of ecology and environmental science more easily after the fundamental theorem for Earth’s surface system modeling(FTESM). The Beijing-Tianjin-Hebei(BTH) region is taken as a case area to conduct empirical studies of algorithms for spatial upscaling, spatial downscaling, spatial interpolation, data fusion and model-data assimilation, which are based on high accuracy surface modelling(HASM), corresponding with corollaries of FTEEM. The case studies demonstrate how eco-environmental surface modelling is substantially improved when both extrinsic and intrinsic information are used along with an appropriate method of HASM. Compared with classic algorithms, the HASM-based algorithm for spatial upscaling reduced the root-meansquare error of the BTH elevation surface by 9 m. The HASM-based algorithm for spatial downscaling reduced the relative error of future scenarios of annual mean temperature by 16%. The HASM-based algorithm for spatial interpolation reduced the relative error of change trend of annual mean precipitation by 0.2%. The HASM-based algorithm for data fusion reduced the relative error of change trend of annual mean temperature by 70%. The HASM-based algorithm for model-data assimilation reduced the relative error of carbon stocks by 40%. 9A global empirical model for mapping zenith wet delays onto precipitable water vapor using GGOS Atmosphere data显示文摘The importance of water vapor in research of global climate change and weather forecast cannot be over emphasized; therefore substantial efforts have been made in exploring the optimal methods to measure water vapor. It is well-established that with a conversion factor, zenith wet delays can be mapped onto precipitable water vapor(PWV). However, the determination of the exact conversion factor depends heavily on the accurate calculation of a key variable, weighted mean temperature of the troposphere(T_m). As a critical parameter in Global Positioning System(GPS) meteorology, T_m has recently been modeled into a global grid known as GWMT. The GWMT_model only requires the location and the day of year to calculate T_m. Despite the advantages that the GWMT_model offers, anomalies still exist in oceanic areas due to low sampling resolution. In this study, we refine the GWMT_model by incorporating the global T_m grid from Global Geodetic Observing System(GGOS) and obtain an improved model, GWMT-G. 10An adjustment method for active reflector of large high-frequency antennas considering gain and boresight显示文摘The design of the Qitai 110 m Radio Telescope(QTT) with large aperture and very high working frequency(115 GHz) was investigated in Xinjiang, China. The results lead to a main reflector with high surface precision and high pointing precision. In this paper, the properties of active surface adjustment in a deformed parabolic reflector antenna are analyzed. To assure the performance of large reflector antennas such as gain and boresight, which can be obtained by utilizing an electromechanical coupling model, and satisfy them simultaneously, research on active surface adjustment applied to a new parabolic reflector as target surface has been done. Based on the initial position of actuators and the relationship between adjustment points and target points, a novel mathematical model and a program thatdirectly calculates the movements of actuators have been developed for guiding the active surface adjustment of large reflector antennas. This adjustment method is applied to an 8 m reflector antenna,in which we only consider gravity deformation. 11Generality and characteristics of genetic and epigenetic changes in newly synthesized allotetraploid wheat lines显示文摘Previous studies have shown rapid and extensive genomic instability associated with early stages of allopolyploidization in wheat.However,these studies are based on either a few pre-selected genomic loci or genome-wide analysis of a single plant individual for a given cross combination,thus making the extent and generality of the changes uncertain.To further study the generality and characteristics of allopolyploidization-induced genomic instability in wheat,we investigated genetic and epigenetic changes from a genome-wide perspective(by using the AFLP and MSAP markers) in four sets of newly synthesized allotetraploid wheat lines with various genome constitutions,each containing three randomly chosen individual plants at the same generation.We document that although general chromosomal stability was characteristic of all four sets of allotetraploid wheat lines,genetic and epigenetic changes at the molecular level occurred in all these plants,with both kinds of changes classifiable into two distinct categories,i.e.,stochastic and directed.The abundant type of genetic change is loss of parental bands while the prevalent cytosine methylation pattern alteration is hypermethylation at the CHG sites.Our results have extended previous studies regarding allopolyploidization-induced genomic dynamics in wheat by demonstrating the generality of both genetic and epigenetic changes associated with multiple nascent allotetraploid wheat lines,and providing novel insights into the characteristics of the two kinds of induced genomic instabilities.Bao Qi 1,Xiaofang Zhong 1,Bo Zhu,Na Zhao,Liying Xu,Huakun Zhang,Xiaoming Yu,Bao Liu Key Laboratory of Molecular Epigenetics of MOE and Institute of Genetics & Cytology,Northeast Normal University,Changchun 130024,China 2010Journal of Genetics and Genomics2010,37,11:5
12Pantoprazole ameliorates liver fibrosis and suppresses hepatic stellate cell activation in bile duct ligation rats by promoting YAP degradation显示文摘Liver fibrosis is one of the most severe pathologic consequences of chronic liver diseases,and effective therapeutic strategies are urgently needed.Proton pump inhibitors(PPIs)are H+/K+-ATPase inhibitors and currently used to treat acid-related diseases such as gastric ulcers,which have shown other therapeutic effects in addition to inhibiting acid secretion.However,few studies have focused on PPIs from the perspective of inhibiting hepatic fibrosis.In the present study,we investigated the effects of pantoprazole(PPZ),a PPI,against liver fibrosis in a bile duct ligation(BDL)rat model,human hepatic stellate cell(HSC)line LX-2 and mouse primary HSCs(pHSCs),and explored the potential mechanisms underlying the effects of PPZ in vitro and in vivo.In BDL rats,administration of PPZ(150 mg·kg^(−1)·d^(−1),i.p.for 14 d)significantly attenuated liver histopathological injury,collagen accumulation,and inflammatory responses,and suppressed fibrogenesis-associated gene expression including Col1a1,Acta2,Tgfβ1,and Mmp-2.In LX-2 cells and mouse pHSCs,PPZ(100–300μM)dose-dependently suppressed the levels of fibrogenic markers.We conducted transcriptome analysis and subsequent validation in PPZ-treated LX-2 cells,and revealed that PPZ inhibited the expression of Yes-associated protein(YAP)and its downstream targets such as CTGF,ID1,survivin,CYR61,and GLI2.Using YAP overexpression and silencing,we demonstrated that PPZ downregulated hepatic fibrogenic gene expression via YAP.Furthermore,we showed that PPZ promoted the proteasome-dependent degradation and ubiquitination of YAP,thus inhibiting HSC activation.Additionally,we showed that PPZ destabilized YAP by disrupting the interaction between a deubiquitinating enzyme OTUB2 and YAP,and subsequently blocked the progression of hepatic fibrosis.Zhen-ning Lu Wei-xiao Niu Na Zhang Mao-xu Ge Yun-yang Bao Yu Ren Xiu-li Guo Hong-wei He 2021Acta Pharmacologica Sinica2021,42,11:5

14Interferon-induced Transmembrane Protein 3 Prevents Acute Influenza Pathogenesis in Mice显示文摘Objective Interferon-induced transmembrane protein 3(IFITM3)is an important member of the IFITM family.However,the molecular mechanisms underlying its antiviral action have not been completely elucidated.Recent studies on IFITM3,particularly those focused on innate antiviral defense mechanisms,have shown that IFITM3 affects the body’s adaptive immune response.The aim of this study was to determine the contribution of IFITM3 proteins to immune control of influenza infection in vivo.Methods We performed proteomics,flow cytometry,and immunohistochemistry analysis and used bioinformatics tools to systematically compare and analyze the differences in natural killer(NK)cell numbers,their activation,and their immune function in the lungs of Ifitm3-/-and wild-type mice.Results Ifitm3-/-mice developed more severe inflammation and apoptotic responses compared to wild-type mice.Moreover,the NK cell activation was higher in the lungs of Ifitm3-/-mice during acute influenza infection.Conclusions Based on our results,we speculate that the NK cells are more readily activated in the absence of IFITM3,increasing mortality in Ifitm3-/-mice.SUN Qiang LEI Na LU Jian GAO Rong Bao LI Zi LIU Li Qi SUN Ying GUO Jun Feng WANG Da Yan SHU Yue Long 2020Biomedical and Environmental Sciences2020,33,5:4

16Analysis of a Community-based Intervention to Reduce Heat-related Illness during Heat Waves in Licheng,China:a Quasi-experimental Study显示文摘Objective To reduce health-related threats of heat waves, interventions have been implemented in many parts of the world. However, there is a lack of higher-level evidence concerning the intervention efficacy. This study aimed to determine the efficacy of an intervention to reduce the number of heat-related illnesses. Methods A quasi-experimental design was employed by two cross-sectional surveys in the year 2014 and 2015, including 2,240 participants and 2,356 participants, respectively. Each survey was designed to include one control group and one intervention group, which conducted in Licheng, China. A representative sample was selected using a multistage sampling method. Data, collected from questionnaires about heat waves in 2014 and 2015, were analyzed using a difference-in-difference analysis and cost effectiveness analysis. Outcomes included changes in the prevalence of heat-related illnesses and cost-effectiveness variables. Results Relative to the control participants, the prevalence of heat-related illness in the intervention participants decreased to a greater extent in rural areas than in urban areas(OR=0.495 vs. OR=1.281). Moreover, the cost-effectiveness ratio in the intervention group was less than that in the control group(US$15.06 vs. US$15.69 per participant). Furthermore, to avoid one additional patient, the incremental cost-effectiveness ratio showed that an additional US$14.47 would be needed for the intervention compared to when no intervention was applied. Conclusion The intervention program may be considered a worthwhile investment for rural areas that are more likely to experience heat waves. Meanwhile, corresponding improving measures should be presented towards urban areas. 17Observation seasonal variation of intraocular pressure in young healthy volunteers显示文摘AIM:To investigate fluctuation of intraocular pressure(IOP)and seasonal variation of 24-hour IOP during one year in healthy participantsMETHODS:Totally 13 young healthy volunteers participated in this study.IOP was measured with Canon TX-20 at about 8:00-9:00 a.m.from Monday to Friday every week for a whole year.They also underwent 24-hour IOP examination every three months.Blood pressure,heart rate,temperature,humidity,atmosphere pressure,sunshine duration and other environment parameters were recorded.RESULTS:The yearly fluctuation curve showed IOP in the summer months were lower than other seasons.In the multivariable generalized estimating equation analysis,IOP had a negative correlation with both temperature and sunshine duration(P<0.05).There also was a seasonal effect on 24-hour IOP.However,all intraclass correlation coefficients values of minimum,maximum and average of the 24-hour IOP and each individual IOP were less than 0.30.CONCLUSION:IOP is trend to be higher in cold days than warm days.IOP have negative association with both environmental temperature and duration of sunshine.On a season-to-season basis,24-hour IOP is not highly reproducible in healthy volunteers.Na Liao Yan-Qian Xie Guang-Yun Mao Fan-Jun Bao Zhong Lin Hui-Lyu Jiang Yuan-Bo Liang 2022International Journal of Ophthalmology(English edition)2022,15,1:3
18Distribution characteristics and indicator significance of Dechloranes in multi-matrices at Ny-lesund in the Arctic显示文摘In recent years, Dechloranes have been widely detected in the environment around the world.However, understanding and knowledge of Dechloranes in remote regions, such as the Arctic,remain lacking. Therefore, the concentrations of 5 Dechloranes in surface seawater, sediment,soil, moss, and dung collected from Ny-lesund in the Arctic were measured with the concentrations 93 pg/L, 342, 325, 1.4, and 258 pg/g, respectively, which were much lower than those in Asian and European regions. The mean ratios of anti-Dechlorane Plus(DP) to total DP(fanti) in seawater, sediment, soil, moss, dung, and atmospheric samples were 0.36, 0.21, 0.18,0.27, 0.66, and 0.43, respectively. Results suggested that the main source of DP in seawater,sediment, soil, and moss was long-range atmospheric transport.
