1Anti-SARS-CoV-2 activities in vitro of Shuanghuanglian preparations and bioactive ingredients显示文摘Human infection with severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)causes coronavirus disease 2019(COVID-19)and there is no cure currently.The 3CL protease(3CLpro)is a highly conserved protease which is indispensable for CoVs replication,and is a promising target for development of broad-spectrum antiviral drugs.In this study we investigated the anti-SARS-CoV-2 potential of Shuanghuanglian preparation,a Chinese traditional patent medicine with a long history for treating respiratory tract infection in China.We showed that either the oral liquid of Shuanghuanglian,the lyophilized powder of Shuanghuanglian for injection or their bioactive components dose-dependently inhibited SARS-CoV-23CLpro as well as the replication of SARS-CoV-2 in Vero E6 cells.Baicalin and baicalein,two ingredients of Shuanghuanglian,were characterized as the first noncovalent,nonpeptidomimetic inhibitors of SARS-CoV-23CLpro and exhibited potent antiviral activities in a cell-based system.Remarkably,the binding mode of baicalein with SARS-CoV-23CLpro determined by X-ray protein crystallography was distinctly different from those of known 3CLpro inhibitors.Baicalein was productively ensconced in the core of the substrate-binding pocket by interacting with two catalytic residues,the crucial S1/S2 subsites and the oxyanion loop,acting as a“shield”in front of the catalytic dyad to effectively prevent substrate access to the catalytic dyad within the active site.Overall,this study provides an example for exploring the in vitro potency of Chinese traditional patent medicines and effectively identifying bioactive ingredients toward a specific target,and gains evidence supporting the in vivo studies of Shuanghuanglian oral liquid as well as two natural products for COVID-19 treatment.Hai-xia Su Sheng Yao Wen-feng Zhao Min-jun Li Jia Liu Wei-juan Shang Hang Xie Chang-qiang Ke Hang-chen Hu Mei-na Gao Kun-qian Yu Hong Liu Jing-shan Shen Wei Tang Lei-ke Zhang Geng-fu Xiao Li Ni Dao-wen Wang Jian-ping Zuo Hua-liang Jiang Fang Bai Yan Wu Yang Ye Ye-chun Xu 2020Acta Pharmacologica Sinica2020,41,9:32
2Pharmacokinetics and pharmacodynamics of Da-Cheng-Qi decoction in the liver of rats with severe acute pancreatitis显示文摘AIM To explore the pharmacokinetics and pharmacodynamics of Da-Cheng-Qi decoction(DCQD) in the liver of rats with severe acute pancreatitis(SAP) based on an herbal recipe tissue pharmacology hypothesis.METHODS Healthy male Sprague-Dawley rats were randomly divided into a sham operation group(SOG); a model group(MG); and low-, median- and high-dose treatment groups(LDG, MDG, and HDG, respectively). Different dosages(6, 12 and 24 g/kg for the LDG, MDG, and HDG, respectively) of DCQD were administered to the rats with SAP. The tissue concentrations of aloeemodin, rhein, emodin, chrysophanol, honokiol, rheo chrysophanol, magnolol, hesperidin, naringenin and naringin in the liver of the treated rats were detected by high-performance liquid chromatography tandem mass spectrometry. Alanine transaminase(ALT) and aspartate transaminase(AST) in serum, inflammatory mediators in the liver and pathological scores were evaluated.RESULTS The major components of DCQD were detected in the liver, and their concentrations increased dosedependently. The high dose of DCQD showed a maximal effect in ameliorating the pathological damages, decreasing the pro-inflammatory mediators tumor necrosis factor-α and interleukin(IL)-6 and increasing anti-inflammatory mediators IL-4 and IL-10 in the liver. The pathological scores in the pancreas for the MG were significantly higher than those for the SOG(P < 0.05). DCQD could reduce the pathological scores in the pancreas and liver of the rats with SAP, especially in the HDG. 3Effect of Substructure on Toughness of Lath Martensite/Bainite Mixed Structure in Low-Carbon Steels显示文摘The quantitative analysis of substructure in the martensite/bainite mixed structure, which is obtained from low-carbon NiCrMoV steels under different cooling conditions, was made by means of optical microscope (OM), scanning electron microscope (SEM), electron backscatter diffraction (EBSD), and transmission electron microscope (TEM), in order to research the effect on toughness. The test results indicate that the toughness of the steel is enhanced with the decrease in the packet and block size under the condition of the same prior austenite grain size mixed with different ratios of martensite and bainite while the lath width is about 0.38 μm. The calculation shows that both the packet and block boundaries have the same hindering effect on crack extension. Furthermore, the effect of the block width on impact energy is much larger than that of the packet. 4Effect of Magnesium on Inclusion Formation in Ti-Killed Steels and Microstructural Evolution in Welding Induced Coarse-Grained Heat Affected Zone显示文摘Effects of Mg on the chemical component and size distribution of Ti-bearing inclusions favored grain refine- ment of the welding induced coarse-grained heat affected zone (CGHAZ),with enhanced impact toughness in Ti-kill- ed steels,which were examined based on experimental observations and thermodynamic calculations.The results in- dicated that the chemical constituents of the inclusions gradually varied from the Ti-O+Ti-Mg-O compound oxide to the Ti-Mg-O+MgO compound oxide and the single-phase MgO,as the Mg content increased from 0.002 3% to 0.006%.A trace addition of Mg (approximately 0.002%) led to the refinement of Ti-bearing inclusions by creating the Ti-Mg-O compound oxide and provided favorable size distribution of the inclusions for acicular ferrite transforma- tion with a high nucleation rate in the CGHAZ,and a high volume fraction of acicular ferrite was obtained in the CGHAZ with enhanced impact toughness.Otherwise,a high content of Mg (approximately 0.006%) produced a single-phase MgO,which was impotent to nucleate an acicular ferrite,and a microstructure comprised of a ferrite side plate and a grain boundary ferrite developed in the CGHAZ.The experimental results were confirmed by ther- modynamic calculations.CHAI Feng YANG Cai-fu SU Hang ZHANG Yong-quan XU Zhou 2009Journal of Iron and Steel Research(International)2009,16,1:22
5Manufacturing Technology and Application Trends of Titanium Clad Steel Plates显示文摘Some of the major manufacturing processes and corresponding mechanical properties of titanium clad steel plates were analyzed, and the consequences of research, manufacturing, and application of titanium clad steel plates in both markets of China and overseas were also summarized. As an economical and environmentally friendly technology, the roll bonding process is expected to become the next-generation mainstream process for the manufacturing of titanium clad steel plate. Some of the crucial and most important technical problems of this particular process, including vacuum sealing technology, surface treatment process technology, application of a transition layer, and rolling process, were discussed along with the advantageous mechanical properties and life-cycle economy of these plates processed by this technology. 6Collagen type Ⅱ suppresses articular chondrocyte hypertrophy and osteoarthritis progression by promoting integrin β1-SMAD1 interaction显示文摘Hypertrophic differentiation is not only the terminal process of endochondral ossification in the growth plate but is also an important pathological change in osteoarthritic cartilage.Collagen type II(COL2A1)was previously considered to be only a structural component of the cartilage matrix,but recently,it has been revealed to be an extracellular signaling molecule that can significantly suppress chondrocyte hypertrophy.However,the mechanisms by which COL2A1 regulates hypertrophic differentiation remain unclear.In our study,a Col2a1 p.Gly1170Ser mutant mouse model was constructed,and Col2a1 loss was demonstrated in homozygotes.Loss of Col2a1 was found to accelerate chondrocyte hypertrophy through the bone morphogenetic protein(BMP)-SMAD1 pathway.Upon interacting with COL2A1,integrinβ1(ITGB1),the major receptor for COL2A1,competed with BMP receptors for binding to SMAD1 and then inhibited SMAD1 activation and nuclear import.COL2A1 could also activate ITGB1-induced ERK1/2 phosphorylation and,through ERK1/2-SMAD1 interaction,it further repressed SMAD1 activation,thus inhibiting BMP-SMAD1-mediated chondrocyte hypertrophy.Moreover,COL2A1 expression was downregulated,while chondrocyte hypertrophic markers and BMP-SMAD1 signaling activity were upregulated in degenerative human articular cartilage.Our study reveals novel mechanisms for the inhibition of chondrocyte hypertrophy by COL2A1 and suggests that the degradation and decrease in COL2A1 might initiate and promote osteoarthritis progression.Chengjie Lian Xudong Wang Xianjian Qiu Zizhao Wu Bo Gao Lei Liu Guoyan Liang Hang Zhou Xiaoming Yang Yan Peng Anjing Liang Caixia Xu Dongsheng Huang Peiqiang Su 2019Bone Research2019,7,1:17
7The HuangZaoSi Maize Genome Provides Insights into Genomic Variation and Improvement History of Maize显示文摘Maize is a globally important crop that was a classic model plant for genetic studies. Here, we report a 2.2 Gb draft genome sequence of an elite maize line, HuangZaoSi (HZS). Hybrids bred from HZS-improved lines (HILs) are planted in more than 60% of maize fields in China. Proteome clustering of six completed sequeneed maize genomes show that 638 proteins fall into 264 HZS-specific gene families with the majority of contributions from tandem duplication events. Resequencing and comparative analysis of 40 HZSrelated lines reveals the breeding history of HILs. More than 60% of identified selective sweeps were clustered in identity.by.descent conserved regions, and yield-related genes/QTLs were enriched in HZS characteristic selected regions. Furthermore, we dem on strated that HZS-specific family genes were not uniformly distributed in the genome but enriched in improvement/function.related genomic regions. 8Aloe-emodin exerts cholesterol-lowering effects by inhibiting proprotein convertase subtilisin/kexin type 9 in hyperlipidemic rats显示文摘Hyperlipidemia(HPL)characterized by metabolic disorder of lipids and cholesterol is one of the important risk factors for cardiovascular diseases.Proprotein convertase subtilsin/kexin type 9(PCSK9)is a potent circulating regulator of LDL through its ability to induce degradation of the low-density lipoprotein cholesterol receptor(LDLR)in the lysosome of hepatocytes.Aloe-emodin(AE)is one of potentially bioactive components of Chinese traditionalmedicine Daming capsule.In this study we evaluated the HPL-lowering efficacy of AE in both in vivo and in vitro HPL models.High-fat diet-induced rats were treated with AE(100 mg/kg per day,ig)for 6 weeks.We found that AE administration significantly decreased the levels of total cholesterol(TC)and LDL in the serum and liver tissues.Moreover,AE administration ameliorated HPL-induced hepaticlipid aggregation.But AE administration did not significantly inhibit HMG-CoA reductase activity in the liver of HPL rats.A cellular model of HPL was established in human hepatoma(HepG2)cells treated with cholesterol(20 μg/mL)and 25-hydroxycholesterol(2 μg/mL),which exhibited markedly elevated cholesterol levels.The increased cholesterol levels could be reversed by subsequent treatment with AE(30 μM).In both the in vivo and in vitro HPL models,we revealed that AE selectively suppressed the stero-regulatory element-binding protein-2(SREBP-2)and hepatocyte nuclear factor(HNF)1α-mediated PCSK9 signaling,which in turn upregulated LDL receptor(LDLR)and promoted LDL uptake.This study demonstrates that AE reduces cholesterol content in HPL rats by inhibiting the hepatic PCSK9/LDLR pathway.Zhen-li Su Peng-zhou Hang Juan Hu Yu-yang Zheng Han-qi Sun Jing Guo Ke-yu Liu Zhi-min Du 2020Acta Pharmacologica Sinica2020,41,8:14

10Microstructure and Precipitation Behavior in HAZ of V and Ti Microalloyed Steel显示文摘Three steels containing 0.05%C-0.1%V-0.01%N (steel V-LN),0.05%C-0.1%V-0.02%N (steel V-HN),and 0.05%C-0.1%V-0.02%N-0.01%Ti (steel V-HN-Ti),which were all essentially vanadium microalloyed steels,were subjected to simulating the microstructure of a coarse grained heat affected zone (CGHAZ). The process involved reheating to 1 350 ℃,rapid cooling to room temperature,and varying the welding heat input from 15 kJ/cm to 54 kJ/cm,including four cooling rates of t8/5 equal to 7.5 s,20 s,40 s,100 s,and the relationship of heat input to t8/5 was calculated by Quiksim software. The microstructure and precipitation of vanadium and titanium carbon nitrides are studied. The results indicate that the microstructure consists of granular bainite and some side plate ferrite in the grain boundary when the steels are produced with the highest heat input. As the heat input decreased,numerous polygonal ferrites and grain boundary ferrites appeared,and the size apparently increased. When the steel contained high nitrogen,it was considerably easier to form martensite-austenite island,which was even worse for the toughness and other properties of the steel. For the limitation of cooling time,vanadium carbon nitrides could not precipitate sufficiently,but as titanium was added,the unmelted or precipitated TiN on cooling absorbed some fraction of nitrogen in the matrix and made more precipitate positions for the round V(C,N),and thus several useful round particles could be seen in titanium-contained steel,and most of them were around TiN. 11Pharmacokinetics and pharmacodynamics of Shengjiang decoction in rats with acute pancreatitis for protecting against multiple organ injury显示文摘AIM To explore the pharmacokinetics and pharmacodynamics of Shengjiang decoction(SJD) in rats with acute pancreatitis(AP) for protecting against multiple organ injury.METHODS An AP model was established by retrograde perfusion of 3.5% sodium taurocholate into the biliopancreatic duct, and a control group(CG) received 0.9% sodium chloride instead. Twelve male Sprague-Dawley rats were randomly divided into a CG treated with SJD(CG + SJD) and a model group treated with SJD(MG + SJD), both of which were orally administered with SJD(5 g/kg) 2 h after surgery. Blood samples were collected via the tail vein at 10, 20, and 40 min and 1, 2, 3, 4, 6, 8, and 12 h after a single dose of SJD to detect its main components using high-performance liquid chromatography-tandem mass spectrometry. The pharmacokinetic parameters were compared. In the pharmacodynamic experiment, 18 male SpragueDawley rats were randomly divided into a CG, an AP model group(MG), and an SJD treated AP group(SJDG). Serum amylase, lipase, and inflammatory cytokines were measured, and heart, lung, liver, spleen, pancreas, kidney, and intestine tissues were collected for pathological examination.RESULTS The MG + SJD displayed significantly shorter mean residence time(MRT) and higher clearance(CL) for emodin and aloe-emodin; significantly shorter time of maximum concentration and T1/2 and a lower area under curve(AUC) for aloe-emodin; a significantly higher AUC and lower CL for rhein; and longer MRT and lower CL for chrysophanol than the CG + SJD. In the pharmacodynamic experiment, the amylase, interleukin(IL)-6, IL-10, and tumor necrosis factor(TNF)-α levels in the MG were higher than those in the CG(P < 0.05). After the herbal decoction treatment, the SJDG had higher IL-10 and lower TNF-α levels than the MG(P < 0.05). The MG had the highest pathological scores, and the pathological scores of the lung, pancreas, kidney, and intestine in the SJDG were significantly lower than those in the MG(P < 0.05).CONCLUSION AP may have varying effects on the pharmacokinetics of the major SJD components in rats. 12On design of physical random access channel in high-speed railway LTE systems显示文摘For the uplink in the long-term evolution(LTE)systems,the physical random access channel(PRACH)adopts Zadoff–Chu(ZC) sequences as preamble sequences,and each user can choose a unique cyclic shift for user identification.In high-mobility scenario,the Doppler frequency shift raises up the false alarm probability by inducing the correlation peak offsetting at the receiver side.Traditional detection method utilizes multiple search windows for searching the potential uplink users.When the Doppler frequency shift is larger than the subcarrier spacing,the energy of correlation peak leaks outside the search window and increases the false alarm probability.In this paper,we present a novel design for PRACH preamble sequence for high-speed railway scenario.Through numerical observation,we find out that the offset of the correlation peaks only depends on the initial root sequence number and the length of ZC sequences.Therefore,the root number of ZC sequences should be carefully designed to generate a preamble sequence to fight against the Doppler frequency shift.Consequently,those offsets of the correlation peaks are concentrated within a single search window,resulting in lower complexity of detection and lower probability of false alarm for random access in LTE uplink.Gang Wu Hang Yuan Wei Guo Su Hu Shaoqian Li 2014Chinese Science Bulletin2014,59,35:10
13Precipitation Behavior of V-N Microalloyed Steels during Normalizing显示文摘The precipitation behavior of V-N microalloyed steel during normalizing process was studied by physicochemical phase analysis and transmission electron microscopy(TEM). The effect of precipitation behavior on mechanical properties was investigated by theoretical calculations. The results showed that 32.9% of V(C,N) precipitates remained undissolved in the austenite during the soaking step of the normalizing process. These precipitates prevented the growth of the austenite grains. During the subsequent cooling process, the dissolved V(C,N) re-precipitated and played a role in precipitation strengthening. The undissolved V(C,N) induced intragranular ferrite nucleation and refined the ferrite grains. 14Effect of Tempering Temperature on Microstructure and Mechanical Properties of Steel Containing Ni of 9%显示文摘Mechanical properties of quenching,intercritical quenching and tempering (QLT) treated steel containing Ni of 9% were evaluated from specimens subject to various tempering temperatures. The detailed microstructures of steel containing Ni of 9% at different tempering temperatures were observed by optical microscope (OM) and transmission electron microscope (TEM). The volume fraction of austenite was estimated by XRD. The results show that high strength and cryogenic toughness of steel containing Ni of 9% are obtained when the tempering temperature are between 540 and 580 ℃. The microstructure keeps the dual phase lamellar structure after the intercritical quenching and there is cementite created in the Ni-rich constituents when tempering temperature is 540 ℃. When tempering temperatures are between 560 and 580 ℃,the reversed austenites (γ′) grow up and the dual phase lamellar structure is not clear. The γ′ becomes instable at 600 ℃. When tempered at temperature ranging from 500 to 520 ℃,the increase of dislocation density in the lamellar matrix makes both tensile strength and yield strength decrease. When tempered at 540 ℃ and higher temperature,the yield strength decreases continuously because the C and alloying elements in the matrix are absorbed by the cementite and the γ′,so the yield ratio is decreased by the γ′. There are two toughness mechanisms at different tempering temperatures. One is that the precipitation of cementite absorbs the carbon in the steel which plays a major role in improving cryogenic toughness at lower temperature. Another is that the γ′ and the purified matrix become major role at higher tempering temperature. When the tempering temperature is 600 ℃,the stability of γ′ is decreased quickly,even the transformation takes place at room temperature,which results in a sharp decrease of Charpy-V impact energy at 77 K. 15Strengths, weaknesses, opportunities and threats (SWOT) analysis of reinitiation into methamphetamine abusers: qualitative findings from an exploration of methamphetamine abusers in Shanghai, China显示文摘Background Despite high relapse rate among methamphetamine (MA) abusers, there still have been little empirical data to date detecting the risk factors related to craving and relapse from the perspective of MA abusers. Therefore, the aim of this study was to use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors that could facilitate or impede individuals' abstinence in a real-life context from the perspectives of MA abusers. Aim To use qualitative research methods exploring the strengths, weaknesses, opportunities and threat factors relating to individuals' abstinence in a real-life context. Method Semistructured interviews were conducted with 32 MA abusers recruited in Shanghai guided by open-ended questions on narrating the real-life catalysts and inhibitors related to craving, initiation, relapse and abstinence. All data were transcribed verbatim and analysed using a strengths, weaknesses, opportunities and threats (SWOT) analysis. Results The results of the SWOT analysis revealed that contextual factors including peer influence, prevalence and availability of MA, familiar venue of MA use, discrimination, sexual behaviours, alcohol, emotional states and their attitudes towards smoking MA were important factors that contribute to reinitiation and relapse. Surveillance systems, antidrug social workers, vocational skills trainings, moving to another city and family responsibility might serve as counter measures targeti叩 those mentioned weaknesses and threats above. Conclusion This SWOT analysis highlights the complex nature of relapse. 16A Model for Precipitation Kinetics in Vanadium Microalloyed Steel显示文摘Small dispersoid particles inhibit recrystallization which is critical in controlling the grain structure of many high strength low alloy steels. A general kinetic model has been developed to predict precipitation of V(C, N) in vanadium microalloyed steels with a series of carbon and nitrogen contents. The solubility product and driving force of carbonitrides precipitated in austenite as well as the interfacial energy and other parameters can be evaluated to predict Nucleation rates-Temperature (NrT) and Precipitation-Time-Temperature (PTT) diagram. By using stress relaxation tests and fitting with Avrami equation, it is possible to draw PTT diagrams. The predictions of the model coincide with results of experimental investigation on V(C, N) precipitation in austenite. 20Daytime HONO formation in the suburban area of the megacity Beijing, China显示文摘Nitrous acid(HONO),as a primary precursor of OH radicals,has been considered one of the most important nitrogencontaining species in the atmosphere.Up to 30%of primary OH radical production is attributed to the photolysis of HONO.However,the major HONO formation mechanisms are still under discussion.During the Campaigns of Air Quality Research in Beijing and Surrounding Region(CAREBeijing2006)campaign,comprehensive measurements were carried out in the megacity Beijing,where the chemical budget of HONO was fully constrained.The average diurnal HONO concentration varied from 0.33 to 1.2 ppbv.The net OH production rate from HONO,POH(HONO)net,was on average(from 05:00 to 19:00)7.1×106 molecule/(cm3 s),2.7 times higher than from O3 photolysis.This production rate demonstrates the important role of HONO in the atmospheric chemistry of megacity Beijing.An unknown HONO source(Punknown)with an average of 7.3×106molecule/(cm3 s)was derived from the budget analysis during daytime.Punknown provided four times more HONO than the reaction of NO with OH did.The diurnal variation of Punknown showed an apparent photo-enhanced feature with a maximum around 12:00,which was consistent with previous studies at forest and rural sites.Laboratory studies proposed new mechanisms to recruit NO2 and J(NO2)in order to explain a photo-enhancement of of Punknown.In this study,these mechanisms were validated against the observation-constraint Punknown.The reaction of exited NO2 accounted for only 6%of Punknown,and Punknown poorly correlated with[NO2](R=0.26)and J(NO2)[NO2](R=0.35).These results challenged the role of NO2 as a major precursor of the missing HONO source.YANG Qiang SU Hang LI Xin CHENG YaFang LU KeDing CHENG Peng GU JianWei GUO Song HU Min ZENG LiMin ZHU Tong ZHANG YuanHang 2014Science China Chemistry2014,57,7:8
