1Efficacy and safety of a novel anti-HER2 therapeutic antibody RC48 in patients with HER2-overexpressing,locally advanced or metastatic gastric or gastroesophageal junction cancer:a single-arm phase II study显示文摘Background:Current treatment options for human epidermal growth factor receptor 2(HER2)-overexpressing gastric cancer at third-line have shown limited clinical benefit.Further,there is no specific treatment for HER2 immunohistochemistry(IHC)2+and fluorescence in-situ hybridization-negative patients.Here,we report the efficacy and safety of a novel anti-HER2 antibody RC48 for patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer.Methods:Patients with HER2-overexpressing(IHC 2+or 3+),locally advanced or metastatic gastric or gastroesophageal junction cancer who were under at least second-line therapy were eligible and received RC482.5 mg/kg alone every 2 weeks.The primary endpoint was the objective response rate(ORR)assessed by an independent review committee.Secondary endpoints included progressionfree survival(PFS),overall survival(OS),duration of response,time to progression,disease control rate,and safety.Results:Of 179 patients screened,125 were eligible and received RC48 treatment.The ORR was 24.8%(95%confidence interval[CI]:17.5%-33.3%).The median PFS and OS were 4.1 months(95%CI:3.7-4.9 months)and 7.9 months(95%CI:6.7-9.9 months),respectively.The most frequently reported adverse events were decreased white blood cell count(53.6%),asthenia(53.6%),hair loss(53.6%),decreased neutrophil count(52.0%),anemia(49.6%),and increased aspartate aminotransferase level(43.2%).Serious adverse events(SAEs)occurred in 45(36.0%)patients,and RC48-related SAEs were mainly decreased neutrophil count(3.2%).Seven patients had adverse events that led to death were not RC48-related.Conclusions:RC48 showed promising activity with manageable safety,suggesting potential application in patients with HER2-overexpressing,advanced gastric or gastroesophageal junction cancer who have previously received at least two lines of chemotherapy.Zhi Peng Tianshu Liu Jia Wei Airong Wang Yifu He Liuzhong Yang Xizhi Zhang Nanfeng Fan Suxia Luo Zhen Li Kangsheng Gu Jianwei Lu Jianming Xu Qingxia Fan Ruihua Xu Liangming Zhang Enxiao Li Yuping Sun Guohua Yu Chunmei Bai Yong Liu Jiangzheng Zeng Jieer Ying Xinjun Liang Nong Xu Chao Gao Yongqian Shu Dong Ma Guanghai Dai Shengmian Li Ting Deng Yuehong Cui Jianmin Fang Yi Ba Lin Shen 2021Cancer Communications2021,41,11:29

7High-precision Dating and Geological Significance of Chang 7 Tuff Zircon of the Triassic Yanchang Formation, Ordos Basin in Central China显示文摘The Ordos Basin, as the second largest petroliferous basin of China, contains abundant oil and gas resources, oil shale, and sandstone-type uranium mineral resources. Chang 7 shale is not only the major source rock of the Mesozoic petroliferous system of the Basin, but is also crucial in determining the space-time distribution relationship of the shale section for the effective exploration and development of the Basin's oil and gas resources. To obtain a highly precise age of the shale development section, we collected tuff samples from the top and bottom profile of the Chang 7 Member, Yishi Village, Yaoqu Town, Tongchuan District, on the southern margin of the Ordos Basin and performed high-precision chemical abrasion(CA)–isotope dilution(ID)–thermal ionization mass spectrometry(TIMS) zircon U-Pb dating on the basis of extensive laser ablation inductively coupled plasma mass spectrometry(LA-ICP-MS) zircon U-Pb dating data. Our results show the precise ages of the top and bottom zircon in the Chang 7 shale to be 241.06±0.12 Ma and 241.558±0.093 Ma, respectively. We first obtained Chang 7 age data with Grade 0.1-Ma precision and then determined the age of the shale development in the Chang 7 Member to be the early-Middle Triassic Ladinian. This result is supported by paleontological evidence. The deposition duration of the Chang 7 shale is 0.5 Ma with an average deposition rate of the shale section being 5.3 cm/ka. 8Long non-coding RNA HULC as a diagnostic and prognostic marker of pancreatic cancer显示文摘BACKGROUND Long non-coding RNA(lncRNA)is abnormally expressed in various malignant tumors.In recent years,it has been found that IncRNA HULC is increasingly expressed in pancreatic cancer tissues and is involved in the development and progression of pancreatic cancer.However,the clinical value of serum HULC in pancreatic cancer remains unclear,and there are few studies on how HULC regulates the biological function of pancreatic cancer cells.AIM To determine the value of lncRNA HULC in the diagnosis and prognosis of pancreatic cancer,and its possible biological potential.METHODS Sixty patients with pancreatic cancer and sixty patients with benign pancreatic diseases admitted to Xiangya Hospital,Central South University were assigned to the pancreatic cancer group and the benign disease group,respectively,and another 60 healthy subjects were enrolled as the normal group during the same period.HULC-siRNA and NC-siRNA were transfected into pancreatic cancer cells.Quantitative real-time polymerase chain reaction was performed to determine the expression of HULC in tissues,serum,and cells.Western Blot was carried out to determine the expression ofβ-catenin,c-myc,and cyclin D1 in cells,and the cell counting kit-8,flow cytometry,and Transwell assay were conducted to determine the proliferation,apoptosis and invasion of cells.RESULTS Highly expressed in the tissues and serum of pancreatic cancer patients,HULC showed good clinical value in distinguishing between patients with pancreatic cancer,patients with benign pancreatic diseases and healthy subjects.HULC was related to pathological parameters including tumor size,T staging,M staging and vascular invasion,and the area-under-the-curve for evaluating these four expression of HULC had a significantly higher 3-year overall survival(OS)and 5-year OS than those with high expression.T staging,M staging,vascular invasion,and HULC were independent prognostic factors affecting the 3-year OS of patients with pancreatic cancer.Inhibition of HULC expression prevented the proliferation and invasion of pancreatic cancer cells,promoted apoptosis,and inhibited the expression of Wnt/β-catenin signaling pathway-related proteins,β-catenin,c-myc,and cyclin D1.The Wnt/β-catenin signaling pathway agonist(LiCl)restored proliferation,apoptosis,and invasion of pancreatic cancer cells with inhibited expression of HULC.CONCLUSION HULC is an effective marker for the diagnosis and prognosis of pancreatic cancer,which may affect the biological function of pancreatic cancer cells through the Wnt/β-catenin signaling pathway.Zheng-Lin Ou Zhen Luo Ye-Bin Lu 2019World Journal of Gastroenterology2019,25,46:13

10Validation of the chinese version of the EORTC QLQ-CR29 in patients with colorectal cancer显示文摘AIM To assess the validity and reliability of the European Organization for Research and Treatment of Cancer Quality of Life Questionnaire-Colorectal Cancer 29(EORTC QLQ-CR29) in Chinese patients with colorectal cancer(CRC). METHODS From March 2014 to January 2015, 356 patients with CRC from four different hospitals in China were enrolled in the study, and all patients self-administered the EORTC QLQ-CR29 and the quality of life core questionnaire(EORTC QLQ-C30). Evaluation of the scores was based on the Karnofsky Performance Scale(KPS). The reliability and validity of the questionnaires were assessed by Cronbach's α coefficient, the Spearman correlation test and Wilcoxon rank sum test.RESULTS The EORTC QLQ-CR29 showed satisfactory reliability(α > 0.7), although the urinary frequency and blood and mucus in stool dimensions had only moderate reliability(α = 0.608). The multitrait scaling analyses showed good convergent(r > 0.4) and discriminant validity. Significant differences were obtained for each item in the different KPS subgroups(KPS ≤ 80; KPS > 80). body image and most single-item dimensions showed statistically significant differences in patients with a stoma compared with the rest of the patients. 13Olmutinib(HM61713) reversed multidrug resistance by inhibiting the activity of ATPbinding cassette subfamily G member 2 in vitro and in vivo显示文摘Overexpressing of ATP-binding cassette(ABC) transporters is the essential cause of multidrug resistance(MDR), which is a significant hurdle to the success of chemotherapy in many cancers.Therefore, inhibiting the activity of ABC transporters may be a logical approach to circumvent MDR.Olmutinib is an epidermal growth factor receptor(EGFR) tyrosine kinase inhibitor(TKI), which has been approved in South Korea for advanced EGFR T790 M-positive non-small cell lung cancer(NSCLC). Here,we found that olmutinib significantly increased the sensitivity of chemotherapy drug in ABCG2-overexpressing cells. Furthermore, olmutinib could also increase the retention of doxorubicin(DOX) and rhodamine 123(Rho 123) in ABC transporter subfamily G member 2(ABCG2)-overexpressing cells. In addition, olmutinib was found to stimulate ATPase activity and inhibit photolabeling of ABCG2 with [^(125) I]-iodoarylazidoprazosin(IAAP). However, olmutinib neither altered ABCG2 expression at protein and m RNA levels nor blocked EGFR, Her-2 downstream signaling of AKT and ERK. Importantly,olmutinib enhanced the efficacy of topotecan on the inhibition of S1-MI-80 cell xenograft growth. All the results suggest that olmutinib reverses ABCG2-mediated MDR by binding to ATP bind site of ABCG2 and increasing intracellular chemotherapeutic drug accumulation. 14Abnormal expression of bHLH3 disrupts a flavonoid homeostasis network,causing differences in pigment composition among mulberry fruits显示文摘Mulberry fruits with high concentrations of anthocyanins are favored by consumers because of their good taste,bright color,and high nutritional value.However,neither the regulatory mechanism controlling flavonoid biosynthesis in mulberry nor the molecular basis of different mulberry fruit colors is fully understood.Here,we report that a flavonoid homeostasis network comprising activation and feedback regulation mechanisms determines mulberry fruit color.In vitro and in vivo assays showed that MYBA-bHLH3-TTG1 regulates the biosynthesis of anthocyanins,while TT2L1 and TT2L2 work with bHLH3 or GL3 and form a MYB-bHLH-WD40(MBW)complex with TTG1 to regulate proanthocyanidin(PA)synthesis.Functional and expression analyses showed that bHLH3 is a key regulator of the regulatory network controlling mulberry fruit coloration and that MYB4 is activated by MBW complexes and participates in negative feedback control of the regulatory network to balance the accumulation of anthocyanins and proanthocyanidins.Our research demonstrates that the interaction between bHLH3 and MYB4 in the homeostasis regulatory network ensures that the fruits accumulate desirable flavonoids and that this network is stable in pigmentrich mulberry fruits.However,the abnormal expression of bHLH3 disrupts the balance of the network and redirects flavonoid metabolic flux in pale-colored fruits,resulting in differences in the levels and proportions of anthocyanins,flavones,and flavonols among differently colored mulberry fruits(red,yellow,and white).The results of our study reveal the molecular basis of the diversity of mulberry fruit colors.Han Li Zhen Yang Qiwei Zeng Shibo Wang Yiwei Luo Yan Huang Youchao Xin Ningjia He 2020Horticulture Research2020,7,1:8

18Validity of the Chinese Version of the Patient-Generated Subjective Global Assessment (PG-SGA) in Lung Cancer Patients显示文摘The prevalence of malnutrition is high among oncology patients in China. Although the Patient- Generated Subjective Global Assessment (PG-SGA) is widely recognized as an important nutritional assessment tool, it has not been validated for Chinese cancer patients. The purpose of this study was to conduct an analysis of nutrition screening and assessment data to validate the Chinese version of the PG-SGA in Chinese patients with lung cancer. This was an observational, cross-sectional study of 2,000 consecutive adult patients with lung cancer treated at several tertiary hospitals in China. Anthropometric and patient descriptive data were collected. The PG-SGA generated a score for the nutritional risk and a global rating for the nutritional status. The internal consistency, external consistency, and construct validity were evaluated, and a known-groups comparison and principal component analyses were conducted to evaluate the reliability and validity of the PG-SGA. The known-groups comparison demonstrated the ability of the PG-SGA to differentiate lung cancer patients of different nutritional and body mass index (BMI) groups. A confirmatory factor analysis supported the original four-factor structure of the PG-SGA. In terms of the internal and external consistency, the PG-SGA demonstrated intra-class correlation coefficients (ICC) up to 0.987. Alternative form validity was also demonstrated between Box 4 of the PG-SGA and the Karnofsky Performance Status score, with excellent external consistency (r = 0.972, P < 0.001). The construct validity was supported, and selected questionnaire dimensions were evident in the principal component analysis. Significant Spearman correlations (P < 0.001) were demonstrated. The Chinese version of the PG-SGA demonstrated significant reliability and sufficient exploratory properties to support its validity. It seems to be a valid tool that can be used to access the nutritional status of Chinese lung cancer patients. 20Silencing MYH9 blocks HBx-induced GSK3βubiquitination and degradation to inhibit tumor stemness in hepatocellular carcinoma显示文摘MYH9 has dual functions in tumors.However,its role in inducing tumor stemness in hepatocellular carcinoma(HCC)is not yet determined.Here,we found that MYH9 is an effective promoter of tumor stemness that facilitates hepatocellular carcinoma pathogenesis.Importantly,targeting MYH9 remarkably improved the survival of hepatocellular carcinoma-bearing mice and promoted sorafenib sensitivity of hepatocellular carcinoma cells in vivo.Mechanistic analysis suggested that MYH9 interacted with GSK3βand reduced its protein expression by ubiquitin-mediated degradation,which therefore dysregulated theβ-catenin destruction complex and induced the downstream tumor stemness phenotype,epithelial–mesenchymal transition,and c-Jun signaling in HCC.C-Jun transcriptionally stimulated MYH9 expression and formed an MYH9/GSK3β/β-catenin/c-Jun feedback loop.X protein is a hepatitis B virus(HBV)-encoded key oncogenic protein that promotes HCC pathogenesis.Interestingly,we observed that HBV X protein(HBX)interacted with MYH9 and induced its expression by modulating GSK3β/β-catenin/c-Jun signaling.Targeting MYH9 blocked HBX-induced GSK3βubiquitination to activate theβ-catenin destruction complex and suppressed cancer stemness and EMT.Based on TCGA database analysis,MYH9 was found to be elevated and conferred poor prognosis for hepatocellular carcinoma patients.In clinical samples,high MYH9 expression levels predicted poor prognosis of hepatocellular carcinoma patients.These findings identify the suppression of MYH9 as an alternative approach for the effective eradication of CSC properties to inhibit cancer migration,invasion,growth,and sorafenib resistance in HCC patients.Our study demonstrated that MYH9 is a crucial therapeutic target in HCC.Xian Lin Ai-min Li Yong-Hao Li Rong-Cheng Luo Yu-Jiao Zou Yi-Yi Liu Chen Liu Ying-Ying Xie Shi Zuo Zhan Liu Zhen Liu Wei-Yi Fang 2020Signal Transduction and Targeted Therapy2020,5,1:6
