| 1 | Blocking FSH inhibits hepatic cholesterol biosynthesis and reduces serum cholesterol显示文摘Menopause is associated with dyslipidemia and an increased risk of cardio-cerebrovascular disease.The classic view assumes that the underlying mechanism of dyslipidemia is attributed to an insufficiency of estrogen.In addition to a decrease in estrogen, circulating follicle-stimulating hormone (FSH)levels become elevated at menopause.In this study,we find that blocking FSH reduces serum cholesterol via inhibiting hepatic cholesterol biosynthesis.First,epidemiological results show that the serum FSH levels are positively correlated with the serum total cholesterol levels,even after adjustment by considering the effects of serum estrogen,in addition,the prevalence of hypercholesterolemia is significantly higher in peri-menopausal women than that in premenopausal women.Furthermore,we generated a mouse model of FSH elevation by intraperitoneally injecting exogenous FSH into ovariectomized (OVX)mice,in which a normal level of estrogen (E2)was maintained by exogenous supplementation. Consistently,the results indicate that FSH,independent of estrogen,increases the serum cholesterol level in this mouse model. | 4 | Outcomes in refractory diffuse large B-cell lymphoma:results fromamulticenter real-world study in China显示文摘Background:Diffuse large B-cell lymphoma(DLBCL)patients refractory to rituximab-based immunochemotherapy have a dismal prognosis.However,the definition of refractory DLBCL remains inconsistent and no large cohort study data is available from Asian countries.To validate the definition and outcomes of refractory DLBCL in China,we conducted a multicenter,retrospective cohort study.Methods:The REtrospective AnaLysis of Treatment REspoNse of refractory DLBCL(REAL-TREND)study was performed using real-world data from 8 centers in China.DLBCL patients with curative intent were included in the REAL-TREND dataset.Overall survival(OS)was estimated using the Kaplan-Meier method and compared by the log-rank test.Due to heterogeneity in response rates among different centers,the response rates of refractory patients were pooled using random-effect models.Multivariate survival analysis was performed using the Cox regression model.Results:A total of 2778 DLBCL patients diagnosed between January,2010 and December,2015 were enrolled to this study.After validating previous definitions,the SCHOLAR-1 study was most suitable to define refractory DLBCL.The estimated 5-year cumulative incidence of refractory patients was 20%(95% confidence Interval[CI]=18%-22%).After the determination of refractory disease,overall response rate and complete remission rate were 30%(95%CI=22%-38%)and 9%(95%CI=4%-15%),respectively.Patients with either no response to immunochemotherapy or relapse within 12 months after stem-cell transplantation had inferior survival with a median OS of 5.9 months(95%CI=5.5-7.1 months)and 2-year OS rate of 16%(95%CI=12%-20%).International prognostic index score 4-5(hazard ratio[HR]=2.22;95%CI=1.47-3.35),central nervous systemrelapse(HR=1.43;95%CI=1.04-1.97),and best response status(HR=2.68;95%CI=1.42-5.03 for partial remission.HR=5.97,95%CI=3.21-11.11 for stable disease/progressive disease)were independent unfavorable prognostic factors.Conclusions:This is the first large-scale Asian cohort study focusing on outcomes of refractory DLBCL.The definition of the SCHOLAR-1 study identifies patients with homogenously inferior survival,thus is appropriate to select refractory DLBCL.Due to poor clinical outcomes in the rituximab era,patients with refractory DLBCL may be potential candidates for novel treatment modalities. | Shuo Wang Li Wang Jianda Hu Wenbin Qian Xi Zhang Yu Hu Qi Zhu Bobin Chen Depei Wu Chung-Chou H.Chang Pengpeng Xu Xiaoyun Zheng Juying Wei Yao Liu Guohui Cui Yong Tang Yan Ma Haiwen Huang Hongmei Yi Weili Zhao | 2021 | Cancer Communications2021,41,3: | 10 |

| 9 | SERK Receptor-like Kinases Control Division Patterns of Vascular Precursors and Ground Tissue Stem Cells during Embryo Development in Arabidopsis显示文摘During embryo development, the vascular precursors and ground tissue stem cells divide to renew them-selves and produce the vascular tissue, endodermal cells, and cortical cells. However, the molecular mech-anisms regulating division of these stem cells have remained largely elusive. In this study, we show that loss of function of SOMATIC EMBRYOGENESIS RECEPTOR-UKE KINASE (SERK) genes results in aberrant em-bryo development. Fewer cortical, endodermal, and vascular cells are generated in the embryos of serk1 serk2bak1 triple mutants. WUSCHEL-RELATED HOMBOBOX5 (WOXS) is ectopically expressed in vascular cells of serkl serk2 bak1 embryos. The first transverse division of vascular precursors in mid-globular em-bryos and second asymmetric division of ground tissue stem cells in early-heart embryos are abnormally altered to a longitudinal division. The embryo defects can be partially rescued by constitutively activated mitogen-activated protein kinase (MAPK) kinase kinase YODA (YDA) and MAPK kinase MKK5. | 12 | Eocene Basins on the SE Tibetan Plateau: Markers of Minor Offset along the Xuelongshan–Diancangshan–Ailaoshan Structural System显示文摘The offset of geological bodies provides robust evidence of displacement along a fault or ductile shear zone. The amount of displacement along the Xuelongshan–Diancangshan–Ailaoshan structural system, southeastern Tibetan Plateau, is uncertain because of the lack of offset geological markers. This NNW–SSE-trending system is developed in three isolated metamorphic complexes and interjacent nonmetamorphosed rocks. They are expected to record similar post-Eocene strain, although their structural patterns should be distinct. Geological mapping in the area between the Xuelongshan and Diancangshan metamorphic complexes has revealed a small Eocene basin, the Madeng Basin, located to the west of the structural system. The sedimentary and volcanic successions of the Madeng Basin are comparable to those of the Jianchuan Basin, which is located to the east of the structural system. Zircon U–Pb geochronological and bulk geochemical data demonstrate that the volcanic rocks of both basins formed during 37–34 Ma and share the same geochemical features. These data suggest that the Madeng and Jianchuan basins previously constituted a single basin, with the distribution of high-K volcanic rocks in the basins defining an ENE–WSW-trending volcanic belt that shows a limited dextral offset of ≤20 km across the Xuelongshan–Diancangshan–Ailaoshan structural system. Therefore, the northern segment of the structural system records no evidence of large-scale lateral movement/displacement. OBJECTIVE: To observe the effect of Tongxinluo on cognitive impairment in a mouse model with vascular dementia and the changes of Aβ deposition andβ-secretase 1 (BACE1) expression. DESIGN: Randomized controlled study. SETTING: State Key Laboratory of Pharmaceutical Biotechnology of Nanjing University and Affiliated Drum Tower Hospital of Nanjing University Medical School. MATERIALS: The experiment was carried out in the State Key Laboratory of Pharmaceutical Biotechnology of Nanjing University and Affiliated Drum Tower Hospital of Nanjing University Medical School from March 2006 to January 2007. A total of 36 healthy Kunming mice, 18 of each gender, were chosen. The study was conducted in accordance with the National Regulations of Experimental Animal Administration, and all animal experiments were approved by the Committee of Experimental Animal Administration of Nanjing University. Tongxinluo was provided by Shijiazhuang Yiling Pharmaceutical Co., Ltd. METHODS: All mice were randomly divided into 6 groups, including naive control (n=6), sham-operated control (n=6) and experimental groups treated with different doses of Tongxinluo (0.2, 0.4, and 0.6 g/kg/d; n=6 for each group) or vehicle (n=6). Five groups were subjected to bilateral common carotid arteries (2-VO) occlusion to produce a vascular dementia model (no occlusion was performed in sham-operated group). The mice in the Tongxinluo treatment groups were intragastricly administered daily with a Tongxinluo suspension (40 g/L in distilled water) at doses of 0.2, 0.4 or 0.6 g/kg/d from day 1 to day 30 post-surgery. The animals in vehicle, sham-operated and naive groups were administered an equal volume of distilled water. MAIN OUTCOME MEASURES: ①Escape latency time determined in all groups of mice before and after 2-VO occlusion by Morris water maze. ②Changes in BACE1 mRNA expression in the hippocampi of mice among the six groups by RT-PCR assay, and BACE1 and Aβ protein expression in the hippocampi of mice by Western blot. RESULTS: All 36 mice were involved in the final analysis. ① No difference was detected in escape latency time to a hidden platform among all groups in water maze test before surgery (P > 0.05) At 30 days after 2-VO occlusion, the vehicle animals exhibited a significantly longer latency in finding the hidden platform compared to that of sham-operated and naive animals (P < 0.01). The prolonged escape latency was significantly reduced by oral administration of 0.4 g or 0.6 kg/day (P < 0.01, P < 0.05). BACE1 mRNA and protein expression in vehicle animals were much higher than in sham-operated and naive animals (P < 0.01). The ischemia-induced increases in BACE1 mRNA and protein level were attenuated by all three doses of Tongxinluo treatment (P < 0.01), and the 0.4 g/kg/d treatment was the most effective. Aβ protein expression in vehicle animals after 2-VO occlusion were much higher than in sham-operated and na?ve animals (P < 0.01). 2-VO occlusion-induced Aβ generation was significantly attenuated by all doses of Tongxinluo treatment, with the most effective dose being 0.4 g/kg/d (P < 0.01). CONCLUSION: BACE1 mRNA levels and protein levels of BACE1 and Aβare reduced in the hippocampi of vascular dementia model mice by all three doses of Tongxinluo treatment, with the most effective dose being 0.4 g/kg/d. | 18 | Enhancing (de)hydrogenation kinetics properties of the Mg/MgH_(2) system by adding ANi_(5)(A=Ce,Nd,Pr,Sm,and Y) alloys via ball milling显示文摘Magnesium(Mg)-based alloys have already been widely studied as the hydrogen storage materials because of their high reversible hydrogen storage capacity,low cost,light weight,etc.However,the poor de/hydrogenation kinetic properties dramatically hinder the practical applications.In this work,the MgH_(2)-ANi_(5)(A=Ce,Nd,Pr,Sm,and Y) composites were prepared by a high-energy ball milling method.which can effectively refine the particle size thus improving the kinetic properties.Experimental results reveal that the MgH_(2)-ANi_(5) composites mainly consist of Mg_(2)NiH_(4),MgH_(2) and rare earth(RE) hydride,which will be dehydrogenated to form Mg_(2)Ni,Mg and stable RE hydride reversibly.Accordingly,the asmilled MgH_(2)-ANi_(5)(A=Ce,Nd,Pr,Sm,and Y) composites with various A-elements can respectively contribute to a reversible hydrogen storage capacity of 6.16 wt%,5.7 wt%,6.21 wt%,6.38 wt%,and 6.5 wt%at a temperature of 300℃,and show much better kinetic properties in comparison to the pure MgH_(2) without any additive.In-situ formed Mg_(2) Ni and stable RE hydride(such as CeH_(2.73) and YH_(2)) might act as effective catalysts to significantly improve the hydrogen storage properties of MgH_(2).The present work provides a guideline on improving the kinetic properties of the Mg-based hydrogen storage alloys. | Wenfang Liao Wenbin Jiang Xu-Sheng Yang Hui Wang Liuzhang Ouyang Min Zhu | 2021 | Journal of Rare Earths2021,39,8: | 3 |
